Self-assembly, as a main method of forming supramolecular complexes, is significant to polymeric materials and life sciences. Hydrogen-bonded complexes are classified according to their multiplicity and associating ways in this article. The self-assembly properties and research of hydrogen-bonded complexes are comprehensively reviewed. Meanwhile, theoretical studies on hydrogen-bonded complexes of our research group are introduced.

Abstract:Macromolecular self-assembly (MSA) has been an active and fruitful research field since the 1980s, especially in this new century, which is promoted by the remarkable developments in controlled radical polymerization in polymer chemistry, etc. and driven by the demands in bio-related investigations and applications. In this review, we try to summarize the trends and recent progress in MSA in relation to biomimetic chemistry and bio-inspired materials. Our paper covers representative achievements in the fabrication of artificial building blocks for life, cell-inspired biomimetic materials, and macromolecular assemblies mimicking the functions of natural materials and their applications. It is true that the current status of the deliberately designed and obtained nano-objects based on MSA including a variety of micelles, multicompartment vesicles, and some hybrid and complex nano-objects is at their very first stage to mimic nature, but significant and encouraging progress has been made in achieving a certain similarity in morphologies or properties to that of natural ones. Such achievements also demonstrate that MSA has played an important and irreplaceable role in the grand and long-standing research of biomimetic and bio-inspired materials, the future success of which depends on mutual and persistent efforts in polymer science, material science, supramolecular chemistry, and biology.

Future developments in the field of biomedical applications of layer-by-layer assembly are also discussed.

Abstract:The complexity and diversity of biomacromolecules make them a unique class of building blocks for generating precise assemblies. They are particularly available to a new generation of biomaterials integrated with living systems due to their intrinsic properties such as accurate recognition, self-organization, and adaptability. Therefore, many excellent approaches have been developed, leading to a variety of quite practical outcomes. Here, we review recent advances in the fabrication and application of artificially precise assemblies by employing proteins and carbohydrates as building blocks, followed by our perspectives on some of new challenges, goals, and opportunities for the future research directions in this field.

Abstract:The pursuit of precision in polymer synthesis and assembly has been the Holy Grail of macromolecular science. Nowadays, precision macromolecules have come to the interface of organic chemistry, polymer science, and biotechnology. We take an interdisciplinary approach to achieving precision at both molecular and supramolecular level in the hope of gaining unique functions. Specifically, sequential "click" chemistry has been utilized to enable modular, robust and efficient construction of giant molecules; cellular synthesis of proteins with complex topologies and/or mechanical bonds have been accomplished using genetically encoded peptide-protein chemistry. These precise macromolecules exhibit intriguing and versatile self-assembly behaviors both in solution and in bulk. Not only does the assembly exhibit a remarkable sensitivity to the primary chemical structures, it also leads to unconventional phases in soft matter, including Frank-Kasper phases and quasicrystals. Based on these results and other peoples' work, we reflect on the role of "precision" in the interplay among chemical structure, physical structure, and function, which stimulates us to further pursue precision macromolecules thoughtfully in specific context to the extent that is both necessary and possible.

Abstract:Peptide self-assemblies show intriguing and tunable physicochemical properties, and thus have been attracting increasing interest over the last two decades. However, the micro/nano-scale dimensions of the self-assemblies severely restrict their extensive applications. Inspired by nature, to genuinely realize the practical utilization of the bio-organic super-architectures, it is beneficial to further organize the peptide self-assemblies to integrate the properties of the individual supermolecules and fabricate higher-level organizations for smart functional devices. Therefore, cumulative studies have been reported on peptide microfabrication giving rise to diverse properties. This review summarizes the recent development of the microfabrication of peptide self-assemblies, discussing each methodology along with the diverse properties and practical applications of the engineered peptide large-scale, highly-ordered organizations. Finally, the current limitations of the state-of-the-art microfabrication strategies are critically assessed and alternative solutions are suggested.

Abstract:<p>Biomolecular self-assembly is a key process used by life to build functional materials from the "bottom up." In the last few decades, bioengineering and bionanotechnology have borrowed this strategy to design and synthesize numerous biomolecular and hybrid materials with diverse architectures and properties. However, engineering biomolecular self-assembly at solid–liquid interfaces into predesigned architectures lags the progress made in bulk solution both in practice and theory. Here, recent achievements in programming self-assembly of peptides, proteins, and peptoids at solid–liquid interfaces are summarized and corresponding applications are described. Recent advances in the physical understandings of self-assembly pathways obtained using in situ atomic force microscopy are also discussed. These advances will lead to novel strategies for designing biomaterials organized at and interfaced with inorganic surfaces.</p>

Abstract:Understanding of new materials at the molecular level has become increasingly critical for a new generation of nanomaterials for nanotechnology, namely, the design, synthesis and fabrication of nanodevices at the molecular scale. New technology through molecular self-assembly as a fabrication tool will become tremendously important in the coming decades. Basic engineering principles for microfabrication can be learned by understanding the molecular self-assembly phenomena. Self-assembly phenomenon is ubiquitous in nature. The key elements in molecular self-assembly are chemical complementarity and structural compatibility through noncovalent interactions. We have defined the path to understand these principles. Numerous self-assembling systems have been developed ranging from models to the study of protein folding and protein conformational diseases, to molecular electronics, surface engineering, and nanotechnology. Several distinctive types of self-assembling peptide systems have been developed. Type I, "molecular Lego" forms a hydrogel scaffold for tissue engineering; Type II, "molecular switch" as a molecular actuator; Type III, "molecular hook" and "molecular velcro" for surface engineering; Type IV, peptide nanotubes and nanovesicles, or "molecular capsule" for protein and gene deliveries and Type V, "molecular cavity" for biomineralization. These self-assembling peptide systems are simple, versatile and easy to produce. These self-assembly systems represent a significant advance in the molecular engineering for diverse technological innovations.

Abstract:Macromolecular self-assembly refers to the assembly of synthetic polymers, biomacromolecules and supra-molecular polymers. Through macromolecular self-assembly, the fabrication of ordered structures at different scales, the control of the dynamic assembly process and the integrations of advanced functions can be realized. Macromolecular self-assembly and nanotechnology research in China has developed rapidly, from the early periods of follow-up at low to high level and progress into a stage of innovation and creation. This review selects some representative progresses achieved recently, aiming to reflect the current status of macromolecular self-assembly and nanotechnology research in China.

Abstract:Molecular assembly (MA) is an essential approach to create novel substances and new functional materials beyond molecules. Improving the controllability of MA processes and the functionality of assemblies is the core target in this field. However, due to the limitation of methods and theories for complex MA systems, most of current researches are based on the “black box” paradigm, which can mostly focus on the initial and final state of MA. Such a paradigm has become a bottleneck that restricts the development of MA. Here, we review the current theoretical methods and models of MA systems from the molecular to subcellcular level. We try to measure the complexity of MA research from the dimension of matter, energy and information, and gradually explore the current status, opportunities and breakthrough of the theoretical research of MA: in the dimension of matter, we illuminate that the pathways of MA processes are multi-step, multi-pathway and multi-scale; in the integrated dimension of energy and matter, we demonstrate that MA systems are usually accompanied by the entropy-driven phenomena or the complementation of entropy and enthalpy. We further discuss how the MA systems far from the equilibrium can form an order spatiotemporal dissipated structure; in the integrated dimension of information, energy and matter, we demonstrate how the synergetic processes of positive or negative feedback facilitates the emergence of complex physiological functions of MA systems. To address these challenges in the development of MA theoretical researches, we probably need to establish a larger theoretical framework and synergistically develop the research methods of MA from the three dimensions of matter, energy and information. It seems to help us to explore its fundamental laws of MA comprehensively, then establish the new theory and promote the development of assisting methods with high efficiency, and finally elevate the complexity and functionality of MA system, which may provide new perspectives and methods for life science and soft matter science.

Abstract:In the past fifty years we have witnessed great advance in the research field of chemical self-assembly.In the future,studies on chemical self-assembly will absolutely continue to be one of the most active research fields of chemical science.How far can we push chemical self-assembly in the coming decades? The answer is probably "no answer".But there are so many challenges which relate to chemical self-assembly and wait for chemists to address in the future.In my personal opinion,new advances or breakthroughs on tackling these challenges may be considered as the hallmarks where we have reached.Herein I would like to list some essential aspects concerning these challenges.To some extent,life is the product of chemical self-assembly and represents its most advanced form.Thus,since its original stage,chemical self-assembly has largely borrowed the inspiration of life in assembling defined and functional structures.One important aspect for self-assembly research is the creation of so-called "artificial" systems that are able to mimic one or more essential behaviors or functions of life.We have already developed artificial enzymes,self-replication,photosynthetic systems and life,and so on by making use of self-assembly.However,we are not able to avoid enzyme poisoning,caused by increased binding of the products to their enzymes.DNA polymerase-like synthetic architectures are not available that would enable the catalysis of an artificial self-replication.Advance on photosynthesis research is still far away from practical application for producing clear energy.Even most advanced self-assembled architectures are only able to exhibit very simple characters relating to life.Clearly,there exists a huge space for the development of artificial self-assembled systems that mimic the behavior of life or biological species and eventually provide useful functions.Self-assembly will strengthen its presence when it begins to produce more and more practical products for the human society.Currently,recognition-based devices have been used for public security,and several liposome drug products are also commercially available for improving drug treatment.However,applying self-assembly strategy to the development of new advanced materials is just at infancy stage.We may very much expect the generation of novel energy materials and smart self-healing materials in the future.In the context of molecular materials,the success of the so-called materials genome should,to a great extent,rely on the practicing of self-assembly principle for rational molecular design.With respect to fundamental research of self-assembly,we may say that the prospect is beyond imagination,from theory to structure.The ultimate vision may be the achievement of controllable operation on molecular arrangement from the nano scale to the micro-,meso-,and macroscale.

Abstract:If there is a secret recipe that enables living cells to build themselves from individual molecules, it is likely to be hierarchical self-organization. Here, we summarize recent progress in synthetic self-assembly analogous to subcellular structures, including flattened sacs, crystalline membranes, reconfigurable coacervate droplets, semiflexible filaments, and asters. Simplicity is the key of these synthetic systems-they can reproduce the architecture and, sometimes, functions of seemingly complicated biological systems with surprisingly minimal constituents, underlying the overwhelming importance of fundamental physicochemical mechanisms over specific molecular details. Beyond molecular self-assembly on a microscale, we expect integration of the assembled structures to function in unison and synergy as the next step towards cell imitation.

Molecular recognition and self-assembly are always the fundament of information storage, duplication and transferring in nature, and therefore based on this, the fabrication of specific supramolecular systems with target biological functions will be crucial important in revealing the nature and process of life. Based on our previous studies, three concerning fields are summarized in the present paper: (1) The mimic of glutathione peroxide (GPX) and the recognition between it and substrate; (2) Tissue regeneration material and bioactivated scaffold; (3) self-assembly and disassembly of virus-like particle (VLP).

Differing from micelles of block and graft copolymers, our micelles don't have any chemical bonds between the core and shell. Abstract:Biomolecular self-assembly provides a facile way to synthesize functional nanomaterials. Due to the unique structure and functions of biomolecules, the created biological nanomaterials via biomolecular self-assembly have a wide range of applications, from materials science to biomedical engineering, tissue engineering, nanotechnology, and analytical science. In this review, we present recent advances in the synthesis of biological nanomaterials by controlling the biomolecular self-assembly from adjusting internal interactions and external stimulations. The self-assembly mechanisms of biomolecules (DNA, protein, peptide, virus, enzyme, metabolites, lipid, cholesterol, and others) related to various internal interactions, including hydrogen bonds, electrostatic interactions, hydrophobic interactions, π–π stacking, DNA base pairing, and ligand–receptor binding, are discussed by analyzing some recent studies. In addition, some strategies for promoting biomolecular self-assembly via external stimulations, such as adjusting the solution conditions (pH, temperature, ionic strength), adding organics, nanoparticles, or enzymes, and applying external light stimulation to the self-assembly systems, are demonstrated. We hope that this overview will be helpful for readers to understand the self-assembly mechanisms and strategies of biomolecules and to design and develop new biological nanostructures or nanomaterials for desired applications.

Abstract:Theoretical work related to the self-assembly of organic materials was dealt with, and the various mechanisms leading to self-assembly, such as transition metal mediated self-assembly, constraint induced self-assembly, covalent bond based self-assembly and van der Waals interaction driven self-assembly, etc., were discussed. The formation of ordered structures could be attributed to the competition between short range attractive forces and long-range repulsion, which was arising from dipole interaction or may result from a different mechanism based on a purely repulsive isotropic short-range pair potential with two characteristic length scales. Such mechanism could be exploited in the study of self-assembly process. First principles SAPT(DFT) interaction energy calculations, combined with the Williams-Stone-Misquitta method, offer the ability to improve the molecular dynamics (MD) accuracy which could in turn be used in the prediction of crystal structures and self-assembly tendency. The combination of experimental and theoretical studies could open new breakthroughs over the design, synthesis, and characterization of self-assembled materials.

Abstract:<p>Natural systems are typically featured with tremendous molecular complexity and construct exquisite architectures and functional materials through precise spatial and temporal control. Supramolecular self-assembly from peptides and proteins is believed to be the key player. Thus, knowledge of the fundamental mechanisms driving peptides into vast functional hierarchical structures will certainly help the rational control of self-assembly process, leading to more precise structural organization and functional optimization. In this review, we briefly summarize the recent progress of this burgeoning field mainly from two directions: peptide-based self-assembly and co-assembly with other functional molecules, each part is further divided into two subparts and representative examples are given for each subpart according to their development timeline. At the end of each part, brief summaries of the closely related applications are outlined. For the closing remarks, we conclude with our own understanding of the area and perspectives are given based on recent developments. Overall, this review could be suitable for both new readers to gain a comprehensive overview of the area and experienced readers to get a summary of the development in short peptide self-assembly from particles to functional materials.</p>

Abstract:Peptide self-assembly is the process by which peptide molecules aggregate into low dimensional (1D, 2D) or 3D ordered materials with potential applications ranging from drug delivery to electronics. Short peptides are particularly good candidates for forming supramolecular assemblies due to the relatively simple structure and ease of modulating their self-assembly process to achieve required material properties. The experimental resolution of fibrous peptide-based nanomaterials as 3D atomic coordinates remains challenging. For surface-mediated peptide assembly in particular, it is typically not feasible to resolve multiple conformationally distinct surface bound peptide structures by experiment. The mechanisms of peptide self-assembly also remain elusive due to the interchange of complex interactions and multiple time and length scales involved in the self-assembly process. Peptide self-assembly in solution, or mediated by surfaces, is driven by specific interactions between the peptides and water, competing interactions within the peptide and/or between peptide aggregate units and, in the latter case, an interplay of the interactions between peptides and solvent molecules for adsorption onto a proximal surface. Computational methodologies have proven beneficial in elucidating the structures formed during peptide self-assembly and the molecular mechanisms driving it, and hence have scope in facilitating the development of functional peptide-based nanomaterials for medical or biotechnological applications. In this perspective, computational methods that have provided molecular insights into the mechanisms of formation of peptide biomaterials, and the all-atom-resolved structures of peptide assemblies are presented. Established and recently emerged molecular simulation approaches are reviewed with a focus on applications relevant to peptide assembly, including all-atom and coarse-grained “brute force” molecular dynamics methods as well as the enhanced sampling methodologies: umbrella sampling, steered and replica exchange molecular dynamics, and variants of metadynamics. These approaches have been shown to contribute all-atom details not yet available experimentally, to advance our understanding of peptide self-assembly processes and biomaterial formation. The scope of this review includes a summary of the current state of the computational methods, in terms of their strengths and limitations for application to self-assembling peptide biomaterials.
