Abstract:The effective management of bacterial infections that are resistant to multiple drugs remains a substantial clinical challenge. The eradication of drug-resistant bacteria and subsequent promotion of angiogenesis are imperative for the regeneration of the infected wounds. Here, a novel and facile peptide containing injectable hydrogel with sustained antibacterial and angiogenic capabilities is developed. The antibacterial peptide that consists of 11 residues (CM11, WKLFKKILKVL) is loaded onto acrylate-modified gelatin through charge interactions. A vascular endothelial growth factor mimetic peptide KLT (KLTWQELYQLKYKGI) with a GCG (Gly-Cys-Gly) modification at the N-terminal is covalently coupled through a visible light-induced thiol-ene reaction. In this reaction, the acrylate gelatin undergoes cross-linkage within seconds. Based on the physical/chemical double crosslinking strategy, the bioactive peptides achieve sustained and sequential release. The results show that the hydrogel significantly inhibits methicillin-resistant Staphylococcus aureus (MRSA) growth through the rapid release of CM11 peptides at early stage; it forms obvious growth inhibition zones against pathogenic bacterial strains. Moreover, cell counting kit-8 assay and scratch test confirm that the CM11/KLT-functionalized hydrogels promote cell proliferation and migration through the later release of KLT peptides. In a mouse skin wound infected with self-luminous MRSA, the CM11/KLT-functionalized hydrogels enhance wound healing, with rapidly bacterial infection reduction, lower expression of inflammatory factors, and neovascularization promotion. These results suggest that the rationally designed, sustained and sequential release CM11/KLT-functionalized hydrogels have huge potential in promoting the healing of multi-drug resistant bacterial infected wounds.

Abstract:Photodynamic therapy (PDT) is a promising treatment against multiantibiotic-resistant bacteria with the advantage of a low tendency towards antibiotic resistance. Due to their high PDT efficiencies and superior chemical stabilities, fullerenes have been proposed as effective photosensitizers for the photodynamic inactivation of bacteria. However, the biomedical applications of fullerenes are hindered by their limited aqueous solubility and apparent tendency to undergo aggregation. Herein, we report a hybrid supramolecular hydrogel prepared by the peptide-modulated self-assembly of fullerenes for targeted and sustained photodynamic antibacterial therapy. Aggregation of the fullerene in the hydrogel is largely inhibited through the non-covalent interactions between the peptide and the fullerene. Consequently, the PDT efficiency of the peptide-fullerene hydrogel is highly improved as compared to the untreated fullerene. The incorporation of fullerene profoundly improves the mechanical properties of the hydrogel, making the peptide-fullerene hydrogel a better injectable formulation for biomedical applications. In vitro and in vivo antibacterial results indicate that the peptide-fullerene hydrogels can effectively inhibit multiantibiotic-resistant Staphylococcus aureus and promote wound healing. This study offers a promising paradigm to adapt self-assembling small peptides with integration of multiple functions for biomedical applications.

Abstract:Supramolecular antimicrobial peptide hydrogels can be developed to treat antimicrobial resistant skin infections in terms of a membrane‐disruption and immune‐regulation cascade effect. This work sheds light on a new strategy to combat the antimicrobial resistant bacteria and provides mechanistic insights into the superbug killing and wound healing. Infections from antimicrobial resistant pathogens have been considered as lethal threats to human health if antibiotic treatments become ineffective. Antimicrobial peptides (AMPs) show broad‐spectrum antimicrobial activity against resistant strains via a membrane disruptive mechanism without developing further resistance and are promising candidates to combat resistant microbes. However, currently developed small molecular AMPs are limited by weak antimicrobial efficacy and potential cytotoxicity. Herein, an antimicrobial AMP hydrogel based on supramolecular self‐assembly is developed. The hydrogel shows strong antimicrobial activity and high potency against antimicrobial resistant microbes. It also possesses high cytocompatibility and immunoregulative activity. The synergistic effects lead to its significant healing of the skin abscess disease induced by the methicillin‐resistant Staphylococcus aureus infection. This study not only sheds light on the design strategies and immune‐related antimicrobial actions of supramolecular AMP hydrogels, but also advances the development of self‐assembly peptide platform to fight against antimicrobial resistant infections.

Hydrogel Loaded with Peptide-Containing Nanocomplexes: Symphonic Cooperation of Photothermal Antimicrobial Nanoparticles and Prohealing Peptides for the Treatment of Infected Wounds

Abstract:Current treatment for chronic infectious wounds is limited due to severe drug resistance in certain bacteria. Therefore, the development of new composite hydrogels with nonantibiotic antibacterial and pro-wound repair is important. Here, we present a photothermal antibacterial composite hydrogel fabricated with a coating of Fe2+ cross-linked carboxymethyl chitosan (FeCMCS) following the incorporation of melanin nanoparticles (MNPs) and the CyRL-QN15 peptide. Various physical and photothermal properties of the hydrogel were characterized. Cell proliferation, migration, cycle, and free-radical scavenging activity were assessed, and the antimicrobial properties of the hydrogel were probed by photothermal therapy. The effects of the hydrogel were validated in a model of methicillin-resistant Staphylococcus aureus (MRSA) infection with full-thickness injury. This effect was further confirmed by changes in cytokines associated with inflammation, re-epithelialization, and angiogenesis on the seventh day after wound formation. The MNPs demonstrated robust photothermal conversion capabilities. The composite hydrogel (MNPs/CyRL-QN15/FeCMCS) promoted keratinocyte and fibroblast proliferation and migration while exhibiting high antibacterial efficacy, effectively killing more than 95% of Gram-positive and Gram-negative bacteria. In vivo study using an MRSA-infected full-thickness injury model demonstrated good therapeutic efficacy of the hydrogel in promoting regeneration and remodeling of chronically infected wounds by alleviating inflammatory response and accelerating re-epithelialization and collagen deposition. The MNPs/CyRL-QN15/FeCMCS hydrogel showed excellent antibacterial and prohealing effects on infected wounds, indicating potential as a promising candidate for wound healing promotion.
Injectable Photothermal PDA/Chitosan/β‐Glycerophosphate Thermosensitive Hydrogels for Antibacterial and Wound Healing Promotion

Abstract:Controlling infections while reducing the use of antibiotics is what doctors as well as researchers are looking for. As innovative smart materials, photothermal materials can achieve localized heating under light excitation for broad‐spectrum bacterial inhibition. A polydopamine/chitosan/β‐glycerophosphate (PDA/CS/β‐GP) temperature‐sensitive hydrogel with excellent antibacterial ability was synthesized in the study. Initially, the hydrogel has good biocompatibility. In vitro experiments reveal its non‐cytotoxic property when co‐cultured with gingival fibroblasts and non‐hemolytic capability. Concurrently, the in vivo biocompatibility is confirmed through liver and kidney blood markers and staining of key organs. Crucially, the hydrogel has excellent photothermal conversion performance, which can realize the photothermal conversion of hydrogel up to 3 mm thickness. When excited by near‐infrared light, localized heating is attainable, resulting in clear inhibition impacts on both Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli), with the inhibition rates of 91.22% and 96.69%, respectively. During studies on mice's infected wounds, it is observed that the hydrogel can decrease S. aureus' presence in the affected area when exposed to near‐infrared light, and also lessen initial inflammation and apoptosis, hastening tissue healing. These findings provide valuable insights into the design of antibiotic‐free novel biomaterials with good potential for clinical applications. This article is protected by copyright. Abstract:The development of moisturizing, antibacterial, and biocompatible multifunctional hydrogels is essential to protect skin and promote skin defects recovery. Gelatin has admired potential to be applied for skin care as a hydrogel in virtue of its hydrophilic biocompatible and biodegradable properties. In this study, triclosan-grafted gelatin and photo-cross-linkable methacrylated gelatin were synthesized and then combined to construct the semi-interpenetrating network and antibacterial hydrogels with the aid of a visible blue light. The antimicrobial test demonstrated that the resulting hydrogel obtained excellent inactivation capacity against E. coli, S. aureus, T. rubrum, and C. albicans with sterilizing rates of 99.998%, 99.998%, 99.19%, and 99.64%, respectively. In addition, the cytotoxicity, hemolysis, skin irritation, and rat skin wound healing experiments proved the good biocompatibility of the hydrogel. Therefore, this investigation sheds light on the development of multifunctional hydrogels in skin care.

Abstract:The abuse of antibiotics greatly increases the risk of bacterial resistance. Therefore, it is crucial to develop an efficient nonantibiotic antibacterial treatment strategy. As a result, a self-healable and injectable nanocomposite hydrogel with synergistic antibacterial peptide-photothermal (PTT)-photodynamic (PDT) antibacterial activities was designed and fabricated. The nanocomposite hydrogel was prepared through the facile mixing of hydroxypropyl chitosan-graft-ε-poly(l-lysine) (HPCS-g-EPL), oxidized dextran (OD), and iron-doped carbon dots (Fe-CDs) with the formation of Schiff base bond between HPCS-g-EPL and OD. With the hydrolysis of an amide bond in the hydrogel, EPL grafted onto HPCS will be released continuously. Under near-infrared irradiation for 10 min, Fe-CDs can act as photothermal therapeutic agents to generate heat (approximately 50 °C) and photodynamic therapeutic agents to rapidly catalyze the decomposition of hydrogen peroxide to produce hydroxyl radicals. After the treatment of synergistic antibacterial treatment, the antibacterial rate against Escherichia coli and Staphylococcus aureus can reach more than 99%. Therefore, the antibacterial peptide (EPL)-PTT-PDT synergistic antibacterial therapy strategy without antibiotics has broad prospects in the field of bacterial infection treatment.

Abstract:Infection-induced chronic wounds cause prolonged pains, a high risk of amputation, and even increased mortality in immunocompromised patients. Here we report an antibacterial microneedle (MN) patch, which features high degradability in biological fluids and gelatinase-responsive release of an antibacterial photothermal peptide AMP-Cypate. We first synthesize gelatin nanoparticles (GNPs) and then conjugate the AMP-Cypate to afford composite AMP-Cypate@GNPs. The proteinaceous nanoparticles can responsively release AMP-Cypate in the presence of gelatinase, an enzyme secreted specifically by Staphylococcus aureus (S. aureus). AMP-Cypate@GNPs were then deposited in the tips of MNs fabricated by PVP and recombinant human type III collagen (Col III) to devise the antibacterial MN/AMP-Cypate@GNP patches. When applied to the infection site, MNs break through the epidermis and the stratum corneum, dissolve in the infected dermis, reach the bacterial colony or biofilm, release AMP-Cypate@GNPs, and exert a gelatinase-responsive photothermal therapy under near-infrared (NIR) irradiation to kill the pathogen S. aureus. In a rat model of staphylococcal infection-induced chronic wounds mimicking the condition of diabetic foot ulcer, the antibacterial MN/AMP-Cypate@GNP patches eradiated the bacterial infection and resulted in complete healing of the wounds, proving its potential application in the treatment of chronic wound infections and diabetic foot ulcers.

Abstract:As a pathogenic worldwide pathogen, Staphylococcus aureus (S. aureus) has brought great challenges to the prevention and treatment of diseases in the community. Although antimicrobial peptides show potent antibacterial activity because of their unique bactericidal mechanism, their inherent cationic toxicity cannot be ignored. Utilizing the related characteristics of S. aureus to achieve targeted therapy can reduce the toxicity to host cells to a great extent. Herein, we designed Cy7-labeled antimicrobial peptide (GLFVDK(-Cy7)GKRWWKWWRRGPLGVRGC) with S. aureus targeting recognition peptide (GLFVD) and gelatinase cleavage site (PLGVR), which together with SH-PEG was anchored to gold nanostars (AuNS) by the Au–thiol bond to form a photothermal nanoprobe (APA) targeting S. aureus. The size of the generated APA nanoprobe was 201 ± 3.7 nm, and it was nonfluorescent in the physiological environment due to the fluorescence resonance energy transfer between the AuNS core and Cy7. In vitro results showed that in the gelatinase environment secreted by S. aureus, antimicrobial peptides were released from AuNS and restored the fluorescence of Cy7 in situ, targeting and priority killing of S. aureus. In the mouse wound model of double bacterial infection, APA effectively removed S. aureus from the infected site and promoted the rapid healing of infected wounds through gelatinase-responsive drug release and photothermal synergistic antibacterial treatment.

Abstract:As a non-invasive and non-specific therapeutic approach, photodynamic therapy (PDT) has been used to treat antibiotic-resistant bacteria with encouraging efficacy. Inspired by light, the photosensitizers can produce excessive reactive oxygen species (ROS) and, thus, effectively destroy or kill bacteria. Cyanine (Cy), a traditional photosensitizer for PDT, has the advantages of low cytotoxicity and high ROS yield. Yet, the water solubility and photostability for Cy are poor, which substantially limit its antibacterial efficiency and clinical translation. Herein, we combined Cy with carbomer gel (CBMG) to form a photodynamic Cy-CBMG hydrogel. In this system, Cy was evenly dispersed in CBMG, and CBMG significantly improved the water solubility and photostability of Cy via electrostatic interactions. The developed Cy-CBMG gel had less photodegradation under laser irradiation and thus can effectively elevate ROS accumulation in bacteria. The Cy-CBMG compound presented remarkable ROS-induced killing efficacy against methicillin-resistant Staphylococcus aureus (93.0%) and extended-spectrum β-lactamase-producing Escherichia coli (88.7%) in vitro. Moreover, as a potential wound dressing material, the Cy-CBMG hydrogel exhibited excellent biocompatibility and effective antimicrobial ability to promote wound healing in vivo. Overall, this work proposed a practical strategy to synthesize a photosensitizer–excipient compound to enhance the photophysical property and antibacterial efficacy for PDT.

Abstract:An injectable, self‐healing and degradable hydrogel (CPDT) possesses good photodynamic and photothermal effects, and can effectively inhibit activity of S. aureus, E. coli and MRSA under laser irradiation with 660 and 808 nm. Hydrogel dressings play an important role in the treatment of bacterial infection. In this study, carboxymethyl chitosan (CM‐CS) was cross‐linked with aldehyde polyglycol (DF‐PEG), and then polydopamine (pDA) and 5,10,15, 20‐tetrakis (4‐aminophenyl) porphyrin (TAPP) were grafted to prepare a CM‐CS/DF‐PEG/pDA/TAPP hydrogel (CDPT). The results showed that the CDPT hydrogel had good adhesion, injectability, self‐healing, degradability, and biocompatibility. Under near‐infrared irradiation, the CDPT hydrogel has excellent photothermal and photodynamic effects and can effectively inhibit activity of Staphylococcus aureus (S. aureus), Escherichia coli (E. coli) and antibiotic‐resistant, methicillin‐resistant Staphylococcus aureus (MRSA). In summary, CDPT hydrogel has potential applications as antibacterial material.

Abstract:Bacterial infection of wounds is an escalating medical problem, issuing threats to both global public health and personal health. Photothermal antibacterial technology as a novel sterilization strategy has outstanding sterilization efficiency, high safety and low risk of emergence of drug-resistant bacteria. By combining inherent antibacterial activity and light-assisted antibacterial treatment, developing novel multifunctional dressings with synergistic high-efficiency antibacterial effects and also promoting wound healing possesses attractive advantages in the field of treating bacterial wound infections in clinical care. Herein, a multifunctional hydrogel formed by in situ photo-cross linking was designed and prepared by first grafting methacrylic anhydride as a photosensitizer onto chitosan, and then introducing oxidatively synthesized polydopamine (PDA). The physicochemical characterizations of the synthesized hydrogels demonstrated their tunability certainly associated with PDA concentration, including pore size, water swelling, rheological properties and in vitro degradability. In addition, the composite hydrogels exhibited good adhesion, anti-oxidation and photothermal properties due to the existence of PDA. Within 10 min upon exposure to 808 nm near-infrared (NIR) light irradiation, this hydrogel system displayed outstanding antibacterial activity against Staphylococcus aureus with almost 100% killing efficiency, of which rapid efficient sterilization plays a significant role in wound healing. Moreover, the hydrogel is capable of cytocompatibility and has low toxicity to murine fibroblasts (L929 and NIH/3T3). In the full-thickness wound defect infection model in mice, the wound closure ratio, inflammatory response, fibroblasts, neovascularization and epithelialization were measured. Animal experiments also reveal that the hydrogel assisted with NIR laser irradiation can inhibit effectively infection at an early stage and accelerate the wound healing process. In , this novel multifunctional injectable hydrogel exhibits excellent swelling capacity, bio-adhesion, antioxidant property, photothermal activity, efficient antibacterial property and facilitates skin healing, which has great promising application as a medical dressing biomaterial in infected wound care fields.

Abstract:Bacterial biofilm-associated infection was one of the most serious threats to human health. However, effective drugs for drug-resistance bacteria or biofilms remain rarely reported. Here, we propose an innovative strategy to develop a multifunctional antimicrobial agent with broad-spectrum antibacterial activity by coupling photosensitizers (PSs) with antimicrobial peptides (AMPs). This strategy capitalizes on the ability of PSs to generate reactive oxygen species (ROS) and the membrane-targeting property of AMPs (KRWWKWIRW, a peptide screened by an artificial neural network), synergistically enhancing the antimicrobial activity. In addition, unlike conventional aggregation-caused quenching (ACQ) photosensitizers, aggregation-induced emission (AIE) PSs show stronger fluorescence emission in the aggregated state to help visualize the antibacterial mechanism. In vitro antibacterial experiments demonstrated the excellent killing effects of the developed agent against both Gram-positive (G + ) and Gram-negative (G – ) bacteria. The bacterial-aggregations induced ability enhanced the photoactivatable antibacterial activity against G – bacteria. Notably, it exhibited a significant effect on destroying MRSA biofilms. Moreover, it also showed remarkable efficacy in treating wound infections in mice in vivo . This multifunctional antimicrobial agent holds significant potential in addressing the challenges posed by bacterial biofilm-associated infections and drug-resistant bacteria.

Versatile Hydrogel Dressing with Skin Adaptiveness and Mild Photothermal Antibacterial Activity for Methicillin‐Resistant Staphylococcus Aureus‐Infected Dynamic Wound Healing

Abstract:A versatile hydrogel conferred skin adaptiveness on dynamic wound healing, including flexible mechanical properties, strong tissue adhesion and excellent self‐healing ability. Moreover, the design of specific bacterial capture, membrane puncture and enhanced near‐infrared permeability, produced a safe and efficient mild PTT antibacteria against methicillin‐resistant Staphylococcus aureus and accelerated the tissue regeneration of MRSA‐infected wounds. Bacterial infection often induces chronic repair of wound healing owing to aggravated inflammation. Hydrogel dressing exhibiting intrinsic antibacterial activity may substantially reduce the use of antibiotics for infected wound management. Hence, a versatile hydrogel dressing (rGB/QCS/PDA–PAM) exhibiting skin adaptiveness on dynamic wounds and mild photothermal antibacterial activity is developed for safe and efficient infected wound treatment. Phenylboronic acid‐functionalized graphene (rGB) and oxadiazole‐decorated quaternary carboxymethyl chitosan (QCS) are incorporated into a polydopamine–polyacrylamide (PDA–PAM) network with multiple covalent and noncovalent bonds, which conferred the hydrogel with flexible mechanical properties, strong tissue adhesion and excellent self‐healing ability on the dynamic wounds. Moreover, the glycocalyx‐mimicking phenylboronic acid on the surface of rGB enables the hydrogel to specifically capture bacteria. The enhanced membrane permeability of QCS enhanced bacterial vulnerability to photothermal therapy(PTT), which is demonstrated by efficient mild PTT antibacteria against methicillin‐resistant Staphylococcus aureus in vitro and in vivo at temperatures of <49.6 °C. Consequently, the hydrogel demonstrate accelerated tissue regeneration on MRSA‐infected wound in vivo, with an intact epidermis, abundant collagen deposition and prominent angiogenesis. Therefore, rGB/QCS/PDA–PAM is a versatile hydrogel dressing exhibiting inherent antibacterial activity and has considerable potential in treating wounds infected with drug‐resistant bacteria.

Abstract:It is of paramount importance to develop novel approaches for combating bacterial resistance and the integration of different antibacterial mechanisms is essential to achieve synergistic bactericidal efficiency while reducing the associated side effects. Herein, amphiphilic antimicrobial copolymers derived from poly-l-lysine (PLL), black phosphorus quantum dots (BPQDs) as near-infrared (NIR) sensitizer, and S-nitrosocysteamine (SNO) as nitric oxide (NO) donor, are assembled into PELI@BPQD-SNO nanoparticles through electrostatic interactions. Amphiphilic copolymers with isopentanyl grafts on PLL at a ratio of 50% achieve an optimal balance between antibacterial activity and hemolysis rate. Photothermal effect of BPQDs leads to NIR-responsive release of NO and the combination with amphiphilic copolymers mutually enhances long-term inhibition of bacterial growth. In an S. aureus-infected subcutaneous abscess model, the bactericidal rate of PELI@BPQD-SNO/NIR treatment reaches nearly 99.6%, which is significantly higher than those without NO release (38%) or amphiphilic copolymers (24%) or NIR irradiation (17%). PELI@BPQD-SNO/NIR treatment shows full recovery of infected wounds, efficient retardation of inflammatory cells, and reconstruction of blood vessels similar to those of healthy skin. Therefore, the electrostatic assembly demonstrates a promising strategy to deliver charged therapeutic agents and the photoactivated release of NO and amphiphilic copolymers achieves synergistic antibacterial efficacy without using any antibiotics.

Abstract:Wound infections are a compelling health issue caused by the invasion and proliferation of pathogens in wound sites. Antibiotic-loaded hydrogels are widely used to achieve anti-infectious wound healing. However, due to the quick emergence of drug-resistant bacteria, such as methicillin-resistant Staphylococcus aureus (MRSA), wound infection has been a formidable challenge to human health. To address MRSA-infected wounds, an antibacterial peptide polymer-loaded hyaluronic acid (HA) hydrogel (Gel-HA@P) is prepared. The peptide polymer is designed to mimic host defense peptides as the antibiotic alternative showing potent antibacterial activity, low susceptibility to drug resistance and good stability against proteolysis. HA is biocompatible and biodegradable hydrogel substrate as a primary constituent of the extracellular matrix and suitable for cell migration and wound healing. Gel-HA@P shows potent activity against MRSA in vitro and in vivo, low toxicity during the treatment and promotes the wound healing in vivo. This design has proven to be an effective and antibiotic-free strategy to enable the healing of MRSA-infected wounds.

Abstract:Antibacterial hydrogels are attracting extensive attention in soft tissue repair and regeneration, including bacteria-infected-wound healing. The abuse of antibiotics leads to drug resistance. Recent developments have demonstrated that the delivery of inorganic bactericidal agents in hydrogels can drive the wound healing process; however, this approach is complicated by external light stimuli, cytotoxicity, nondegradability, and sophisticated fabrication. Herein, an inherent antibacterial, bioresorbable hydrogel was developed by the spontaneous self-aggregation of amphiphilic, oxadiazole-group-decorated quaternary ammonium salts (QAS)-conjugated poly(ε-caprolactone)-poly(ethylene glycol)-poly(ε-caprolactone) (PCEC-QAS) micellar nanoantimicrobials for methicillin-resistant Staphylococcus aureus (MRSA)-infected cutaneous wound healing. The PCEC-QAS hydrogel showed a stable gel state within the temperature range of 5–50 °C and antibacterial efficacy against both Gram-negative and -positive bacteria in vitro and in vivo. Additionally, the PCEC-QAS hydrogel facilitated the cell spreading, proliferation, and migration without cytotoxicity. An in vivo degradation and skin defect healing study suggested the PCEC-QAS hydrogel was totally absorbed without local or systemic toxicity and could promote wound repair in the absence of drugs, cytokines, or cells. Significantly, this hydrogel accelerated the regeneration of a MRSA-infected full-thickness impaired skin wound by successfully reconstructing an intact and thick epidermis similar to normal mouse skin. Abstract:Photodynamic therapy (PDT) emerges as a promising strategy for combating bacteria with minimal drug resistance. However, a significant hurdle lies in the ineffectiveness of most photosensitizers against Gram-negative bacteria, primarily attributable to their characteristic impermeable outer membrane (OM) barrier. To tackle this obstacle, we herein report an amphipathic peptide-photosensitizer conjugate (PPC) with intrinsic outer membrane disruption capability to enhance PDT efficiency against Gram-negative bacteria. PPC is constructed by conjugating a hydrophilic ultrashort cationic peptide to a hydrophobic photosensitizer. PPC could efficiently bind to the OM of Gram-negative bacteria through electrostatic adsorption, and subsequently disrupt the structural integrity of the OM. Mechanistic investigations revealed that PPC triggers membrane disruption by binding to both lipopolysaccharide (LPS) and phospholipid leaflet in the OM, enabling effective penetration of PPC into the Gram-negative bacteria interior. Upon light irradiation, PPC inside bacteria generates singlet oxygen not only to effectively decrease the survival of Gram-negative bacteria P. aeruginosa and E. coli to nearly zero in vitro, but also successfully cure the full-thickness skin infection and bacterial keratitis (BK) induced by P. aeruginosa in animal models. Thus, this study provides a broad-spectrum antibacterial phototherapeutic design strategy by the synergistic action of membrane disruption and PDT to combat Gram-negative bacteria.
