Fate of Cell Components of Allogeneic Tendon Grafts

Abstract:Objective To determine the time-dependent donor cell survival in an extra-articular environment following allogeneic tendon transplantation. Methods Achilles' tendon defects of 36 rats were repaired by allogeneic tendon graft. The recipient animals were euthanized at 1, 4 and 8 weeks following transplantation (12 animals per time interval) and subsequently had DNA analysis. DNA microsatellite repeats markers were used to distinguish between the cellular DNA of two different strains of rats. DNA fragments were amplified by polymerase chain reaction and analyzed by denaturing high performance liquid chromatography technique. Results The extra-articular survival of donor cells started to diminish as early as 1 week after transplantation as indicated by decreased content of donor DNA. Donor DNA was detectable in 4 of the 12 animals. At 4 weeks, donor DNA could be detected only in 2 of the 12 rats. No donor DNA was detected in all 12 animals at 8 weeks after transplantation. Conclusion Viable donor cells in cryopreserved tendon allografts did not survive longer than 8 weeks following transplantation in an extra-articular environment.
