Abstract:Increasing evidence indicates that major depressive disorder (MDD) is associated with cognitive as well as mood disturbances. To evaluate cognitive function and white matter structure, resting-state brain function in first-episode, treatmentnaive patients with MDD. To explore brain structure and function mechanisms of cognitive impairment in MDD. 46 Han Chinese MDD patients aged 18–45 year and 46 controls were assessed by a series of validated test procedures.Then, 30 patients and 30 controls were obtained by MRI scan.White matter abnormalities evaluated using diffusion tensor imaging (DTI) were analyzed using tract based spatial statistics (TBSS) and resting-state brain function was evaluated using regional homogeneity (ReHo) analysis. Cognitive impairment in patients with MDD was demonstrated by reduced accuracy in the Wisconsin Card Sorting test (WSCT) and to a lesser extent the Continuous Performance test (CPT) and Trail Making tests (TMT). White matter abnormalities found in the left cerebellum, and resting-state abnormalities present in the left inferior parietal gyrus, left anterior cingulate nucleus and left hippocampal gyrus were associated with impaired performance in the WSCT and CPT tests. We also showed that poor WSCT performance was associated with increased interconnectivity between the left ventral anterior cingulate nucleus and the medial frontal lobe areas. The present study indicates cognitive disturbances in patients with MDD are associated with white matter and resting-state changes and altered interconnections in specific brain areas.

Abstract:Background: Several neurobiological models of anxiety disorder posit a primary role for dysfunction of the amygdala and anterior cingulate cortex (ACC). This study tests the hypothesis that patients with generalized anxiety disorder (GAD) have abnormal white matter microstructure in the amygdala and ACC, as inferred from diffusion tensor imaging, compared with healthy controls.Methods: Subjects were 16 right-handed, first-episode, treatment-naive GAD patients without comorbid disorders and 26 matched, healthy comparison controls. All subjects underwent diffusion tensor imaging and structural magnetic resonance imaging brain scanning. Fractional anisotropy (FA), a robust intravoxel measure of water self-diffusion, was compared between groups on a voxel-by-voxel basis. Associations between clinical ratings of symptom severity (i.e., the Hamilton Anxiety Scale and the Penn State Worry Questionnaire) and FA were assessed.Results: Compared with healthy volunteers, patients demonstrated significantly higher FA in the right amygdala white matter and lower FA in the caudal ACC/mid-cingulate cortex white matter. Higher right amygdala FA correlated significantly with higher Hamilton Anxiety Scale scores and higher Penn State Worry Questionnaire scores.Limitations: The sample size was modest and may contribute to false positive effects.Conclusions: These findings provide the first evidence of an abnormality in white matter microstructure that involves the amygdala and the cingulate cortex in the pathogenesis of GAD, and are consistent with neurobiological models that posit a defect in emotion-related brain regions. Aberrant morphological brain networks in first-episode, treatment-naive adolescents with major depressive disorder

Abstract:Abstract Previous studies have shown that major depressive disorder (MDD) is associated with disrupted topological organizations of large-scale brain networks. However, the disruptions and their clinical and cognitive relevance are not well established for morphological brain networks in adolescent MDD. In this study, twenty-five first-episode, treatment-naive adolescents with MDD and nineteen healthy controls underwent T1-weighted MRI and a battery of neuropsychological tests. Individual morphological brain networks were constructed separately based on 4 morphological features, whose topological organizations were quantified by graph-based approaches. Permutation testing and partial correlation were used to examine between-group differences and clinical and cognitive relevance of the differences, respectively. Finally, support vector machine was used to classify the patients from controls. Compared with the controls, the patients exhibited topological alterations mainly in cortical thickness-based networks characterized by higher nodal centralities in parietal (left PriMary Sensory Cortex) but lower centralities in temporal (left ParaBelt Complex, right Perirhinal Ectorhinal Cortex, right Area PHT and right Ventral Visual Complex) regions. Moreover, decreased nodal centralities of some temporal regions were correlated with cognitive dysfunction and course of illness of the patients. These results were largely reproducible for binary and weighted network analyses. Finally, topological properties of the cortical thickness-based networks were able to distinguish the patients from controls with 87.6% accuracy. In short, adolescent MDD is associated with abnormal local organizations of morphological brain networks, which provide potential biomarkers for diagnosing and monitoring the disease.

Abstract:Objective The aim of this study was to explore the microstruetural integrity of whole brain white matter with diffusion tensor imaging(DTI)in patients with major depressive disorder(MDD). Methods Fourteen first-episode,medication-nave young adult patients with MDD and 14 matched healthy controls received DTI scan,then were analyzed by voxel-based analysis.Results The patients showed significantly lower fractional anisotropy(FA)values than healthy controls in the white matter of the fight middle frontal gyrus(x=36,y=49,z=10;t=4.89,cluster=258),left lateral occipitotemporal gyrus (x=-42,y=-56,z=-1;t=5.00,cluster=96),and the angular gyrus(x=24,y=-47,z=41; t=5.23,cluster=120)and sub-gyral(x=42,y=-65,z=27;t=4.56,cluster=76)in right parietal lobe(P0.001).There were no significant correlations between the FA value in any brain area and depressive severity as well as illness course.Furthermore,compared with healthy controls,no regions with significantly higher FA values were found in patients with MDD.Conclusion These findings suggest that abnormalities of brain white matter may be actually present early in the course of MDD and support that white matter lesions which disrupt the neural circuits may be involved in mood regulation.
Alterations of white matter integrity in adults with major depressive disorder: a magnetic resonance imaging study.

Abstract:Objective: The purpose of our study was to investigate alterations of white matter integrity in adults with major depressive disorder (MDD) using magnetic resonance imaging (MRI). Methods: We performed diffusion tensor imaging with a 3T MRI scanner on 45 patients with major depression and 45 healthy controls matched for age, sex and education. Using a voxel-based analysis, we measured the fractional anisotropy (FA), and we investigated the differences between the patient and control groups. We examined the correlations between the microstructure abnormalities of white matter and symptom severity, age of illness onset and cumulative illness duration, respectively. Results: We found a significant decrease in FA in the left hemisphere, including the anterior limb of the internal capsule and the inferior parietal portion of the superior longitudinal fasciculus, in patients with MDD compared with healthy controls. Diffusion tensor imaging measures in the left anterior limb of the internal capsule were negatively related to the severity of depressive symptoms, even after we controlled for age and sex. Conclusion: Our findings provide new evidence of microstructural changes of white matter in non-late-onset adult depression. Our results complement those observed in late-life depression and support the hypothesis that the disruption of cortical-subcortical circuit integrity may be involved in the etiology of major depressive disorder.

Abstract:Abnormalities of the white matter (WM) tracts integrity in brain areas involved in emotional regulation have been postulated in major depressive disorder (MDD). However, there is no diffusion tensor imaging (DTI) study in patients with treatment-responsive MDD at present. DTI scans were performed on 22 patients with treatment-responsive MDD and 19 well-matched healthy subjects. Tract-based spatial statistics (TBSS) approach was employed to analyze the scans. Voxel-wise statistics revealed four brain WM tracts with lower fractional anisotropy (FA) in patients compared to healthy subjects: the bilateral internal capsule, the genu of corpus callosum, the bilateral anterior corona radiata, and the right external capsule. FA values were nowhere higher in patients compared to healthy subjects. Our findings demonstrate that the abnormalities of the WM tracts, major in the projection fibers and corpus callosum, may contribute to the pathogenesis of treatment-responsive MDD.
Two Patterns of White Matter Abnormalities in Medication-Naive Patients With First-Episode Schizophrenia Revealed by Diffusion Tensor Imaging and Cluster Analysis.

Abstract:IMPORTANCE Accumulating evidence supports the hypothesis that cerebral white matter abnormalities are involved in the pathophysiology of schizophrenia; however, findings from in vivo neuroimaging studies have been inconsistent. Besides confounding factors, including age, illness duration, and medication effects, an additional cause for the inconsistent results may be heterogeneity in the nature of white matter alterations associated with the disorder. OBJECTIVE To investigate whether different patterns of white matter abnormalities exist in a large cohort of medication-naive patients with first-episode schizophrenia and the relationship between such patterns and clinical parameters. DESIGN, SETTING, AND PARTICIPANTS A cross-sectional diffusion tensor imaging study of 113 medication-naive patients with first-episode schizophrenia and 110 demographically matched healthy control individuals. The study was conducted in the mental health center of West China Hospital, Sichuan University, Chengdu, China, from January 2006 to June 2014. MAIN OUTCOMES AND MEASURES The patterns of white matter abnormalities revealed by tract-specific analysis in conjunction with hierarchical clustering. RESULTS With diffusion features extracted from 18 fiber tracts, cluster analysis revealed 2 patterns of abnormalities. One pattern (42.5% of patient sample) showed widespread white matter abnormalities compared with matched healthy control individuals, while another pattern (57.5% of patient sample) only showed circumscribed regional white matter abnormalities, mainly in the left superior longitudinal fasciculus. Patients in these subgroups did not differ in demographic features; however, negative symptoms were more severe in patients with widespread white matter abnormalities. CONCLUSIONS AND RELEVANCE Two distinct patterns of white matter abnormalities exist at the early phase of schizophrenia, with those having global abnormalities experiencing more severe negative symptoms. The finding that distinct subgroups of patients with schizophrenia have different forms of white matter pathology may reflect qualitatively distinct genetic influences or neurodevelopmental alterations and thus represents a promising strategy for resolving neurobiological heterogeneity in the schizophrenia syndrome.
Association between segmental alterations of white matter bundles and cognitive performance in first-episode, treatment-naïve young adults with major depressive disorder

Abstract:Background: Cumulative evidence has consistently shown that white matter (WM) disruption is associated with cognitive decline in geriatric depression. However, limited research has been conducted on the correlation between these lesions and cognitive performance in untreated young adults with major depressive disorder (MDD), particularly with the specific segmental alterations of the fibers. Method: Diffusion tensor images were performed on 60 first-episode, treatment-naïve young adult patients with MDD and 54 matched healthy controls (HCs). Automated fiber quantification was applied to calculate the tract profiles of fractional anisotropy (FA), mean diffusivity (MD), axial diffusivity (AD), and radial diffusivity (RD) to evaluate the WM microstructural organization. Correlation analysis was performed to find the associations between the diffusion properties and cognitive performance. Results: Compared with HCs, patients with MDD exhibited predominantly different diffusion properties in bilateral uncinate fasciculus (UF), corticospinal tracts (CSTs), left superior longitudinal fasciculus and anterior thalamic radiation. The FA of the temporal cortex portion of right UF was positively correlated with working memory. The MD of the temporal component of left UF was negatively correlated with working memory and positively correlated with symptom severity. Additionally, a positive correlation between the MD of left CST and the psychomotor speed, negative correlation between the MD of left CST and the executive functions and complex attentional processes were observed. Conclusions: Our study validated the alterations in spatial localization of WM microstructure and its correlations with cognitive performance in first-episode, treatment-naïve young adults with MDD. This study added to the knowledge of the neuropathological basis of MDD.

Abstract:Background: Almost half of patients with major depressive disorder (MDD) also have clinically meaningful levels of anxiety. Anxious depression is a distinct clinical subtype of MDD, which has poor response to pharmacotherapy; however, the neural mechanisms behind are largely unknown. In the present study, we explored the white matter (WM) integrity traits of anxious depression in first-episode and medication-free (medication-naive and medication washout) Chinese young adult patients by detecting differences in diffusion tensor imaging (DTI) with the tract-based spatial statistics (TBSS) method. Subjects and methods: DTI was obtained from 39 first-episode, medication-free anxious depressive patients, 45 nonanxious depressive patients, and 50 demographically similar healthy controls. All subjects underwent clinical assessments. TBSS was carried out to investigate the difference in WM integrity among three groups within DTI parameter maps. WM integrity was measured using fractional anisotropy (FA), mean diffusivity, axial diffusivity, and radial diffusivity (RD). The correlations between WM integrity and clinical features were also computed. Results: When compared with nonanxious patients, lower FA values in anxious depressive patients were found in multiple regions of the brain, mainly involving left uncinate fasciculus (UF), superior longitudinal fasciculus (SLF), and forceps major and minor. Higher RD in forceps major and minor and SLF were also detected. The decreased FA values and increased RD values correlated with both anxiety level and depression level in the pooled depressive group. Conclusion: The anxious depressive patients had more abnormalities in WM integrity at the early phase than the nonanxious group. Alternations in WM integrity in fiber pathways, including SLF, UF, and forceps major and minor, may play a critical role in the neuropathology of anxious depression and might help to identify anxious MDD from nonanxious MDD. Further study with larger sample size, larger age range, and longitudinal design is needed to confer a robust inference to better understand the dynamic neurological change and neuropathology of WM integrity in anxious MDD.

Abstract:Background: Previous structural imaging studies found evidence of brain morphometric changes in major depression (MD) patients, but they rarely excluded compounding effects of some important factors, such as medication and brain degeneration. This study sought to explore the brain morphometric changes of first-episode, drug-naive, non-late-life adult MD patients with optimized voxel-based morphometry (VBM) method. Methods: Twenty-three first-episode, drug-naive, non-late-life adult depressed patients and 23 healthy control subjects were enrolled in this study. Subjects underwent high-resolution magnetic resonance imaging, and optimized VBM was performed to analyze the morphometric data. A partial correlation model was used to analyze associations of morphometric changes with Hamilton Depression Rating Scale scores and illness duration. Results: Depressed patients showed significant gray matter volume reduction in the bilateral limbic system, especially in hippocampus. These changes did not significantly correlate with symptom severity or illness duration. Conclusions: Our findings provided new evidence of gray matter deficits in first-episode, drug-naive, non-late-life adult MD patients. It supported that the reduction of hippocampal volume is a trait for MD patients and further highlighted the important role of the limbic system, particularly hippocampus in the pathophysiology of MD.
Disrupted Intrinsic Functional Connectivity of the Cognitive Control Network Underlies Disease Severity and Executive Dysfunction in First-Episode, Treatment-Naive Adolescent Depression.

Abstract:Background: Previous neuroimaging studies have showed that imbalanced functional integration of distributed large-scale brain networks is associated with pathophysiological characteristics of major depressive disorder (MDD). However, the association between network integrative disturbances and clinical features and cognitive functions remains largely unclear in adolescent MDD. This study investigated the neural correlates of abnormal functional connectivity networks with clinical and cognitive characteristics in adolescent MDD. Methods: Twenty-eight first-episode, treatment-naive adolescents with MDD and 24 well-matched healthy controls (HCs) underwent functional magnetic resonance imaging (fMRI) and a battery of cognitive tests. A seed-based functional connectivity (FC) approach was used to depict connectivity patterns of the cognitive control network (CCN), affective network (AN) and default mode network (DMN), whose between-group differences were correlated with clinical variables and cognitive functions in the patients. Results: Compared with the HCs, the MDD patients exhibited impaired executive functions. The FC analysis revealed lower CCN FC with the temporal, parietal and frontal regions and the limbic system, higher AN FC with the temporal and occipital regions and the cerebellum, and lower DMN FC with the cerebellum and insula. Interestingly, the decreased CCN FC was related to disease severity (with the inferior frontal gyrus) and executive dysfunctions (with the middle cingulate gyrus and supramarginal gyrus) in the patients. Limitations: The main limitations were the relatively small sample size and suboptimal imaging parameters. Conclusion: Functional alteration of CCN during the developmentally sensitive period may be important in the neurobiology of adolescent MDD.
Abnormal voxel-wise whole-brain functional connectivity in first-episode, drug-naïve adolescents with major depression disorder

Abstract:Major depression disorder (MDD) is one of the most common psychiatric disorders. Previous studies have demonstrated structural and functional abnormalities in adult depression. However, the neurobiology of adolescent depression has not been fully understood. The aim of this study was to investigate the intrinsic dysconnectivity pattern of voxel-level whole-brain functional networks in first-episode, drug-naïve adolescents with MDD. Resting-state functional magnetic resonance imaging data were acquired from 66 depressed adolescents and 47 matched healthy controls. Voxel-wise degree centrality (DC) analysis was performed to identify voxels that showed altered whole-brain functional connectivity (FC) with other voxels. We further conducted seed-based FC analysis to investigate in more detail the connectivity patterns of the identified DC changes. The relationship between altered DC and clinical variables in depressed adolescents was also analyzed. Compared with controls, depressed adolescents showed lower DC in the bilateral hippocampus, left superior temporal gyrus and right insula. Seed-based analysis revealed that depressed adolescents, relative to controls, showed hypoconnectivity between the hippocampus to the medial prefrontal regions and right precuneus. Furthermore, the DC values in the bilateral hippocampus were correlated with the Hamilton Depression Rating Scale score and duration of disease (all P < 0.05, false discovery rate corrected). Our study indicates abnormal intrinsic dysconnectivity patterns of whole-brain functional networks in drug-naïve, first-episode adolescents with MDD, and abnormal DC in the hippocampus may affect the association of prefrontal-hippocampus circuit. These findings may provide new insights into the pathophysiology of adolescent-onset MDD.

White Matter Abnormalities and Changes in First-onset Major Depressive Disorder by Antidepressant Treatment Investigated with Voxel-based Diffusion Tensor Imaging

Abstract:Purpose:To detect white matter abnormalities in patients with first-onset major depressive disorder(MDD) and identify changes caused by antidepressant treatment.Methods:Thirteen patients with first onset MDD and 14 normal age-matched volunteers underwent diffusion tensor imaging(DTI) and all the patients underwent scanning twice during an 8-week period.Fractional anisotropy(FA) maps were processed using SPM2 to make voxel-wise comparison of anisotropy in whole brain between the two groups.Results: Significant reductions in anisotropy were found in the white matter of genu of corpus callosum,genu of the right internal capsule and right inferior longitudinal fasciculus in patients(P<0.001 corrected).Symptomatic improvement after antidepressant was associated with reversion of the FA reductive areas(P<0.001 corrected).Conclusion:Changes in FA maps of genu of corpus callosum,genu of the right internal capsule and right inferior longitudinal fasciculus associated with symptomatic improvement indicate that DTI may be a useful surrogate marker of antidepressant treatment response.

Abstract:BACKGROUND: Resting-state fMRI is a novel approach to measure spontaneous brain activity in patients with major depressive disorder (MDD). Although most resting-state fMRI studies have focused on the examination of temporal correlations between low-frequency oscillations (LFOs), few studies have explored the amplitude of these LFOs in MDD. In this study, we applied the approaches of amplitude of low-frequency fluctuation (ALFF) and fractional ALFF to examine the amplitude of LFOs in MDD. METHODOLOGY/PRINCIPAL FINDINGS: A total of 36 subjects, 18 first-episode, treatment-naive patients with MDD matched with 18 healthy controls (HCs) completed the fMRI scans. Compared with HCs, MDD patients showed increased ALFF in the right fusiform gyrus and the right anterior and posterior lobes of the cerebellum but decreased ALFF in the left inferior temporal gyrus, bilateral inferior parietal lobule, and right lingual gyrus. The fALFF in patients was significantly increased in the right precentral gyrus, right inferior temporal gyrus, bilateral fusiform gyrus, and bilateral anterior and posterior lobes of the cerebellum but was decreased in the left dorsolateral prefrontal cortex, bilateral medial orbitofrontal cortex, bilateral middle temporal gyrus, left inferior temporal gyrus, and right inferior parietal lobule. After taking gray matter (GM) volume as a covariate, the results still remained. CONCLUSIONS/SIGNIFICANCE: These findings indicate that MDD patients have altered LFO amplitude in a number of regions distributed over the frontal, temporal, parietal, and occipital cortices and the cerebellum. These aberrant regions may be related to the disturbances of multiple emotion- and cognition-related networks observed in MDD and the apparent heterogeneity in depressive symptom domains. Such brain functional alteration of MDD may contribute to further understanding of MDD-related network imbalances demonstrated in previous fMRI studies.

Abstract:Identifying data-driven biotypes of major depressive disorder (MDD) has promise for the clarification of diagnostic heterogeneity. However, few studies have focused on white-matter abnormalities for MDD subtyping. This study included 116 patients with MDD and 118 demographically-matched healthy controls assessed by diffusion tensor imaging and neurocognitive evaluation. Hierarchical clustering was applied to the major fiber tracts, in conjunction with tract-based spatial statistics, to reveal white-matter alterations associated with MDD. Clinical and neurocognitive differences were compared between identified subgroups and healthy controls. With fractional anisotropy extracted from 20 fiber tracts, cluster analysis revealed 3 subgroups based on the patterns of abnormalities. Patients in each subgroup versus healthy controls showed a stepwise pattern of white-matter alterations as follows: subgroup 1 (25.9% of patient sample), widespread white-matter disruption; subgroup 2 (43.1% of patient sample), intermediate and more localized abnormalities in aspects of the corpus callosum and left cingulate; and subgroup 3 (31.0% of patient sample), possible mild alterations, but no statistically significant tract disruption after controlling for family-wise error. The neurocognitive impairment in each subgroup accompanied the white-matter alterations: subgroup 1, deficits in sustained attention and delayed memory; subgroup 2, dysfunction in delayed memory; and subgroup 3, no significant deficits. Three subtypes of white-matter abnormality exist in individuals with major depression, those having widespread abnormalities suffering more neurocognitive impairments, which may provide evidence for parsing the heterogeneity of the disorder and help optimize type-specific treatment approaches.

Abstract:Puberty is a vulnerable period for the onset of major depressive disorder (MDD) due to considerable neurodevelopmental changes. Prior diffusion tensor imaging (DTI) studies in depressed youth have had heterogeneous participants, making assessment of early pathology challenging due to illness chronicity and medication confounds. This study leveraged whole-brain DTI and graph theory approaches to probe white matter (WM) abnormalities and disturbances in structural network topology related to first-episode, treatment-naïve pediatric MDD. Participants included 36 first-episode, unmedicated adolescents with MDD (mean age 15.8 years) and 29 age- and sex-matched healthy controls (mean age 15.2 years). Compared to controls, the MDD group showed reduced fractional anisotropy in the internal and external capsules, unveiling novel regions of WM disruption in early-onset depression. The right thalamus and superior temporal gyrus were identified as network hubs where betweenness centrality changes mediated links between WM anomalies and depression severity. A diagnostic model incorporating demographics, DTI, and network metrics achieved an AUROC of 0.88 and a F1 score of 0.80 using a neural network algorithm. By examining first-episode, treatment-naïve patients, this work identified novel WM abnormalities and a potential causal pathway linking WM damage to symptom severity via regional structural network alterations in brain hubs.

Abstract:Previous structural imaging studies have found evidence of brain morphometric changes in patients with major depressive disorder (MDD), but these studies rarely excluded compounding effects of certain important factors, such as medications and long duration of illnesses. Furthermore, the neurobiological mechanism of the macroscopic findings of structural alterations in MDD patients remains unclear. In this study, we utilized magnetization transfer imaging, a quantitative measure of the macromolecular structural integrity of brain tissue, to identify biophysical alterations, which are represented by a magnetization transfer ratio (MTR), in MDD patients. To ascertain whether MTR changes occur independent of volume loss, we also conduct voxel-based morphometry (VBM) analysis. The participants included 27 first-episode, drug-naive MDD patients and 28 healthy controls matched for age and gender. Whole-brain voxel-based analysis was used to compare MTR and gray matter volume across groups and to analyse correlations between MTR and age, symptom severity, and illness duration. The patients exhibited significantly lower MTR in the left superior parietal lobule and left middle occipital gyrus compared with healthy controls, which may be related to the attentional and cognitive dysfunction in MDD patients. The VBM analysis revealed significantly increased gray matter volume in right postcentral gyrus in MDD patients. These findings in first-episode, drug-naive MDD patients may reflect microstructural gray matter changes in the parietal and occipital cortices close to illness onset that existed before volume loss, and thus potentially provide important new insight into the early neurobiology of depression.

Abstract:In current time, the crowd of depression has increased rapidly across colleges and universities in China. They often suffer anhedonia, social failure, drug abuse and so on. The recent reports points out that the brain reward system showed damaged in patients with depression, so the identification of the dysfunction in the brain reward system is really crucial. By analyzing brain magnetic resonance imaging (MRI) data, this article aimed to identify the gray matter volume (GMV) abnormalities in brain reward system between first-episode medication-naive patients with major depressive disorder (MDD) and healthy controls (HCs). 14 medication-naive participants with MDD aged 19-24 years (6 males, 8 females) and 18 healthy controls aged 19-24 years (9 males, 9 females) were recruited. We used voxel-based morphometry (VBM) to analyze brain imaging data. Then, two sample t-test was applied to detect GM abnormalities in MDD compared to HCs. This study found that MDD showed increased gray matter volume (GMV) in putamen, precuneus and amygdala in right hemisphere compared to HCs. Furthermore, MDD showed decreased GMV in left rectus, right orbital medial prefrontal cortex (omPFC), left superior temporal gyrus (STG) and left insula compared to HCs. However, no significant changes were found in caudate nucleus. These experimental results show that the depression disorder causes extensive damage in reward system and subcortical brain regions, and the alterations in the rectus, the omPFC, STG, precuneus and amygdala, may be characters of MDD in first episode.

Abstract:BackgroundResting-state functional magnetic resonance imaging has been widely used for the assessment of brain functional network, yet with inconsistent results. The present study aimed to investigate intranetwork and internetwork connectivity differences between patients with major depressive disorder (MDD) and healthy controls at the integrity, network and edge levels of 8 well-defined resting state networks.MethodsThirty patients with MDD and sixty-three healthy control subjects were recruited in this study.ResultsCompared with healthy controls, patients with MDD showed increased node degree in the right amygdala and putamen, increased connectivity strength in the deep gray matter network (DGN) and increased functional connectivity in intranetwork and internetwork. Meanwhile, MDD showed decreased connectivity strength in visual network-DGN pair.LimitationsThe sample size was small, and all patients in this study were of Asian ethnicity, especially Han individuals.ConclusionsThese findings demonstrate that MDD cases and healthy controls may have divergent intranetwork and internetwork connectivity at an early stage without confounding influence of medication. These differences may underlie cognitive and behavioral alterations in patients with MDD. And these differences may help with the discrimination of patients and healthy people at an early stage of MDD. More studies in the future are warranted to assist in the diagnosis of this burdensome disease.

Abstract:The white matter differences between the first episode major depression and the recurrent major depression in, Chinese young and middle-aged populations were explored using three-dimensional structural Magnetic Resonance Imaging (3D) and Diffusion tensor imaging (DTI) technologies. Compared with the first episode major depressive patients, the recurrent depressive patients show heavy reduction in fractional anisotropy (FA) values. However, there are no significant differences in the while matter concentration (WMC). It demonstrates that the recurrent major depressive patients have more severe abnormalities in while matter. It's consistent with the clinical phenomena that the prognosis would be worse after a long duration and multi attack times.
