TARGETED CYBRIDIZATION IN CITRUS AND TRANSCRIPT PROFILING OF A MALE STERILE CYBRID PUMMELO CONTAINING CYTOPLASM FROM SATSUMA MANDARIN

Abstract:Seedlessness is an elite fruit quality trait. In our program, by symmetric fusion of embryogenic callus protoplasts of Satsuma mandarin and mesophyll protoplasts of elite seedy cultivars, diploid cybrid plants containing sterile cytoplasm from Satsuma were regenerated and characterized from several fusions. The leaf parents included Hirado Buntan and Shatian pummelos, and Bingtang, Early gold, Taoye and Hongjiang sweet oranges. Encouragingly, the cybrid pummelo between Satsuma Guoqing No. 1 (G1) and Hirado Buntan pummelo (HBP) showed male sterility. Chromosome karyotype analysis was conducted to further confirm results of our previous molecular maker analysis, which indicated that the cybrid (G1+HBP) contained the nuclear genome of HBP. RNA-seq was performed to assess gene expression profiles of G1+HBP and HBP; a large number genes were differentially expressed between the two types, such as nuclear genes involved in nucleic acid binding and response to hormone synthesis, and genes required for floral organ identification. In addition, agreeable with the flower morphology of the cybrid, expression levels of PISTILLATA (PI) and APETALA3 (AP3) were reduced in stamen-like structures, though they were restricted to correct floral whorls. These finding indicated that citrus class-B MADS-box genes likely constituted 'perfect targets' for CMS retrograde signaling. Correlation analysis of transcriptome and proteome data showed that posttranscriptional regulation played an important role in floral bud development and might contribute to male sterility of G1+HBP.
