Abstract:Objective To determine the feasibility and application value of quantifying intratumoral microvascular perfusion with contrast-enhanced gray-scale harmonic ultrasonography (CEUS) at low mechanical index. Methods A nude murine subcutaneously implanted hepatocelluar carcinoma model was established.CEUS were performed at 27 days (27D),31 days (31D),38 days (38D),59 days (59D) and 72days (72D) after tumors implantation,respectively.Dynamic images were analyzed off-line by a special computer software to perform the time-intensity curves (TIC) of the entire tumor.Parameters were acquired from the TICs and compared with tumor growth. Results The tumor volumes increased as time went by.TIC parameters of a2, a3,PI,△SI,AREA and BF have statistical difference among different measurement time point (P<0.05).The value of a2 decreased as the tumor grew,however, the value of PI,△SI,AREA and BF increased,and decreased at the last time point due to the necrosis in tumors. Conclusion Low MI contrast-enhanced gray-scale harmonic ultrasonography shows intratumoral microvascularity accurately.In addition,TIC parameters reflect changes of intratumoral microvascular perfusion quantatively.Therefore,quantification of intratumoral perfusion flow by CEUS offers a noninvasive imaging method to monitor tumor vascular response to antiangiogenic theraphy..
Characteristics of VEGFR2-targeted Lipid Microbubbles Contrast-Enhanced Ultrasound in Nude Mice with Subcutaneous Colonic Carcinoma Xenografts: Compared with Lipid Microbubbles

Abstract:Objective To explore the difference of the contrast-enhanced ultrasound (CEUS) and quantitative parameters between lipid mierobubbles and VEGFR2-targeted lipid microbubbles in nude mice model with subcutaneous orthotopic transplantation colon cancer.Methods 30 Balb/C nude mice with high VEGFR2 expressed subcutaneous colonic carcinoma xenografts were enrolled in this study.CEUS was performed in each mouse with targeted and non targeted lipid microbubbles respectively.Qontrast software was used to analyze the dynamic image quantitatively.The difference of the CEUS image and quantitative parameters between the two kinds of microbubbles was analyzed.Results Two kinds of microbubbles were characterized by perfusion from peripheral to the middle of the centripetal in the arterial phase by routine visual examination,there were no significant difference of the effect of enhancement.In the venous phase the lipid microbubble were presented as hypo-enhancement,but the VEGFR2-targeted lipid microbubbles were presented as inhomogeneous hyper-enhancement.There were no significant difference of the parameters of peak intensity,arrive time,time to peak,regional blood volume and regional blood flow (P ＞0.05).However,compared with the lipid microbubbles,the regression time and the clearance time of the VEGFR2-targeted lipid microbubbles had significant delay(P ＜0.05,P ＜0.01).The area under the curve in the VEGFR2-targeted lipid microbubbles had significant higher than lipid microbubbles(P ＜0.01).Conclusions The VEGFR2-targeted lipid microbubbles can specially attach to the specific site in neovascular endothelium of tumor and can enhance ultrasound imaging persistently,which may provide an important method for the evaluation of tumor neovascularization and monitoring therapeutic effect of anti-angiogenesis.
Study on the Therapeutic Efficacy of Subcutaneous Colon Cancer Xenograft Tumor with Ultrasound Destruction of VEGFI-carrying Microbubbles

Abstract:Objective To estimate the therapeutic efficacy of ultrasound destruction of vascular endothelial growth factor inhibitor (VEGFI)-carrying lipid microbubbles in colon cancer in nude mice.Methods Colon tumor xenografts cells were inoculated subcutaneously in eighteen Balb/c nude mice and tumor (5-20 mm) developed,all the mice were randomly divided into three groups:Group A (6 mice),as controls,underwent contrast-enhanced ultrasound (CEUS) examination and sham ultrasound radiation.Group B (6 mice) were given lipid microbubbles combined with ultrasound radiation.Group C (6mice)underwent VEGFI-carrying lipid microbubbles injection combined with ultrasound radiation.Contrastenhanced ultrasound (CEUS) examination was performed before and after ultrasound radiation.Enhanced peak intensity(PI),regional blood volume (RBV),regional blood flow (RBF) were recorded and compared.Results There was no significant difference in PI,RBV,RBF before radiation among the three groups (P＞0.05).PI,RBV,RBF in group A were higher 1 day after sham ultrasound radiation,the difference was not statistically significant (P ＞0.05),1 week later PI,RBV,RBF were higher than those before radiation,the difference was statistically significant (P ＜0.05).1 day after ultrasound radiation,PI,RBV,RBF in group B decreased,the difference was statistically significant (P ＜ 0.05),while after 1 week,PI,RBV,RBF increased respectively,comparing with those before ultrasound radiation,the difference was not statistically significant (P ＞0.05).The above indexes were significantly decreased in group C 1 day and 1 week after ultrasound radiation with statistically significant (P ＜ 0.05).Conclusions Ultrasound destruction of VEGFI-carrying lipid microbubbles can improve the therapeutic efficacy of colon cancer in nude mice.

Abstract:The development and metastasis of uterine tumors depend highly on tumor angiogenesis. Multiphase dynamic contrast-enhanced magnetic resonance imaging can quantitatively describe the hemodynamic changes of uterine tumors based on a variety of tracer kinetic models and time-signal curves and by simulating the distribution of contrast inside and outside the blood vessels. Functional parameters can accurately and noninvasively assess tumor angiogenesis. It provides a non-invasive functional evaluation method for the differential diagnosis,staging,response evaluation,and prognostic prediction of uterine tumors.

Abstract:Background: Antiangiogenesis is a promising therapy for advanced hepatocellular carcinoma (HCC), but the effects are difficult to be evaluated. Pazopanib (GW786034B) is a pan-vascular endothelial growth factor receptor inhibitor, the antitumor effects or antiangiogenic effects haven't been investigated in HCC.Methods: In vitro direct effects of pazopanib on human HCC cell lines and endothelial cells were evaluated. In vivo antitumor effects were evaluated in three xenograft nude mice models. In the subcutaneous HCCLM3 model, intratumoral blood perfusion was detected by contrast-enhanced ultrasonography (CEUS), and serial quantitative parameters were profiled from the time-intensity curves of ultrasonograms.Results: In vitro proliferation of various HCC cell lines were not inhibited by pazopanib. Pazopanib inhibited migration and invasion and induced apoptosis significantly in two HCC cell lines, HCCLM3 and PLC/PRF/5. Proliferation, migration, and tubule formation of human umbilical vein endothelial cells were inhibited by pazopanib in a dose-dependent manner. In vivo tumor growth was significantly inhibited by pazopanib in HCCLM3, HepG2, and PLC/PRF/5 xenograft models. Various intratumoral perfusion parameters changed over time, and the signal intensity was significantly impaired in the treated tumors before the treatment efficacy on tumor size could be observed. Mean transit time of the contrast media in hotspot areas of the tumors was reversely correlated with intratumoral microvessel density.Conclusions: Antitumor effects of pazopanib in HCC xenografts may owe to its antiangiogenic effects, and the in vivo antiangiogenic effects could be evaluated by quantitative CEUS.

Abstract:Objective To investigate the feasibility of contrast-enhanced ultrasonography(CEUS) in quantitative assessment of angiogenesis in patients with hepatocellular carcinoma(HCC).Methods Thirtythree patients with HCCs confirmed by pathology underwent CEUS in one week preoperatively.The presets and contrast dosage were fixed in all the studies.The new quantitative software with GAMMA fitting technique was used to analyze the dynamic images offline.The quantitative parameters such as baseline intensity,increased signal intensity(ASI),decent curvature(a2),up slope rate(a3),arrive time(AT), time to peak(TTP), accelerate time(ACT),peak intensity(PI),and area under curve(AREA) were calculated.The pathological specimen was stained with CD34 antibady and microvessel density (MVD) was calculated automatically.The correlation between parameters of CEUS and MVD was analyzed statistically.Results The parameters of ASI, a2, a3, AT, TTP, PI and ACT in the tumor were significantly different from those in liver parenehyma ( P＜0.01 ).The value of a2 in the tumor was correlated with MVD, the standardized ASI and AREA were correlated with MVD (P＜0.05).Conclusions The quantitative analysis with CEUS reflects the microvascular perfusion flow objectively.It provides a noninvasive imaging method to assess the angiogenesis in HCCs.

Abstract:Ultrasound microvessel imaging (UMI), when applied with ultrafast planewave acquisitions, has demonstrated superior blood signal sensitivity in comparison to conventional Doppler imaging. Here we propose a high spatial resolution and ultra-sensitive UMI that is based on conventional line-by-line high-frequency ultrasound imagers and singular value decomposition (SVD) clutter filtering for the visualization and quantification of tumor microvasculature and perfusion. The technique was applied to a chicken embryo tumor model of renal cell carcinoma that was treated with two FDA-approved anti-angiogenic agents at clinically relevant dosages. We demonstrate the feasibility of 3D evaluation with UMI to achieve highly sensitive detection of microvasculature using conventional line-by-line ultrasound imaging on a preclinical and commercially available high-frequency ultrasound device without software or hardware modifications. Quantitative parameters (vascularization index and fractional moving blood volume) derived from UMI images provide significantly improved evaluation of anti-angiogenic therapy response as compared with conventional power Doppler imaging, using histological analysis and immunohistochemistry as the reference standard. This proof-of-concept study demonstrates that high-frequency UMI is a low-cost, contrast-agent-free, easily applicable, accessible, and quantitative imaging tool for tumor characterization, which may be very useful for preclinical evaluation and longitudinal monitoring of anti-cancer treatment.

Abstract:Objective To evaluate the tumor growth by 2-dimensional ultrasound and the tumor angiogenesis by contrast-enhanced power Doppler. Methods Under the guidance of ultrasound, 0.2 ml of heated liquid agarose was injected to seal the aperture after injection of VX2 tumor fragment suspension 0.3 ml (10 7-10 8 cells/ml) into left lobe of the liver for fifteen New Zealand white rabbits. Tumor growth was monitored by ultrasound. Three weeks after implanting VX2 tumor into liver, microbubble contrast agent intravenous bolus injection, blood flow signals were detected by power Doppler and mean color vessel density (MCVD) was calculated for every tumor. Biological features of tumors were examined by routine histology and the value of microvessel density (MVD) of the tumor tissues was examined by immunohistochemistry. Results ①Under the ultrasound, most tumors were isoechoic, some tumors were hypoechoic or slightly hyperechoic, with surrounding echo halo. ②In all the tumors, blood flow was detected by color Doppler and power Doppler. ③The tumors were poorly demarcated from the surrounding liver parenchyma. Multiple mitotic figures were detected on histology. ④The maximum tumor diameters measured by ultrasound and macroscopic pathology were closely correlated. ⑤There was linear correlation between MCVD and MVD of the tumors. Conclusion Ultrasonography is useful in evaluating liver tumor growth. Contrast-enhanced power Doppler can be used for evaluating tumor angiogenesis in vivo.
Study of Multi-Slice CT Perfusion Imaging on Angiogenesis of VX2 Tumor in Rabbits: Before and after Interventional Therapy

Abstract:Objective To investigate the applicative value of multi-slice CT (MSCT)perfusion imaging on quantitative assessment of tumor angiogenesis before and after interventional therapy and monitoring the early interventional therapeutic effect in malignant soft-tissue tumor.Methods Twenty-four New Zealand white rabbits successfully implanted VX_2 tumor in the unilateral proximal thigh were divided into two groups (twelve per group)at random and performed conventional CT plain scan and perfusion scan 14 days after implantation and 3 days after interventional therapy respectively.The values of blood flow (BF),blood valume(BV),mean transit time (MTT)and premeability surface(PS)of VX_2 tumors and normal muscular tissues were computed and the differences among them were analyzed.Meanwhile, correlative analysis between perfusion parameters and micro-vessel density(MVD)counts,average A value of vascular endothelial growth factor(VEGF)was done.Results Before interventional therapy,the values of BF,BV,MTT and PS of VX_2 tumors in interventional group were(303.3±69.9)ml·100g~(-1)·min~(-1), (7.02±3.10)ml/100 g,(1.99±0.28)s and (65.9±9.4)ml·100g~(-1)·min~(-1)respectively. Compared with the parametric values of normal muscular tissues,there were significant differences between them (F value was 4285.82,1867.46,413.04 and 698.42 respectively,P0.01 ). There were no significant differences of the above parametric values between interventional group and the control group (F value was 2.47,2.03,0.02 and 0.53 respectively,P0.05).The values of MVD counts,average A value of VEGF of VX_2 tumors in interventional group were 50.1±4.1/HP and 0.352±0.011 respectively.There were no significant differences between interventional group and the control group (F value was 0.02 and 0.19 respectively,P0.05).Three days after interventional therapy,the values of BF,BV,MTT,PS,MVD and VEGF of VX_2 tumors in interventional group were (7.5±2.4)ml· 100g~(-1)·min~(-1),(1.20±0.23)ml/100g,(3.29±0.57)s,(4.0±1.5)ml·100g~(-1)·min~(-1), 16.0±2.4/HP and 0.215±0.008 respectively.Compared with the values of pre-interventional therapy and the control group,there were significant differences among them(P0.01).Additionally,the values of BF,BV,PS in VX_2 tumors had a significant positive correlation with MVD counts,average A value of VEGF (r0.7,P0.05),however,the value of MTT had no significant correlation with MVD (P0.05)but had a significant negative correlation with average A value of VEGF(r=-0.78,P0.05).Conclusion MSCT perfusion imaging is a functional imaging technique that is capable of quantitative assessment to tumor angiogenesis,blood perfusion and vascular permeability,which can evaluate the early interventional therapeutic effect in malignant soft-tissue tumor invasively and accurately.
Molecular ultrasound imaging with vascular endothelial growth factor receptor 2-targeted microbubbles in a mouse liver cancer model

Abstract:Objective:To evaluate the feasibility and reproducibility of molecular ultrasound imaging of tumor angiogenesis with vascular endothelial growth factor receptor 2 (VEGFR2)-targeted microbubbles.Methods:VEGFR2-targeted microbubbles were accomplished by anti-VEGFR2 monoclonal antibody conjugation to the microbubble surface using biotin-avidin linkage, which was assessedex vivo using immunolfuorescence. Ten mice bearing subcutaneous human liver tumor were divided into two groups randomly (targeted group,n=5; non-targeted group,n=5) and scanned at 2 weeks after implantation with contrast-enhanced ultrasound (CEUS) using either targeted or non-targeted microbubbles. Following CEUS, time-intensity curves were constructed off-line and perfusion parameters were calculated. The residual bubble signal was determined 5 min after contrast agent injection with destruction-replenishment analysis.Results:Both targeted and non-targeted microbubbles revealed signiifcant enhancementin vivo. A signiifcant difference in area under the curve (AUC) was recorded between targeted group and non-targeted group [(3 940.4±200.9) dB?svs. (2 796.8±452.3) dB?s,P=0.001]. After the destruction sequence, there was a signiifcant difference in the decrease of video intensity between targeted group and non-targeted group [(7.61±2.20) dBvs. (3.74±1.40) dB,P=0.010].Conclusion:Molecular ultrasound imaging with VEGFR2-targeted microbubbles is noninvasive, feasible and reproducible to assess tumor angiogenesis.

Abstract:Tumor neoangiogenesis plays an important role in growth and metastasis of human hepatocellular carcinoma (HCC).Angiogenesis induced by tumor cells is different from the normal vessels.Microvessel density measured by immunohistochemistry is used as the gold standard for assessment of angiogenesis,but it is an invasive diagnostic method and its clinical application is restricted.Digital subtraction angiography and noninvasive imaging examinations such as multi-slice spiral CT,MRI,ultrasound,nuclear medicine diagnosis are widely used in clinic for assessment of angiogenesis induced by tumor cells,which can play an important role in clinical diagnosis of HCC and assessment of the treatment effect of inhibiting angiogenesis induced by tumor cells.Molecular images such as optical imaging and synchrotron radiation X-ray imaging are gradually used in the experimental study of assessment of angiogenesis induced by tumor cells.

Abstract:PurposeTo evaluate the effectiveness of contrast-enhanced susceptibility-weighted imaging with ultrasmall superparamagnetic iron oxide (USPIO-enhanced SWI) in the assessment of intratumoral vascularity in hepatocellular carcinoma (HCC). Materials and MethodsOrthotopic xenograft HCC nude mouse models were established first and magnetic resonance imaging (MRI) examinations were performed on a 1.5T MR scanner 28 days later. Three groups of mice, 10 in each, were imaged using unenhanced and USPIO-enhanced SWI at doses of 4, 8, and 12mg Fe/kg. Intratumoral susceptibility signal intensity (ITSS) was scored. ITSS-to-tumor contrast-to-noise ratio (ITSST-CNR) was measured. These measurements were compared between unenhanced and USPIO-enhanced SWI at each dose and differences in the measurements between different dose groups were estimated. Correlation between ITSS and tumor microvessel density (MVD) was analyzed. ResultsCompared with unenhanced SWI, significantly higher ITSS was identified on USPIO-enhanced SWI at doses of 8mg Fe/kg (Z=-2.000, P=0.046) and 12mg Fe/kg (Z=-2.333, P=0.020). Significantly higher ITSST-CNR was found on USPIO-enhanced SWI than that on unenhanced SWI (P < 0.05). Significantly higher ITSST-CNR at a dose of 8mg Fe/kg was observed than that at 4mg Fe/kg (Z=-3.326, P=0.001). Positive correlation between ITSS on USPIO-enhanced SWI at a dose of 8mg Fe/kg and tumor MVD was demonstrated (r=0.817, P=0.004). ConclusionUSPIO-enhanced SWI at a dose of 8mg Fe/kg greatly improves the detection of intratumoral vascularity in a xenograft HCC model. J. Magn. Reson. Imaging 2016;44:288-295.

Abstract:BackgroundThis study aimed to determine whether synchrotron radiation (SR)-based X-ray in-line phase-contrast imaging (IL-PCI) can be used to investigate the morphological characteristics of tumor neovascularization in a liver xenograft animal model.MethodsA human hepatocellular carcinoma HCCLM3 xenograft model was established in nude mice. Xenografts were sampled each week for 4 weeks and fixed to analyze tissue characteristics and neovascularization using SR-based X-ray in-line phase contrast computed tomography (IL-XPCT) without any contrast agent.ResultsThe effect of the energy level and object–to-detector distance on phase-contrast difference was in good agreement with the theory of IL-PCI. Boundaries between the tumor and adjacent normal tissues at week 1 were clearly observed in two-dimensional phase contrast projection imaging. A quantitative contrast difference was observed from weeks 1 to 4. Moreover, 3D image reconstruction of hepatocellular carcinoma (HCC) samples showed blood vessels inside the tumor were abnormal. The smallest blood vessels measured approximately 20 μm in diameter. The tumor vascular density initially increased and then decreased gradually over time. The maximum tumor vascular density was 4.29% at week 2.ConclusionIL-XPCT successfully acquired images of neovascularization in HCC xenografts in nude mice.

Enhancement of Tumor Perfusion and Antiangiogenic Therapy in Murine Models of Clear Cell Renal Cell Carcinoma Using Ultrasound-Stimulated Microbubbles

Abstract:Objective To explore the effect of ultrasound-stimulated microbubble cavitation (USMC) on enhancing antiangiogenic therapy in clear cell renal cell carcinoma. Materials and Methods We explored the effects of USMC with different mechanical indices (MIs) on tumor perfusion, 36 786-O tumor-bearing nude mice were randomly assigned into four groups: (i) control group, (ii) USMC 0.25 group (MI = 0.25), (iii) USMC 1.4 group (MI = 1.4) (iv) US 1.4 group (MI = 1.4). Tumor perfusion was assessed by contrast-enhanced ultrasound (CEUS) before the USMC treatment and 30 min, 4h and 6h after the USMC treatment, respectively. Then we evaluated vascular normalization(VN) induced by low-MI (0.25) USMC treatment, 12 tumor-bearing nude mice were randomly divided into two groups: (i) control group (ii) USMC 0.25 group. USMC treatment was performed, and tumor microvascular imaging and blood perfusion were analyzed by MicroFlow imaging (MFI) and CEUS 30 min after each treatment. In combination therapy, a total of 144 tumor-bearing nude mice were randomly assigned to six groups (n = 24): (i) control group, (ii) USMC 1.4 group, (iii) USMC 0.25 group, (iv) bevacizumab(BEV) group, (v) USMC 1.4 +BEV group, (vi) USMC 0.25 +BEV group. BEV was injected on the 6th, 10th, 14th, and 18th d after the tumors were inoculated, while USMC treatment was performed 24 h before and after every BEV administration. We examined the effects of the combination therapy through a series of experiments. Results Tumor blood perfusion enhanced by USMC with low MI (0.25)could last for more than 6h, inducing tumor VN and promoting drug delivery. Compared with other groups, USMC 0.25 +BEV combination therapy had the strongest inhibition on tumor growth, led to the longest survival time of the mice. Conclusion The optimized USMC is a promising therapeutic approach that can be combined with antiangiogenic therapy to combat tumor progression.

Abstract:1333 Objectives Anti-angiogenic therapy (AAT) has become the major anticancer treatment strategies and there is an urgent need to monitor AAT response by noninvasive, functional molecular imaging with PET/CT since the tumor shrinkage maybe absent for AAT. This study aimed to monitor Sunitinib-based AAT response in mouse U87MG tumor models using 18F-FLT and 18F-FDG MicroPET/CT, comparatively. Methods U87MG tumor models in female nude mice were established and separated into two groups. Both groups were treated by the intragastrical injection of Sunitinib at a dose of 80 mg/kg for 7 days. The MicroPET/CT imaging of both 18F-FDG and 18F-FLT were acquired using MicroPET/CT scanner (Inveon) at day 0, 1, 3, 7 and 13 after AAT. The regions of interest within tumor tissues were delineated and the maximum Stanardized Uptake Values (SUVmax) were applied for the quantitative analysis. The tumor tissues were collected to perform Ki-67, Glut-1 and CD31 staining for immunohistochemical analysis. Data were expressed as mean ± SD and analyzed using One-Way ANOVA test and Pearson correlation analysis. Results Both 18F-FLT and 18F-FDG MicroPET/CT imaging were applied into monitoring AAT response in mouse U87MG tumor models. SUVmax of 18F-FLT uptake post AAT decreased significantly at day 1 (1.57 ± 0.06, P = 0.009), 3 (1.70 ± 0.10, P = 0.034), 7 (1.33 ± 0.15, P = 0.001), then raised at day 13 (1.87 ± 0.38, P = 0.162), comparing with baseline uptake at day 0 (2.13 ± 0.23). Notably, there were no significant differences for 18F-FLT uptakes at days post AAT. However, for 18F-FDG uptakes, there were no significant changes between post and prior AAT, with the exception that an increased SUVmax was observed at day 3 (2.33 ± 0.47), comparing with that at day 0 (1.68 ± 0.43, P = 0.013). Moreover, AAT was confirmed by CD31 staining, and the probe uptakes were correlated with the positive radios of Ki-67 and Glut-1 in tumor tissues. Conclusions MicroPET/CT imaging with 18F-FLT could be superior to that with 18F-FDG for monitoring anti-angiogenic therapy response. Evaluation of Antiangiogenic and Antiproliferative Effects of Sorafenib by Sequential Histology and Intravoxel Incoherent Motion Diffusion-Weighted Imaging in an Orthotopic Hepatocellular Carcinoma Xenograft Model.

Abstract:Purpose: To investigate the effectiveness of intravoxel incoherent motion (IVIM) in the assessment of the therapeutic efficacy of sorafenib in an orthotopic hepatocellular carcinoma (HCC) xenograft model.Materials and Methods: Thirty-five HCC nude mouse models were established. IVIM was performed on a 1.5T MR scanner at baseline (n = 55) and serially at 7, 14, and 21 days after sorafenib treatment. The apparent diffusion coefficient (ADC(total)), true diffusion coefficient (D), pseudodiffusion coefficient (D*), and perfusion fraction (f) at these timepoints were measured and compared between the treated (n = 51) and control group (n = 1). Differences in measurements among different timepoints were evaluated. Correlations between IVIM parameters and histologic features including necrotic fraction (NF) and microvessel density (MVD) were analyzed.Results: Compared to the control group, ADC(total) and D were significantly higher at each timepoint (P=0.009), while f significantly decreased at 7 days (P = 0.009) and increased at 21 days (P = 0.028) in the treated group. Serial measurements in the treated group showed that both ADC(total) and D increased significantly at 7, 14, and 21 days compared to baseline (P < 0.005), while f significantly declined at 7 days (P=0.016) and increased at 21 days (P=0.009). Significant correlations were found between ADCtotal and NF (r=0.811, P < 0.001), D and NF (r=0.838, P < 0.001), and between f and NF (r = 0.528, P = 0.017) in the treated group.Conclusion: IVIM may provide useful biomarkers for evaluating the therapeutic effects of sorafenib on HCC.
Monitoring microvascular tortuosity and velocity distribution response of 4T1 tumor to the DOX-NDs treatment using super-resolution ultrasound imaging

Abstract:Super-resolution ultrasound (SRUS) imaging has been gradually employed for the non-invasive evaluation of tumor intervention, while the comprehensive indicators such as vessel density (VD), tortuosity (VT) and velocity (VV) could easily conceal the local changes of the heterogeneous tumor microvasculature. Herein, our study attempts to construct the novel monitor manner for the local microvascular changes and verify its non-invasion quantitative evaluation of tumor response during intervention. Tumors were delineated into central and peripheral regions based on B-mode images. VD, regional mean VT, and VV indices were quantified for subsequent comparisons of tumor angiogenesis and vessel perfusion over time. SRUS images depicted a gradual reduction in tumor vascularity during treatment. Traditional quantitative data indicated solely a decline in central VD, with marginal VD and VT exhibiting negligible variations. Conversely, a distinct leftward shift in the VT distribution center was evident for peripheral vasculature. Moreover, the VV distribution illustrated a progressive rise in the count of rapidly flowing vessels in the periphery, implying a gradual shift of the tumor's blood supply center from the core to the outskirts. These findings offer a fresh perspective for utilizing SRUS imaging in evaluating intervention responses of highly heterogeneous tumors and guiding more precise therapeutic strategies.
Contrast-enhanced Ultrasound in evaluating of angiogenesis and tumor staging of nasopharyngeal carcinoma in nude mice

Abstract:Objective: To explore the use of Contrast-enhanced Ultrasound (CEUS) in evaluating angiogenesis in a xenograft nasopharyngeal carcinoma (NPC) model in nude mice and the evolution of CEUS parameters according to the growth of NPC. Methods: Nude mice were divided into three groups according to experiments conducted at various times from tumor implantation (8 mice/group; group A: 4 weeks from implantation; group B:6 weeks from implantation; group C:8 weeks from implantation). CNE-2 cells were transplanted in 24 nude mice and CEUS evaluations of the tumors were performed at 4, 6 or 8 weeks from implantation. CEUS parametric perfusion images and pathological findings were recorded. R version 3.4.4 software was used to analyze the CEUS parameters and pathological findings. Results: One-way anova analysis indicated statistically significant differences among the three groups with the parameters of peak intensity (PI) (p<0.001), area wash in (AWI) (p<0.001), area wash out (AWO) (p<0.001) and tumor volumes (p<0.001).Pearson correlation coefficient analysis indicated that microvessel density (MVD) was correlated with tumor volume (r = 0.644, p = 0.001), PI (r = 0.904, p<0.0001), AWI (r = 0.547, p = 0.008) and AWO (r = 0.744, P<0.0001). Tumor volume was correlated with MVD (r = 0.644, p = 0.001), PI (r = 0.625, p = 0.002), AWI (r = 0.528, p = 0.012) and AWO (r = 0.784, p<0.001). The percentage of necrosis in histological sections was correlated with the percentage of CEUS unperfused area (r = 0.446,p = 0.038). Spearman rank correlation coefficient analysis indicated that vascular endothelial growth factor (VEGF) was correlated with PI (r = 0.462, P = 0.032). Welch t test indicated PI, AWI and AWO parameters were significantly lower than that of kidneys (p<0.001, p = 0.009, p = 0.005). Conclusions: The CEUS parameters PI, AWI and AWO indirectly reflect the MVD and the tumor volume in our model of subcutaneous transplanted NPC in nude mice, providing precious information on angiogenesis and tumor growth. VEGF may play a role in promoting angiogenesis of NPC.

Abstract:Objective To investigate the value of contrast enhanced ultrasound(CEUS) with microbubbles targeted to vascular endothelial growth factor receptor type 2 (VEGFR2) for imaging tumor angiogenesis in murine tumor models.Methods Established human renal cell carcinomas(RCC) subcutaneous xenograft tumor model in nude mice,microbubbles targeted to VEGFR2(MBV) was prepared,control microbubbles(MBC) and MBV were injected respectively in each mouse,the intensity of each microbubble was compared,the expressions of VEGFR2 in tumors were tested by immunohistochemistry.Results CEUS imaging showed the intensity of MBV and MBC was (6.50±1.43)dB,(2.59±0.99)dB,respectively.There was significantly higher intensity when using MBV compared with MBC (P<0.01).The time to wash out was longer in MBV contrast group compared with MBC group.Immunohistochemistry showed VEGFR2 was highly expressed in novel vessel walls.Conclusions Contrast microbubbles targeted to VEGFR2 can specially enhance the images of RCC and has tremendous significance in the assessment of angiogenesis.
