Effects of melatonin on the voltage-gated calcium channels in hippocampal pyramidal neurons of new-born rats in vitro

Abstract:Objective To investigate the effects of different concentrations of melatonin on voltsge-gated calcium channels.Methods Hippocampal pyramidal neurons were isolated from new-born Wistsr rats ( < 12 h) and cultured for 7-12 days. The whole-cell configuration of the patch clamp technique was used to record the calcium channel current (Icx2+). Fifty-seven neurons were divided into 7 groups and exposed to different concentrations of melatonin, 1 nmol/L, 10 nmol/L, 100 nmol/L, 1 μmol/L, 10 μmol/L, 100 μmol/L and 1 mmol/L. The effects of melatonin on the amplitudes and kinetics of Ca2+ currents were investigated. Results The calcium channel in hippocampal pyramidal neurons was L-type calcium channel with activation threshold at about (-52±10) mV, slow inactivation and rundown phenomenon. The effect of melatonin on calcium channel was rapid and reversible. Except for 10 and 100 nmol/L the other concentrations of melatonin had no influences on the kinetics characteristics of the I-V curve. All concentrations of melatonin increased Ca2+ current in different degrees. Melatonin 1 nmol/L, 10 umol/L and 100 umol/L gradually increased the amplitude of Ca2+ current and melatonin 100 nmol/L had the maximum effect. The degree of increased amplitude of Ca2+ current decreased with melatonin 1 μmol/L, 10 μmol/L, 100 μmol/L and 1 mmol/L. Conclusion Melatonin I nmol/L -1 mmol/L can gradually increase Ca2+ currents in cultured hippocampal pyramidal neurons of new-born rats and melatonin 100 umol/L has the maximum effect.
