Fish Collagen Promotes Proliferation and Inhibits the Inflammation in Human Epidermal Keratinocytes in Vitro

Abstract:As a new collagen-based material, fish collagen has gradually captured people's attention in recent years. At present, fish collagen has been widely used in the fields of food, cosmetics and medicine, but regarding it research in the field of biomaterials, especially as an anti-inflammatory wound dressing, has not yet been carried out. This study elaborated on fish collagen and human epidermal keratinocytes (HaCaT cells). The effect of fish collagen on the proliferation of human epidermal keratinocytes was studied by employing the MTT (3-[4, 5-dimethylthiazol-2-yl]-2, 5 diphenyl tetrazolium bromide) assay. The effect of fish collagen on the secretion of inflammation factors Interleukin-6 (IL-6), Interleukin-8 (IL-8) and Tumor necrosis factor (TNF)-ą in human epidermal keratinocytes was studied using the Enzyme-Linked Immunosorbent Assay (ELISA); furthermore, the effect of fish collagen on the expression of inflammation-related genes IL-6, IL-8 and TNF-ą in human epidermal keratinocytes was studied by Quantitative Real-Time PCR (qRT-PCR). The results showed that fish collagen can promote the proliferation of HaCaT cells, inhibit the excessive secretion of LPS-stimulated inflammatory factors IL-6, IL-8 and TNF-ą, and also inhibit the expression of inflammation-related genes IL-6, IL-8 and TNF-ą. The above results indicate that fish collagen has the potential to act as an active additive for anti-inflammatory wound dressing.
