Abstract:Objective To construct a prokaryotic expression vector of recombinant human apolipoprotein A5(apoA5)for the preparation of specific antibody to apoA5.Methods The total RNA was extracted from human hepatoma tissue as template,and reverse transcription polymerase chain reaction(RT-PCR)was performed to amplify the full length of coding region of ApoA5 gene.The cloning vector pPCR-Script ampSK(+)-apoA5 and expression vector pET16B-apoA5 including the objective fragment were constructed,and the correctness of recombinant plasmid was verified by enzyme incision,electrophoresis and induced expression.Results A specific fragment with 1 100 bp was amplified by PCR,and cloned into pPCR-Script-ampSK(+).The homology of sequence was confirmed by comparing to GeneBank.The electrophoresis of recombinant plasmid pET16B-apoA5 after enzyme incision displayed 2 DNA fragments with 5 700 bp and 1 100 bp.The magnitude of expressed apoA5 induced by IPTG was as expected.Conclusions The prokaryon expression vector of recombinant human apoA5 was constructed successfully.
Prokaryotic Expression of Human Apolipoprotein E and Analyses of the Molecular Interaction Between ApoE and PrP Proteins in Vitro

Abstract:In order to study the possible molecular interaction between PrP protein and the prokaryotic-expressed apolipoprotein E,the full-length sequence of human ApoE gene was amplified with RT-PCR using the mRNA from cell line SH-SY5Y as the template,and inserted into a prokaryotic-expressing plasmid pET32a.The purified fusion protein was emulsified with complete or incomplete Freund's adjuvant and injected subcutaneously into rabbits.The interaction between ApoE and PrP was evaluated with ELISA and immunoprecipitation assays.SDS-PAGE showed that the molecular weight of the expressed fusion protein HIS-ApoE was about 54000. ELISA assay revealed that the reacting titer of the prepared antiserum reached to 1∶(406900),while Western blot confirmed that the prepared antiserum was able to recognize both the recombinant ApoE protein and the native ApoE from various mammalian's liver tissues.Using special ApoE-coated ELISA and immunoprecipitation tests,the activities of molecular interaction between ApoE and PrP were detected.The binding domain lies within the N-terminus of PrP protein.
Molecular Cloning, Expression and Polymorphism of the Porcine Apolipoprotein A5 Gene in a Jinhua × Pietrain F2 Reference Population

Abstract:As a newly described member of the apolipoprotein gene family, apolipoprotein A5 (APOA5) has been suggested to play a key role in the triglyceride metabolism in both human and mice. The aim of this study was to identify the porcine (Sus scrofa) APOA5 gene, determine its mRNA and its mutations that are associated with lipid accumulation. The porcine APOA5 cDNA was amplified by reverse transcriptase polymerase chain reaction using the information of the mouse or other mammals. It had been determined that the open reading frame of the porcine APOA5 gene consists of 1092 bp, which encodes a predicted protein composed of 363 amino acids with a similarity to bovine (80.43%) and to human (78.47%). The expression analysis indicated that the porcine APOA5 gene was expressed in hypophysis, fat and liver. Twelve single nucleotide polymorphisms (SNPs), including 4 SNPs in the 5′ end, 1 SNP in second intron, 1 SNP in third exon and 6 SNPs in the 3′ end, were identified in the porcine APOA5 gene and genotyped on the Jinhua × Pietrain F2 reference population, it had revealed that the SNP of C1834T was significantly associated with average backfat thickness and leaf fat weight (P < 0.01 and P < 0.05, respectively). In conclusion, this study has got basic information of the porcine APOA5 gene and provides evidence that the APOA5 gene could be a potential candidate gene for fat deposition.
Establishment of a human APOA1 transgenic mouse model

Abstract:Objective To establish a human APOA1 transgenic mouse model.Methods The transgenic plasmid was constructed by inserting human APOA1 gene into the downstream of the human ubiquitin C(Ubc) promoter.The transgenic mice were produced by microinjection and the genotyping was detected by PCR.The expression level of the gene was determined by Western blotting.The levels of blood lipids of the mice of different ages were detected using a biochemical analyzer.Results Two human APOA1 gene transgenic mouse lines with different expression levels were established.The expression levels of human APOA1 gene in blood,liver,heart,kidney,spleen and blood vessels of the transgenic mice were significantly higher than those from wild type mice.The blood biochemical assay showed that the level of HDL-cholesterol in the transgenic mice was significantly higher and the level of TG was lower than that of the wild type mice.Conclusions The transgenic mice with overexpression of human APOA1 gene have been successfully established.It may be a useful animal model for further research of hyperlipidemia and related angiocardiopathy.

Abstract:Abstract Apolipoprotein C3 (apoC3) and apolipoprotein A5 (apoA5), encoded by APOA1/C3/A4/A5 gene cluster, are two critical regulators of plasma triglyceride (TG) metabolism. Deficiency of apoC3 or apoA5 led to significant decreased or increased plasma TG levels, respectively. Recent studies indicated apoC3 and apoA5 also played roles in plasma remnant cholesterol, high density lipoprotein (HDL) and hepatic TG metabolisms. Moreover, large scale population genetic studies indicated that loss of function mutations in APOC3 and APOA5 gene conferred decreased and increased risk of coronary artery disease (CAD), respectively. This manuscript mainly reviewed existing evidences suggesting the opposite role of apoC3 and apoA5 in lipid metabolism and CAD risk, and discussed the potential correlation between these two apolipoproteins.

Apolipoprotein A5 regulates intracellular triglyceride metabolism in adipocytes

Abstract:It has previously been demonstrated that apolipoprotein A5 (apoA5) can be internalized by human adipocytes and significantly decreases intracellular triglyceride content. In the present study, endocytosis of apoA5 by adipocytes under different conditions, and the underlying mechanism by which apoA5 regulates cellular triglyceride storage, was investigated. The results revealed that the apoA5 protein was detected in human subcutaneous abdominal adipose tissues. In addition, the uptake of apoA5 was attenuated in human obese adipose tissues and in cultured adipocytes with hypertrophy or insulin resistance. Low‑density lipoprotein receptor protein 1 (LRP1) knockdown in adipocytes resulted in a decrease in internalized apoA5 content, suggesting that LRP1 serves a role in apoA5 uptake. Treatment of adipocytes with apoA5 decreased the expression of the lipid droplet‑associated proteins such as cidec and perilipin. ApoA5‑treated adipocytes demonstrated an increase in lipolysis activity and expression of uncoupling protein 1, which is the molecular effector of thermogenesis in brown adipocytes. These results suggested that decreased triglyceride accumulation in adipocytes induced by apoA5 may be associated with enhanced lipolysis and energy expenditure, which may result from reduced expression of cidec and perilipin. In conclusion, the present study demonstrated a novel role of apoA5 in regulating the intracellular triglyceride metabolism of adipocytes. The results of the present study suggested that apoA5 may serve as a potential therapeutic target for the treatment of obesity and its related disorders.
Cloning and Characterization of a Novel Apolipoprotein Gene, Apolipoprotein AV, in Tree Shrews

Abstract:Apolipoprotein AV (apoAV) modulates plasma triglyceride levels, which is an independent risk factor for cardiovascular disease. ApoAV is also involved in atherosclerosis lesion formation. In order to systematically evaluate the apolipoprotein-related gene profile in tree shrew, a model for its insusceptibility to atherosclerosis, we performed apoAV cloning and characterization. The full-length cDNA of apoAV was identified using SMART-RACE. ApoAV cDNA sequence revealed two transcripts, 1,948 and 1,397 base pairs, due to alternative polyadenylation. These two transcripts share the same open reading frame (ORF), which encodes a 369-amino acid protein with high identity to human apoAV (75 %), including a 23-amino acid N-terminal signal peptide. ApoAV is expressed exclusively in the liver. Mature apoAV was expressed in E. coli BL21(DE3) and purified by Ni-chelated resin. Lipoprotein lipase activity was significantly stimulated by this recombinant protein. The full-length ORF of apoAV was cloned into pDsRed-monomer-N1 vector with a red fluorescent protein tag and was primarily localized in cytoplasm of hepG2 cells. The successful cloning, expression and localization of apoAV in tree shrew has laid down the foundation for further investigation on its structure and functions.
The relationship between serum apolipoprotein A5 and coronary heart disease

Abstract:Objective To analyze the relationship between serum apolipoprotein A5(apoA5) and lipid as well as coronary heart disease(CHD),and to observe the effect of apoA5 on the pathogenesis of CHD.Methods One hundred and fifty CHD patients and 104 healthy controls were enrolled.Serum lipid was detected by a biochemistry analyzer and serum apoA5 was quantitatively measured by a sandwich enzyme-linked immunosorbent assay.Results The concentration of serum apoA5 in stable angina patients was(209.8±52.6) ng/ml,lower than that in healthy controls(231.5± 57.0) ng/ml,P<0.05.Even adjusted by serum lipid,age and other factors,the difference was still statistically significant.Serum apoA5 concentration in female stable angina patients were higher than that in male patients [(239.9±53.1) ng/ml vs(194.5±41.7) ng/ml,P<0.05].apoA5 was negatively correlated with age,and the concentrations of TG,TC,and lipoprotein(a)(r=-0.24,-0.27,-0.19,-0.17,respectively),and positively correlated with HDL-C(r=0.36).Serum apoA5 concentration(248.6±54.4) ng/ml of ACS group was higher than that in stable angina group(209.8±52.6) ng/ml,P<0.01.Conclusion The data demonstrate that serum apoA5 concentration was negatively correlated with serum TG,Lp(a),and CHD.Serum apoA5 concentration may be an independent protective predictor of CHD.

Abstract:OBJECTIVE:Acute monocytic leukemia (AML)-M5 is the common type of acute myeloid leukemias in children. Studies have shown that there are abundant lipopolysaccharide (LPS) receptor (designated as CD14) molecules on the cell membrane of M5 cells and they play an important role in the diagnosis of M5, since they can be recognized and bound by mouse-anti-human CD14 monoclonal antibody (McAb). However, mouse-originated antibodies are largely not suitable for clinical application due to the severe side effects, thus "humanized antibody" is desired. As the first step for developing humanized antibody, the construction and expression of single chain antibody (scFv) with functional protein are necessary. The present study aimed to express ZCH-7-2F9 ScFv (scFv(2F9)) in eukaryotic cells and obtain the scFv(2F9) protein with a high biological activity.METHODS:Four primers were synthesized to construct the eukaryotic expressional vector, which included SfiI and EcoRI enzyme cleaving site, 6 x His and stop code TGA sequences. scFv(2F9) gene was amplified through splicing by overlap extension (SOE) using the high fidelity Taq polymerase. Positive recombinants (pSectag2A/scFv(2F9)) were identified through enzyme cleaving and sequenced before expression and were transformed into Chinese hamster ovary (CHO) cells for expression. Western-Blot and flow cytometry (FCM) were carried out to determine the relative molecular mass (Mr) and binding activity of scFv(2F9).RESULTS:The cloned scFv(2F9) gene was identified to be functional by sequencing and expressing. The interested protein could be detected in the culture supernatant of transformed CHO cells with an Mr of 31 000. The blocking test showed that the positive cell percentages, the mean fluorescence intensity (MFI) and the peak of channel (peak Ch) were reduced by 90.02%, 63.30% and 63.38%, respectively after blocking with scFv(2F9), while those were 4.55%, 10.09% and 5.02% after blockage using the supernatant from the CHO cells transfected with blanked vector pSectag2A.CONCLUSIONS:The scFv(2F9) against human CD14 antigen was successfully expressed in eukaryotic cells and showed a high biological activity, which may be useful for the further studies on its humanized antibodies.
Effects of High Plasma Triglyceride Caused by ApoC Ⅲ Transgene on Ab-Dominal Aortic Aneurysm Induced by Elastase in LDLR-/-mice

Abstract:AIM:To investigate the effects of high plasma triglyceride (TG) caused by apolipoprotein C Ⅲ( ApoC Ⅲ) transgene on the occurrence and development of abdominal aortic aneurysm ( AAA) .METHODS:The animal models of hypercholesterolemia and hypercholesterolemia combined with hypertriglyceridemia were established by feeding high-fat diet to LDLR-/-and ApoC Ⅲ+LDLR-/-mice, respectively.AAA was induced in these mice by pancreatic elastase. By evaluating the incidence of AAA, relative maximal abdominal aortic diameter, disruption of the elastic lamellar structure and expression of matrix metalloproteinases ( MMPs) in the aorta walls of the AAA, the occurrence and development of AAA were compared in LDLR-/-and ApoC Ⅲ+LDLR-/-mice fed with either chow diet or high-fat diet.In addition, an in vitro TNF-α-induced aneurysmal microenvironment model on vascular smooth muscle cells ( VSMC) was used to study the impact of triglyceride-rich lipoproteins ( TRLs) from mice with normal or high contents of ApoCⅢon elastin protein expres-sion.RESULTS:Feeding the high-fat diet aggravated the severity of AAA in the LDLR-/-mice.ApoC Ⅲ+LDLR-/-mice fed with high-fat diet had less severe AAA than LDLR-/-mice fed with high-fat diet.TRLs inhibited degradation of VSMC elas-tin protein induced by TNF-α, and in vitro TRLs from the mice with high content of ApoC Ⅲ, compared to those with nor-mal content of ApoC Ⅲ, had better inhibitory effect on the degradation of elastin.CONCLUSION:High plasma TG caused by ApoC Ⅲtransgene alleviates AAA of the LDLR-/-mice induced by elastase and high-fat diet.The effect is probably attrib-uted to the hypertriglyceridemia caused by ApoC Ⅲtransgene.

Abstract:Clinical and epidemiological investigations confirm that patients with loss-of-function mutations (R19X, etc.) in Apolipoprotein CIII (ApoCIII) showed beneficial lipid profile including decreased plasma triglyceride and increased high density lipoprotein (HDL) levels. However, whether HDL level would be reduced in hypertriglyceridemia (HTG) induced by high ApoCIII expression has not been demonstrated yet. Here we showed, ApoCIII transgenic mice (ApoCIIItg) displayed severe HTG and had significantly lower HDL level. Analysis of apolipoproteins in lipoprotein fractions by SDS-PAGE revealed marked decrease of apolipoprotein AI (ApoAI) in HDL in transgenic mice compared with the wild type mice (WT) as controls. Further examination demonstrated that hepatic but not intestinal ApoAI mRNA was significantly reduced. Therefore, the decreased ApoAI synthesis might be accounted for the lower plasma HDL level in ApoCIII transgenic mice.
Cloning and expression of apolipoprotein E3 and its variant apoE2 and apoE4

Abstract:In order to obtain three isoforms of apolipoprotein E (apoE), the cDNA encoding apoE3 was obtained by RT-PCR from normal human liver tissue. Site-directed mutagenesis was used to obtain the cDNAs encoding apoE2 and apoE4 isoforms. The 3 cDNAs were subcloned into vector pGEM-3Z and verified by DNA sequencing. The expression recombinant which can express the target protein as a (His) 6-tagged fusion was constructed by subcloning apoE cDNA into vector pT7-PL. The purified proteins were gained by Ni-NTA column. The SDS-PAGE results revealed the 6 His fusion proteins (apoE2, apoE3 and apoE4) were correctly expressed and purified successfully.
[Study on Polymorphism in the Apolipoprotein A5 Gene in Patients with Premature Coronary Heart Disease].

Abstract:OBJECTIVE:To investigate the association between the apolipoprotein A5(APOA5) -1131T/C polymorphism and premature coronary heart disease in northern Chinese Han population.METHODS:Using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and polyacrylamide gel electrophoresis (PAGE), we analyzed the genotype and allele distribution in 140 patients with premature coronary heart disease diagnosed by coronary angiography and 156 healthy controls. The levels of serum lipid profiles were also studied by biochemical methods.RESULTS:The allele frequency of APOA5-1131T/C polymorphism in the premature coronary heart disease group was significantly higher (43.2% vs. 33.0%, P=0.011) than that in the control group. Compared with TT homozygotes, CC homozygotes exhibited a 2.809-fold (95% CI 1.331-5.927) increased risk of developing premature coronary heart disease. Logistic regression analysis found that this correlation was independent of sex, age, body mass index (BMI), smoking history as well as serum total cholesterol(TC), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) levels; In premature coronary heart disease group, the triglyceride(TG) level in CC homozygotes was significantly higher than those in TC heterozygotes or TT homozygotes.CONCLUSION:The APOA5-1131T/C polymorphism has influence on serum TG level, and the APOA5-1131C allele is associated with the development of premature coronary heart disease in northern Chinese Han population.
The Associated Study of ApolipoproteinA5 Gene Polymorphisms with the Degrees of Coronary Artery Stenosis in Coronary Heart Disease Patients

Abstract:Aim To investigate the association of apolipoproteinA5(ApoA5) gene polymorphisms of patients with coronary heart disease(CHD)and the degree of coronary artery stenosis. Methods Using polymerase chain reaction-restriction fragmentpolymorphisms of patients with CHD and the degree of length polymorphism methods,the genotypes of 260 patients with CHD confirmed by coronary angiography were analyzed,the levels of coronary artery stenosis were characterized by the numbers of diseased vessels and Gensini scores. Results Serum triglyceride level of the patients with ApoA5-1131CC genotype and c.553T allele carriers were obviously higher than that of the patients with ApoA5 c.553GG genotype and-1131T allele carriers,but the Gensini scores were not significantly different among them,and the distribution for the genotyopes of ApoA5 genetic variants were also not statistically different among variable number of coronary diseased vessels.The number of coronary diseased vessels and Gensini scores had strongly positive correlation with the presence of diabetes(r=0.141 and 0.143,P<0.05),and obviously negative correlation with the level of serum high density lipoprotein cholesterol(r=-0.129 and-0.164,P<0.05). Conclusions ApoA5 gene polymorphisms are remarkably associated with the level of triglyceride,but not the extent of coronary artery stenosis among CHD patients.

Abstract:Objective To clone,express and purify human PrP-like protein Shadoo(SHO),and to prepare polyclonal antibody against SHO.Methods Total RNAs of different tissues of hamster were extracted and the transcription status of SHO gene was comparatively evaluated by a RT-PCR methodology.Full-length human SHO gene was amplified by specific PCR and cloned into a HIS-GST(fusion protein) expression vector after verified by sequence analysis.Human SHO fusion protein was expressed and purified from Escherichia coli SHO-specific antibody was prepared by immunizing rabbits with the purified SHO protein.The immuno-reactivities of the prepared antibody with recombinant and endogenous SHO protein were analyzed with Western blot.Results The transcription level of SHO gene in different tissues varied remarkably.The significant SHO transcripts were observed in brain tissues.SDS-PAGE assays yielded that a roughly 37 ku SHO protein was expressed and purified.The prepared polyclonal antibody against SHO protein reacted well with the genetic engineering expressed and endogenous SHO proteins.Conclusion SHO fusion protein has been expressed and purified successfully and this study supplies basis of studying the relationship between SHO and prion protein.
High-efficient bacterial production of human ApoA-I amyloidogenic variants

Abstract:Apolipoprotein A-I (ApoA-I)-related amyloidosis is a rare disease caused by missense mutations in the APOA1 gene. These mutations lead to protein aggregation and abnormal accumulation of ApoA-I amyloid fibrils in heart, liver, kidneys, skin, nerves, ovaries, or testes. Consequently, the carriers are at risk of single- or multi-organ failure and of need of organ transplantation. Understanding the basic molecular structure and function of ApoA-I amyloidogenic variants, as well as their biological effects, is, therefore, of great interest. However, the intrinsic low stability of this type of proteins makes their overexpression and purification difficult. To overcome this barrier, we here describe an optimized production and purification procedure for human ApoA-I amyloidogenic proteins that efficiently provides between 46 mg and 91 mg (depending on the protein variant) of pure protein per liter of Escherichia coli culture. Structural integrity of the amyloidogenic and native ApoA-I proteins were verified by circular dichroism spectroscopy and intrinsic fluorescence analysis, and preserved functionality was demonstrated by use of a lipid clearance assay as well as by reconstitution of high-density lipoprotein (HDL) particles. In conclusion, the use of the described high-yield protein production system to obtain amyloidogenic ApoA-I proteins, and their native counterpart, will enable molecular and cellular experimental studies aimed to explain the molecular basis for this rare disease.

Abstract:. The angiopoietins are an important family of growth factors specific for vascular endothelium. Most of them bind to the TIE2 receptor and are related to regulation of angiogenesis. During large-scale DNA sequencing of the human fetal brain cDNA library, we cloned a novel human angiopoietin-like cDNA and termed it human angiopoietin-like 5 ( ANGPTL5 ). Like other members of the angiopoietin family, ANGPTL5 -deduced protein also has an N-terminal cleavable signal peptide, a predicted coiled-coil domain, and a fibrinogen-like domain. The search against the human genome database indicated that ANGPTL5 maps to 11q22. Expression analysis of ANGPTL5 shows that it is mainly expressed in adult human heart.

Abstract:Apolipoprotein AI (ApoAI) is an important apolipoprotein in plasma and is known to have various physiological functions suitable for pharmaceutical applications. Human blood has been the only source of this protein for research and large-scale applications. To obtain large amounts of ApoAI a Pichia pastoris expression system was first used to obtain a high level of expression of secreted, recombinant protein. The human gene encoding ApoAI was inserted into the secretion vector pPIC9K and used to transform P. pastoris GS115. AP16, a high expression transformant with high G418 resistance, was obtained. After induction with methanol, the expression level of rhApoAI (recombinant human ApoAI) was 160 mg/L in a 14L fermentor. RhApoAI was purified by cold acetone precipitation followed by Q-Sepharose Fast Flow ion exchange column chromatography with 60% recovery. The N-terminal amino acid sequence and molecular weight (mass spec.) of rhApoAI are identical to native human ApoAI. Purified rhApoAI has specific binding activity with liver cells SMC7721 and binding can be inhibited by native human ApoAI.

Abstract:Apolipoprotein AV (APOA5) deficiency causes hypertriglyceridemia in mice and humans. For years, the cause remained a mystery, but the mechanisms have now come into focus. Here, we review progress in defining APOA5's function in plasma triglyceride metabolism. Biochemical studies revealed that APOA5 binds to the angiopoietin-like protein 3/8 complex (ANGPTL3/8) and suppresses its ability to inhibit the activity of lipoprotein lipase (LPL). Thus, APOA5 deficiency is accompanied by increased ANGPTL3/8 activity and lower levels of LPL activity. APOA5 deficiency also reduces amounts of LPL in capillaries of oxidative tissues (e.g., heart, brown adipose tissue). Cell culture experiments revealed the likely explanation: ANGPTL3/8 detaches LPL from its binding sites on the surface of cells, and that effect is blocked by APOA5. Both the low intracapillary LPL levels and the high plasma triglyceride levels in Apoa5 −/− mice are normalized by recombinant APOA5. Carboxyl-terminal sequences in APOA5 are crucial for its function; a mutant APOA5 lacking 40-carboxyl-terminal residues cannot bind to ANGPTL3/8 and lacks the ability to change intracapillary LPL levels or plasma triglyceride levels in Apoa5 −/− mice. Also, an antibody against the last 26 amino acids of APOA5 reduces intracapillary LPL levels and increases plasma triglyceride levels in wild-type mice. An inhibitory ANGPTL3/8-specific antibody functions as an APOA5-mimetic reagent, increasing intracapillary LPL levels and lowering plasma triglyceride levels in both Apoa5 −/− and wild-type mice. That antibody is a potentially attractive strategy for treating elevated plasma lipid levels in human patients.

Endogenous ApoA-I expression in macrophages: A potential target for protection against atherosclerosis

Abstract:ApoA-I is a major protein component of high-density lipoprotein (HDL) that is widely known for regulating cholesterol trafficking and inflammatory and immune responses and for protecting against atherosclerosis. ApoA-I is generally considered to be synthesized in the liver (hepatocytes) and small intestine (enterocytes). However, computer analysis of ApoA-I has shown that the ApoA-I gene may be expressed in not only hepatocytes and enterocytes but also monocyte-macrophage cells, dendritic cells (DCs) and T cells. ApoA-I expression has been detected in THP-1 monocytes and macrophages, peripheral blood mononuclear cells (PBMCs) from postmenopausal women, human PBMC-derived monocytes and macrophages, mouse peritoneal macrophages, etc. Endogenous ApoA-I in macrophages has anti-inflammatory and cholesterol efflux effects. However, our understanding of the detailed roles of macrophage-synthesized ApoA-I is still at an early stage and very limited. More experiments are needed to elucidate the exact roles of endogenous ApoA-I in macrophages. Several lines of evidence indicate that recombinant exogenous human ApoA-I in mouse macrophages increases cholesterol efflux and thus reduces atherosclerosis development. Considering the antiatherogenic effect of exogenous ApoA-I overexpression in mouse macrophages, better understanding the role and mechanisms underlying macrophage-synthesized ApoA-I in atherosclerosis will enable macrophage-synthesized ApoA-I therapy to open new avenues for reducing the risk of atherosclerosis.
