Abstract:Demodecidae are the most specialized parasitic mites of mammals; they typically inhabit the skin, but they have been found in other tissues and organs. They can cause demodecosis (a disease which is hazardous and difficult to cure) in humans, domestic animals and livestock. They are parasites with high host and topical specificity. They have been found for most orders of mammals, and they are common in the populations of numerous host species. Therefore, they not only constitute an important subject of veterinary and medical study, but also comprise an excellent model for faunistic and parasitological analyses concerning different aspects of functioning and evolution of the host–parasite relationship. The current level or knowledge of demodecid mites is irregular and fragmentary, and numerous questions require elaboration and ordering, from the taxonomic diversity to geographic distribution and relations with hosts. Such data may be of use i.a. for the development of more efficient and reliable diagnostic methods, as well as understanding the etiology and pathogenesis mechanisms of demodecosis, currently a contentious issue. The present paper lists all formally-described valid species of demodecid mites, together with other functioning specific names, verified and with comments on their status. This is significant for correct species identification and demodecosis diagnostics. The list has been drawn up on the basis of data acquired in the period 1842−2020. It contains 122 valid species of parasite, including their hosts and geographic distribution, data on parasitism, as well as only the second record of Demodex sciurinus in Eurasian red squirrel Sciurus vulgaris in over 100 years since its initial discovery.
Searching for common patterns in parasite ecology: species and host contributions to beta-diversity in helminths of South African ungulates and fleas of South American rodents

Abstract:We searched for common patterns in parasite ecology by investigating species and host contributions to the beta-diversity of infracommunities (=assemblages of parasites harboured by a host individual) in helminths of three species of South African ungulates and fleas of 11 species of South American rodents, assuming that a comparison of patterns in distinctly different parasites and hosts would allow us to judge the generality or, at least, commonness of these patterns. We used data on species' composition and numbers of parasites and asked whether (i) parasite species' attributes (life cycle, transmission mode, and host specificity in helminths; possession of sclerotized combs, microhabitat preference, and host specificity in fleas) or their population structure (mean abundance and/or prevalence) and (ii) host characteristics (sex and age) affect parasite and host species' contributions to parasite beta-diversity (SCBD and HCBD, respectively). We found that parasite species' morphological and ecological attributes were mostly not associated with their SCBD. In contrast, parasite SCBD, in both ungulates and rodents, significantly increased with either parasite mean abundance or prevalence or both. The effect of host characteristics on HCBD was detected in a few hosts only. In general, parasite infracommunities' beta-diversity appeared to be driven by variation in parasite species rather than the uniqueness of the assemblages harboured by individual hosts. We conclude that some ecological patterns (such as the relationships between SCBD and parasite abundance/prevalence) appear to be common and do not differ between different host-parasite associations in different geographic regions, whereas other patterns (the relationships between SCBD and parasite species' attributes) are contingent and depend on parasite and host identities.

Abstract:Mastomys natalensis and M. coucha are commensal rodent species endemic to Africa. A recent taxonomic revision within Mastomys leaves the parasite–host list of M. natalensis questionable and that of M. coucha incomplete. The current study aimed to develop a better understanding of the ectoparasite diversity associated with the 2 distinct but closely related rodent species and to explore the influence of host and habitat type on ectoparasite infestations. Between 2014 and 2020, 590 rodents were trapped in 3 habitat types (village, agriculture and natural) across a wildlife-human/domestic animal interface. In total 48 epifaunistic species (45 ectoparasitic and 3 predatory) represented by 29 genera from 4 taxonomic groups (fleas, lice, mites and ticks) were recorded. Only 50% of the epifauna were shared between the 2 rodent species, with mites the most speciose taxon in both host species. The abundance of epifaunistic individuals, and also those of mites and fleas, were significantly higher on male M. natalensis, while ticks were significantly higher on reproductively active M. natalensis. For both rodent species, infestations by most epifaunistic taxa (on M. natalensis) and some taxa (on M. coucha) were significantly lower in the village as opposed to the less disturbed agricultural and natural habitat types. The study highlights the importance of host life history, even in closely related rodent species, in shaping parasite profiles and a loss of parasite diversity in more extreme anthropogenic habitats.

Abstract:The Amblyomma marmoreum complex includes afrotropical species, such as Amblyomma sparsum , a three-host tick that parasitizes reptiles, birds, and mammals, and is a recognized vector of Ehrlichia ruminantium . However, the lack of morphological, genetic and ecological data on A. sparsum has caused considerable confusion in its identification. In this study, we used microscopy and metagenomic approaches to analyze A. sparsum ticks collected from a puff adder snake ( Bitis arietans ) in southwest Senegal (an endemic rickettsioses area) in order to supplement previous morphological descriptions, provide novel genomic data for the A. marmoreum complex, and describe the genome of a novel spotted fever group Rickettsia strain. Based on stereoscope and scanning electron microscopy (SEM) morphological evaluations, we provide high-quality images and new insights about punctation and enameling in the adult male of A. sparsum to facilitate identification for future studies. The metagenomic approach allowed us assembly the complete mitochondrial genome of A. sparsum , as well as the nearly entire chromosome and complete plasmid sequences of a novel Rickettsia africae strain. Phylogenomic analyses demonstrated a close relationship between A. sparsum and Amblyomma nuttalli for the first time and confirmed the position of A. sparsum within the A. marmoreum complex Our results provide new insights into the systematics of A. sparsum and A. marmoreum complex, as well as the genetic diversity of R. africae in the Afrotropical region. Future studies should consider the possibility that A. sparsum may be a vector for R. africae .

Comparative Prevalence and Intensity of Endoparasites in a Dynamic Boreal Ungulate Community

Abstract:Surveillance of endoparasites at the host community level is rarely reported for ungulates. Yet, changes in the composition and abundance of species in ungulate assemblages, coupled with environmental and climate change, bring into focus the need for baseline data on endoparasite occurrence in host species at the community level. We investigated the prevalence and intensity of eggs of endoparasites in feces of a dynamic boreal ungulate community in Yukon, Canada, that included reintroduced bison (Bison bison), as well as introduced elk (Cervus canadensis), naturally colonizing mule deer (Odocoileus hemionus), and resident populations of caribou (Rangifer tarandus), moose (Alces americanus), and thinhorn sheep (Ovis dalli). We also examined the change in endoparasite prevalence and intensity in bison fecal samples collected eight years apart. The prevalence of eggs detected in feces differed across species for most endoparasite groups. We also provide new records of several endoparasites in novel hosts or new geographic records. We detected a substantially greater prevalence and intensity of trichostrongyle-type eggs in bison feces between samples collected eight years apart. Our data emphasize the need for targeted pathogen surveillance programs to monitor the movement of various ungulate and associated endoparasites. This is particularly pertinent since our data potentially supports evidence for the continued northward expansion and host switching of protostrongylid species, which may have health implications for animals at a new interface.

Sympatry of Amblyomma eburneum and Amblyomma variegatum on African buffaloes and prevalence of pathogens in ticks

Abstract:The Amblyomma genus is represented on the African continent by 24 species, out of which 17 are known to occur in different ecological niches of southern Africa. Amblyomma, known for their aggressive hunting behaviour and aptitude as pathogen vectors, are of main concern to travellers, mainly in rural and conservation areas of Africa. In this study, we highlight the overlapping distribution of Amblyomma eburneum and Amblyomma variegatum found on African buffaloes (Syncerus caffer) at Coutada 11, Central Mozambique. In total, 1,039 Amblyomma ticks were collected and morphologically identified using taxonomic keys, and genomic DNA was extracted. They were subjected to reverse line blotting for pathogen identification followed by molecular analysis (COI sequencing) of both tick species. Pathogens such as Ehrlichia ruminantium, Anaplasma centrale, Theileria sp., Babesia sp. and Rickettsia africae were detected, of which R. africae is zoonotic. Ehrlichia ruminantium, R. africae, Theileria mutans and Theileria velifera are well-established pathogens transmitted by Amblyomma ticks; however, Anaplasma spp. and Babesia spp. are not, suggesting residual parasite DNA in the bloodmeal. Little is mentioned in the literature about A. eburneum, including its role as a vector and reservoir for pathogens. In Mozambique A. eburneum is currently restricted to wildlife but the spread of the tick may be observed given the climate change that is occurring. The infection rates for the pathogens in both Amblyomma tick species were lower than expected, but this may be due to the low host density in the forest niche and the innate immunity of these hosts. With the propensity of ticks of the Amblyomma genus to form parapatric distributions, the mechanisms that allows for the overlapping distribution of these two Amblyomma species while maintaining tick species identity is of great interest.

Abstract:During the present study, species boundaries in the velvet worm Peripatopsis birgeri species complex in KwaZulu-Natal, South Africa, were examined with the use of DNA sequence data, gross morphology, scanning electron microscopy (SEM) and ecological niche modelling. Mitochondrial CO1 and nuclear 18S rRNA sequence data were used to generate a phylogeny using both a Bayesian inference and maximum likelihood approaches. Both the CO1 sequence data and the combined DNA sequence topology ( CO1 + 18S rRNA) revealed the presence of two geographically distinct clades within the P. birgeri species complex. Results from three species delimitation methods (GMYC, PTP and ASAP), were incongruent in determining the number of species within the species complex and generally overestimated the number of operational taxonomic units. The latter result can be attributed to the marked genetic structure within the species complex, a result corroborated by the CO1 haplotype network and statistically significant F ST values. Divergence time estimation suggests that the two clades diverged during the Plio/Pleistocene. Niche modelling revealed that the two clades were differentially impacted by bioclimatic variables and occupied different habitats. Gross morphological characteristics, such as leg pair numbers and colour revealed no fixed differences between the two clades, however, a creamy white headband/collar occurred exclusively in specimens of clade 1 ( P. birgeri ). In contrast, SEM revealed fixed dorsal and ventral scale rank differences between the two clades. The results corroborate the presence of a novel species within the P. birgeri s.s. species complex. Peripatopsis birgeri s.s. is revised and restricted to the southern Drakensberg and a new species, P. polychroma sp. nov. Nematodes and the effect of seasonality in grassland habitats of South Africa

Abstract:Nematodes in South Africa have mainly been studied for their diversity and agricultural importance. However, the ecological status of nematodes and the effect of seasonal variation in local grasslands remain unknown. For this reason, a nematode study was conducted in the Telperion Nature Reserve and represented the first ecological study in a natural grassland area in South Africa. In total, 104 soil samples were collected during four consecutive seasons from 2015 until 2016 in three habitats, viz. (i) open grassland, (ii) shrubland with rocky outcrops, and (iii) riparian zone. From these the nematode community structure and soil ecosystem status were studied. In total, 93 genera from 50 families were recorded with herbivores and bacterivores being the most abundant trophic groups in all three habitats. Linear mixed models revealed that season had an overwhelmingly dominant impact on the condition, food web status, and functioning of the soil ecosystems with pairwise comparisons indicating that significantly higher values were recorded during winter. Interestingly, this seasonal shift can largely be attributed to fluctuations in the populations of only a few nematode groups (namely Aporcelaimellus, Dorylaimidae, Iotonchus, and Mononchus) with high colonizer-persister values. Although the reason for the higher abundance of specific nematode groups recorded during the winter is not explicitly clear, it is possibly linked to reduced competition from other soil fauna. This study clearly shows that further investigations are required to better understand the dynamics of grassland ecosystems.
Identification of ticks and tick-borne pathogens of wildlife necropsy cases submitted to the SANBI National Zoological Gardens, South Africa

Abstract:Ticks are arachnid blood-feeding parasites, which infest livestock, wildlife, and humans, transmitting medically and veterinary significant pathogens. Their biodiversity and distribution in wild animals remains complex. This study analysed archived tick samples (n = 48) from the South African Biodiversity Institute (SANBI) Wildlife Biobank utilizing morphology and genetic analyses of the 16S rRNA and COI (DNA barcoding) mitochondrial genes to identify ticks collected among 13 vertebratesavian, reptilian, and mammalian host species. The specimens came from nine localities including nature reserves and captive facilities (zoological garden) in South Africa, Namibia, and Botswana. These ticks were also assessed for associated pathogens with the reverse line blot (RLB) hybridization assay. Seven tick genera, Amblyomma, Hyalomma, Haemaphysalis, Ixodes, Rhipicephalus, Rhipicentor, and Otobius were identified, with Amblyomma being the most prevalent (22.9 %) in our sample set. Obtained sequences were 95-100 % similar to published records of tick species collected from wild and domestic animals, as well as those collected from vegetation, from different southern African areas. However, tick specimens (n = 3) identified morphologically as Hyalomma truncatum, Rhipicephalus e. evertsi, and R. simus, were, on a molecularly level, more closely related to their sister taxa (H. glabrum, R. e. mimeticus, and R. gertrudae, respectively) suggesting a need for taxonomic verification. With the RLB hybridization assay, six samples reacted with the Ehrlichia/Anaplasma genus-specific probe, while two reacted with the Theileria/Babesia genus-specific probe. Sequencing of the RLB amplicons targeting the 18S rRNA gene (n = 2) indicated 100 % similarity to Hepatozoon fitzsimonsi, while one was closely related to He. ingwe with 99.39 % similarity. The results show that wildlife harbour different tick species, and pathogen detection identified novel genotypes, indicating wildlife as potential pathogens reservoirs. This study enhances our understanding of tick biodiversity, distribution and highlights wildlife's role in harbouring diverse tick species and novel pathogens.
Dynamics of Reduviidae Species, Predators of Podagrica decolorata, in West of Côte d'Ivoire

Abstract:Background and Objective: Okra is an important vegetable crop in Côte d'Ivoire. In the okra crop, there are species of Reduviidae that are predators of insect pests. These Reduviidae occupy a very important place in biological control because of their biocontrol potential. The objective of the study was to investigate the dynamics of the Reduviidae species, predators of Podagrica decolorata. Materials and Methods: This study was conducted during the dry and rainy seasons of 2019 in the city of man located in the west of Côte d'Ivoire. Colour traps and mowing nets were used to capture the different species of Reduviidae to determine the influence of abiotic factors and the populations of the species Podagrica decolorata on the dynamics of Reduviidae species. Results: A study on the frequency of occurrence revealed two constant species which are Rhynocoris rapax and Rhynocoris albopilosus with frequencies of occurrence of 63.46 and 75%, respectively. In dry and rainy seasons, Rhynocoris albopilosus was higher numbers at each phenological stage. The study revealed the influence of abiotic factors (temperature, rainfall and humidity) on the population of Reduviidae species. The study also showed highly significant positive correlations between the population of Reduviidae and that of the pest, P. decolorata in the dry season (r = +0.76 and p<0.001) and in the rainy season (r = +0.47 and p<0.001). Conclusion: The study revealed a total of six Reduviidae species whose numbers varied during the two seasons as potential predators of Podagrica decolorata.

Abstract:Research on amphibian parasites has been on the increase because of the noticeable decline of amphibians. Studies on these parasites have been limited to their prevalence and infection patterns, and there is still a dearth of knowledge on the phenotypic variations. To characterise and investigate the genetic variability and relatedness of amphibian nematode parasites, two nematode parasites–suspected Cosmocerca sp. and suspected Rhabdias sp.–of the toad Schlerophrys regularis were investigated. One hundred and sixty-eight S. regularis were collected from three study areas: Ikorodu, Ojota and Badagry. Intestines were dissected to obtain the enteric parasites, and lungs were washed in saline solution. Parasites were labelled according to the location of collection (Badagry 5A-5I, Ikorodu 5J–5O, Ojota 5P and 5S) and were then preserved for further identification. Eighteen parasites were identified morphologically using standard keys and by amplification of the 18S small subunit (SSU) ribosomal DNA sequence. Phylogenetic analysis was done using the Neighbour-Joining method. From the sequence homology and phylogenetic analysis, the suspected Cosmocerca sp. shared the closest genetic relatedness with Aplectana chamaeleonis isolate AC3 with 100% similarities while the suspected Rhabdias sp. shared the closest genetic relatedness with Rhabditoides regina isolate DF5012 with 100% similarities. Based on location, four Aplectana chamaeleonis (5E, 5F, 5G, 5H) and five Rhabditoides regina (5A, 5B, 5C, 5D, 5I) were from Badagry, four Aplectana chamaeleonis (5J, 5M, 5N, 5O) and two Rhabditoides regina (5K, 5L) from Ikorodu, one Aplectana chamaeleonis (5S) and two Rhabditoides regina (5P, 5Q) from Ojota. We report that Rhabditoides regina , a parasite known to be associated with insects, was recovered in the lungs of toads. Further investigations to resolve the complexities of superfamily Cosmocercoidea are recommended. Relatedness was not influenced by collection location as parasites were not unique to a specific environment.

Abstract:Nematodes constitute the most significant, most numerous, and diversified set of multicellular organisms on the earth. They live in various environments and exhibit a wide range of behavioural patterns. In the soil food web, they can be found at various trophic levels as herbivores (plant parasitic nematodes), bacterivores, fungivores, omnivores, and predators. As there were fewer studies on nematode ecology in the Udupi region, the present study aims to study the community structure and functional diversity of soil nematodes. Soil samples were collected following opportunistic random sampling employing a soil auger and were stored at 40C until transported to the laboratory. Nematodes were isolated from soil, killed, fixed, dehydrated, and displayed on a glass slide after isolation. The standard keys were used to identify the individual to genera level. 62 genera of soil nematodes belonging to 26 families and 7 orders were identified. Predator were the most prevalent communities. Various statistical indices for assessing nematode population ecology and nematodes specific indices were also calculated and it indicated a significant abundance of large plant parasitic nematodes. The region exhibits low levels of labile organic carbon and nutrient enrichment (Enrichment Index (EI):14.06 to 21.22). Despite this, the soil food web in the region is well-structured, indicated by Structure Index (SI) (85.51 to 89.74). Prevalence of fungal decomposition dominance and the soil appears to be minimally disturbed, as indicated by high channel index values and low Basal Index (BI) values, respectively

Abstract:A study of gastrointestinal nematodes in roe deer was carried out in the regional hunting reserves of Riaño and Mampodre, Province of León, Spain, to provide information on their prevalence and intensity of infection in relation to the sampling areas, age of the animals, and body weight. Through a regulated necropsy of the animals, all of them harbored gastrointestinal nematodes in their digestive tract, with a mean intensity of parasitism of 638 ± 646.1 nematodes/infected animal. Eleven genera were found and 18 species of gastrointestinal nematodes were identified, three of them polymorphic: Trichostrongylus axei, Trichostrongylus vitrinus, Trichostrongylus capricola, Trichostrongylus colubriformis, Haemonchus contortus, Spiculopteragia spiculoptera/Spiculopteragia mathevossiani, Ostertagia leptospicularis/Ostertagia kolchida, Ostertagia (Grosspiculopteragia) occidentalis, Teladorsagia circumcincta/Teladorsagia trifurcate, Marshallagia marshalli, Nematodirus europaeus, Cooperia oncophora, Capillaria bovis, Oesophagostomum venulosum, and Trichuris ovis. All of them have already been cited in roe deer in Europe, but Marshallagia marshalli, Capillaria bovis, and Ostertagia (Grosspiculopteragia) occidentalis are reported for the first time in Spain in this host. The abomasum was the intestinal section, where the prevalence (98.9%) and mean intensity (x¯ = 370.7 ± 374.4 worms/roe deer; range 3-1762) were significantly higher, but no statistically significant differences were found when comparing the sampling areas and age of animals. The animals with lower body weight had a higher parasite load than those in better physical condition, finding, in this case, statistically significant differences (p = 0.0020). Seven genera and 14 species were identified. In the small intestine, 88% of the animals examined presented gastrointestinal nematodes, with an average intensity of x¯ = 131.7 ± 225.6 parasites/infected animal, ranging between 4-1254 worms. No statistically significant differences were found when the three parameters studied were compared. Four genera and seven species were identified. In the large intestine/cecum, 78.3% of the examined roe deer presented adult worms, with an average intensity of 6.3 ± 5.5 worms/infected animal; range 1-26 worms. Only statistically significant differences were observed when considering the mean intensity of parasitism and the sampling area (p = 0.0093). Two genera and two species were identified. Several of the species found in the study were studied molecularly, and with the sequences obtained compared with those deposited in GenBank, phylogenetic trees were prepared to determine their taxonomic status. Using coprological techniques, the existing correlation in the shedding of gastrointestinal nematode eggs in roe deer was investigated with that of semi-extensive sheep farms in the same study area to verify the existence of cross-transmission of these parasites between wild and domestic animals. The high values found in the studied parameters show that northern Spain is an area of high-intensity infection for roe deer.
Temporal and spatial dynamics of gastrointestinal parasite infection in Père David’s deer

Abstract:Background The Père David’s deer ( Elaphurus davidianus ) population was established from only a small number of individuals. Their genetic diversity is therefore relatively low and transmissible (parasitic) diseases affecting them merit further attention. Parasitic infections can affect the health, survival, and population development of the host. However, few reports have been published on the gastrointestinal parasites of Père David’s deer. The aims of this study were: (1) to identify the intestinal parasites groups in Père David’s deer; (2) to determine their prevalence and burden and clarify the effects of different seasons and regions on various indicators of Père David’s deer intestinal parasites; (3) to evaluate the effects of the Père David’s deer reproductive period on these parasites; (4) to reveal the regularity of the parasites in space and time. Methods In total, 1,345 Père David’s deer faecal samples from four regions during four seasons were tested using the flotation (saturated sodium nitrate solution) to identify parasites of different genus or group, and the McMaster technique to count the number of eggs or oocysts. Results Four groups of gastrointestinal parasites were found, of which strongyles were dominant; their prevalence and burden were significantly higher than other groups. Significant temporal and spatial effects on gastrointestinal parasitic infection were found. Parasite diversity, prevalence, parasite burden, and aggregation were the highest in summer. Among the four regions, parasite diversity, prevalence, and burden were the highest in the Dongting Lake area. In addition, parasite diversity and burden during the reproductive period of Père David’s deer was significantly higher than during the post-reproductive period. Conclusions The summer season and the reproductive period of Père David’s deer had great potential for parasite transmission, and there is a high risk of parasite outbreaks in the Dongting Lake area.

Natural infection of white grubs (Coleoptera: Scarabaeidae) with entomopathogenic nematodes in the KwaZulu-Natal province of South Africa

Abstract:White grubs are root feeding larvae of beetles (Coleoptera: Scarabaeidae) that are sporadic pests in agriculture and can lead to economic damage. The grubs feed on the roots of plants, while the adult beetle can bore into underground stems, as well as cause defoliation of plants. Sporadic incidence of larvae with symptoms of nematode infections were detected in wattle and sugarcane plantations in the KwaZulu-Natal province of South Africa. The larvae with infection symptoms were isolated, washed, and put on water traps to collect infective juveniles of possible nematode infections. Three species of entomopathogenic nematodes (EPNs) were isolated from the white grub larvae. These included Steinernema bertusi isolated from Maladera sp. 4., Oscheius myriophila from Maladera sp. 4 and Schizonchya affinis, and Steinernema fabii isolated from Maladera sp. 4., Pegylis sommeri, and S. affinis. Of these S. fabii was the most common species in the sample (87%). This is the first report of such a high diversity of locally occurring EPNs found naturally associated with white grub species in this region of South Africa.

Abstract:The preservation of the world's biodiversity for future generations has been a global objective for many years, with the establishment of the International Union for Conservation of Nature (IUCN) Red List of Threatened Species in 1964. However, the conservation of parasites is a more recent development and, due to the difficulty in obtaining data and studying some of the parasitic species, comes its own series of challenges. Using parasites of cyprinid hosts (one critically endangered, one endangered and three near threatened) collected from South Africa's Cape Fold freshwater ecoregion (CF) as a case study, this paper discusses the challenges and possible solutions for implementing a fish parasite conservation project. Novel data on the fish parasites (1819 metazoan parasite individuals, representing the Acanthocephala, Cestoda, Copepoda, Digenea, Monogenea and Nematoda) of the CF are provided from the five endemic hosts, Cheilobarbus serra (Peters, 1864), Labeobarbus seeberi (Gilchrist et Thompson, 1913), Pseudobarbus phlegethon (Barnard, 1938), Sedercypris calidus (Barnard, 1938), and Sedercypris erubescens (Skelton, 1974). Conservation statuses for selected parasite taxa are also proposed based on the conservation statuses of the fish hosts, according to the Conservation Assessment Methodology for Animal Parasites (CAMAP).

Abstract:Most data on species associations and vector potential of mosquitoes in relation to arboviral infections in South Africa date back from the 1940s to late 1990s. Contextual information crucial for disease risk management and control, such as the sampling effort, diversity, abundance, and distribution of mosquitoes in large parts of South Africa still remains limited. Adult mosquitoes were collected routinely from two horse farms in Gauteng Province; two wildlife reserves in Limpopo Province, at Orpen Gate in Kruger National Park (KNP) and Mnisi Area in Mpumalanga Province between 2014-2017, using carbon dioxide-baited light and tent traps. Mosquito diversity and richness are greater in untransformed natural and mixed rural settings. In untransformed wilderness areas, the most dominant species were Culex poicilipes, Anopheles coustani, and Aedes mcintoshi, while in mixed rural settings such as the Mnisi area, the two most abundant species were Cx. poicilipes and Mansonia uniformis. However, in peri-urban areas, Cx. theileri, Cx. univittatus, and Cx. pipiens sensu lato were the most dominant. Aedes aegypti, Ae. mcintoshi, Ae. metallicus, Ae. vittatus, Cx. pipiens s.l., Cx. theileri, and Cx. univittatus had the widest geographical distribution in northern South Africa. Also collected were Anopheles arabiensis and An. vaneedeni, both known malaria vectors in South Africa. Arbovirus surveillance and vector control programs should be augmented in mixed rural and peri-urban areas where the risk for mosquito-borne disease transmission to humans and domestic stock is greater.

Abstract:Captive reindeer in German zoos and wildlife parks live outside their natural geographic range and are exposed to a variety of viral, bacterial and protozoan pathogens, some host-specific and some which they are not exposed to in their native habitat. Reindeer blood samples and ticks collected in 2013 from 123 reindeer at 16 different zoological facilities were available from a previous study. The aims of this study were to assess the serological status of these animals with regards to various microorganisms as well as to test ticks (Ixodes ricinus) and blood samples for the presence of Anaplasma spp. DNA in order to evaluate the exposure of captive reindeer in Germany to a variety of pathogens. A total of 119 or 118 serum samples were screened (ELISA) and antibodies were detected (seropositive/tested, prevalence, confidence interval) against alphaherpesvirus (24/119, 20.3%, CI: 13.9–28.3), bluetongue virus (BTV; 4/119, 3.4%, CI: 1.0–8.7), malignant catarrhal fever related gammaherpesvirus (MCFV-related gammaherpesvirus; 7/119, 5.9%, CI: 2.7–11.9), pestivirus (5/118, 4.2%, CI: 1.6–9.8), Schmallenberg virus (SBV; 70/118, 59.3%, CI: 50.3–67.8), smooth Brucella spp. (1/118; 0.9%, CI: 0–5.1), Neospora caninum (5/118, 4.2%, CI: 1.6–9.8), and Toxoplasma gondii (62/119, 52.1%, CI: 43.2–60.9). These results suggested the exposure of reindeer to all tested pathogens. Moreover, real-time PCR for Anaplasma phagocytophilum targeting the partial msp2 gene was performed on DNA extracted from whole blood samples from reindeer (n = 123) and from ticks (n = 49) collected from 22 reindeer in seven different facilities. In addition to the real-time PCR, a semi-nested PCR for the partial groEL gene, and a nested PCR targeting the partial 16S rRNA gene were performed. DNA of A. phagocytophilum was detected in 17 reindeer (13.8%) and 15 ticks (30.6%). Three of the five reindeer with ticks having A. phagocytophilum DNA also had such DNA in blood. These results indicate that captive reindeer can be exposed to several ruminant pathogens that they hitherto had no known exposure to through their natural geographical distribution and habitats as shown for Culicoides-borne BTV and SBV. Further, captive reindeer may serve as reservoir hosts for pathogens circulating in local domestic, captive, and wild ruminant species and populations and arthropod vectors.
