Effects of Substituents X and Y on the NMR Chemical Shifts of 2-(4-X Phenyl)-5-y Pyrimidines

Abstract:Pyrimidine derivatives are an important class of heterocyclic compounds with various biological activities and excellent photoelectric properties, which have attracted considerable attention in pharmaceutical, agricultural and material fields. In this work, a set of substituted 2-(4-X phenyl)-5-Y pyrimidines with various substituents (X = NO2, CF3, F, Cl, H, CH3, OH, OCH3, NH2; Y= NO2, F, Cl, Br, H, CH3, OCH3) were synthesized, and based on which the substituent effects in the long conjugated system were investigated. Nuclear magnetic resonance (NMR) chemical shift is directly related to the electron density around the nucleus, which is influenced by the substituent directly or indirectly connected to the atom. Hammett parameters (sigma, sigma(F), sigma(R)), electronegativity (chi), the excited-state substituent effect parameter (sigma(ex)(CC)), the cross interaction of substituents (Delta sigma(2)) and the heavy atom effect indicator (D) were employed as the molecular descriptors to characterize the substituent effects. Good quantitative structure-property relationship (QSPR) models against NMR chemical shifts have been built for delta(H-4,H-6), delta(C-2), delta(C-4,C-6), delta(C-5), delta(C-1') and delta(C-4') with three to five molecular descriptors. The physical meaning of each model was illustrated in detail. Leave-one-out cross validation confirms good prediction performance of each model. This study provides a deep insight into the substituent effects on the electron distribution in the polar conjugated system.
