Abstract:We reported a density functional theory (M06–2X) study of the possible mechanisms operating in a chemo- and stereoselective [3+3] annulation reaction of 2-bromoenal with β - ketothioamide catalyzed by a chiral NHC catalyst. DFT computations suggest the catalytic reaction involves nucleophilic addition, [ 1 , 2 ]-proton transfer, debromination, [ 1 , 3 ]-proton transfer, Michael addition, [ 1 , 5 ]-proton transfer, ring closure and catalyst dissociation, in which Michael addition is the chemo- and stereoselectivity-determining step and provides the SR -configured sipro-piperidinone predominantly. This result is consistent with the stereoselectivity observed in experiment. Nonconvalent interaction (NCI) analysis reveals that the origin of stereoselectivity is the different weak interactions (C–H···π, LP···π and π···π). The computational results should deepen our understanding of NHC catalysis, and may thus contribute to the design of new catalytic systems.

Abstract:An intramolecular hydrogen bond-promoted activation of imines represents a new strategy in annulation reactions catalyzed by N-heterocyclic carbenes (NHCs). In this study, the mechanism and the origin of stereoselectivity in the NHC-catalyzed annulation of enals and imines are suggested and investigated theoretically. The computational results show that the reaction between a Breslow intermediate and an intramolecular hydrogen bond-activated imine is the rate- and stereoselectivity-determining step and results in the preferential generation of RS-configured products. The presence of intramolecular hydrogen bond is crucial for the reaction, because the formation of such bond significantly enhances the electrophilicity of the imine and thus facilitate the reaction. Non-covalent interaction (NCI) analysis shows that the CH•••π, CH•••O, π•••π, and lone pair (LP)•••π interactions contribute significantly to stabilizing the lower-energy transition state, thus inducing to the preferential formation of the RS-configured product.

Abstract:We have investigated an N-heterocyclic carbene (NHC)-catalyzed annulation reaction in order to determine the mechanisms and origins of stereoselectivity. DFT results show that the catalytic formation of spiro-glutarimides involves coordination of 2-bromoenal to NHC, followed by a bicarbonate aided proton transfer to form the Breslow intermediate. Subsequent debromination and proton transfer forms the α,β-unsaturated acylazolium intermediate. Then the reaction proceeds via Michael addition, proton transfer and intramolecular cyclization to deliver the cyclic intermediate, which finally releases NHC affording the spiro-glutarimide product. The Michael addition resulting in a new carbon−carbon bond is found to be the stereochemistry-controlling step and generates the SR-configurational product observed in experiment. The important role of noncovalent interactions (such as C−H•••O, C−H•••π, π•••π and lone pair (LP)•••π) in controlling stereoselectivity is elucidated by noncovalent interaction (NCI) analysis. Moreover, analysis of the global reactivity indexes (GRIs) and the computed C−Br bond lengths revealed that NHC plays an important role in activating C−Br bond.

Abstract:Using density functional theory (DFT), a systematic computation has been conducted on the reaction between an enal and an azadiene in the presence of N-heterocyclic carbene (NHC) to explore the catalytic mechanisms and origins of stereoselectivity. Several possible pathways leading to benzofuroazepinone have been investigated. We found that the preferred pathway proceeds via six steps, i.e. (1) the nucleohilic addition wherein the NHC adds to the enal, (2) 1,2- proton transfer allows to form the Breslow intermediate, (3) the carbon−carbon bond formation, (4) 1,3- proton transfer, (5) intramolecular cyclization and (6) catalyst regeneration that eventually leads to the RR-configurational cycloadduct benzofuroazepinone. The DFT results agree well with the experimentally observed stereoselectivity. We also found that HOAc plays a vital role as proton shuttle in proton transfer steps. The 1,3-proton transfer was the stereoselectivity-determining step. Furthermore, we conducted global reactivity index (GRI), distortion/interaction and noncovalent interaction (NCI) analyses to identify the role of the NHC in the reaction as well as the origin of the experimentally observed stereochemistry.

Abstract:The α,β-unsaturated acyl azolium has emerged as a central reactive intermediate for reaction discovery using N-heterocyclic carbene catalysis during the past decades, whereas, the mechanistic studies on transformations of carbonyl compounds involving α,β-unsaturated acyl azolium as key intermediates are rare. In the present study, the mechanism of activation/transformation of enals catalyzed by NHC is theoretically studied using DFT method. Based on the calculations, Michael-type addition process is the stereoselectivity-determining step with SR-configured product being generated preferentially. Furthermore, NCI and AIM analyses have been performed to disclose the origin of stereoselectivity, by which the larger number and stronger noncovalent interactions are the key for stabilizing the low-energy transition state and thus leading to the stereoselectivity inducing.

Abstract:A systematic density functional theory (DFT) computation has been performed to understand the mechanism and stereoselectivity involved in the [3 + 4] annulation reaction between 2-bromoenal and 3-formylindol-2-methyl-malonate catalyzed by N-heterocyclic carbene (NHC). The optimal reaction path consists of nine steps: (1) nucleophilic addition of NHC to 2-bromoenal, (2) formation of the Breslow intermediate via Bronsted acid Et3N•H+-mediated proton transfer, (3) C–Br bond cleavage, (4) formation of acyl intermediate via a proton transfer promoted by Et3N, (5) C–C bond formation via Michael addition, (6) the base (Et3N) mediated proton transfer, (7) protonation of α-carbon atom, (8) intramolecular cyclization and (9) regeneration of NHC. Michael addition (Step 5) is found to be the stereocontrolling step. Non-covalent interactions (NCI) analysis suggests that the preference of S-configured azepino[1,2-a]indoles can be traced to the cumulative effects of C–H•••O, C–H•••N, C–H•••π and π•••π interactions. Notably, the NHC catalyst plays an important role in the C–Br bond activation, which is in contrast to other NHC-catalyzed cyclization reactions.

Abstract:The mechanism, chemoselectivity and stereoselectivity in NHC-catalyzed reaction between enal and pyrrole for the synthesis of 5,6-dihydroindolizine were studied using DFT calculations. The cycle for catalytically generation of 5,6-dihydroindolizine proceeds via seven steps: (1) addition of the NHC to enal, (2) formation of Breslow intermediate through [1,2]-proton transfer, (3) oxidation, (4) Michael addition, (5) [2+2] cycloaddition, (6) liberation of NHC and (7) decarboxylation. Our results show that the presence of DMAP·H+ lowers the barrier for [1,2]-proton transfer. In addition, the NHC·H+ plays a key role in decarboxylation. Michael addition which involves forming a new C–C bond was identified to be the chemo- and stereoselectivity-determining step, leading to the experimentally observed 5S,6R-dihydroindolizine. Analysis of the noncovalent interactions revealed that the observed stereoselectivity is attributed to the differential weak interactions (CH···π, LP···π and CH···CH) involved in the transition states during the Michael addition step. The computational results not only rationalize experimental observations but also provide some useful information for future design of new catalytic processes.

Abstract:The mechanisms and chemo- and regioselectivities of Ru(II)-catalyzed decarboxylative C-H alkenylation of aryl carboxylic acids with alkynes were investigated with density functional theory (DFT) calculations. The catalytic cycle involves sequential carboxylate-directed C-H activation, alkyne insertion, decarboxylation and protonation. The facile tether-assisted decarboxylation step directs the intermediate toward the desired decarboxylative alkenylation, instead of typical annulation and double alkenylation pathways. The decarboxylation barrier is very sensitive to the tether length, and only the seven-membered ring intermediate can selectively undergo the designed decarboxylation, suggesting a tether-dependent chemoselectivity. This tether-dependent chemoselectivity also applies to the alkyl tethers. In addition, the polarity of solvent is found to control the chemoselectivity between the decarboxylative alkenylation and [4 + 2] annulation. Solvent with low polarity (toluene) favors the decarboxylation pathway, leading to the decarboxylative alkenylation. Solvent with high polarity (methanol) favors the ionic stepwise C-O reductive elimination pathway, leading to the [4 + 2] annulation. To understand the origins of regioselectivity with asymmetric alkynes, the distortion/interaction analysis was applied to the alkyne insertion transition states, and led to a predictive frontier molecular orbital model. The asymmetric alkynes selectively use the terminal with the larger HOMO orbital coefficient to form the C-C bond in the insertion step.

Abstract:Axially chiral C2-arylquinoline has been successfully constructed via asymmetric heteroannulation of alkynes catalyzed by chiral phosphoric acid with high yield and high enantioselectivity. Inspired by this intriguing work, theoretical calculations have been carried out, and the detailed reaction mechanism has been elaborated, in which the whole reaction can be divided into steps including hydrogen transfer, C-N bonding, annulation reaction and the final dehydration processes. The initial hydrogen-transfer reaction has two possible pathways, while the subsequent C-N bonding process has eight possible pathways. Then, after the annulation reaction and the final dehydration processes, the major product and byproduct were formed. QTAIM and IGMH analyses were used to illustrate the role of weak intermolecular interactions in the catalytic process, and the distortion/interaction and EDA analyses provided a deeper understanding of the origin of enantioselectivity. The calculated results are consistent with the experimental results. This work would provide valuable insights into asymmetric reactions catalyzed by chiral phosphoric acid.

Abstract:N-heterocyclic carbenes (NHCs) are effective organocatalysts, which are widely used in various cycloaddition reactions. Among them, the [3 + 3] cycloaddition reaction type can be used for the synthesis of six-membered ring, which is also one of the most important methods for the formation of carbon–carbon bonds in organic chemistry. In this paper, a possible reaction pathway for the stereoselective [3 + 3] cycloaddition of bromoenal and β-tetralone have been investigated at the M06-2X/6–31G(d,p)/IEFPCM(THF) level. Our suggested mechanism includes binding of the NHC to bromoenal, 1,2-proton transfer, debromination, 1,3-proton shift, Michael addition, protonation, deprotonation and cyclisation, and NHC regeneration. The stereochemistry of the reaction is determined by the Michael addition step, and the favourable pathway generates the R-configurational dihydropyranone. Furthermore, through the use of global reactivity indexes (GRIs), we are able to demonstrate that the nucleophilicity of the bromoenal is increased by the NHC. The mechanistic insights gained in this work should be helpful in the rational design of potential catalysts for analogous reactions.

Abstract:The mechanism and origins of selectivities in [Ni(NHC)]-catalyzed intramolecular (5 + 2) cycloadditions and homo-ene reactions of vinylcyclopropanes (VCPs) and alkynes have been studied using density functional theory. The preferred mechanism involves oxidative alkyne-alkene cyclization to form a metallacyclopentene intermediate, in contrast to a cyclopropane cleavage pathway in the reaction with Rh(I) catalysts. The selectivity between the (5 + 2) and homo-ene products is determined in the subsequent competing reductive elimination and β-hydride elimination steps. Two similar-sized N-heterocyclic carbene (NHC) ligands, SIPr and ItBu, yielded reversed product selectivity, favoring the (5 + 2) and homo-ene products respectively. This is attributed to the anisotropic steric environment of these NHC ligands, which positions the bulky substituents on the ligand toward different directions and leads to distinct steric control in the reductive elimination and β-hydride elimination transition states.

The reaction may tolerate a broad range of functional groups and has been applied successfully to the synthesis of many natural products and new chiral compounds.

Abstract:A strategy for the N-heterocyclic carbene (NHC) catalyzed asymmetric synthesis of 3,5-diaryl substituted cyclohexenones has been developed via oxidative [4+2] annulation of enals and alkenylisoxazoles. It is the first example of using NHC organocatalysis in a double vinylogous Michael type reaction, a challenging but highly desirable topic. This unprecedented protocol affords good yields as well as high to excellent diastereo- and enantioselectivities.
