Abstract:Memory research remains focused on just a few brain structures—in particular, the hippocampal formation (the hippocampus and entorhinal cortex). Three key discoveries promote this continued focus: the striking demonstrations of enduring anterograde amnesia after bilateral hippocampal damage; the realization that synapses in the hippocampal formation are plastic e.g., when responding to short bursts of patterned stimulation ("long-term potentiation" or LTP); and the discovery of a panoply of spatially-tuned cells, principally surveyed in the hippocampal formation (place cells coding for position; head-direction cells, providing compass-like information; and grid cells, providing a metric for 3D space). Recent anatomical, behavioral, and electrophysiological work extends this picture to a growing network of subcortical brain structures, including the anterior thalamic nuclei, rostral midline thalamic nuclei, and the claustrum. There are, for example, spatially-tuned cells in all of these regions, including cells with properties similar to place cells of the hippocampus proper. These findings add new perspectives to what had been originally been proposed—but often overlooked—half a century ago: that damage to an extended network of structures connected to the hippocampal formation results in diencephalic amnesia. We suggest these new findings extend spatial signaling in the brain far beyond the hippocampal formation, with profound implications for theories of the neural bases of spatial and mnemonic functions.

Abstract:Key Points Retrieving a memory seems to involve a set of distinct processes, with different neural substrates, that are coordinated to orchestrate the act of remembering. Recent advances in cognitive science, including brain-imaging techniques, have provided evidence for the nature of these processes. Frontal cortical areas are central to the retrieval attempt. Anterior frontal-polar cortex is activated specifically during retrieval attempts for both verbal and non-verbal material, whereas more posterior frontal cortex is activated during both retrieval and processing of verbal material. Posterior frontal activation becomes stronger as remembering becomes more difficult. Overall, frontal cortex seems to be important for strategic aspects of retrieval. A network of left-sided cortical areas, including left parietal cortex, shows activity that correlates with retrieval success. This may act as a signal to inform a person that information is from the past rather than the present. The content of a memory is probably encoded as a 'reactivation' of those areas of cortex that are activated by sensory perception in the same modality, although the systems used for perception and retrieval do not completely overlap. It is unclear to what extent early sensory cortices are activated by retrieval. Retrieval may be initiated when top–down modulation from frontal cortex interacts with sensory representation of cues to trigger reactivation of the cortical networks that represent a memory, with a network including left parietal cortex possibly signalling that the representation is old. A sense of familiarity may occur when frontal and parietal regions are activated without detailed reactivation of sensory areas, whereas detailed recollection may rely on more extensive activity to represent the content of the memory.

Abstract:Episodic memories comprise diverse attributes of experience distributed across neocortical areas. The hippocampus is integral to rapidly binding these diffuse representations, as they occur, to be later reinstated. However, the nature of the information exchanged during this hippocampal-cortical dialogue remains poorly understood. A recent study has shown that the secondary motor cortex carries two types of representations: place cell-like activity, which were impaired by hippocampal lesions, and responses tied to visuo-tactile cues, which became more pronounced following hippocampal lesions. Using two-photon Ca2+ imaging to record neuronal activities in the secondary motor cortex of male Thy1-GCaMP6s mice, we assessed the cortical retrieval of spatial and non-spatial attributes from previous explorations in a virtual environment. We show that, following navigation, spontaneous resting state reactivations convey varying degrees of spatial (trajectory sequences) and non-spatial (visuo-tactile attributes) information, while reactivations of non-spatial attributes tend to precede reactivations of spatial representations surrounding hippocampal sharp-wave ripples.
Unsilenced inhibitory cortical ensemble gates remote memory retrieval

Abstract:Acquired information can be consolidated to remote memory for storage but persists in a dormant state until its retrieval. However, it remains unknown how dormant memory is reactivated. Using a combination of simultaneous two-photon calcium imaging and holographic optogenetics in the anterior cingulate cortex (ACC) in vivo, we discover a subset of GABAergic neurons that are specifically associated with dormant memory retrieval. These interneurons display persistent activity and inter-neuronal synchronization at the remote memory stage. In the absence of natural contextual cues, directly activating these interneurons reliably recalls cortical ensembles relevant to remote memory retrieval with context specificity. Conversely, targeted volumetric inactivation of these neurons suppresses context-induced memory retrieval. Our results reveal an unexpected role of unsilenced inhibitory cortical ensembles in causally gating the retrievability of dormant remote memory.

Dentate gyrus is needed for memory retrieval

Abstract:The hippocampus is crucial for acquiring and retrieving episodic and contextual memories. In previous studies, the inactivation of dentate gyrus (DG) neurons by chemogenetic- and optogenetic-mediated hyperpolarization led to opposing conclusions about DG's role in memory retrieval. One study used Designer Receptors Exclusively Activated by Designer Drugs (DREADD)-mediated clozapine N-oxide (CNO)-induced hyperpolarization and reported that the previously formed memory was erased, thus concluding that denate gyrus is needed for memory maintenance. The other study used optogenetic with halorhodopsin induced hyperpolarization and reported and dentate gyrus is needed for memory retrieval. We hypothesized that this apparent discrepancy could be due to the length of hyperpolarization in previous studies; minutes by optogenetics and several hours by DREADD/CNO. Since hyperpolarization interferes with anterograde and retrograde neuronal signaling, it is possible that the memory engram in the dentate gyrus and the entorhinal to hippocampus trisynaptic circuit was erased by long-term, but not with short-term hyperpolarization. We developed and applied an advanced chemogenetic technology to selectively silence synaptic output by blocking neurotransmitter release without hyperpolarizing DG neurons to explore this apparent discrepancy. We performed in vivo electrophysiology during trace eyeblink in a rabbit model of associative learning. Our work shows that the DG output is required for memory retrieval. Based on previous and recent findings, we propose that the actively functional anterograde and retrograde neuronal signaling is necessary to preserve synaptic memory engrams along the entorhinal cortex to the hippocampal trisynaptic circuit.

Abstract:We investigated memory encoding and retrieval during a quasinaturalistic spatial-episodic memory task in which subjects delivered items to landmarks in a desktop virtual environment and later recalled the delivered items. Transition probabilities and latencies revealed the spatial and temporal organization of memory. As subjects gained experience with the town, their improved spatial knowledge led to more efficient navigation and increased spatial organization during recall. Subjects who exhibited stronger spatial organization exhibited weaker temporal organization. Scalp-recorded electroencephalographic signals revealed spectral correlates of successful encoding and retrieval. Increased theta power (T+) and decreased alpha/beta power (A−) accompanied successful encoding, with the addition of increased gamma (G+) accompanying successful retrieval. Logistic regression classifiers trained on spectral features reliably predicted mnemonic success in held-out sessions. Univariate and multivariate electroencephalographic analyses revealed a similar spectral T+A−G+ of successful memory. These findings extend behavioral and neural signatures of successful encoding and retrieval to a naturalistic task in which learning occurs within a spatiotemporal context. Abstract:Recent results suggest that social memory requires the dorsal hippocampal CA2 region as well as a subset of ventral CA1 neurons. However, it is unclear whether dorsal CA2 and ventral CA1 represent parallel or sequential circuits. Moreover, because evidence implicating CA2 in social memory comes largely from long-term inactivation experiments, the dynamic role of CA2 in social memory remains unclear. Here, we use pharmacogenetics and optogenetics in mice to acutely and reversibly silence dorsal CA2 and its projections to ventral hippocampus. We show that dorsal CA2 activity is critical for encoding, consolidation, and recall phases of social memory. Moreover, dorsal CA2 contributes to social memory by providing strong excitatory input to the same subregion of ventral CA1 that contains the subset of neurons implicated in social memory. Thus, our studies provide new insights into a dorsal CA2 to ventral CA1 circuit whose dynamic activity is necessary for social memory.

Abstract:Memory formation is typically divided into phases associated with encoding, storage, consolidation, and retrieval. The neural determinants of these phases are thought to differ. This study first investigated the impact of the experience of novelty in rats incurred at a different time, before or after, the precise moment of memory encoding. Memory retention was enhanced. Optogenetic activation of the locus coeruleus mimicked this enhancement induced by novelty, both when given before and after the moment of encoding. Optogenetic activation of the locus coeruleus also induced a slow-onset potentiation of field potentials in area CA1 of the hippocampus evoked by CA3 stimulation. Despite the locus coeruleus being considered a primarily noradrenergic area, both effects of such stimulation were blocked by the dopamine D1/D5 receptor antagonist SCH 23390. These findings substantiate and enrich the evidence implicating the locus coeruleus in cellular aspects of memory consolidation in hippocampus.

Abstract:We develop of a line of work initiated by Curto and Itskov towards understanding the amount of information contained in the spike trains of hippocampal place cells via topology considerations. Previously, it was established that simply knowing which groups of place cells fire together in an animal's hippocampus is sufficient to extract the global topology of the animal's physical environment. We model a system where collections of place cells group and ungroup according to short-term plasticity rules. In particular, we obtain the surprising result that in experiments with spurious firing, the accuracy of the extracted topological information decreases with the persistence (beyond a certain regime) of the cell groups. This suggests that synaptic transience, or forgetting, is a mechanism by which the brain counteracts the effects of spurious place cell activity.
Distinct Contribution of Granular and Agranular Subdivisions of the Retrosplenial Cortex to Remote Contextual Fear Memory Retrieval

Abstract:The retrieval of recent and remote memories are thought to rely on distinct brain circuits and mechanisms. The retrosplenial cortex (RSC) is robustly activated during the retrieval of remotely acquired contextual fear memories (CFMs), but the contribution of particular subdivisions [granular (RSG) vs agranular retrosplenial area (RSA)] and the circuit mechanisms through which they interact to retrieve remote memories remain unexplored. In this study, using both anterograde and retrograde viral tracing approaches, we identified excitatory projections from layer 5 pyramidal neurons of the RSG to the CA1 stratum radiatum/lacunosum-moleculare of the dorsal hippocampus and the superficial layers of the RSA in male mice. We found that chemogenetic or optogenetic inhibition of the RSG-to-CA1, but not the RSG-to-RSA, pathway selectively impairs the retrieval of remote CFMs. Collectively, our results uncover a specific role for the RSG in remote CFM recall and provide circuit evidence that RSG-mediated remote CFM retrieval relies on direct RSG-to-CA1 connectivity. The present study provides a better understanding of brain circuit mechanisms underlying the retrieval of remote CFMs and may help guide the development of therapeutic strategies to attenuate remote traumatic memories that lead to mental health issues such as post-traumatic stress disorder.SIGNIFICANCE STATEMENT The RSC is implicated in contextual information processing and remote recall. However, how different subdivisions of the RSC and circuit mechanisms through which they interact to underlie remote memory recall remain unexplored. This study shows that granular subdivision of the RSC and its input to hippocampal area CA1 contributes to the retrieval of remote contextual fear memories. Our results support the hypothesis that the RSC and hippocampus require each other to preserve fear memories and may provide a novel therapeutic avenue to attenuate remote traumatic memories in patients with post-traumatic stress disorder.
Retrieval of contextual memory can be predicted by CA3 remapping and is differentially influenced by NMDAR activity in rat hippocampus subregions

Abstract:Episodic memory is essential to navigate in a changing environment by recalling past events, creating new memories, and updating stored information from experience. Although the mechanisms for acquisition and consolidation have been profoundly studied, much less is known about memory retrieval. Hippocampal spatial representations are key for retrieval of contextually guided episodic memories. Indeed, hippocampal place cells exhibit stable location-specific activity which is thought to support contextual memory, but can also undergo remapping in response to environmental changes. It is unclear if remapping is directly related to the expression of different episodic memories. Here, using an incidental memory recognition task in rats, we showed that retrieval of a contextually guided memory is reflected by the levels of CA3 remapping, demonstrating a clear link between external cues, hippocampal remapping, and episodic memory retrieval that guides behavior. Furthermore, we describe NMDARs as key players in regulating the balance between retrieval and memory differentiation processes by controlling the reactivation of specific memory traces. While an increase in CA3 NMDAR activity boosts memory retrieval, dentate gyrus NMDAR activity enhances memory differentiation. Our results contribute to understanding how the hippocampal circuit sustains a flexible balance between memory formation and retrieval depending on the environmental cues and the internal representations of the individual. They also provide new insights into the molecular mechanisms underlying the contributions of hippocampal subregions to generate this balance.

Abstract:Episodic memory involves learning and recalling associations between items and their spatiotemporal context. Those memories can be further used to generate internal models of the world that enable predictions to be made. The mechanisms that support these associative and predictive aspects of memory are not yet understood. In this study, we used an optogenetic manipulation to perturb the sequential structure, but not global network dynamics, of place cells as rats traversed specific spatial trajectories. This perturbation abolished replay of those trajectories and the development of predictive representations, leading to impaired learning of new optimal trajectories during memory-guided navigation. However, place cell assembly reactivation and reward-context associative learning were unaffected. Our results show a mechanistic dissociation between two complementary hippocampal codes: an associative code (through coactivity) and a predictive code (through sequences).

Abstract:Recollecting a past episode involves remembering not only what happened but also when it happened. We used positron emission tomography (PET) to directly contrast the neural correlates of item and temporalorder memory. Subjects studied a list of words and were then scanned while retrieving information about what words were in the list or when they occurred within the list. Item retrieval was related to increased neural activity in medial temporal and basal forebrain regions, whereas temporal-order retrieval was associated with activations in dorsal prefrontal, cuneus/precuneus, and right posterior parietal regions. The dissociation between temporal and frontal lobe regions confirms and extends previous lesion data. The results show that temporal-order retrieval involves a network of frontal and posterior brain regions.

Abstract:How the human brain reconstructs, step-by-step, the core elements of past experiences is still unclear. Here, we map the spatiotemporal trajectories along which visual object memories are reconstructed during associative recall. Specifically, we inquire whether retrieval reinstates feature representations in a copy-like but reversed direction with respect to the initial perceptual experience, or alternatively, this reconstruction involves format transformations and regions beyond initial perception. Participants from two cohorts studied new associations between verbs and randomly paired object images, and subsequently recalled the objects when presented with the corresponding verb cue. We first analyze multivariate fMRI patterns to map where in the brain high- and low-level object features can be decoded during perception and retrieval, showing that retrieval is dominated by conceptual features, represented in comparatively late visual and parietal areas. A separately acquired EEG dataset is then used to track the temporal evolution of the reactivated patterns using similarity-based EEG–fMRI fusion. This fusion suggests that memory reconstruction proceeds from anterior frontotemporal to posterior occipital and parietal regions, in line with a conceptual-to-perceptual gradient but only partly following the same trajectories as during perception. Specifically, a linear regression statistically confirms that the sequential activation of ventral visual stream regions is reversed between image perception and retrieval. The fusion analysis also suggests an information relay to frontoparietal areas late during retrieval. Together, the results shed light onto the temporal dynamics of memory recall and the transformations that the information undergoes between the initial experience and its later reconstruction from memory.

Hippocampal-parietal cortex causal directed connectivity during human episodic memory formation: Replication across three experiments

Abstract:Hippocampus-parietal cortex circuits are thought to play a crucial role in memory and attention, but their neural basis remains poorly understood. We employed intracranial EEG from 96 participants (51 females) to investigate the neurophysiological underpinning of these circuits across three memory tasks spanning verbal and spatial domains. We uncovered a consistent pattern of higher causal directed connectivity from the hippocampus to both lateral parietal cortex (supramarginal and angular gyrus) and medial parietal cortex (posterior cingulate cortex) in the delta-theta band during memory encoding and recall. This connectivity was independent of activation or suppression states in the hippocampus or parietal cortex. Crucially, directed connectivity from the supramarginal gyrus to the hippocampus was enhanced in participants with higher memory recall, highlighting its behavioral significance. Our findings align with the attention-to-memory model, which posits that attention directs cognitive resources toward pertinent information during memory formation. The robustness of these results was demonstrated through Bayesian replication analysis of the memory encoding and recall periods across the three tasks. Our study sheds light on the neural basis of casual signaling within hippocampus-parietal circuits, broadening our understanding of their critical roles in human cognition.

Time-dependent reorganization of brain circuitry underlying long-term memory storage

Abstract:Retrograde amnesia observed following hippocampal lesions in humans and animals is typically temporally graded1,2, with recent memory being impaired while remote memories remain intact, indicating that the hippocampal formation has a time-limited role in memory storage3,4. However, this claim remains controversial because studies involving hippocampal lesions tell us nothing about the contribution of the hippocampus to memory storage if this region was present at the time of memory retrieval5,6. We therefore used non-invasive functional brain imaging using (14C)2-deoxyglucose uptake to examine how the brain circuitry underlying long-term memory storage is reorganized over time in an intact brain. Regional metabolic activity in the brain was mapped in mice tested at different times for retention of a spatial discrimination task. Here we report that increasing the retention interval from 5 days to 25 days resulted in both decreased hippocampal metabolic activity during retention testing and a loss of correlation between hippocampal metabolic activity and memory performance. Concomitantly, a recruitment of certain cortical areas was observed. These results indicate that there is a time-dependent reorganization of the neuronal circuitry underlying long-term memory storage, in which a transitory interaction between the hippocampal formation and the neocortex would mediate the establishment of long-lived cortical memory representations.

Abstract:Events occurring close in time are often linked in memory, providing an episodic timeline and a framework for those memories. Recent studies suggest that memories acquired close in time are encoded by overlapping neuronal ensembles, but the role of dendritic plasticity mechanisms in linking memories is unknown. Using activity-dependent labeling and manipulation approaches, longitudinal one- and two-photon imaging of somatic and dendritic compartments, and computational modeling, we show that memory linking is not only dependent on ensemble overlap in the retrosplenial cortex, but also on branch-specific dendritic allocation mechanisms. The same dendritic segments are preferentially activated by two linked memories, and spine clusters added after each of the two linked memories are allocated to the same dendritic segments. Our results demonstrate a causal mechanistic role for dendritic plasticity in memory integration and reveal a novel set of rules that govern how linked and independent memories are allocated to dendritic compartments.

Abstract:It is now widely accepted that remembering the past and imagining the future rely on a number of shared processes and recruit a similar set of brain regions. However, memory and future thinking place different demands on a range of processes. For instance, although remembering should lead to early associative retrieval of event details, event construction may be slower for future events, for which details from different memories are combined. In order to shed light on the question of how the brain distinguishes between memories and future thoughts, we investigated the differences in the electrophysiological correlates of the vivid elaboration of future and past events. In the slow cortical potentials of 24 healthy human participants, differences during early elaboration were observed at temporo-parietal and parieto-occipital electrode sites, presumably reflecting differential recruitment of sensory and semantic detail retrieval. Additional differences emerged over the right pre-frontal cortex during later elaboration, which could be linked to differential retrieval demands. In conclusion, the time course differences, which presumably reflect the varying recruitment of sub-processes engaged during mental time travel, will help to understand the mechanisms with which the brain separates memories from future thoughts.

Abstract:NeuroscienceSleep affects memories via several mechanisms. Izawa et al. identified a possible new pathway in the brain: REM sleep–active hypothalamic melanin-concentrating hormone (MCH)–producing neurons, which, among others, project to the hippocampus. Surprisingly, genetic ablation of MCH neurons increased memory performance in mice. Conversely, pharmacogenetic activation of MCH neurons impaired memory. In vitro physiological experiments showed that activation of MCH fibers in hippocampal slices suppressed spiking activity of pyramidal cells.
