Abstract:Polyamine biosynthesis is controlled primarily by ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (AdoMetDC). Antisense ODC and AdoMetDC sequences were cloned into an adenoviral vector (AdODC-AdoMetDCas). To evaluate the effect of recombinant adenovirus Ad-ODC-AdoMetDCas which can simultaneously express both antisense ODC and S-adenosylmethionine decarboxylase (AdoMetDC), the human esophageal squamous carcinoma cell line Eca109, was infected with Ad-ODC-AdoMetDCas as well as with control vector. Viable cell counting, determination of polyamine concentrations, cell cycle analysis, and Matrigel invasion assays were performed in order to assess the properties of tumor growth and invasiveness. Our study demonstrated that adenovirus-mediated ODC and AdoMetDC antisense expression inhibits tumor cell growth through a blockade of the polyamine synthesis pathway. This inhibitory effect cannot be reversed by the administration of putrescine. Tumor cells were arrested at the G 1 phase of the cell cycle after gene transfer and had reduced invasiveness. Our study suggests that as a new anticancer reagent, the recombinant adenovirus Ad-ODC-AdoMetDCas holds promising hope for the therapy of esophageal cancers.

Abstract:BACKGROUND & OBJECTIVE Esophageal carcinoma is one of the most common malignant tumors in China. It is reported that the content and biosynthesis of polyamine in esophageal cancer is markedly increased. This study was to investigate inhibitory effects of both antisense ornithine decarboxylase (ODCas) and S-adenosylmethionine bi-antisense (AdoMetDCas) on polyamine biosynthesis, cell proliferation and invasion of esophageal cancer cell line Eca109. METHODS An adenoviral vector Ad-ODC-AdoMetDCas containing antisense sequences of ODCse and AdoMetDCas was used. Cell proliferation was observed by counting viable cells or BrdU labeling. Protein expressions of ODC and AdoMetDC and the polyamine content in Eca109 cells were measured by Western blot and high performance liquid chromatography (HPLC), respectively. Invasion of Eca109 cells in vitro was detected using Matrigel invasion assay. Furthermore, the anti-proliferation effect of Ad-ODC-AdoMetDCas on Eca109 cell xenografts in nude mice was evaluated. RESULTS Ad-ODC-AdoMetDCas significantly inhibited proliferation of Eca109 cells(P<0.05) and protein expressions of ODC and AdoMetDC. Transfection of Ad-ODC-AdoMetDCas significantly decreased synthesis of three polyamines in Eca109 cells, which were putrescine (Put), spermidine (Spd) and spermine (Spm) (P<0.05). Ad-ODC-AdoMetDCas dramatically suppressed invasiveness of Eca109 cells in vitro(P<0.05). In addition, compared with Ad-GFP, Ad-ODC-AdoMetDCas significantly suppressed the growth of Eca109 cell xenografts in nude mice. CONCLUSION Ad-ODC-AdoMetDCas significantly inhibits cell proliferation and invasion in esophageal cancer Eca109 cells.

Abstract:Objective:To study the inhibitory effects of Ad-ODC-AdoMetDCas,a recombinant adenovirus with biantisense RNA of ornithine decarboxylase(ODC)and s-adenosylmethionine decarboxylase(AdoMetDC),on cell proliferation and invasion of esophageal cancer cells.Methods:The Ad-ODC-AdoMetDCas was used to infect esophageal cancer Eca109 cells.Western blotting was used to examine the protein expression of ODC and AdoMetDC in Eca109 cells.The content of polyamine in Eca109 cells was determined by HPLC.The inhibition on the proliferation of Eca109 cells was investigated by viable cell counting.Matrigel invasion assay was used to study the invasion ability of Eca109 cells.Furthermore,the anti-tumor effect of Ad-ODC-AdoMetDCas was evaluated in a nude mouse xenograft model.Results:Western blotting assay showed that Ad-ODC-AdoMetDCas inhibited the expression of ODC and AdoMetDC;HPLC indicated that the contents of putrescine,spermidine and spermine were markedly decreased in Eca109 cells after infection with Ad-ODC-AdoMetDCas(P0.01).Viable cell counting showed that Ad-ODC-AdoMetDCas significantly inhibited the proliferation of Eca109 cells(P0.01).Matrigel invasion assay showed that Ad-ODC-AdoMetDCas significantly decreased the invasion ability of Eca109 cells in vitro(P0.01).Meanwhile,experiments with nude mouse xenograft model demonstrated that Ad-ODC-AdoMetDCas had significant anti-tumor ability in vivo(P0.05,P0.01).Conclusion:Ad-ODC-AdoMetDCas can significantly inhibit the proliferation and invasion of esophageal cancer,which makes it a potential therapy for the treatment of esophageal cancer.

Abstract:OBJECTIVE:To study the inhibitory effects of antisense bicistronic recombinant adenovirus vector of ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (Ad-ODC-AdoMetDCas) on polyamine biosynthesis,proliferation and invasion of lung cancer cells.METHODS:Adenovirus-mediated gene transduction efficiency was assessed with counting GFP-positive cells using trypan blue. Western Blot and HPLC were used to detect ODC and S-AdoMetDC expression and polyamine content in A-549 cells respectively. Viable cell counting and cell cycle analysis were adopted to evaluate cell growth and cell cycle distribution, and A-549 cell invasion in vitro was detected with Matrigel invasion assay.RESULTS:Approximate 75% of A-549 cells were infected with Ad-ODC-AdoMetDCas when multiplicity of infection reached 50. Our study demonstrated that Ad-ODC-AdoMetDCas vector-mediated gene transfer inhibited tumor cell growth through the blockade of polyamine synthesis pathway. The tumor cells were arrested at cell cycle G1 phase after gene transfer. Gene transferred tumor cells were shown to possess markedly decreased invasiveness.CONCLUSION:Ad-ODC-AdoMetDCas has significant inhibitory effects on lung cancer cell proliferation and invasion and bears therapeutic potential for the treatment of lung cancer.

Ad-ODC-AdoMetDCas could inhibit Eca109 cell growth and invasive ability. TUNEL proved that Ad-ODC-AdoMetDCas can lead to cell apoptosis. Characterized morphology was observed by electron microscope (chromatin condensation, nuclear disintegration, formation of apoptotic bodies). It was suggested Ad-ODC-AdoMetDCas has significant inhibitory effects on esophageal cancer cell proliferation , leads to cell apoptosis and bears therapeutic potential for the treatment of esophageal cancer.

Abstract:Objective: To investigate the inhibitory effect of Ad-ODC-AdoMetDCas which can simultaneously express both antisense ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (AdoMetDC) on colorectal cancer cell growth and detect the possible mechanism. Methods: Human colorectal cancer cells HT-29 were cultured in RPMI 1640 medium and were infected with Ad-ODC-AdoMetDCas, MTS was used to assess the HT-29 cell growth. Cell cycle progression was detected by flow cytometry analysis. ODC, AdoMetDC and cell cycle regulated proteins levels were measured by Western blot analysis. The mRNA level of cyclin D1 was measured by RT-PCR. Results: The recombinant adenovirus Ad-ODC-AdoMetDCas downregulated the expression of ODC and AdoMetDC in HT-29 cells. Ad-ODC-AdoMetDCas significantly inhibited the cell growth and induced G1 arrest. Cyclin DI protein and mRNA levels were decreased by Ad-ODC-AdoMetDCas infection. Conclusion: Downregulation of ODC and AdoMetDC mediated by Ad-ODC-AdoMetDCas induces G1 arrest in HT-29 cells by suppressing cyclin DI expression. As a new anticancer reagent, Ad-ODC-AdoMetDCas has potential therapeutic effect on colorectal cancers.

Abstract:To investigate the in vitro inhibitory effect of rAd-ODC/Ex3as on human esophageal carcinoma cells. The infection rate of rAd-ODC/Ex3as was measured with the aid of GFP expression. Western Blot technique was used to observe the inhibition of ODC expression in infected tumor cells. The malignant phenotype of Eca109 cell line was assessed by growth curve. TUNEL was used to analyze cell apoptosis. Approximate 65% of Eca109 cell line were infected with rAd-ODC/Ex3as when MOI reached 50. The expression of ODC was inhibited in the infected tumor cells. rAd-ODC/Ex3as could inhibit Eca109 cell line growth and invasive ability at 20 of MOI. TUNEL proved that rAdODC/Ex3as can lead to cell apoptosis. rAd-ODC/Ex3as could inhibit effectively the expression of ODC gene and the growth of of esophageal squamous carcinoma cell line Eca109 in vitro, and induce apoptosis. It may be one of the promising medicines for antisense gene therapy in esophageal cancer.

Abstract:AIM:To construct a recombinant adenovirus that can simultaneously express both antisense ornithine decarboxylase (ODC) and adenosylmethionine decarboxylase ( AdoMetDC ) and detect its inhibitory effect on the intracellular polyamine pool and colorectal cancer cell growth.METHODS:A 205-bp cDNA of AdoMetDC was reverse-inserted into recombinant pAdTrack-ODCas vectors and recombined with pAdEasy-1 vectors in AdEasy-1 cells. Positive clones were selected and transfected into the packaging cell HEK293 after they were linearized by PacI. Green fluorescent protein expression was used to monitor the process of adenovirus packaging. The ODC and AdoMetDC protein levels were identified by western blotting, and intracellular polyamine content was detected by reverse-phase high performance liquid chromatography. A viable cell count was used to determine the growth of HT-29 cells with or without exogenous polyamine.RESULTS:Sequencing confirmed that AdoMetDC cDNA was successfully ligated into the pAdTrack-ODCas vector. GFP expression in 293 cells during virus packing and amplification was observed by fluorescence microscopy. Western blotting demonstrated that both ODC and AdoMetDC were downregulated by Ad-ODC-AdoMetDCas, and consequently 3 kinds of polyamine (putrescine, spermidine and spermine) were reduced to very low levels. HT-29 cell growth was significantly inhibited as compared with control conditions, and growth arrest was not reversed by exogenous putrescine.CONCLUSION:The successfully constructed recombinant adenovirus, Ad-ODC-AdoMetDCas, blocked polyamine synthesis and has therapeutic potential for treating colorectal cancer in vitro.

Abstract:OBJECTIVE:To construct the recombinant adenovirus encoding antisense c-myc fragment and to investigate its effect on the chemotherapy sensitivity of osteosarcoma MG-63 cells to cisplatin.METHODS:The recombinant adenovirus (Ad-Asc-myc) encoding antisense c-myc fragment was constructed by cloning c-myc cDNA of about 720 base pairs in a reverse direction into adenovirus vector, then undergoing recombination, amplifying and being complemented in vivo. The osteosarcoma MG-63 cells were transfected by the Ad-Asc-myc in vitro, and Wright staining, Acridine Orange staining, Western Blot, MTT, Flow Cytometry (FCM) were used to study cell morphology, expression of c-myc protein, tumor cell proliferation in vitro, apoptosis and cell cycle change.RESULTS:Ad-Asc-myc encoding antisense c-myc fragment was obtained with the titer of 2 x 10(9) pfu/ml. Ad-Asc-myc down-regulated the expression of c-myc protein after transfected MG-63 cells for 48 h, combined with the treatment of 2.0, 5.0 microg/ml cisplatin for 2 h could inhibit tumor cells proliferation in vitro by 33.4% and 54.2% respectively, which were significantly difference compared with control recombinant adenovirus (Ad-LacZ) groups (P < 0.05). Acridine Orange staining and FCM analysis showed that Ad-Asc-myc could induce apoptosis of transfected cells, which was enhanced by the treatment of cisplatin cell. Cycle analysis showed that obvious G2/M phase arrested in transfected cells.CONCLUSION:Ad-Asc-myc increases the chemotherapy sensitivity of osteosarcoma MG-63 cells to cisplatin as well as induced apoptosis.

Abstract:Aims: Studies on cancer biology have shown that overexpression of oncogenes (with or without functional loss of tumor suppressor genes), which is responsible for the progression of human malignancies via a multistep process, may be reduced by antisense technology. Caffeine enhances the effect of cisplatin (CDDP) chemotherapy on osteosarcoma cells. We constructed the recombinant adenovirus (Myc-AS) encoding the antisense c-myc fragment and investigated the synergic effect of caffeine and Myc-AS on the in vitro sensitivity of osteosarcoma MG-63 cells to cisplatin. Methods: The recombinant adenovirus (Myc-AS) encoding the antisense c-myc fragment was constructed by cloning c-myc cDNA of about 750 bp in a reverse direction into adenovirus vector, then undergoing recombination, amplification and complementation in vivo. Myc-AS and caffeine were used either alone or in combination with CDDP to treat osteosarcoma MG-63 cells in vitro. Western blot, MTT, flow cytometry (FCM) and electron microscopy were used to evaluate the expression of c-myc protein, tumor cell proliferation in vitro and apoptosis and to perform cell cycle analysis. Results: Myc-AS encoding antisense c-myc fragment was obtained with a titer of 2 x 10(9) pfu/ml. Myc-AS downregulated the expression of c-myc protein after transfecting MG-63 cells for 48 h, induced tumor cell apoptosis and inhibited tumor cell proliferation in vitro. Myc-AS or caffeine can enhance the cytotoxic effects of 2.0 and 5.0 mu g/ml CDDP on MG-63 cells. Moreover, the significantly enhancing effect of the Myc-AS-caffeine combination on CDDP chemotherapy of MG-63 cells was not restricted to apoptosis but also decreased tumor cell proliferation in vitro. Expression of the apoptosis-associated bcl-2 gene was downregulated and bax was upregulated, with no changes in E2F-1 expression. FCM analysis showed that CDDP treatment induced a block in S phase, and caffeine reversed this block and accelerated cell progression through the S phase. Conclusions: Myc-AS can induce obvious G2/M phase arrest in transfected cells. Abstract:Objective: To generate recombinant adenovirus that could simultaneously express ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (AdoMetDC) antisenses specifically in prostate cancer cells, and evaluate its inhibitory effect on prostate cancer in vivo. Methods: Fragments of ODC and AdoMetDC genes were generated by PCR, cloned into the pPGL-PSES, and then recombined with pAdEasy-1 vectors in AdEasy-1 cells. Ad-PSES-ODC-AdoMetDCas virus was produced in HEK293 cells. Following transfection with Ad-PSES-ODC-AdoMetDCas, the levels of ODC or AdoMetDC were determined by RT-PCR and western blot assays. The effect of Ad-PSES-ODC-AdoMetDCas treatment on tumor formation and growth was evaluated in xenograft models of prostate cancers in vivo. Results: The plasmid pAdEasy-PSES-ODC-AdoMetDCas was successfully constructed and the recombinant Ad-PSES-ODC-AdoMetDCas adenovirus was produced. Transfection with Ad-PSES-ODC-AdoMetDCas adenovirus significantly inhibited the expression of ODC and AdoMetDC genes specifically in prostate DU145 cells, but not H1299, HT29 and HepG2 cancer cells, and disrupted the ability of DU145 cells to form solid prostate cancer in vivo. Intratumoral treatment with Ad-PSES-ODC-AdoMetDCas adenovirus significantly inhibited the growth of engrafted prostate tumors in vivo. Conclusion: The recombinant Ad-PSES-ODC-AdoMetDCas adenovirus specifically reduces the expression of both ODC and AdoMetDC genes in prostate cells and may be used for treatment of prostate cancers at the clinic.

Abstract:To explore the function of ornithine decarboxylase in esophageal squamous cell carcinoma progression and test the effectiveness of anti-ornithine decarboxylase therapy for esophageal squamous cell carcinoma. In this study, we examined the expression pattern of ornithine decarboxylase in esophageal squamous cell carcinoma cell lines and tissues using immunohistochemistry and Western blot analysis. Then we investigated the function of ornithine decarboxylase in ESCC cells by using shRNA and an irreversible inhibitor of ornithine decarboxylase, difluoromethylornithine. To gather more supporting pre-clinical data, a human esophageal squamous cell carcinoma patient-derived xenograft mouse model (C.B-17 severe combined immunodeficient mice) was used to determine the antitumor effects of difluoromethylornithine in vivo. Our data showed that the expression of the ornithine decarboxylase protein is increased in esophageal squamous cell carcinoma tissues compared with esophagitis or normal adjacent tissues. Polyamine depletion by ODC shRNA not only arrests esophageal squamous cell carcinoma cells in the G2/M phase, but also induces apoptosis, which further suppresses esophageal squamous cell carcinoma cell tumorigenesis. Difluoromethylornithine treatment decreases proliferation and also induces apoptosis of esophageal squamous cell carcinoma cells and implanted tumors, resulting in significant reduction in the size and weight of tumors. The results of this study indicate that ornithine decarboxylase is a promising target for esophageal squamous cell carcinoma therapy and difluoromethylornithine warrants further study in clinical trials to test its effectiveness against esophageal squamous cell carcinoma.

Abstract:c-myc, an oncogene, plays an important role in regulation of cell proliferation, and has been found to be amplified and overexpressed in osteosarcoma. Moreover, it can promote cell transformation, and induce metastasis. This study was to construct recombinant adenovirus encoding antisense c-myc, and to investigate its effects on osteosarcoma cell lines MG-63 (p53-deficient) and U2OS (with wild type p53).Recombinant adenovirus Ad-As-c-myc encoding antisense c-myc was constructed by gene reconstruction technique, defined by polymerase chain reaction (PCR), and transfected into human osteosarcoma cell lines MG-63 and U2OS. Western blot, acridine orange staining, reverse transcription-PCR (RT-PCR), and flow cytometry (FCM) were used to detect expression of c-myc, proliferation, apoptosis, and cell cycle of MG-63 and U2OS cells.Ad-As-c-myc encoding antisense c-myc was obtained with the titer of 2x10(9) pfu/ml. Ad-As-c-myc significantly inhibited proliferation of both cell lines, while U2OS with wild type p53 was more susceptible to Ad-As-c-myc. Expression of c-myc mRNA was down-regulated in 2 cell lines 48 h after transfection of Ad-As-c-myc. Acridine orange staining and FCM analysis showed that Ad-As-c-myc induced apoptosis of both cell lines, cell cycle analysis showed obvious G(2)/M phase arrest in MG-63 cells,and G1 phase arrest in U2OS cells after transfection of Ad-As-c-myc.Ad-As-c-myc could induce apoptosis through both P53-dependent and P53-independent pathways, and inhibit proliferation of osteosarcoma cells.
Polyamine Depletion by ODC‐AdoMetDC Antisense Adenovirus Impairs Human Colorectal Cancer Growth and Invasion in Vitro and in Vivo

Abstract:Background Polyamine biosynthesis is controlled primarily by ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (AdoMetDC). Polyamine concentrations are elevated in colorectal cancer. Depletion of polyamine content in colorectal cancer by chemotherapy is related to tumor regression and impaired tumorigenicity. The current study evaluates the therapeutic effects of antisense ODC and AdoMetDC sequences on colorectal cancer in vitro and in vivo.Methods Antisense ODC and AdoMetDC sequences were cloned into an adenoviral vector (Ad-ODC-AdoMetDCas). The human colon cancer cell lines HT-29 and Caco-2 were infected with Ad-ODC-AdoMetDCas as well,, as with control vector. Viable cell counting, determination of polyamine concentrations, cell cycle analysis, and Matrigel invasion assays were performed in order to assess properties of tumor growth and invasiveness. Furthermore the antitumor effects of Ad-ODC-AdoMetDCas were also, evaluated in vivo in a nude mouse tumor model.Results Our study demonstrated that adenovirus-mediated ODC and AdoMetDC antisense expression inhibits tumor cell growth through a blockade of the polyamine synthesis pathway. This inhibitory effect cannot be reversed by the administration of putrescine. Tumor cells were arrested at the G, phase of the cell cycle after gene transfer and had reduced invasiveness. The adenovirus also induced tumor regression in established tumors in nude mice.Conclusions Our study suggests that Ad-ODC-AdoMetDCas has antitumor activity and therapeutic potential for the treatment of colorectal cancer. Abstract:Objective: To study the effects of different concentrations of antisense oligodeoxynucleotides (ASODN) on human pancreatic cancer cell line PaTu8988s. Method: Human pancreatic cancer cell line PaTu8988s in exponential growth stage was used to study the effect of different drug concentrations on the cell line in the presence of different concentrations (0 μg/ml, 5 μg/ml, 25 μg/ml, 50 μg/ml, 10 μg/m, 200 μg/ml) of ASODN and sense oligodeoxynucleotides (SODN). Cell counts and 3-[4,5-dimethylthiazolzyl]-2,5-diphenyl tetrazolium bromide (MTT) assays were carried out. Results: The inhibitory rate on the cell line PaTu8988s was 98.73%, 95.76%, 69.49%, 33.05% and 0 for ASODM concentrations of 200 μg/ml, 100 μg/ml, 50 μg/ml, 25 μg/ml and 5 μg/ml at 48 hours. Conclusions: K-ras complementary ASODN can inhibit the growth of human pancreatic cancer cell line PaTu8988s by 30.05% to 98.73%. This is likely to contribute to the specificity of the K-ras mRNA complementary capped ASODN sequential codon. Non-specific effect and side effect of ASOND were observed for instance, the greater the concentration is, the earlier the peak of inhibitory rate is reached. In concentration, of 25 μg/ml to 100 μg/ml ASODN showed a dose-effect correlation.

Abstract:C-myc is an oncogene with the important role of cell proliferation controller. It has been found to be amplified and overexpressed in osteosarcoma. Moreover, it can promote cell transformation and induce metastatic features. Some studies showed that overexpression of c-myc could induce resistance in response to antineoplastic agents. Currently, we constructed the recombinant adenovirus (Ad-Asc-myc) encoding antisense c-myc fragment and investigated its effect on the in vitro sensitivity of osteosarcoma MG-63 cells to cisplatin(CDDP). The osteosarcoma MG- 63 cells were transfected by the Ad-Asc-myc in vitro, and Western Blot, MTT assay, RT-PCR, flow cytometry (FCM), and transmission electron microscopy (TEM) were used to study expression of c-myc and caspase- 3 protein, tumor cell proliferation in vitro, cell apoptotic morphology and cell cycle change. Ad-Asc-myc encoding antisense c-myc fragment was obtained with the titer of 2.0 x 10(9) pfu/ml. Ad-Asc-myc downregulated the expression of c-myc protein after transfected MG-63 cells for 48 hours, combined with the treatment of 2.0, 5.0 mu g/ml cisplatin for 2 hours can inhibited tumor cells proliferation in vitro by 33.4 and 54.2 percent, respectively, which had significant difference compared with control recombinant adenovirus (Ad-LacZ) groups (P < 0.05). RT-PCR revealed that Ad-Asc-myc downregulated expression of bcl-2 and upregulated expression of Bax, and no appreciable changes were observed in the expression of E2F-1. Detection of caspase-3 protein TEM, and FCM analysis showed that Ad-Asc-myc could induce apoptosis of transfected cells, which was enhanced by the treatment of cisplatin. Cell cycle analysis showed that obvious G(2)/M phase arrested in transfected cells. In conclusion, Ad-Asc-myc increased the in vitro sensitivity of osteosarcoma MG-63 cells to cisplatin as well as induced apoptosis.

Abstract:To evaluated the effect of recombinant adenovirus Ad-ODC-AdoMetDCas which can simultaneously express both antisense ornithine decarboxylase (ODC) and Sadenosylmethionine decarboxylase (AdoMetDC) on cell cycle distribution in colorectal cancer cell and investigated underlying regulatory responses, human colorectal cancer cells HT-29 were cultured in RPMI 1640 medium and infected with Ad-ODC-AdoMetDCas. Cell cycle progression was detected by flow cytometry analysis. The expression levels of cell cycle regulated proteins were measured by Western blot analysis. The mRNA level of cyclin D1 was measured by RT-PCR. And a luciferase reporter plasmid of cyclin D1 promoter was constructed to observe the effect of Ad-ODC-AdoMetDCas on cyclin D1 promoter activity. The results showed that recombinant adenovirus Ad-ODC-AdoMetDCas significantly induced G1 arrest, decreased levels of cyclin D1 protein and mRNA and suppressed the promoter activity. Ad-ODC-AdoMetDCas also inhibited nuclear translocation of beta-catenin. In conclusion, downregulation of ODC and AdoMetDC mediated by Ad-ODC-AdoMetDCas transfection induces G(1) arrest in HT-29 cells and the arrest was associated with suppression of cyclin D1 expression and inhibition of beta-catenin nuclear translocation. As a new anticancer reagent, the recombinant adenovirus Ad-ODC-AdoMetDCas holds promising hope for the therapy of colorectal cancers.
Inhibitory Effect of EGFR Antisense Oligodeoxynucleotide on Human Hepatoma Cell Line.

Abstract:BACKGROUND:Hepatocellular carcinoma (HCC) is one of the most common cancer-related causes of death worldwide. The epidermal growth factor receptor (EGFR) is highly expressed in many human tumors and provides a new target for anticancer drug development. The aim of the present study was to explore the effect of EGFR antisense oligodeoxynucleotide on human HCC.METHODS:SMMC-7721 cells in culture were treated with 10 mumol/L antisense-odn for 24, 48, 72 hours respectively and MTT assay was adopted to determine the proliferation of tumor cells in vitro. About 2 x 10(6) SMMC-7721 cells with or without pretreatment(30 micromol/L oligodeoxynucleotide) were inoculated into subcutaneous flap of 21 nude mice, of which 7 were treated with EGFR antisense-odn, 7 with scrambled oligodeoxynucleotide (scrambled-odn), and 7 not treated in vivo.RESULTS:In vitro, after 24, 48, 72 hours the inhibitory rate of proliferation of SMMC-7721 cells treated with EGFR antisense-odn was 8%, 32%, and 34% respectively. In vivo after 8 weeks, no palpable tumor was found in 1/7 mice receiving cells pretreated with antisense-odn, whereas 7/7 untreated mice and 6/7 mice treated with scrambled-odn developed palpable tumors. Tumor growth in antisense-odn treated mice was significantly inhibited in comparison with that of those untreated (P < 0.01) or treated with scrambled-odn (P < 0.05).CONCLUSIONS:Antisense oligodeoxynucleotide acts as a specific growth inhibitor on SMMC-7721 in a sequence specific and time-dependent manner. EGFR antisense-odn can significantly inhibit the proliferation of human hepatoma cell in vitro as well as in vivo, indicating that EGFR may play an important role in the development of hepatoma and will be a new target for its treatment.

Abstract:In this study, we detected the expression of phosphylated ERK1/2 in human salivary adenoid cystic carcinoma (SACC) by immunohistochemical method, the effect of various concentrations of PD98059 on the growth of ACC-M cells (a cell line of human adenoid cystic carcinoma of the salivary glands) by MTT assay, and the effect of PD98059 on the lung metastasis of inoculated ACC-M cells in nude mice. The results showed that the phosphylated ERK1/2 was overexpressed in SACC, the ACC-M cell line proliferation was inhibited by PD98059, and the lung metastasis level was reduced by PD98059, indicating that ERK may be a key molecule in targeted therapy of adenoid cystic carcinoma.
