Molecular imaging techniques visualise neurotransmitter systems and cellular processes, shedding light on disease mechanisms. AI algorithms analyse vast datasets, simulate neural networks, and even decode neural signals for brain-machine interfaces. This has the potential for personalised medicine and ground-breaking treatments. The future holds immense promise. Abstract:Neurodegenerative disorders represent a significant and growing global health challenge, necessitating continuous advancements in diagnostic tools for accurate and early detection. This work explores the recent progress in Magnetic Resonance Imaging (MRI) techniques and their application in the realm of neurodegenerative disorders. The introductory section provides a comprehensive overview of the study's background, significance, and objectives. Recognizing the current challenges associated with conventional MRI, the manuscript delves into advanced imaging techniques such as high-resolution structural imaging (HR-MRI), functional MRI (fMRI), diffusion tensor imaging (DTI), and positron emission tomography-MRI (PET-MRI) fusion. Each technique is critically examined regarding its potential to address theranostic limitations and contribute to a more nuanced understanding of the underlying pathology. Abstract:Alzheimer's disease (AD) is an irreversible neurodegenerative disorder that destroys cognitive functions. Recently, a number of high-profile clinical trials based on the amyloid cascade hypothesis have encountered disappointing results. The failure of these trials indicates the necessity for novel therapeutic strategies and disease models. In this review, we will describe how recent advances in stem cell technology have shed light on a novel treatment strategy and revolutionized the mechanistic investigation of AD pathogenesis. Current advances in promoting endogenous neurogenesis and transplanting exogenous stem cells from both bench research and clinical translation perspectives will be thoroughly summarized. In addition, reprogramming technology-based disease modeling, which has shown improved efficacy in recapitulating pathological features in human patients, will be discussed.

Abstract:Language, as an information medium created by advanced organisms, has always been a concern of neuroscience regarding how it is represented in the brain. Decoding linguistic representations in the evoked brain has shown groundbreaking achievements, thanks to the rapid improvement of neuroimaging, medical technology, life sciences and artificial intelligence. In this work, we present a taxonomy of brain-to-language decoding of both textual and speech formats. This work integrates two types of research: neuroscience focusing on language understanding and deep learning-based brain decoding. Generating discernible language information from brain activity could not only help those with limited articulation, especially amyotrophic lateral sclerosis (ALS) patients but also open up a new way for the next generation's brain-computer interface (BCI). This article will help brain scientists and deep-learning researchers to gain a bird's eye view of fine-grained language perception, and thus facilitate their further investigation and research of neural process and language decoding.

Abstract:Neuropsychiatric disorders affect hundreds of millions of patients and families worldwide. To decode the molecular framework of these diseases, many studies use human postmortem brain samples. These studies reveal brain-specific genetic and epigenetic patterns via high-throughput sequencing technologies. Identifying best practices for the collection of postmortem brain samples, analyzing such large amounts of sequencing data, and interpreting these results are critical to advance neuropsychiatry. We provide an overview of human brain banks worldwide, including progress in China, highlighting some well-known projects using human postmortem brain samples to understand molecular regulation in both normal brains and those with neuropsychiatric disorders. Finally, we discuss future research strategies, as well as state-of-the-art statistical and experimental methods that are drawn upon brain bank resources to improve our understanding of the agents of neuropsychiatric disorders.

Abstract:Considerable information about mental states can be decoded from noninvasive measures of human brain activity. Analyses of brain activity patterns can reveal what a person is seeing, perceiving, attending to, or remembering. Moreover, multidimensional models can be used to investigate how the brain encodes complex visual scenes or abstract semantic information. Such feats of “brain reading” or “mind reading,” though impressive, raise important conceptual, methodological, and ethical issues. What does successful decoding reveal about the cognitive functions performed by a brain region? How should brain signals be spatially selected and mathematically combined to ensure that decoding reflects inherent computations of the brain rather than those performed by the decoder? We highlight recent advances and describe how multivoxel pattern analysis can provide a window into mind-brain relationships with unprecedented specificity, when carefully applied. However, as brain-reading technology advances, issues of neuroethics and mental privacy will be important to consider.

Abstract:Most leading research in basic and clinical neuroscience has been carried out by functional magnetic resonance imaging (fMRI), which detects the blood oxygenation level dependent signals associated with neural activities. Among new fMRI applications, brain decoding is an emerging research area, which infers mental states from fMRI signals. Brain decoding using fMRI includes classification, identification, and reconstruction of brain states. It is generally conducted using multi-voxel pattern analysis based on neuroscientific evidence that brain functions are mediated by distributed activation patterns. Brain decoding techniques have been successful in diverse applications such as the brain computer interface, patient monitoring, and neurofeedback. These techniques have expanded our understanding of how the brain encodes distinct information. In the current paper, we reviewed recent fMRI-based brain decoding techniques and applications. We also discussed the potential implications of brain decoding in neuroscience.

Abstract:In 2022 Alzheimers Disease Report, World Health Organization (WHO) states that Alzheimers disease and dementia have become the seventh most common causes of death in the U.S. However, there is currently no cure for Alzheimers disease and dementia, therefore research into effective treatments for Alzheimers disease is imminent. This paper introduces the incidence, pathogenesis, pathological mechanisms, and treatment of dementia and Alzheimers disease, and describes the latest research direction for Alzheimers disease - deep brain electrical signal stimulation. Although deep brain electrical signal stimulation is not commonly used to improve and treat cognitive function in Alzheimers disease at this stage of the disease, it is a very promising research direction for repairing or re-stimulating brain function. This paper suggests that it may be possible to use the emerging technology of brain-computer interfaces (BCI), equipped with deep brain electrical stimulation systems, to synchronize the input and output of electrical signals to achieve the purpose of precisely determining the electrical stimulation points. Brain-computer interface technology can also be linked to artificial intelligence technology to build personalized brain models, completing the generalization of therapeutic technology.

Abstract:Abstract In recent years, brain research has indisputably entered a new epoch, driven by substantial methodological advances and digitally enabled data integration and modelling at multiple scales— from molecules to the whole brain. Major advances are emerging at the intersection of neuroscience with technology and computing. This new science of the brain combines high-quality research, data integration across multiple scales, a new culture of multidisciplinary large-scale collaboration and translation into applications. As pioneered in Europe’s Human Brain Project (HBP), a systematic approach will be essential for meeting the coming decade’s pressing medical and technological challenges. The aims of this paper are to: develop a concept for the coming decade of digital brain research, discuss this new concept with the research community at large, to identify points of convergence, and derive therefrom scientific common goals; provide a scientific framework for the current and future development of EBRAINS, a research infrastructure resulting from the HBP’s work; inform and engage stakeholders, funding organisations and research institutions regarding future digital brain research; identify and address the transformational potential of comprehensive brain models for artificial intelligence, including machine learning and deep learning; outline a collaborative approach that integrates reflection, dialogues and societal engagement on ethical and societal opportunities and challenges as part of future neuroscience research.

Abstract:The complexity of the brain has attracted scientists from all over the world.Much effort has been paid to explore the mecha-nisms from genetics to molecules,from cells to circuits,and from Caenorhabditis elegans to humans.Brain research contributes to the development of new technologies,especially artificial intelli-gence(AI).According to the International Data Corporation,the global revenue of the AI market is expected to exceed $500 billion by 2023 and $900 billion by 2026,highlighting the great impor-tance of brain research for social progress.Treatments for brain disorders also get optimized,reducing disease burden and eco-nomic loss.Take dementia,for example:according to data from the World Health Organization,in 2019 the global cost for treat-ment in this field was estimated at $1.3 trillion,and this amount is expected to rise to $1.7 trillion by 2030.Despite its importance for health,economy,and society,a systematic understanding of the brain is still lacking,possibly due to the fragmentation of research strategies.Therefore,many countries have carried out brain projects,following a unified research strategy supported by national governments.In contrast to studies that are conducted by individual researchers and groups,brain projects are character-ized by systematic research directions,large funding,and across-the-aboard participation.The expectation is that brain projects will achieve major breakthroughs in a short period of time.

Abstract:The pandemic is transforming neurology. Long COVID will linger, neurologic diseases will increase, and technology, artificial intelligence, and new virtual worlds will usher a new age of the brain and new roles for neurologists. The pandemic has compelled international collaboration, greatly increased communications, and accelerated drug and vaccines approvals. It also dramatized the close interconnection of cognitive, mental, and social health and their relevance to building back better health, education, work, and leisure. Brain health is the key to health, productivity, and well-being. Neurologists are best placed to lead brain knowledge integration and application through the unifying theme of brain health by becoming advocates, healers, and guardians of the brain.

Abstract:Brain health is essential for physical and mental health, social well-being, productivity, and creativity. Current neurological research focuses mainly on treating a diseased brain and preventing further deterioration rather than on developing and maintaining brain health. The pandemic has forced a shift toward virtual working environments that accelerated opportunities for transdisciplinary collaboration for fostering brain health among neurologists, psychiatrists, psychologists, neuro and socio-behavioral scientists, scholars in arts and humanities, policymakers, and citizens. This could shed light on the interconnectedness of physical, mental, environmental, and socioeconomic determinants of brain disease and health. We advocate making brain health the top priority worldwide, developing common measures and definitions to enhance research and policy, and finding the cause of the decline of incidence of stroke and dementia in some countries and then applying comprehensive customized cost-effective prevention solutions in actionable implementation units. Life cycle brain health offers the best single individual, communal, and global investment.

Brain-Computer Interface (BCI) technology, which facilitates direct communication between the brain and external devices, emerges as an innovative key to unlocking neurological conditions, demonstrating significant promise in this context. This comprehensive review uniquely synthesizes the latest advancements in BCI research across multiple neurological disorders, offering an interdisciplinary perspective on both clinical applications and emerging technologies. We explore the progress in BCI research and its applications in addressing various neurological conditions, with a particular focus on recent clinical studies and prospective developments. Initially, the review provides an up-to-date overview of BCI technology, encompassing its classification, operational principles, and prevalent paradigms. It then critically examines specific BCI applications in movement disorders, disorders of consciousness, cognitive and mental disorders, as well as sensory disorders, highlighting novel approaches and their potential impact on patient care. This review reveals emerging trends in BCI applications, such as the integration of artificial intelligence and the development of closed-loop systems, which represent significant advancements over previous technologies. The review concludes by discussing the prospects and directions of BCI technology, underscoring the need for interdisciplinary collaboration and ethical considerations. It emphasizes the importance of prioritizing bidirectional and high-performance BCIs, areas that have been underexplored in previous reviews. Abstract:Brain science accelerates the study of intelligence and behavior, contributes fundamental insights into human cognition, and offers prospective treatments for brain disease. Faced with the challenges posed by imaging technologies and deep learning computational models, big data and high-performance computing (HPC) play essential roles in studying brain function, brain diseases, and large-scale brain models or connectomes. We review the driving forces behind big data and HPC methods applied to brain science, including deep learning, powerful data analysis capabilities, and computational performance solutions, each of which can be used to improve diagnostic accuracy and research output. This work reinforces predictions that big data and HPC will continue to improve brain science by making ultrahigh-performance analysis possible, by improving data standardization and sharing, and by providing new neuromorphic insights.

More recently, the availability of large-scale imaging and multi-omics datasets from the human brain has afforded the opportunity to the discovering of common genetic variants contributing to the structural and functional IDPs of the human brain. By integrative analyses with functional multi-omics data from the human brain, a set of critical genes, functional genomic regions and neuronal cell types have been identified as significantly associated with brain IDPs. Here, we review the recent advances in the methods and applications of multi-omics integration in brain imaging analysis. We highlight the importance of functional genomic datasets in understanding the biological functions of the identified genes and cell types that are associated with brain IDPs. Moreover, we summarize well-known neuroimaging genetics datasets and discuss challenges and future directions in this field.
