Abstract:Soil microorganisms play an important role in the ecosystem, and have a certain relationship with the continuous cropping obstacles, which are common with sweet potato. However, there are few reports on the effects of continuous cropping of sweet potato on the microbial community structure in the rhizospheric soil. Here, we investigated the effects of continuous cropping of sweet potato on the fungal community structure in rhizospheric soil, in order to provide theoretical basis for prevention and control of continuous cropping obstacles. This study used X18 and Y138 varieties as experimental materials. Soil samples were collected during the early period of planting and harvest in two consecutive years, and fungi were analyzed using Illumina Miseq. Results showed that the fungi diversity and richness in rhizospheric soil of X18 and Y138 were significantly increased after continuous cropping; the most dominant fungi phylum was Ascomycota, which decreased significantly after continuous cropping. In addition, the content of beneficial fungi such as Chaetomium was reduced, while that of harmful fungi such as Verticillium, Fusarium, and Colletotrichum were increased. The composition of X18 and Y138 fungal community in the same sampling period after continuous cropping was similar, although that of the same sweet potato variety significantly differed with the sampling period. Overall, our results indicate that continuous cropping alters the fungal community structure of the sweet potato rhizospheric soil, such that the content of beneficial fungi decrease, while that of harmful fungi increase, thereby increasing soil-borne diseases and reducing the yield and quality of sweet potato. Furthermore, these effects are different for different sweet potato varieties. Thus, during actual production, attention should be paid to maintain the stability of sweet potato rhizospheric soil micro-ecology through rotation or application of microbial fertilizers and soil amendments to alleviate continuous cropping obstacles.

Abstract:Changes in soil chemical properties, phenolic compounds and bacterial community structure and diversity in rhizosphere and bulk soils under 2 to 10 years continuous cropping of strawberry (Fragaria orientalis Lozinsk.) and tomato (Lycopersicon esculentum Mill.), was examined under green house conditions. Soil bacterial community composition was analysed using T-RFLP technique combined with 16S rRNA gene clone library construction. Results showed that soil pH, organic matter, extractable P and cations in both rhizosphere and bulk soils decreased with increasing years of continuous cropping. The phenolic compounds content of rhizosphere soils increased, especially p-hydroxybenzoic acid. Members affiliated to Proteobacteria were predominant in rhizosphere soils, followed by Bacteroidetes and Acidobacteria. Dominant genera were Devosia, Bradyrhizobium and Sphingomonas of Alphaproteobacteria. content The bacterial diversity in rhizosphere soils was lower under continuous cropping than under rotation cropping and negatively correlated with vanillin content. Continuous cropping of strawberry and tomato, chemically deteriorated the soils and increased the phenolic compounds content, thereby affecting the bacterial community composition.

Abstract:Plant rhizosphere-associated bacterial communities play key roles in affecting host health in response to diverse biotic stresses. Currently, the effect of continuous cropping of potato on soil bacterial communities and physiochemical parameters has not been well documented. Herein, we compared bacterial composition and diversity in rotationally and continuously (5, 10, and 30 years) cropped soils, and clarified the correlations between soil properties and the bacterial communities revealed by Illumina MiSeq sequencing. Our results demonstrated that Proteobacteria, Actinobacteria and Firmicutes were the predominant phyla in all the tested soil samples. While the abundance of Proteobacteria showed an increase, the abundance of Actinobacteria and Firmicutes displayed a reduction with the increase of continuous cropping years. At the genus level, as continuous cropping years increasing, the abundance of Pseudarthrobacter, Bacillus and Pseudomonas decreased, but the abundance of Rhodanobacte, Sphingobium, Mizugakiibacter and Devosia increased. Our results also demonstrated that the abundance of plant growth-promoting rhizobacteria in the rotationally cropped soil was significantly higher than that of continuously cropped soil. Furthermore, our results showed that soil organic matter, available nitrogen, available phosphorus and available potassium were significantly correlated with bacterial community distribution. Overall, our work provides a comprehensive view of altered structure and composition of bacterial communities between the continuously cropped soil and rotationally cropped soil.

Abstract:This study explored variations in the rhizosphere microbial community composition resulting from the continuous cropping of sorghum. We explored the potential growth-promoting effects of Bacillus amyloliquefaciens in manipulating the microbial community composition of the rhizosphere soil under continuous cropping. Illumina MiSeq sequencing was used to compare the composition of the bacterial and fungal communities in the rhizosphere soil from a sorghum–maize rotation system, sorghum continuous cropping system, and sorghum continuous cropping with the application of the biological agent B. amyloliquefaciens. The rhizosphere soil under continuous sorghum cropping had lower bacterial richness and higher fungal diversity than the sorghum–maize rotation rhizosphere soil. The bacterial phyla Acidobacteria and Gemmatimonadetes, and the fungal phylum Basidiomycota, were strongly enriched in the continuous cropping soil. The class Tremellomycetes was the most dominant group, accounting for 19.30% of the total fungal sequences in the continuous cropping soil and 4.51% in sorghum-maize rotation soil. Application of B. amyloliquefaciens significantly decreased the abundance of Tremellomycetes by 8.87% in the continuous cropping soil. Our results indicate that the decrease in sorghum growth and yield under continuous cropping are in coincidence with the alteration of soil microbial community composition, particularly the accumulation of Tremellomycetes in the continuously cropped rhizosphere soil. Our findings contribute to an improved understanding of the effects of continuous sorghum cropping on soil microbiome, and B. amyloliquefaciens can be used to manipulate the microbial communities of the sorghum rhizosphere soil.

Enrichment of Phytopathogen Dominated by Volutella in the Rhizosphere May Be an Important Cause of Continuous Cropping Obstacles in Sweet Potatoes

Abstract:Obstacles related to the continuous cropping of sweetpotato ( Ipomoea batatas (L.) Lam.) restrict its intensive production. However, the effects of continuous cropping on soil properties and/or the rhizosphere microbial community are largely unclear. In this study, we analysed changes in rhizosphere soil chemical properties and microbial communities of sweetpotato across different cultivation years. We observed that the microbial diversity and complexity of the fungal ecological network in rhizosphere soils of sweetpotato were decreased after 5 years of continuous cropping, with significant enrichment in Sphingobium, Gemmatimonas, Volutella, and Neoidriella. Tuber yield, soil pH, and available potassium (AK) content were significantly reduced after continuous cropping, with the soil microbial community having the highest correlation with pH, AK, and ammonium nitrogen (NH -N). Specifically, soil pH and AK were positively correlated with Bacillus and Gaiella, and negatively correlated with some plant-pathogenic fungi ( Volutella and Neoidriella), while NH -N showed the opposite trend. In addition, soil pH, AK, and the relative abundance of Bacillus were positively correlated with tuber yield, while Volutella showed the opposite trend. In summary, the continuous cropping of sweetpotato negatively affects rhizosphere soil health, resulting in imbalanced soil fertility and increased abundance of pathogens. These results improve our understanding of factors driving obstacles faced with the continuous cropping of sweetpotato, enabling future studies and the development of technologies to overcome these obstacles.
[Effects of addition of sorghum stubble rhizosphere soil on growth and rhizosphere microbes of continuous cropping cucumber]

Abstract:We explored the effects of addition of sorghum stubble rhizosphere soil on the growth of continuous cropping cucumber and rhizosphere microbial community in a pot experiment. The diffe-rences in soil bacterial and fungal community composition were analyzed with fluorescence quantitative PCR and high-throughput sequencing technology. There were four treatments: CK (no fertilization), T1(fertilizer only), T2(optimized fertilization), and T3(optimized fertilization + rhizosphere soil of sorghum stubble). The results showed that compared with other treatments, T3 promoted the growth and development of cucumber, and increased the abundance of 16S rRNA and ITS rRNA genes in soil. Compared with the T1 treatment, T2 and T3 significantly increased the richness and diversity of bacterial communities. There was no significant difference in fungal community richness and diversity among different treatments. Adding rhizosphere soil of sorghum stubble changed the composition of bacterial and fungal communities at both phylum and genus levels. For bacteria, it increased the abundances of Acidobacteria and Bacteroides, but decreased that of Proteobacteria, Firmicutes, Nitrospira and Bacillus. For fungi, it increased the abundance of Basidiomycota, Trichoderma and Pseudurotium, but decreased that of Fusarium and Metarhizium. Results of redundancy analysis showed that soil nitrate and organic matter were the key factors affecting the difference of bacterial and fungal community composition, respectively. In conclusion, addition of sorghum stubble rhizosphere soil improved the total abundance of soil microorganisms and bacterial diversity for continuous cropping cucumber. It increased the abundance of beneficial bacteria Trichoderma, reduced that of pathogenic Fusarium, and maintained the survival rate of cucumber, thus provided a feasible solution for alleviating the barriers for the continuous cropping of cucumber.
Continuous Cropping Changes the Composition and Diversity of Bacterial Communities: A Meta-Analysis in Nine Different Fields with Different Plant Cultivation

Abstract:Chinese goldthread (Coptis chinensis Franch.) represents one of the most important medicinal plants with diverse medicinal applications, but it easily suffers from continuous cropping obstacles in the plantation. In this study, we have selected eight different continuously cropped fields with C. chinensis and fallow field, providing detailed information regarding the diversity and composition of the rhizospheric bacterial communities. We have found a significant difference between fallow field (LH) and other continuously cropped fields in soil pH; the total content of nitrogen, phosphorus, and potassium; and soil enzyme activities. The results indicate that continuous cropping had a significant effect on soil physicochemical properties and enzyme activities under different plant cultivations. The relative abundance of bacterial phyla was significantly altered among the fields; for example, proteobacteria and Actinobacteria were observed to be higher in continuous cropping of maize (HY6) and lower in sweet potato continuous cropping (HH). Alpha diversity analysis showed that different plants with different years of continuous cropping could change the diversity of bacterial communities, among which the effect of maize and Polygonum multiflorum continuous cropping were most significant. Principle coordinate analysis (PCoA) showed that continuously cropped C. chinensis (LZ) and cabbage continuously cropped for 2 years (HS) were slightly clustered together and separated from LH and others. The results showed that the similarity of the bacterial community in the same crop rotation was higher, which further indicated that the bacterial community structure was significantly altered by the continuous cropping system and plant species. Our study provides a foundation for future agricultural research to improve microbial activity and increase crops/cash-crops productivity under a continuous cropping system and mitigate continuous cropping obstacles.

Abstract:The continuous cropping of plants can result in the disruption of the soil microbial community and caused significant declines in yields. However, there are few reports on the effects of continuous cropping of sugarcane on the microbial community structure and functional pathway. In the current study, we analyzed the structural and functional changes of microbial community structure in the rhizospheric soil of sugarcane in different continuous cropping years using Illumina Miseq high-throughput sequencing and metagenomics analysis. We collected rhizosphere soils from fields of no continuous cropping history (NCC), 10 years of continuous cropping (CC10), and 30 years of continuous cropping (CC30) periods in the Fujian province. The results demonstrated that continuous sugarcane cropping resulted in significant changes in the physicochemical properties of soil and the composition of soil bacterial and fungal communities. With the continuous cropping, the crop yield dramatically declined from NCC to CC30. Besides, the redundancy analysis (RDA) of the dominant bacterial and fungal phyla and soil physicochemical properties revealed that the structures of the bacterial and fungal communities were mainly driven by pH and TS. Analysis of potential functional pathways during the continuous cropping suggests that different KEGG pathways were enriched in different continuous cropping periods. The significant reduction of bacteria associated with rhizospheric soil nitrogen and sulfur cycling functions and enrichment of pathogenic bacteria may be responsible for the reduction of effective nitrogen and total sulfur content in rhizospheric soil of continuous sugarcane as well as the reduction of sugarcane yield and sugar content. Additionally, genes related to nitrogen and sulfur cycling were identified in our study, and the decreased abundance of nitrogen translocation genes and AprAB and DsrAB in the dissimilatory sulfate reduction pathway could be the cause of declined biomass. The findings of this study may provide a theoretical basis for uncovering the mechanism of obstacles in continuous sugarcane cropping and provide better guidance for sustainable development of the sugarcane.

Abstract:Under the condition of field,set the maize straw returning not returning rotary tillage(CK) and corn stover(XG) and returning the commune(SF) 3 processing,two years of field orientation test in cold area,the short straw returning to the influence of continuous cropping of corn field soil microenvironment was analysed.The results showed that straw returning rotary tillage and returning the commune significantly improved 0 to 30 cm soil enzyme activity and soil bacterial diversity.Straw rotary tillage could improve the soil bacterial diversity in the 0-30cm layer,and the straw returning to the field could improve the soil bacterial diversity in the topsoil (11-30 cm).Straw returning to the commune in soil rich in functional microbes,later can be used as screening functional microbial source of bacteria.
Distinct Characteristics of the Bacterial Community in the Rhizosphere Soils of Sweet Potato Storage Roots and Fibrous Roots Following Long-Term Fertilization

Abstract:Sweet potato (Ipomoea batatas Lam.) has two special root systems, namely, storage roots and fibrous roots. However, the response of the bacterial community characteristics in the rhizosphere soils of the two types of roots to long-term fertilization is not clear. Here, a long-term fertilization experiment was established under wheat-sweet potato crop rotation in 2011. We sampled the rhizosphere soils of storage roots and fibrous roots under three fertilization treatments (i.e., NK, no P; NPK, chemical P fertilizer; and NPKM, inorganic combined with organic fertilizers). The diversity and composition of the bacterial community and the relative abundance of specific bacterial taxa and their relationships with soil properties were investigated at the stage of sweet potato expansion. The results showed that compared to the NK treatment, the NPK treatment significantly increased soil available P (AP) in the rhizosphere of storage roots and fibrous roots by 503 % and 668 %, dissolved organic carbon (DOC) by 25 % and 18 %, and available K (AK) by 3.5 % and 12.3 %, respectively. However, compared to the NK treatment, the Chao1 index was significantly decreased under the NPK treatment in both storage roots and fibrous roots. In addition, compared to storage roots, bacterial alpha-diversity and soil beta-glucosidase and alkaline phosphatase activities significantly improved in the rhizosphere of fibrous roots (P < 0.001) under the NPKM treatment. Chryseobacterium, Acinetobacter and Myroides were the dominant bacterial genera in the rhizosphere soil of storage roots under the NPKM treatment. Furthermore, principal coordinate analysis (PCoA) indicated that the structure of the bacterial community was markedly altered in the rhizosphere soils of storage roots and fibrous roots under the NPKM treatment. Moreover, a complex network of bacterial communities was formed in the rhizosphere of storage roots rather than that of fibrous roots. Based on redundancy analysis (RDA) and structural equation modeling (SEM), the results revealed that the diversity and structure of the bacterial com-munity were primarily driven by the AK concentration in the rhizosphere soil of the storage roots. In summary, long-term fertilization reshaped the bacterial community in the rhizosphere soils of two types of sweet potato roots by altering the properties of the rhizosphere soils.

Abstract:Understanding the response of microbial communities and their potential functions is essential for sustainability of agroecosystems under long-term continuous cropping. However, limited research has focused on investigating the interaction between soil physicochemical factors and microbial community dynamics in agroecosystems under long-term continuous cropping. This study probed into the physicochemical properties, metabolites, and microbial diversity of tobacco rhizosphere soils cropped continuously for 0, 5, and 20 years. The relative abundance of bacterial genera associated with nutrient cycling (e.g., Sphingomonas) increased while potential plant pathogenic fungi and beneficial microorganisms showed synergistic increases with the duration of continuous cropping. Variations in soil pH, alkeline nitrogen (AN) content, and soil organic carbon (SOC) content drove the shifts in soil microbial composition. Metabolites such as palmitic acid, 3-hydroxypropionic acid, stearic acid, and hippuric acid may play a key role in soil acidification. Those results enhance our ability to predict shifts in soil microbial community structure associated with anthropogenic continuous cropping, which can have long-term implications for crop production.

Abstract:Abstract Improving soil structure, fertility, and production is of major concern for establishing sustainable agroecosystems. Further research is needed to evaluate whether different methods of straw returning determine the variations of soil aggregation and the microbial community in aggregates in the long term. In this study, we comparatively investigated the effects of long-term fertilization regimes performed over six years, namely, non-fertilization (CK), chemical fertilization (CF), continuous straw return (CS), and continuous straw-derived biochar amendment (CB), on soil aggregation and bacterial communities in rice-wheat rotation systems. The results showed that straw/biochar application increased soil nutrient content and soil aggregate size distribution and stability at both 0–20 cm and 20–40 cm soil depths, compared with those of CF and CK; CB performed better than CS. CB increased bacterial community diversity and richness in 0–20 cm soil, and evenness in 0–40 cm soil ( p < 0.05); CS had no significant effect on these aspects. Variations in the relative abundance of Actinobacteria, Chloroflexi, Bacteroidetes, Nitrospirae, Gemmatimonadetes, and Latescibacteria in specific aggregates confirmed the different effects of straw/biochar on bacterial community structure. The partial least squares discrimination analysis and permutation multivariate analysis of variance revealed that fertilization, aggregate size fractions, and soil depth affected the bacterial community, although their effects differed. This study suggests that CB may reduce chemical fertilizer usage and improve the sustainability of rice-wheat cropping systems over the long term, with a better overall outcome than CS.

Effects of Sweet Potato Intercropping in Banana Orchard on Soil Microbial Population Diversity

Abstract:PurposeThis study was purposely designed to understand the effects of intercropping banana and sweet potato on soil microbial community. The research question addressed was what were the differences in population number, population diversity and dominant population of soil microorganisms between interplant bananas with sweet potatoes and banana monoculture.MethodsThe Illumina MiSeq high-throughput sequencing technology was used to detect and analyse the population composition and structure of soil microorganisms in banana field.ResultsThe results showed that from May to September, the number of soil bacterial population in intercropping sweet potato was 5.54-28.67% higher than that in monoculture, and the richness and diversity of the population were significantly or extremely significantly higher than that in monoculture. The number of dominant bacterial population was less than that in monoculture, and the relative abundance of non dominant population was 10.58 - 58.81% higher than that in monoculture. The number, abundance and diversity of soil fungal populations in intercropping were higher than those in monoculture.ConclusionsThe intercropping of banana and sweet potato has a significant effect on regulating the composition structure of soil microbial population and improving the abundance and diversity of microbial population. There has a great significance to improve the micro ecological environment of banana root soil and promote the stable and sustainable development of banana industry.

Abstract:Continuous cropping of soybean causes soil degradation and decreased soybean yield in Northeast PR China. However, the mechanism underlying the effect on soil fungal communities and the relationship between these effects and cropping systems have not yet been fully elucidated. In this study, we comparatively investigated the abundance, diversity, and structures of fungal communities in the bulk and rhizospheric soils of soybean grown in two cropping systems, that is, continuous cropping (CC) of soybean and cropping rotation (CR) with maize using real-time Polymerase Chain Reaction (PCR) and Illumina MiSeq sequencing methods. The results showed that the rhizospheric soil fungal abundance was significantly higher in CC than in CR, but the opposite trend was observed in bulk soils. CC decreased the fungal alpha diversity in both bulk and rhizospheric soils compared with CR. The fungal community compositions were distinctly different between CC and CR in both bulk and rhizospheric soils, and the composition and diversity in bulk soils were predominantly affected by the soil carbon to nitrigen (C/N) ratio. The soil fungal functional groups varied between the two cropping systems; CC increased the relative abundances of the potentially pathogenic fungi Fusarium oxysporum and Lectera longa and beneficial fungi Mortierella sp. and Paecilomyces lilacinus. In contrast, CR increased the relative abundance of Ustilago maydis, which is a pathogen that causes corn smut. Our findings showed that CC increased the abundances of plant pathogens, which is harmful to soybean growth; in contrast, the abundances of some beneficial fungi also increased, which suggested that suppressive soils might be developed after long-term CC.
