Su1092 CAUSAL INFERENCE ANALYSIS OF THE TRANSCRIPTOME IDENTIFIED DIFFERENTIAL INNATE IMMUNITY AND INFLAMMATORY PATHWAY BETWEEN CROHN'S DISEASE AND ULCERATIVE COLITIS

Abstract:Background: Inflammatory bowel diseases (IBD) mainly consist Crohn's disease (CD) and ulcerative colitis (UC).The pathological differences between CD and UC remain unclear.Here we aim to identify differential pathological characteristics of CD and UC using causal inference analysis of the transcriptome.Method: Kruskal-Wallis and Dunn test were performed to identify differentially expressed genes (DEGs) between IBD group (CD, N = 10; UC, N = 15) and control group (N = 13), respectively.KEGG enrichment pathway analysis was then implemented to determine differentially expressed pathways (DEPs) between CD and UC.Last, based on DEPs' network, causal inference analysis was performed to identify key genes differentially expressed between CD and UC.Result: Compared with control group, there were 1352 and 2081 DEGs in CD and UC group, respectively.DEPs analysis showed that inflammation-related pathways differ between CD and UC, such as NOD-like receptor signaling pathway, staphylococcus aureus infection, IL-17 signaling pathway, TNF signaling pathway and chemokine signaling pathway.27 genes in DEP's network were identified as key differential genes between CD and UC by causal inference analysis.Among these genes, GBP5, IFNG and CCL3, related to innate immune and inflammatory reaction, were significantly upregulated in CD compared with UC group.Similarly, the down-stream gene of GBP5, NLRP3, an inflammasome assembly coding gene, was highly expressed in CD.Therefore, the elevated expression of GBP5-NLRP3 is specific for CD.Conclusion: Our results suggested that CD is more active in innate immunity and inflammatory responses than UC.Our findings also identified potential markers to distinguish CD from UC, and may serve as potential therapeutic targets for CD.
