Abstract:Two new guaianolides, namely, 3β,8α-dihydroxy-13-methoxyl-4(14),10(15)-dien-(1αH,5αH,6β H,11βH)-12,6-olide (1) and 3β,8α-dihydroxy-13-methoxyl-4(14),10(15)-dien-(1αH,5αH,6β H,11αH)-12,6-olide (2), together with six known guaianolides, 8-hydroxyzaluzanin C, austricin, chlorojanerin, cynaropicrin, chlorohyssopifolin E and chlorohyssopifolin A were isolated from the aerial parts of Saussurea alata. The structures were established mainly based on spectral analysis, especially 2D NMR techniques.

Abstract:A new steroidal glycoside (1), (25R)-14α, 17α-hydroxyspirost-5-en-3β-yl 3-O-α-L-rhamnpyranosyl-(1 → 2)-β-D-glucopyranosyl-(1 → 3)-β-D-glucopyranoside, together with three known steroidal glycosides, (25R)-3β-hydroxyspirost-5-en-1β-yl-3-O-α-L-rhamnopyranosyl-(1 → 2)-O-β-D-xylopyranosyl-(1 → 3)-α-L-arabinopyranoside (2), Cixi-ophiopogon B (3) and Cixi-ophiopogon A (4), were obtained from the tuberous roots of Ophiopogon japonicus (Liliaceae). Compound 2 was isolated from the Ophiopogon genus for the first time. Their structures were identified on the basis of extensive mass and nuclear magnetic resonance spectroscopic analysis.

Abstract:A new guaiane-type sesquiterpene glycoside, torilolone 8-O-beta-D-glucopyranoside (1), together with a known analogue compound, torilolone 11-O-beta-D-glucopyranoside (2), was isolated from the fruits of Daucus carota L. Their chemical structures were elucidated on the basis of MS, NMR spectroscopic analyses coupled with chemical degradation. The cytotoxic activities of two isolated compounds were evaluated against two human gastric cancer cell lines BGC-823 and AGS using MTT assay.

Abstract:Two new guaiane-type sesquiterpene glycosides, atractylodes guaianoside I ( 1 ) and atractylodes guaianoside II ( 2 ), along with eleven known sesquiterpenes ( 3 – 13 ), were isolated from Atractylodes Japonica Koidz. ex Kitam. The structures of these two new compounds were elucidated by extensive spectroscopic analyses and mass spectrometric techniques. And their absolute configurations were confirmed by electronic circular dichroism (ECD) calculations. In addition, the isolated compounds were evaluated for their cytotoxic activities against three cancer cell lines HT-29, A-549, and MCF-7. Results showed that new compound 2 had inhibitory effects on three types of cancer cells, while new compound 1 only had inhibitory effects on HT-29 and MCF-7.

Abstract:Three new compounds, (5 beta,9 beta)-guaia-6,10(14)-dien-9-ol (=rel-(1R,3aS,5R,8aR)-1,2,3,3a,4,5,6,8a-octahydro-1-methyl-4-methylene-7-(1-methylethyl)azulen-5-ol; 1), 6-acetyl-8-methoxy-2,3-dimethylchromen-4-one (=6-acetyl-8-methoxy-2,3-dimethyl-4H-1-benzopyran-4-one; 2), and (2S)-3'-hydroxy5',7-dimethoxyflavanone (=(2S)-2,3-dihvdro-2-(3-hydroxy-5-methoxyphenyl)-7-methoxy-4H-1-benzopyran-4-one; 3) were isolated from the roots and rhizomes of Ligularia macrophylla, together with seven known compounds. Their structures and configurations were elucidated by spectroscopic methods, including 2D-NMR techniques.

Abstract:Two new guaianolides (1 and 2) were isolated from the aerial parts of Ajania fruticulosa along with a triterpene (alpha-amyrin), two plant sterols (beta-sitosterol and daucosterol), four flavonols [santin (3), axillarin (4), centaureidin, and 5,7,4'-trihydroxy-3, 3'-dimethoxyflavone], and five sesquiterpenes [ketoplenolide B, 9beta-hydroxyeudesma-4,11(13)-dien-12-oic acid, 9beta-acetoxyeudesma-4,11(13)-dien-12-oic acid, 1alpha,4alpha, 10alpha-trihydroxy-9alpha-angeloyloxyguaia-2,11(13)-dien-12, 6alpha-olide, and 3beta,4alpha-dihydroxyguaia-11(13),10(14)-dien-12, 6alpha-olide]. The structures of the new guaianolides were established as 1alpha-hydroperoxy-4beta,8alpha,10alpha, 13-tetrahydroxyguaia-2-en-12,6alpha-olide (1) and 1alpha-hydroperoxy-4alpha, 10alpha-dihydroxy-9alpha-angeloyloxyguaia-2,11(13)-dien-12, 6alpha-olide (2), respectively. Xanthine oxidase assays of all isolates revealed that santin (3) and axillarin (4) inhibited the enzyme with IC(50) values of 36.5 and 36.0 &mgr;M (that of allopurinol used as a positive control in the study was 24.2 &mgr;M), respectively.

Abstract:Using a medium pressure liquid chromatographic system, a new triterpene lactone named glyuranolide [3 beta, 22 alpha-dihydroxy-11-oxo-delta 12-oleanene-27 alpha-methoxy carbonyl-29-oic acid (29, 22 alpha-) lactone.] was isolated from the crude sapogenins of Glycyrrhiza uralensis Fisch. Its structure was elucidated by IR, UV, FAB-MS, and various NMR spectra (including NOE, BBD, INEPT, SR, COSY, NOESY etc.).

Abstract:A new benzoxazolinone glucoside, 4-O-beta-D-glucopyranosyl-benzoxazolin-2(3H)-one, was isolated from Acanthus ilicifolius, a medicinal plant from south China. The complete (1)H and (13)C NMR assignments of the compound were carried out by using DEPT, HMBC, HMQC, NOESY and (1)H--(1)H COSY spectra.

Abstract:Glaucatotones A - I, nine new guaiane-type sesquiterpenoids, along with two reported compounds, namely (1β,5β)-1-hydroxyguaia-4(15),11(13)-dieno-12,5-lactone (10) and pseudoguaianelactone C (11), were isolated from the roots of Lindera glauca. The structures and absolute configurations of these compounds were elucidated by extensive spectroscopic analyses, single-crystal X-ray diffraction, and comparison of experimental and calculated electronic circular dichroism (ECD) data. Structurally, glaucatotone A (1) is characterized as a dihomosesquiterpenoid with an unprecedented 5/5/7/6 ring system. A pair of enantiomers, (±)-glaucatotone B (2a/2b), represent the first rearranged norsesquiterpenoid with a (cyclopentylmethyl)cyclohexane skeleton. 3 is defined as a dinorsesquiterpenoid possessing a 5/7/5 ring system. 4-6 are three guaiane-type norsesquiterpenoids. In vitro bioactivity, 2a selectively inhibited Bcap-37 with IC<sub>50</sub> value of 5.60 μM, and 9 selectively inhibited Du-145 with IC<sub>50</sub> value of 5.52 μM. The anti-inflammatory activity of 1-9 were tested, and of these compounds, 1, 2a, 2b and 7 exhibited potent inhibitory effects.

Abstract:A new lignan glycoside, (7S,8R)-guaiacylglycerol-ferulic acid ether-7-O-β-D-glucopyranoside (1), along with five known phenylpropanoids (2-6) and seven phenylpropanoid glycosides (7-13), were isolated from the stems of Zanthoxylum armatum DC. Their structures were elucidated by spectroscopic analysis. Compounds 2-8 and 11-13 were first isolated from Rutaceae and the others were isolated for the first time from Z. armatum.

Abstract:AIM: To study chemical constituents of the aerial parts of Artemisia anomala (Asteraceae). METHODS: The constituents were isolated with silica gel, ODS column chromatography and semi-preparative HPLC, and their structures were elucidated on the basis of physical characteristics and spectral data. RESULTS: Eight compounds were obtained, and their structures were identified as 3 beta-ethoxytanapartholide (1), (4S*, 5S*)-dihydro-5-[( 1R*, 2S*)-2-hydroxy-2-methyl-5-oxo-3-cyclopenten-1-yl]-3-methylene-4-(3-oxobutyl)-2(3H)-furanone (2), ligucyperonol (3), cyperusol C (4), santamarin (5), 1 alpha, 2 alpha, 3 alpha, 4 alpha, 10 alpha -pentahydroxyguaia-11(13)-ene-12, 6 alpha-olide (6), balanophonin (7), methyl 3-(2'-hydroxy-4'-methoxyphenyl) propanoate (8). CONCLUSION: Compound 1 was a new artifact, 4 and 8 were isolated from the genus Artemisia for the first time, and compounds 2-3, 5-7 were isolated from this plant for the first time.

Abstract:Secoatractylohexone A (1), an unprecedented secoguaiane lactone glycoside featuring 6/7 cores and dihydroxy-9-guaine-3-one 11-O-β-d-glucopyranoside (2), a 9,10-unsaturated guaiene-type glycoside possessing an uncommon scaffold, were isolated from the water-soluble portion of the ethanolic extract of Atractylodes lancea rhizomes together with five known compounds (3–7). The structures of 1 and 2 were elucidated on the basis of extensive spectroscopic data and application of the CD technique. The potential biological activities of secoatractylohexone A were predicted by network pharmacology in silico, the result of which indicated that secoatractylohexone A may be used to treat type II diabetes.

Abstract:Eight undescribed guaianolides (macrocephalolides A-H) and two known analogues (parishin C, artabsinolide E) were isolated from the whole plant of Artemisia macrocephala growing in Xinjiang, China. Their structures were determined on the basis of extensive spectroscopic analysis, with absolute configurations established by comparison of experimental and calculated ECD data, as well as confirmation of single-crystal X-ray diffraction crystallography. Macrocephalolides A-B featured an unusual type of 3-oxa-guaianolide with a cyclopentenone moiety. Macrocephalolides C-E possessed a dihydro-2H-pyran acetal segment, representing an unprecedented 2, 3-secoguaianolide skeleton with 6/7/5 tricyclic ring system in natural sesquiterpenes. The X-ray crystal structures of parishin C and artabsinolide E were reported for the first time.

Abstract:Three new lignan glycosides (1-3) were isolated from the stems of Akebia trifoliata. Their structures were elucidated as (7R,8R,7'R,8'R)3,3',5,5'tetramethoxy-4,4'dihydroxy-7,9':7',9-diepoxylignan-4-O-beta-D-glucopyranoside(1), (7S,8S,8'R)-4,4',9-trihydroxy-3,3',5,5'-tetramethoxy-7,9'-epoxylignan-7'-one 9-O-beta-D-glucopyranoside (2),(7R,8R,8'S)-4,4',9-trihydroxy3,3',5,5'-tetramethoxy-7,9'-epoxylignan-7'-one 9-O-beta-D-glucopyranoside (3) by spectral analyses, primarily NMR, MS and CD. Chemical Structure and Nmr of A New Saponin from Dipsacus Japonicus (Dipsacaceae)

Abstract:A new triterpenoid saponin containing five sugars was isolated from the ethanolic root extract of Dipsacus japonicus Miq., and its structure was established as 3-O-[beta-D-glucopyranosyl (1-->4)] [alpha-L-rhamnopyransoyl (1 --> 3)]-beta-D-glucopyranosyl (1-->3)-alpha-L-rhamnopyranosyl (1-->2)-alpha-L-arabinopyranosyl-oleanoic acid. The sites of glycosylation and the sequence of sugars in the glycoside can be determined unambiguously and total assignment of severely overlapping proton resonance of sugar residues were achieved by a combined use of the 1D-SEMDY and NOE difference spectroscopy in rotating frame techniques, without having recourse to chemical degration or modification. The results showed that these new NMR techniques are very effective and convenient for the structure determination and spectral assignment of this class of compounds.

Abstract:The structures of two flavanonol glycosides isolated from Smilax glabra, named smitilbin and neosmitilbin, have been revised to isoastilbin and neoastilbin, respectively. The revised structures were determined based on intensive studies of chemical interconversion, NMR spectroscopy, and X-ray crystallographic analysis. The latest NMR data were also summarized.
