Abstract:Changes in adenosine triphosphate (ATP) and peroxynitrite (ONOO-) concentrations have been correlated in a number of diseases including ischemia-reperfusion injury and drug-induced liver injury. Herein, we report the development of a fluorescent probe ATP-LW, which enables the simultaneous detection of ONOO- and ATP. ONOO- selectively oxidizes the boronate pinacol ester of ATP-LW to afford the fluorescent 4-hydroxy-1,8-naphthalimide product NA-OH (λex = 450 nm, λem = 562 nm or λex = 488 nm, λem = 568 nm). In contrast, the binding of ATP to ATP-LW induces the spirolactam ring opening of rhodamine to afford a highly emissive product (λex = 520 nm, λem = 587 nm). Due to the differences in emission between the ONOO- and ATP products, ATP-LW allows ONOO- levels to be monitored in the green channel (λex = 488 nm, λem = 500-575 nm) and ATP concentrations in the red channel (λex = 514 nm, λem = 575-650 nm). The use of ATP-LW as a combined ONOO- and ATP probe was demonstrated using hepatocytes (HL-7702 cells) in cellular imaging experiments. Treatment of HL-7702 cells with oligomycin A (an inhibitor of ATP synthase) resulted in a reduction of signal intensity in the red channel and an increase in that of the green channel as expected for a reduction in ATP concentrations. Similar fluorescence changes were seen in the presence of SIN-1 (an exogenous ONOO- donor).

Abstract:In this work, taking full advantage of the intramolecular charge transfer (ICT) mechanism, a hydroxynaphthalimide-based ratiometric two-photon fluorescent probe RTP-PN was synthesized to detect ONOO-. Probe RTP-PN could accurately detect ONOO- in the range of 1.4 nM-1.4 mM with the detection limit of 1.4 nM by a ratiometric fluorescence spectroscopy method. Additionally, probe RTP-PN exhibited an ultrafast response for ONOO- than other various species including H2O2 and ClO-. Abstract:Abstract Background Ex vivo and in vivo detection and imaging of adenosine triphosphate (ATP) is critically important for the diagnosis and treatment of diseases, which still remains challenges up to present. Results We herein demonstrate that ATP could be fluorescently detected and imaged ex vivo and in vivo. In particular, we fabricate a kind of fluorescent ATP probes, which are made of titanium carbide (TC) nanosheets modified with the ROX-tagged ATP-aptamer (TC/Apt). In the constructed TC/Apt, TC shows superior quenching efficiency against ROX (e.g., ~ 97%). While in the presence of ATP, ROX-tagged aptamer is released from TC surface, leading to the recovery of fluorescence of ROX under the 545-nm excitation. Consequently, a wide dynamic range from 1 μM to 1.5 mM ATP and a high sensitivity with a limit of detection (LOD) down to 0.2 μM ATP can be readily achieved by the prepared TC/Apt. We further demonstrate that the as-prepared TC/Apt probe is feasible for accurate discrimination of ATP in different samples including living cells, body fluids (e.g., mouse serum, mouse urine and human serum) and mouse tumor models. Conclusions Fluorescence detection and imaging of ATP could be readily achieved in living cells, body fluids (e.g., urine and serum), as well as mouse tumor model through a new kind of fluorescent ATP nanoprobes, offering new powerful tools for the treatment of diseases related to abnormal fluctuation of ATP concentration.

Abstract:The fluorescent probe, GXY-ADP-2, with xanthene structure as the fluorescent core was designed and prepared for the selective detection of peroxynitrite (ONOO−). ONOO− can be produced endogenously and exogenously and is a strong oxidant with a short half-life. Oxidative modifications of biomolecules, that can be attributed to the formation of ONOO−, occur in the reactions of biomolecules with secondary ONOO−-derived radical oxidants. Therefore, it is very important to develop a specific fluorescent probe for detecting ONOO− to monitor oxidative stress state. The excitation wavelength and emission wavelength of the probe are 689 nm and 739 nm respectively. In the process of co-incubation with ONOO−, generate a new substance with two internal conjugated structures through a special reaction mechanism, one giving the fluorescence with the excitation wavelength of 347 nm and the emission wavelength of 484 nm with the detection limit of 0.12 μM, and the other that with the excitation wavelength of 433 nm and the emission wavelength of 583 nm with the detection limit of 0.077 μM. The linear dynamic range of the probe is 0–5 μM. Its response is not affected by the other reactive oxygen species, thus can sensitively detect ONOO−. In bioimaging experiments with HepG2 cells, the green and blue cell fluorescence signals (583 nm and 433 nm, respectively) were increased, while the red one (739 nm) was significantly reduced, under lipopolysaccharide (LPS) induced oxidative stress, proving that the probe could sensitively detect ONOO− in living cells. This work provides a new tool for the dynamic changes of ONOO− and oxidative stress processes in biological systems.

Abstract:Adenosine triphosphate (ATP), commonly produced in mitochondria, is required by almost all the living organisms; thus fluorescent probes for monitoring mitochondrial ATP levels fluctuation are essential and highly desired. Herein, we report a multisite-binding switchable fluorescent probe, ATP-Red 1, which selectively and rapidly responds to intracellular concentrations of ATP. Live-cell imaging indicated that ATP-Red 1 mainly localized to mitochondria with good biocompatibility and membrane penetration. In particular, with the help of ATP-Red 1, we successfully observed not only the decreased mitochondrial ATP levels in the presence of KCN and starvation state, but also the increased mitochondrial ATP levels in the early stage of cell apoptosis. These results indicate that ATP-Red 1 is a useful tool for investigating ATP-relevant biological processes.

Abstract:Exploring the fluctuation of reactive oxygen species (ROS) and adenosine triphosphate (ATP) in living systems is significant for the early diagnosis of many diseases. However, the simultaneous detection of ROS and ATP is still in challenge. Herein, a supramolecular fluorescent probe (G3⊂CP5) has been designed for two-channel simultaneous detection of H2O2 and ATP. G3⊂CP5 is consisting of the Eu(III)-complex and water-soluble pillar[5]arene (CP5), achieving the supramolecular assembly via synergistic coordinated, host-guest and amphiphilic interactions. The phenylboric acid in ligand can interact with H2O2 and CP5 has host-guest recognition toward ATP. As consequence, the phenylboric acid groups and CP5 are the responsive moieties for H2O2 and ATP, respectively. The optical experiments demonstrated G3⊂CP5 exhibited low limits of detection and could respond to H2O2 and ATP in two channels to reduce mutual interference. Significantly, the probe shows good mitochondrial targeting. The fluorescent sensing of H2O2 and ATP in both Hela cells and RAW264.7 macrophages is also confirmed. As consequence, by taking advantage of the superior luminescence of rare-earth complexes and the precise recognition of pillar[n]arenes, this work prepared a fluorescent probe for simultaneous detection of H2O2 and ATP, providing the opportunities for further accurate biosensing.

Abstract:The simultaneous or continuous detection of multiple targets in complex samples is for various applications, notably in medical diagnostics and environmental monitoring. A novel dual-channel colorimetric and fluorescent method has been developed for the continuous detection of adenosine triphosphate (ATP) and copper ions based on PCN-224@Fe2O3, which exhibits both fluorescent and enhanced peroxidase-like (POD-like) properties. The synthesis of PCN-224@Fe2O3 involved the in-situ growth of ferric oxide on Zr-based metal-organic frameworks (PCN-224). Through catalyzing the colorless compound 3,3 ',5,5 '-tetramethylbenzidine (TMB) to produce a blue product (oxTMB) in the presence of H2O2, PCN-224@Fe2O3 allows for the detection of ATP and copper ions. The unique mechanism involves ATP weakening the POD-like activity of PCN-224@Fe2O3 by forming Zr-O-P bonds with Zr in PCN-224, which also leads to fluorescence recovery. Copper ions, through chelating with ATP, counteract the effect of ATP on PCN-224@Fe2O3, resulting in increased POD-like activity and the amount of oxTMB alongside the quenching of the fluorescence of the system. This facilitates colorimetric "on-off-on" and fluorescence "off-on-off" switch sensing for ATP and Cu2+. The application of PCN-224@Fe2O3 in the detection of ATP and Cu2+ in human serum yielded satisfactory results. This innovative approach not only provides a means to regulate the mimic activity of oxides but also presents a convenient, sensitive, and accurate detection method for ATP and Cu2+, significantly broadening the potential application of MOFs and nanozymes in bioanalysis.

Abstract:As energy currency of life, adenosine triphosphate (ATP) is highly dynamic in living organisms. It is crucial yet challenging to probe intracellular or in-vivo ATP precisely without destruction of the living biological systems. Herein, we have designed novel ATP-responsive ratiometric nanoprobes by employing upconversion nanoparticles (UCNPs) with cypate as antennas. Nuclear magnetic resonance spectroscopy was used to analyze the interfacial interactions among lanthanide nanocrystal, cypate and adenosine phosphates, disclosing the origin of detection specificity. ATP induced interruption of energy transfer pathway in cypate-sensitized UCNPs was further investigated by femtosecond transient absorption and time-resolved photoluminescence spectroscopy. The nanoprobes with dual near-infrared (NIR)-excited ratiometric UCL were successfully applied for quantification of ATP level in living cells under vegetatively growing, starving, and glycolysis-inhibiting states, respectively. Furthermore, the increase of available energy by feeding for zebrafish larvae was confirmed through monitoring the in-vivo ATP variation with the nanoprobes. Such nanoprobes showed great potential in ATP-associated biological research and clinical applications. These findings may gain deep insights into the surface science and photophysics in dye-lanthanide nanocomposites, and open new avenues for the design and bioapplications of lanthanide nanoprobes.
Novel perylene probe-encapsulated metal-organic framework nanocomposites for ratiometric fluorescence detection of ATP

Abstract:Adenosine triphosphate (ATP) plays an important role in various biological processes and the ATP level is closely associated with many diseases. Herein, a novel ratiometric fluorescence assay for ATP was developed based on the excimer-monomer transfer of a perylene probe. By encapsulating a perylene probe, N,N'-bis(6-caproic acid)-3,4:9,10-perylenediimide (PDI), into zeolitic imidazolate framework-8 (ZIF-8) nanocrystals, fluorescent nanocomposites (PDI@ZIF-8) with significant excimer emission of the perylene probe were prepared for the first time. The presence of ATP will trigger the decomposition of PDI@ZIF-8 due to much stronger coordination between ATP and Zn2+ than that of 2-methylimidazole and Zn2+. As a result, the encapsulated PDI probes were released, leading to significantly increased monomer emission accompanying the decrease in the excimer emission. The excimer-monomer transition signal was utilized for ratiometric ATP sensing and its potential application for detecting ATP in cell lysates was also successfully demonstrated.

Abstract:Two boronate fluorescent probes have been developed for the detection of peroxynitrite (TCFB1 and TCFB2). TCFB1 was shown to have a low sensitvity towards peroxynitrite and have a poor solubility in aqueous solution whereas TCFB2 demonstrated high sensitivity towards peroxynitrite and mitochondria localisation with the ability to detect exogenous and endogenous peroxynitrite in live cells (Hep-G2, RAW 264.7, HeLa and A459).

Abstract:Peroxynitrite (OONO(-)) is profoundly implicated in health and disease. The physiological and pathological outcome of OONO(-) is related to its local concentration, and hence, a reliable OONO(-) assay is highly desired. We have developed a FRET-based small-molecule fluorescent probe (PNCy3Cy5), harnessing the differential reactivity of Cy3 and Cy5 toward OONO(-) by fine-tuning. It exhibits high detection sensitivity and yields a ratiometric fluorescent signal. We have exemplified that it can be applied in semiquantitative determination of OONO(-) in living cells. Notably, it specifically localizes in mitochondria, where endogenous OONO(-) is predominantly generated. Thus, PNCy3Cy5 is a promising molecular tool for peroxynitrite biology.

Abstract:Based on the Förster resonance energy transfer (FRET) strategy, a reversible ratiometric fluorescent NIR‐II probe, consisting of xanthenoid dyes and a piperazine ring, is developed for in vivo detection of ATP. CX‐RATP is demonstrated to have the potential for real‐time monitoring of the fluctuations of ATP under changing physiological and pathological environments. It can also achieve in situ imaging of tumor lesions. ATP plays a prominent role in shaping the evolution of immune cell responses to injury, infection, and cancer. However, real‐time monitoring of ATP levels in vivo remains a challenge, due to the lack of reliable tools that work reversibly in deep tissues. Herein, based on the Förster resonance energy transfer (FRET) strategy, a reversible ratiometric second near‐infrared window (NIR‐II) molecular fluorescent probe (CX‐RATP) is designed and synthesized. CX‐RATP exhibits a significant fluorescence ratio change with the existence of 0–10 mM ATP in vitro, which is within the range of physiological and pathological ATP concentrations. Meanwhile, the recognition process can be reversed by apyrase. It can capture the increase and decrease in ATP levels in real time before and after the treatment of acute inflammation. Besides, CX‐RATP is also successfully applied to the hepatocellular carcinoma (HCC) model and achieves in situ imaging of tumor lesions. These results all provide solid proof that CX‐RATP is a powerful tool for in vivo real‐time ATP detection.

Abstract:An ESIPT-based 'AND' logic fluorescence probe (GSH-ABAH) was developed for the simultaneous detection of ONOO- and biological thiols. GSH-ABAH was shown to have good cell permeability and with the addition of just SIN-1 (ONOO- donor) or GSH, no fluorescence response was observed in live cells. Abstract:Peroxynitrite (ONOO-) is a highly reactive substance, and plays an essential part in maintaining cellular homeostasis. It is crucial to monitor the ONOO- level in cells in normal and abnormal states. We introduced a p-dimethylaminophenylether-based fluorescent probe PDPE-PN, which could be synthesized readily. The new probe had prominent sensitivity and specificity, and a fast response towards ONOO-. The spectral performance of probe PDPE-PN was outstanding and the limit of detection was 69 nM. Probe PDPE-PN with low toxicity was applied to detect endogenous/exogenous ONOO- in RAW 264.7 macrophages and zebrafish. Importantly, successful application of the new receptor opens up new ideas for the design of ONOO- probes.

Herein, we develop a near-infrared fluorescence (NIRF) and photoacoustic dual-modal molecular probe (CySO3CF3) composed of a water-soluble hemicyanine dye caged with a trifluoromethyl ketone moiety for in vivo imaging of ONOO-. The trifluoromethyl ketone moiety can undergo a series of ONOO--induced cascade oxidation-elimination reactions, leading to sensitive and specific fluorescence and photoacoustic turn-on responses toward ONOO-; whereas, a zwitterionic structure of the hemicyanine component ensures good water-solubility. Thus, CySO3CF3 not only specifically detects ONOO- in solution and cells with the limit of detection down to 53 nM but also allows for NIRF and photoacoustic dual-modal imaging of ONOO- in the tumors of living mice.
A Fast Detection of Peroxynitrite in Living Cells.

Abstract:Peroxynitrite (ONOO-) plays a crucial role in the regulation of diverse pathophysiological processes, and high level of ONOO- is profound association with numerous diseases. Herein, we developed an anthraquinone-based fluorescent probe L for ONOO- determination by a new recognition mechanism: amido oxidized nitroso-group by ONOO-. Probe L with amine-based recognition receptor is more selective to ONOO- than other reactive oxygen species, including H2O2 and ClO-. Furthermore, ONOO- could be rapidly detected by probe L with a Limit of Detection of 13 nM. More importantly, L could be used to monitor intracellular ONOO- in SMMC-7721 cells. (C) 2020 Elsevier B.V. Zeolitic imidazolate framework-8 encapsulating gold nanoclusters and carbon dots for ratiometric fluorescent detection of adenosine triphosphate and cellular imaging

Abstract:A novel nanoprobe was prepared by encapsulating carbon dots (CDs) and gold nanoclusters (AuNCs) into zeolitic imidazolate framework-8 (ZIF-8) for sensitive detecting adenosine triphosphate (ATP). Under excitation at 360 nm, the obtained CDs/ nanoprobe exhibits dual-emissions at 469 nm and 660 nm, respectively, corresponding to the fluorescence emission of CDs and the aggregation-induced emission enhancement (AIEE) of AuNCs. The framework of ZIF-8 in this probe can be degraded by ATP due to the coordination competition of ATP and 2-Methylimidazole towards zinc ion (Zn 2+ ), resulting in the release of CDs and AuNCs. The following dispersion of CDs would improve efficiencies of the fluorescence excitation and the consequent emission of CDs. On the contrary, the AIEE of AuNCs would be decreased spontaneously after the AuNCs originally restricted in ZIF-8 were allowed to escape. The intensity ratio of fluorescence at 469 nm to that at 660 nm ( I 469 / I 660 ) was conveniently employed as the response signal for representing the amount of ATP. This nanoprobe exhibits excellent sensitivity and selectivity toward ATP, with a limit of detection (LOD) of 0.061 μM. Besides, low cytotoxicity of this nanoprobe facilitates its application as a fluorescent indicator in fluorescence imaging of living cells. Encapsulating two types of fluorescent nanomaterials by a degradable ZIF-8 structure makes the ratiometric fluorescence response of the nanocomposite probe towards the target analyte that destroys the ZIF-8 structure possible, and simplifies the application of the probe.
