Effect of CDC2 depletion on the proliferation and apoptosis in human glioma cell line SHG44:experiment in vitro and in vivo

Abstract:Objective The effects of CDC2 on molecular etiology of human gliomas were investigated.Method Retroviral vectors expressing short hairpin RNAs(shRNAs)against CDC2 were designed and transducted into human glioma cell line SHG44 in vitro and in vivo.Subsequently the resulting phenotypic changes of target gene were investigated as followed.The level of CDC2 mRNA and protein were measured utilizing real-time PCR and Western blotting,respectively.The cell cycles and apoptosis were detected by using flow cytometry.Results Human glioma cell line SHG44 transfected with retrovirus vector expressing shRNAs targeting CDC2 Was arrested in G2/M phase.Proliferation of tumor cells was suppressed.Apoptosis was increased.Weights of human glioma xenografts in nude mice were markedly reduced,the survival of mouse intracranial xenograft models was improved signiticantly.Conclusions CDC2 gene plays an important role in the maintenance of proliferation of human gliomas.Downregulation of CDC2 could prevent the progression of the malignant phenotype of human gliomas.
