1 Propionate Promotes Intestinal Lipolysis and 2 Metabolic Benefits via AMPK / LSD 1 Pathway in Mice 3

Abstract:Dietary fibers and the microbial fermentation products short chain fatty acids promote 19 metabolic benefits, but the underlying mechanisms are poorly understood. Recent studies indicate 20 that intestinal lipid handling is under regulatory control and has broad influence on whole body 21 energy homeostasis. Here we report that dietary inulin and propionate significantly decrease whole 22 body fat mass without affecting food intake in mice fed with chow diet. Inulin and propionate 23 decrease intestinal triglyceride content and induce enterocyte lipolysis genes expression, such as 24 adipose triglyceride lipase (ATGL), hormone sensitive lipase (HSL) and adipose triglyceride lipase 25 (LAL). Both in vivo and in vitro studies show propionate can induce enterocyte lipolysis and 26 decrease lipid content, via direct activation of phosphorylation of AMP-activated protein kinase (p27 AMPK) and lysine-specific histone demethylase 1 (LSD1). Moreover, propionate and inulin can 28 increase intestinal lipid lipolysis under high fat diet (HFD) fed condition and prevent from HFD29 induced obesity. Our study suggests dietary fiber inulin and its microbial fermentation product 30 propionate can induce intestinal lipolysis and decrease whole body fat mass via activation of p31 AMPK and LSD1, which could provide a novel therapeutic target for both prevention and treatment 32 of obesity. 33
