Abstract:A series of non-annulated products: trifluoromethylpyrazolone-tethered trisubstituted methane derivatives were synthesized in high yields in acetonitrile solvent catalyzed by NH4OAc via one-pot, three-component reactions of 2-hydroxy-1,4-naphthoquinone, aromatic aldehydes and 1-aryl-3-trifluoromethyl-5-pyrazolone. Treatment of trifluoromethylpyrazolone-tethered trisubstituted methane derivatives with the dehydrating agent SOCl2/Pyridine in acetonitrile afforded the corresponding annulated fused polyheterocyclic products trifluoromethylated benzo[6,7]chromeno[2,3-c]pyrazole-5,10-dione derivatives. Furthermore, an efficient, straightforward protocol has been developed for the synthesis of trifluoromethylated benzo[6,7]chromeno[2,3-c]pyrazole-5,10-dione derivatives at medium yields via one-pot, two-step, three-component reactions. Meanwhile, the structures of typical products were further confirmed by XRD-analysis.

Abstract:A facile and efficient domino cyclization between 3-aminocoumarins and arylglyoxal monohydrates in the presence of para-toluenesulfonic acid (p-TSA) as a promoter was employed for the synthesis of two libraries of chromeno[3,4-b]pyrrol-4(3H)-one derivatives. Simple workup procedures were used to obtain the products in moderate to excellent yields. The solvents toluene and ethanol served important roles in the selective formation of the pyrrolocoumarin derivatives characterized by their different substitution patterns.

Abstract:A series of poly-heterocycles containing hemi-aminal moieties: 6'-hydroxy trifluoromethylated Spiro [indoline-3,4'-pyrazolo [3,4-b]pyridine] derivatives were efficiently synthesized in ethanol catalyzed by NH4OAc via one-pot, three-component reactions of 3-aminopyrazole, ethyl 4,4,4-trifluoroacetoacetate and isatin. Treatment of this hemi-aminal containing products with the dehydrating reagent SOCl2/Pyridine in acetonitrile afforded the corresponding dehydrating trifluoromethylated polyheterocyclic Spiro [indoline-3,4'-pyrazolo [3,4-b] pyridine] derivatives. Furthermore, a more efficient, straightforward approach to formation of their dehydrating products has been developed via one-pot, three-component reactions catalyzed by hydrochloric acid. Meanwhile, the structures of products were fully confirmed by spectroscopic data, two representative products were further confirmed by XRD analysis, respectively.

Abstract:A variety of 4-(trichloromethyl)pyrido[2',1':3,4]pyrazino[2,1-b]quinazolinones were prepared in moderate to good yields with high regioselectivity through intramolecular 6-endo-dig cyclization and trichloromethylation of N3-alkynyl-2-pyridinyl tethered quinazolinones in chloroform. Mechanistic studies revealed that chloroform might serve as trichloromethyl anion precursor. Furthermore, the reaction could be easily performed at gram scales and an estrone-derived 4-(trichloromethyl)pyrido[2',1':3,4]pyrazino[2,1-b]quinazolinone was prepared over five steps. The present method features broad substrate scope, good functional group tolerance, new dearomatization of pyridine ring, and chloroform as trichloromethylation reagent.

Abstract:Reactions of easily accessible chloropyrimidinyl hydrazones with bromine were investigated. Oxidative cyclization followed by concomitant bromination led to the formation of [1,2,4]triazolo[4,3-c]pyrimidines, which then underwent the Dimroth rearrangement to afford highly functionalized 2-substituted [1,2,4]triazolo[1,5-c]pyrimidines.

Abstract:An efficient and general method for the synthesis of 3-trifluoromethyl-4 trifluoroacetyl pyrazoles from the cycloaddition of sydnones with 1,1,1,5,5,5-hexafluoropentane-2,4-dione has been established. Using ZnI2/bpy as a catalyst in THF, this protocol proved to be general to prepare a variety of 3-trifluoromethyl-4 trifluoroacetyll pyrazoles in good yields. Utility of this method was demonstrated by further transformations of the trifluoroacetyl moiety providing convenient access to 3 trifluoromethyl pyrazole-based carboxylic acid and alcohol derivatives. Abstract:A facile and consecutive one-pot, multi-component reaction for synthesis of trifluoromethylated spirochromeno[2,3-c]-6H-pyrazol-2',5-dione derivatives by the reaction of isatin, cyclohexane-1,3-dione and 1-aryl-3-(trifluoromethyl)-1H-pyrazol-5(4H)-one in the presence of p-toluenesulfonic acid (p-TSA) with good yields was reported. Compared with the stepwise reactions, the p-TSA played dual roles as both catalyst and dehydrating agent in the conversion. The structures of new compounds were determined by spectroscopic methods and X-ray diffraction analysis. In addition, a possible mechanism for the formation of compound 5 was presented. Abstract:A novel cascade reaction was developed for the synthesis of diverse members of a series of benzofuro(3,2-d]pyrimidine derivatives. The process utilizes readily prepared 3-chlorochromenones and various commercially available amidines and their analogues as starting materials. Abstract:8-Bromo-7-chloro[1,2,4]triazolo[4,3-c]pyrimidines were prepared by bromine-mediated oxidative cyclization of the aldehyde-derived hydrazones. The structure of one such product was unambiguously confirmed by single-crystal X-ray analysis. While the C-5 unsubstituted 1,2,4-triazolo[4,3-c]pyrimidine chemotype is extremely susceptible to ring isomerization at ambient conditions, the C-5-substituted analogues were found to be quite stable, permitting isolation in pure form. Nevertheless, they can still be converted into the 1,2,4-triazolo[1,5-c]pyrimidines by base-or acid-promoted Dimroth rearrangement. The presence of halogen functionalities on the pyrimidine nucleus renders the products useful as versatile synthetic intermediates for the easy diversification, as evidenced by palladium-catalyzed Kumada cross-couplings and Buchwald-Hartwig amination, as well as direct aromatic substitution.

Abstract:An effective [3+2] cycloaddition reaction of difluoromethyl or trifluoromethyl hydrazonoyl bromides with alkylidene pyrazolones was disclosed. The method provides an efficient approach to access a variety of highly functionalized fluoroalkyl spiropyrazolones in good yields. This protocol also features some advantages such as easy available and stable substrates, simple operation procedure, atom and step economy. The formation of (cis)- and (trans)-products was discussed.

Abstract:A simple and efficient protocol for direct synthesis of multisubstituted 6-alkyl-4-chromenyl-1-aryl-3,4-dihydro-1 H -pyrrolo[3,4- b ]pyridine-2,5,7(6 H )-trione derivatives is presented by a cascade reaction of 3-formylchromones, Meldrum's acid and α -aminomaleimides as readily accessible starting materials. In this chemoselective, metal catalyst-free approach Michael addition/intramolecular cyclization and elimination of acetone and carbon dioxide occur consecutively. The significant advantages of this strategy are creating a C–N and a C–C bond, short reaction times, excellent yields (68–80%), and a metal-free catalyst.

Abstract:Efficient and convenient one-pot, three-component reactions of pyrrolidone, aromatic aldehydes and 1-phenyl-3-trifluoromethyl-5-pyrazolone afforded highly functionalized trifluoromethylated pyrazol-4-ylpyrrole-2,5-dione derivatives in good yields. The effect of solvents on the reaction efficiency and yield was briefly investigated. The structures of products were determined by spectral methods and X-ray diffraction analysis. The latter showed that the products formed have a strong intramolecular hydrogen bond, which made them particularly stable and the corresponding annulated products were not obtained by treatment with dehydrating reagents.

Abstract:A multicomponent Kröhnke reaction route toward 2‐trifluoromethyl pyridines from reaction of chalcones with easily available and stable 1‐(3,3,3‐trifluoro‐2‐oxopropyl)pyridin‐1‐ium bromide and ammonium acetate is reported. The reaction is found to be general with a range of substituted chalcones, and the applicability of the methodology in gram scale is demonstrated. Preliminary results suggest that this transformation is initiated by 1,4‐Michael addition, followed by inter‐ and intramolecular nucleophilic addition, and elimination.
