Abstract:3220A number of tumor suppressor genes that contribute to the pathogenesis of breast cancers can be inactivated by hypermethylation of CpG islands in their promoter regions, but little is known regarding the significance of the methylation profile for multiple genes. In colon cancer and leukemia, a CpG island methylator phenotype (CIMP) has been defined where multiple genes are coordinately inactivated, but this has not been previously reported in breast cancer. We have screened 9 known tumor suppressor genes (ARHI, RASSF1A, hMLH1, HIN-1, RIL, E-cadherin, p16, 14-3-3 sigma, RIZ1) in 6 breast cancer cell lines, 2 normal breast epithelial cell samples, and 37 pairs of breast cancers and adjacent normal breast tissues using a new technique known as Pyrosequencing Methylation Assays.
