Mutations in DNA Methyltransferases and Demethylases

Abstract:Recent cancer genomic studies have revealed that several enzymes regulating DNA methylation are frequently mutated in cancer, especially in hematopoietic malignancies. Among these enzymes, DNMT3A and TET2 are responsible for “writing” and “erasing” DNA methylation, respectively, whereas IDH1/2 act as important regulators. Functional studies define the important roles of these enzymes in hematopoiesis and neoplastic transformation. Interestingly, mutated IDH1/2 acquire a neomorphic activity to produce a “oncometabolite,” 2-HG, which inhibits the TETs and other dioxygenases involved in several cellular pathways. Clarification of the mechanisms shall advance our understanding of epigenetic regulation of cancer and facilitate development of new therapeutics.
