A sensitive beagle model to evaluate anaphylactoid reactions

Abstract:Background: Anaphylactoid reactions lead to high morbidity and health-care costs every year. A successful animal model is a prerequisite for the research of anaphylactoid reactions. The aim of the study was to detect a sensitive animal model of anaphylactoid reactions. Methods. Animals such as beagle dogs, guinea pigs, and mice were used and intravenously injected with Tween-80, a trigger of anaphylactoid reactions. The behavioral responses were observed. The vascular permeability was assayed by spectrophotometry. Results: The results indicated that serious multisystem symptoms appeared in the dogs after intravenous Tween-80. One dog displayed anaphylactoid symptoms grade III such as scratching the nose and head, shaking the ears, skin rash, gait disturbance; Five dogs displayed grade IV symptoms of pawing the ground, tumbling, sluggishness, somnolence or hypodynamia. Moreover, the skin rash was generalized and remarkable. However, guinea pigs did not display any abnormal behavior after the administration of Tween-80. In addition, there was no significant increase of Evans blue absorbance in mouse ascites and skin after administration of Tween-80. The Evans blue absorbance of muscle in the Tween-80 group was higher than the one in the control group. Conclusions: Beagles dogs are a sensitive animal model of anaphylactoid reactions.
