Abstract:The Alberta Plant Health Lab (APHL) provides plant pest diagnosis and expertise to Alberta's agricultural industry. The lab accepts samples exclusively from agricultural fieldmen, academic institutions, applied research associations and municipal pest management departments, at no cost. A total of 542 samples were processed for diagnosis in the 2019 crop year. Dutch elm disease was not identified in any of the 16 suspect elm samples received. There was a substantial increase in conifer samples diagnosed with needle cast pathogens in 2019. For fusarium head blight in cereals, no sample was identified as positive. METHODS: Samples are submitted to the Alberta Plant Health Lab (APHL) by agricultural fieldmen, academic institutions, applied research associations and municipal pest management departments. Diagnoses are based on a combination of visual examination of symptoms, microscopic observation, culturing on artificial media, PCR/qPCR, DNA barcoding and use of commercial diagnostic kits. Specifically, fungal barcoding was performed by sequencing DNA fragments of internal transcribed spacers (ITS) (White et al. 1990) and/or elongation factor -1 alpha (EF1) (Stielow et al. 2015) and/or beta-tubulin (Stukenbrock et al. 2012). Bacteria are usually identified based on DNA sequencing of 16S ribosomal RNA gene (Klindworth et al. 2013) and/or cpn60 (Links et al. 2012). The diagnosis of the new clubroot pathotype of P5-like was according to Zhou et al. (2018). PCR identification of Fusarium graminearum from submitted cultures was performed following Zuzak et al. (2018). For identification of Fusarium species from plant tissues, protocols described by Demeke et al. (2005) were used. Phytoplasma were detected by PCR using the primer pairs P1/Tint and R16MF2n/R16MR2n (Smart et al. 1996). Confirmation of late blight on potato and tomato was conducted using the Agdia ImmunoStrip kit for Phytophthora species (http://www.agdia.com). For diagnosis of all other diseases, when PCR techniques were used, quantitative PCR (qPCR) preceded conventional PCR and probe-based qPCR preceded SYBR Green based qPCR. The primers and protocols were chosen from the most recent literature and verified by APHL using positive and negative controls. RESULTS: A total of 542 disease diagnoses were completed between January 1 and October 31, 2019. Categories of samples received for diagnosis included cereals (11.4%), canola (6.4%), potato (6.4%), legume (6.3%), tree and fruit (44.7%), vegetable (18.5%) and other (6.3%). The category 'other' covers samples such as flax, hemp, sunflower, thistle and tulip flower. Fungal, oomycete, protist, bacterial and viral pathogens were identified in these samples as the causal agents of disease. In most samples, one or more causal agents were identified. Summaries of symptoms and associated pathogens diagnosed on the submitted samples are provided in Tables 1 to 7 by crop category. The diagnoses reported may not necessarily reflect the major problems encountered during the season in the field, but rather those most prevalent within the samples submitted for testing. Fusarium species were commonly isolated from the various cereal crop samples. Except in one sample of corn, no F. graminearum was isolated. Take-all, tan spot, leaf blotch and leaf blight were also observed. Twenty-one canola samples were confirmed to be infected by Plasmodiophora brassicae. One of those samples was confirmed to be infected by a new pathotype of P. brassicae. Stem cankers caused by Leptosphaeria maculans and L. biglobosa, white mould caused by Sclerotinia sp., powdery mildew caused by Erysiphe sp., and positive phytoplasma were observed on canola samples. Fusarium spp., Pythium spp. and Alternaria spp. causing root rot, seedling blight and leaf spot diseases, respectively, were commonly found on legume crops. Damping off (Phytophthora sojae) and ascochyta blight were found in one soybean sample and one field pea sample, respectively. For potato samples, there was no detection of Dickeya dianthicola, although blackleg (Pectobacterium atrosepticum), powdery scab (Spongospora subterranea) and phytoplasma were detected. Sixteen elm samples were submitted for Dutch elm disease diagnosis and none of them tested positive for this disease. Three samples were infected by Cytospora sp., Nectria sp. and Phaeobotryon rhois, respectively. All the other elm samples were infected by Plenodomus sp. Sydowia polyspora was identified as the causal agent of needle cast and blight in 27 spruce, nine pine and three balsam fir samples. This pathogen is associated with tip dieback as well as needle cast of conifers. Cytospora canker (Cytospora sp.) from elm (one sample), spruce (five samples), balsam fir (two samples) and Saskatoon berry (10 samples); leaf blight (Alternaria spp.) from spruce (37 samples) and pine (three samples); and dieback (Valsa sp. on willow, Pseudomonas syringae on poplar) were also detected. Fire blight (Erwinia amylovora) was detected on pear and crab apple. Alternaria spp. were also commonly detected on conifer needles, oak leaves, willow, Saskatoon berry, apple, raspberry and strawberry. Fruit rot caused by Botrytis spp. was detected in Saskatoon berry, raspberry and strawberry. Fusarium species caused root rot and dry rot of beet (two samples), carrot (five samples), cucumber (six samples), garlic (22 samples), onions (eight samples) and pepper (one sample). Pythium intermedium and Pythium irregulare were detected in beet and cauliflower, respectively. Alternaria spp. were associated with leaf blight (leaf spot) of cucumber (four samples), garlic (17 samples) and tomato (1 sample). Phytoplasma was confirmed in twelve garlic and three onion samples. One zucchini sample had powdery mildew caused by Podosphaera xanthii. Seedling blight and root rot caused by Pythium sp. and Fusarium sp. were found in sunflower and tulip respectively. Alternaria caused leaf blight on thistle, leaf spot on five weed samples and stem blight on flax in one sample. Six samples of thistle showing rust symptoms were infected with Puccinia punctiformis.
