CHANGES OF CHLOROPHYLL AND ANTHOCYANIN CONTENT AND RELATED ENZYME ACTIVITIES OF FLOWER STALK IN CHINESE KALE UNDER DIFFERENT LIGHT INTENSITIES

Abstract:The contents of chlorophyll and anthocyanin and activities of Chlorophyllase (Chlase), Phenylalanine ammonialyase (PAL), Chalcone isomerase (CHI) and UDP glucose-flavonoid3-o-glucosyl transferase (UFGT) in flower stalk epidermis of Chinese kale (Brassica alboglabra Bailley cv. Jianyexia & Zhonghua) were affected by different light intensities (control, 40% shade, and 60% shade). Under the shade, the chlorophyll contents in flower stalk epidermis of increased by 22.8% and 20.2% under 40% and 60% shade in 'Jianyexia' and by 15.2% and 8.9% in 'Zhonghua'. The activities of Chlase in flower stalk epidermis of Chinese kale increased under shade in 2 cultivars possibly due to the Chlase degrading activity of chlorophyll. Similarly, the increase of the chlorophyll content in flower stalk under shade may be due to increased synthesis. The content of anthocyanin decreased under shade by 59.8% and 80.2% under 40% and 60% shade in 'Jianyexia' and by 21.0% and 51.6% in 'Zhonghua'. The activities of PAL increased under shade in 2 cultivars (but decreased slightly in 'Jianyexia' under 40% shade). The activities of CM decreased under shade, but those in 'Zhoughua' only decreased under 40% shade. Under shade the activities of UFGT in 'Jianyexia' decreased whereas those in 'Zhonghua' increased. Because the changes in activities of PAL, CHI and UFGT were not consistent with the change in anthocyanin contents in flower stalk epidermis, PAL, CHI and UFGT might were not regarded as the limiting factors regulating anthocyanin biosynthesis in flower stalk epidermis in Chinese kale.
