Completed Autophagy Protected Airway Epithelial Barrier From Antigens Stimulation

Abstract:Asthma is a complex disease, caused by antigens, virus and environmental pollution in children or adults. Autophagy, progression of self-protection in cells, is a common program when cells are being attacked by certain stress. The association between asthma and autophagy, however, is less demonstrated. Our study focused upon the disruption of airways epithelial barrier in asthma, then we found whether autophagy played a key role in this process. We used HBECs to be treated with HDM for certain time, autophagy could be detected by western blotting, after that, we chose accepted autophagy related genes to determine the association between autophagy and disruption of airway epithelial barrier. Atg5 and atg12 were chosen because these two genes varied upon the time dependent manner. Knocked down the expression of atg5 or atg12, then the level of TSLP, which induce the disruption of airway epithelial barrier, remarkably reduced. While blockade the procession of autophagy by Chloroquine diphosphate salt, the barrier of airway epithelium disrupted which highly distinct to the results being referred above. At the end, we found phosphorylation of Erk1/2 increased in HBECs under the treatment of HDM. Different results of autophagic progression affected the disruption of airway epithelial barrier in different ways, these results demonstrated autophagy was induced in HBECs to protect cells from harmful stimulation but the progression was blockaded, HBECs can not survive from death by the barrier of airway epithelium will be disrupted inevitably.
