Abstract:Background: Abnormalities of the mevalonate pathway, an important cellular metabolic pathway, are common in many diseases including cardiovascular disease. The mevalonate pathway related enzyme expressions in pressure overload-induced cardiac hypertrophy and associated diastolic dysfunction remains largely unknown. This study aims to investigate whether the expression of mevalonate pathway related enzyme is altered during the progression of cardiac hypertrophy and associated diastolic dysfunction induced by pressure overload. Methods: Male Sprague-Dawley (SD) rats were randomly divided into two groups: the suprarenal abdominal aortic coarctation (AAC) group and the sham group. Results: Histological and echocardiographic assessments showed that there was a significant cardiovascular remodeling in the AAC group compared with the sham group after 3 weeks post-operatively, and the left ventricular (LV) diastolic function was reduced at 8 and 14 weeks post-operatively in the AAC group, without any change in systolic function during the study. The tissue of the heart and the abdominal aorta proximal to the coarctation showed over-expression of several enzymes, including 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR), farnesyl diphosphate synthase (FDPS), farnesyltransferase-alpha (FNTA), farnesyltransferase-beta (FNTB), geranylgeranyltransferase type I (GGTase-I) and the activation of their downstream proteins was enhanced. Conclusions: AAC induced compensatory LV hypertrophy to decompensatory diastolic dysfunction, accompanied by altered expression of several key enzymes in the mevalonate pathway. Copyright (C) 2013 S. Quantitative Proteomic Study of Myocardial Mitochondria in Urea Transporter B Knockout Mice.

Abstract:In previous research, we showed that 16‐week‐old urea transporter B (UT‐B) null mice have an atrial–ventricular conduction block, and hypothesized myocardial mitochondrial dysfunction. To investigate the mechanism of this block, we examined the proteomic differences in the myocardial mitochondria of UT‐B null and wild‐type mice with nanoscale LC–MS/MS. Of 26 proteins clearly downregulated in the UT‐B null mice, 15 are involved in complexes I, III, IV, and V of the respiratory chain, which would strongly reduce the activity of the electron transport chain. Excess electrons from complexes I and III pass directly to O2 to generate ROS and deplete ROS‐scavenging enzymes. Myocardial intracellular ROS were significantly higher in UT‐B null mice than in wild‐type mice (p < 0.01), constituting an important cause of oxidative stress injury in the myocardia of UT‐B null mice. The mitochondrial membrane potential (ΔΨm) was also lower in UT‐B null mice than in wild‐type mice (p < 0.05), causing oxidative phosphorylation dysfunction of complex V and insufficient ATP in the myocardial cells of UT‐B null mice. HADHA (a trifunctional protein) and HSP60 were also downregulated in the UT‐B null myocardial mitochondria. These results confirm that mitochondrial dysfunction underlies the pathogenesis of the atrial–ventricular conduction block in UT‐B null mice.
LncRNA TUG1 Alleviates Cardiac Hypertrophy by Targeting Mir‐34a/dkk1/wnt‐β‐catenin Signalling

Abstract:The current study was designed to explore the role and underlying mechanism of lncRNA taurine up‐regulated gene 1 (TUG1) in cardiac hypertrophy. Mice were treated by transverse aortic constriction (TAC) surgery to induce cardiac hypertrophy, and cardiomyocytes were treated by phenylephrine (PE) to induce hypertrophic phenotype. Haematoxylin‐eosin (HE), wheat germ agglutinin (WGA) and immunofluorescence (IF) were used to examine morphological alterations. Real‐time PCR, Western blots and IF staining were used to detect the expression of RNAs and proteins. Luciferase assay and RNA pull‐down assay were used to verify the interaction. It is revealed that TUG1 was up‐regulated in the hearts of mice treated by TAC surgery and in PE‐induced cardiomyocytes. Functionally, overexpression of TUG1 alleviated cardiac hypertrophy both in vivo and in vitro. Mechanically, TUG1 sponged and sequestered miR‐34a to increase the Dickkopf 1 (DKK1) level, which eventually inhibited the activation of Wnt/β‐catenin signalling. In conclusion, the current study reported the protective role and regulatory mechanism of TUG1 in cardiac hypertrophy and suggested that TUG1 may serve as a novel molecular target for treating cardiac hypertrophy.
UT‐B Deficient Mice Develop Cardiac Hypertrophy and Cardiac Dysfunction Associated with Aging

Abstract:Urea transporter B (UT‐B) is a urea transporter protein expressed in kidney, heart, testis, etc. To determine whether UT‐B could play an important role in cardiac function, we evaluated the morphological and functional difference between wild‐type mice and UT‐B null mice by histological analysis, electrocardiogram and left ventricular (LV) hemodynamics measurement. RT‐PCR and immunofluorescence showed UT‐B expression in cardiomyocytes in wild‐type mice and no UT‐B expression in UT‐B null mice. Urea concentration in heart tissue and serum were significantly higher in UT‐B null mice than that in wild‐type mice , and became much higher with aging. Aging UT‐B null mice had significant cardiac hypertrophy with severe interstitium fibrosis compared to age‐matched wild‐type mice. M‐model echocardiographic images of hearts showed increased AWd, AWs, PWd and PWs in 52‐weeks‐old UT‐B null mice compared with age‐matched wild‐type mice. But LVDd and LVDs in UT‐B null mice did not differ from those of wild‐type mice (P > 0.05). E/A ratio was decreased and LVEDP was increased in UT‐B null mice. Our results provide the first evidence that UT‐B deletion causes accumulation of urea in heart tissue, cardiac hypertrophy and progressive cardiac dysfunction in mice, which suggests that UT‐B play an important role in cardiac function. Abstract:The urea transporter UT-B is expressed in multiple tissues including erythrocytes, kidney, brain, heart, liver, colon, bone marrow, spleen, lung, skeletal muscle, bladder, prostate, and testis in mammals. Phenotype analysis of UT-B-null mice has confirmed that UT-B deletion results in a urea-selective urine-concentrating defect (see Chap. 9 ). The functional significance of UT-B in extrarenal tissues studied in the UT-B-null mouse is discussed in this chapter. UT-B-null mice present depression-like behavior with urea accumulation and nitric oxide reduction in the hippocampus. UT-B deletion causes a cardiac conduction defect, and TNNT2 and ANP expression changes in the aged UT-B-null heart. UT-B also plays a very important role in protecting bladder urothelium from DNA damage and apoptosis by regulating the urea concentration in urothelial cells. UT-B functional deficiency results in urea accumulation in the testis and early maturation of the male reproductive system. These results show that UT-B is an indispensable transporter involved in maintaining physiological functions in different tissues.
UGCG modulates heart hypertrophy through B4GalT5-mediated mitochondrial oxidative stress and the ERK signaling pathway

Abstract:Mechanical pressure overload and other stimuli often contribute to heart hypertrophy, a significant factor in the induction of heart failure. The UDP-glucose ceramide glycosyltransferase (UGCG) enzyme plays a crucial role in the metabolism of sphingolipids through the production of glucosylceramide. However, its role in heart hypertrophy remains unknown. In this study, UGCG was induced in response to pressure overload in vivo and phenylephrine stimulation in vitro. Additionally, UGCG downregulation ameliorated cardiomyocyte hypertrophy, improved cardiomyocyte mitochondrial oxidative stress, and reduced the ERK signaling pathway. Conversely, UGCG overexpression in cardiomyocytes promoted heart hypertrophy development, aggravated mitochondrial oxidative stress, and stimulated ERK signaling. Furthermore, the interaction between beta-1,4-galactosyltransferase 5 (B4GalT5), which catalyses the synthesis of lactosylceramide, and UGCG was identified, which also functions as a synergistic molecule of UGCG. Notably, limiting the expression of B4GalT5 impaired the capacity of UGCG to promote myocardial hypertrophy, suggesting that B4GalT5 acts as an intermediary for UGCG. Overall, this study highlights the potential of UGCG as a modulator of heart hypertrophy, rendering it a potential target for combating heart hypertrophy.

Progressive Heart Block and Ventricular Hypertrophy in Mice Lacking Urea Transporter Ut-B

Abstract:Urea transporters B(UT‐B) is membrane proteins that facilitate urea movement across biological membranes, expressed in kidney, erythrocytes, heart, etc. The aim of this study was to determine the phenotype of UT‐B deletion in heart. UT‐B expression in heart was studied by RT‐PCR and Western blot. Surface electrocardiogram recording (lead II) was measured in wildtype mice and UT‐B null mice. Cardiac function was evaluated by echocardiography, left ventricle internal diameters and wall thicknesses were measured at end systole and end diastole (LVPW(d) and LVPW(s)). RT‐PCR and Western blot analysis showed that UT‐B was expressed in heart in wild‐type mice. No UT‐B transcript and protein was found in UT‐B null mice. ECG recording showed that P‐R interval was significantly delayed in UT‐B null mice vs wildtype mice (p < 0.05). Atrial ventricular heart block type and only appeared in 52 weeks old UT‐B null mice. Echocardiography revealed that the LVPW(d) and LVPW(s) in 52 weeks old UT‐B null mice (1.1128 ± 0.0981 and 1.6157 ± 0.1322 mm) were greatly thicker than age matched wild‐type mice (0.8914 ± 0.1433 and 1.2500 ± 0.1786, P < 0.05 respectively). These results provide the first evidence that UT‐B deletion cause progressive heart block in mice and ventricular hypertrophy is involved in the pathogenesis of heart block. *This study was supported by grant 30670852 from the Natural Science Foundation of China.

Abstract:AIM: To investigate alteration of functional receptors for urotensin II in pressure overload-induced cardiac hypertrophy of rats. METHODS: In the rat cardiac hypertrophy model produced by constriction of the abdominal aorta, urotensin II binding sites in myocardial sarcolemma were determined with radioligand assay first day (early group) and 30th day (late group) after operation. RESULTS: Compared with control and sham group, in the early group，the blood pressure increased 54% and 43% respectively(P＜0.01),and heart/body weight ratio unchanged(P＞0.05),UII receptors were up-regulated by 184% and 159%(P＜0.01)respectively, While the affinity to UII was decreased (Kd increased 224% and 206 respectively,P＜0.01).In the late group, the blood pressure increased 85% and 67% (P＜0.01), and heart/body weight ratio increased 18% and 22% (P＜0.05) respectively，than that of control and sham group. UII receptors were down-regulated by 35% and 41%(P＜0.05) respectively while the affinity to UII was increased (Kd decreased 30% and 33% respectively, P＜0.05). CONCLUSION: The biphasic changes of UII receptor in myocardial sarcolemma induced by pressure-overload were observed,increasing in early stage, while decreasing in late stage, and these changes were involved in the development of cardiac hypertrophy.

Abstract:Objective - To explore the possible role of uncoupling protein-2 (UCP2) in cardiomyocyte apoptosis in pressure overload-induced left ventricular hypertrophy (LVH).Methods and results - Pressure overload was developed in 21 Sprague-Dawley rats by thoracic aortic constriction at 12 weeks of age.An equal number of sham-operated, age-matched rats served as controls.Aortic blood pressure (ABP), LVH, myocardial apoptosis index (MAI), and UCP2 mRNA expression were quantified in 7 subgroups of 3 treated and 3 control rats that were killed, at 1, 2, 4, 7, 14, 2 1, or 30 days post-surgery, respectively,. Compared to controls, ABP increased gradually throughout the study in the treated rats; LVH did not develop significantly until 4 days after surgery and increased progressively afterwards. The MAI increased immediately after surgery, reached a plateau from 4 to 7 days, and then declined rapidly; apoptosis was undetectable throughout the study in the cardiornyocytes of the control rats. In treated rats, the expression of UCP2 mRNA in myocardium was upreguiated at 4 days, and developed progressively to the end of the experiment.Conclusions - (i) Apoptosis of cardiornyocytes is an important regulatory mechanism that is involved in the cardiac adaptive response to pressure overload, and (ii) the apoptosis of cardiomyocytes may be suppressed, in part, by UCP2.
Functional Inhibition of Urea Transporter UT-B Enhances Endothelial-Dependent Vasodilatation and Lowers Blood Pressure Via L-arginine-endothelial Nitric Oxide Synthase-Nitric Oxide Pathway

Abstract:Mammalian urea transporters (UTs), UT-A and UT-B, are best known for their role in urine concentration. UT-B is especially distributed in multiple extrarenal tissues with abundant expression in vascular endothelium, but little is known about its role in vascular function. The present study investigated the physiological significance of UT-B in regulating vasorelaxations and blood pressure. UT-B deletion in mice or treatment with UT-B inhibitor PU-14 in Wistar-Kyoto rats (WKYs) and spontaneous hypertensive rats (SHRs) reduced blood pressure. Acetylcholine-induced vasorelaxation was significantly augmented in aortas from UT-B null mice. PU-14 concentration-dependently produced endothelium-dependent relaxations in thoracic aortas and mesenteric arteries from both mice and rats and the relaxations were abolished by N(ω)-nitro-L-arginine methyl ester. Both expression and phosphorylation of endothelial nitric oxide synthase (eNOS) were up-regulated and expression of arginase I was down-regulated when UT-B was inhibited both in vivo and in vitro. PU-14 induced endothelium-dependent relaxations to a similar degree in aortas from 12 weeks old SHRs or WKYs. In summary, here we report for the first time that inhibition of UT-B plays an important role in regulating vasorelaxations and blood pressure via up-regulation of L-arginine-eNOS-NO pathway, and it may become another potential therapeutic target for the treatment of hypertension.
Ndufa7 Plays a Critical Role in Cardiac Hypertrophy

Abstract:Cardiac hypertrophy is a common pathological change in patients with progressive cardiac function failure, which can be caused by hypertrophic cardiomyopathy (HCM), dilated cardiomyopathy (DCM) or arterial hypertension. Despite years of study, there is still limited knowledge about the underlying molecular mechanisms for cardiac hypertrophy. NDUFA7, a subunit of NADH:ubiquinone oxidoreductase (complex I), has been reported to be a novel HCM associated gene. However, the biological role of NDUFA7 in heart remains unknown. In this study, we found that NDUFA7 exhibited high expression in the heart, and its level was significantly decreased in mice model of cardiac hypertrophy. Moreover, we demonstrated that ndufa7 knockdown in developing zebrafish embryos resulted in cardiac development and functional defects, associated with increased expression of pathological hypertrophy biomarkers nppa (ANP) and nppb (BNP). Mechanistic study demonstrated that ndufa7 depletion promoted ROS production and calcineurin signalling activation. Moreover, NDUFA7 depletion contributed to cardiac cell hypertrophy. Together, these results report for the first time that ndufa7 is implicated in pathological cardiac hypertrophy.

Abstract:BackgroundPrevious studies found that urea transporter UT-B is abundantly expressed in bladder urothelium. However, the dynamic role of UT-B in bladder urothelial cells remains unclear. The objective of this study is to evaluate the physiological roles of UT-B in bladder urothelium using UT-B knockout mouse model and T24 cell line.Methodology/principal findingsUrea and NO measurement, mRNA expression micro-array analysis, light and transmission electron microscopy, apoptosis assays, DNA damage and repair determination, and intracellular signaling examination were performed in UT-B null bladders vs wild-type bladders and in vitro T24 epithelial cells. UT-B was highly expressed in mouse bladder urothelium. The genes, Dcaf11, MCM2-4, Uch-L1, Bnip3 and 45 S pre rRNA, related to DNA damage and apoptosis were significantly regulated in UT-B null urothelium. DNA damage and apoptosis highly occurred in UT-B null urothelium. Urea and NO levels were significantly higher in UT-B null urothelium than that in wild-type, which may affect L-arginine metabolism and the intracellular signals related to DNA damage and apoptosis. These findings were consistent with the in vitro study in T24 cells that, after urea loading, exhibited cell cycle delay and apoptosis.Conclusions/significanceUT-B may play an important role in protecting bladder urothelium by balancing intracellular urea concentration. Disruption of UT-B function induces DNA damage and apoptosis in bladder, which can result in bladder disorders.
Expression of Adenine Nucleotide Translocator-1 and Cardiomyocyte Apoptosis in Pressure Overload-Induced Left Ventricular Hypertrophy in Rats

Abstract:Objective To explore the effects of adenine nucleotide translocator-1 (ANT1) on cardiomyocyte apoptosis in pressure overload-induced left ventricular hypertrophy. Methods The pressure overload rat model was established by abdominal aorta coarctation. The observation time points were 1, 2, 4, 7, 14, 21 and 30 days after operation. The ANT1 mRNA expression was evaluated by RT-PCR. The TUNEL method was applied to detect the cardiomyocyte apoptosis. Results 1. Compared with control group, the expression of ANT1 mRNA was upregulated on 4th day after coarctation, and peaked on 7th day, then it returned to control level on 14th day and persisted until 30th day. 2. The apoptosis increased significantly 1 day after operation, and reached a plateau between 4 and 7 day. Afterwards, it decreased continuously to a low level. However, the apoptosis was so rare that it was undetectable in control group. Conclusion Apoptosis is an important regulatory mechanism involved in cardiac adaptive response to pressure overload, and it may be mediated, in part, by ANT1.

Abstract:Objectives MicroRNAs, as one of key regulators for post-transcriptional gene control, have shown to be involved in cardiovascular diseases. This study aimed to investigate the roles of miR-378 in regulation of cardiac hypertrophy and cardiac fibrosis stimulated by pressure overload. Methods To construct cardiac remodelling mice models, pressure overload was imposed on the heart of C57B/L6 mice (8 weeks) by constriction of transverse aortic banding (TAC). Chemically modified oligonucleotides miRNA mimics (miR-378-Agomir) and inhibitors (miR-378-Antagomir) treatment started 24 h after TAC by intravenous injections on two consecutive days. Mice were randomly divided into four groups: 1) the TAC group; 2) the TAC-Agomir group; 3) the TAC- Antagomir group; 4) the Sham operation group. Mice after sham or 2 weeks TAC operation were subjected to hemodynamic and echocardiographic measurement. Tissue and serum miR-378 expressions were detected by TaqMan MicroRNA Assay Kit. ANP, BNP, β-MHC, collagen I and collagen III was measured by Real-time PCR. ERK1/2, p-ERK1/2, MMPs and α-SMA expression were analysed by western blotting. Collagen was assessed by immunohistochemistry. Results We found that miR-378 has a high expression pattern in the heart. After two-week acute pressure overload, the TAC group exhibit prominent cardiac hypertrophic responses and decreased expression of miR-378(p < 0.05) comparing with the sham group. In vivo overexpression of miR-378 in TAC-Agomir group markerly inhibited cardiac hypertrophy, accompanied with downregulation of fetal gene ANP, BNP and β-MHC compared to that of TAC group. Howerver, inhibition of endogenous miR-378 by an infusion of an antagomir caused sustained cardiac hypertrophy, associated with a reinduction of fetal gene expression to a less extent. The p-ERK level was also downregulated in TAC-Agomir group and upregulated in TAC- Antagomir group. In part of cardiac fibrosis, we found that the over-expression of miR-378 of TAC-Agomir group reduced the expression of col I, col III, MMP2, MMP9 and α-SMA(p < 0.05) compared to that of TAC group, whereas down regulation of endogenous miR-378 with anti-miRs promoted collagens and MMPs expression. Conclusions These findings reveal orchestrating regulatory roles for miR-378 in pressure-overload induced cardiac remodeling. It suggests that miR-378 not only suppresses cardiac hypertrophy by regulating fetal genes expression and ERK1/2 signaling pathway but also controls fibrosis-related genes expression in the heart.
Abstract 17329: Myocardial-Specific Overexpression of a Dominant-Negative Mutant of NADPH Oxidase Subunit P67 Phox Attenuates Pressure-Overload-Induced Cardiac Hypertrophy in Adult Transgenic Mice

Abstract:We had reported a transgenic (Tg) mouse model with cardiac-specific overexpression of a dominant-negative (DN) mutant (V204A) of NADPH oxidase (NOX) subunit p67 phox that inhibits NOX activity. In order to test the role of NOX in pressure-overload-induced cardiac hypertrophy in young adult mice, age and body weight (BW)-matched male homozygous Tg and wild type (WT) C57BL/6 mice (10-12 weeks) were subjected to transverse aortic constriction (TAC) or sham operation. Four-week post TAC, 2-D echocardiography (echo) was performed to determine the following cardiac parameters: left ventricular myocardial area (LVMA); LV wall thickness at end diastole; LV internal dimension at end diastole (IDd), LV end-diastolic volume (EDV), LV ejection fraction (EF), LV mass & aortic pressure gradient. Mice were divided into 4 groups (15-24 mice/group) including WT-sham, WT-TAC, Tg-sham & Tg-TAC. The LVEDV, LVIDd & BW were similar among all groups. The LVEF (mean 48-53%) was not statistically different among all groups. The aortic pressure gradient was between 30-60 mmHg in TAC mice and was statistically similar between the Tg and WT groups (45.3 ±1.7 vs. 50.4 ± 2.1 mmHg, p =0.1). Tg-sham showed no significant difference in LVMA, LV mass & wall thickness vs. WT-sham (Table 1). The WT-TAC mice showed significant increase in LVMA, LV mass & wall thickness indicating LV hypertrophy. The Tg-TAC mice also showed mild increase in the above parameters with the LV wall thickness statistically different from Tg-sham. However, these increases in Tg-TAC mice were significantly ( p <0.05) smaller than those in WT-TAC mice, suggesting TAC-induced LV hypertrophy was attenuated in Tg mice. Our echo finding was also confirmed by our results of heart weight/BW ratio, ANF mRNA expression, and histological study of the LV myocardium. In summary, our results showed that selective inhibition of myocardial NOX subunit p67 phox attenuated chronic pressure-overload-Induced cardiac hypertrophy in young adult mice.

Abstract:Increased cardiac angiotensin converting enzyme-1 (ACE1) is found in individuals who carry a deletion in intron 16 of ACE1 gene or in individuals who suffer from cardiac disorders, such as hypertrophy. However, whether a single increase in ACE1 expression leads to spontaneous cardiac defects remains unknown. To determine if the increased cardiac ACE1 actively plays a role or is merely the consequence of pathological changes in the process of cardiac hypertrophy, we generated a transgenic rat model with selective over-expression of human ACE1 in the cardiac ventricles. The left ventricular ACE1 activity is elevated about 50-fold in transgenic rats. Angiotensin-1 perfusion of isolated hearts demonstrated a significant decrease in coronary artery flow compared with non-transgenic littermates, suggesting that the transgenic ACE1 is functional. Neither cardiac hypertrophy nor other morphological abnormalities were observed in transgenic rats under standard living conditions. It was found, however, after induction of hypertension by suprarenal aortic banding, that the degree of cardiac hypertrophy in transgenic rats was significantly higher than that of banded control rats. The expressions of both ANF and collagen III, molecular markers of cardiac hypertrophy, were also increased in banded transgenic rats compared with banded control. Our results suggest that increased cardiac ACE1 does not trigger but augments cardiac hypertrophy.

Abstract:Objectives: To study the role of myocardial adenine nucleotide translocator(ANT) during the occurrenc and development of hypertensive cardiac hypertrophy. Methods: The animal model of pressure load is made by means of abdominal aorta coarctation. The relative content of ANT1 mRNA and ANT2 mRNA in myocardium is detected by RT PCR and gel electrophoresis imaging. Results: ①Abdominal aorta pressure,cardiac weight index and myocardial pathological section of the operated group rats confirm the success of the animal model of pressure load. ②At about 4 days after coarctation, the level of ANT1 mRNA was up regulated but ANT2 mRNA was down regulated,and both reached the peak and valley respectively at 7 days. ③The levels of ANT1 mRNA and ANT2 mRNA are close to that of the sham operated group at 14 days after coarctation. Conclusions: Adenine nucleotide translocator(ANT) participate in the occurrence and development of hypertensive cardiac hypertrophy.And it may be ANT1 that is involved preferentially in the regulation of myocardial apoptosis,whereas the ANT2 might be more engaged in energy supply.
The Urea Transporter UT-A1 Plays a Predominant Role in a Urea-Dependent Urine-Concentrating Mechanism

Abstract:Urea transporters are a family of urea-selective channel proteins expressed in multiple tissues that play an important role in the urine-concentrating mechanism of the mammalian kidney. Previous studies have shown that knockout of urea transporter (UT)-B, UT-A1/A3, or all UTs leads to urea-selective diuresis, indicating that urea transporters have important roles in urine concentration. Here, we sought to determine the role of UT-A1 in the urine-concentrating mechanism in a newly developed UT-A1?knockout mouse model. Phenotypically, daily urine output in UT-A1?knockout mice was nearly 3-fold that of WT mice and 82% of all-UT?knockout mice, and the UT-A1?knockout mice had significantly lower urine osmolality than WT mice. After 24-h water restriction, acute urea loading, or high-protein (40%) intake, UT-A1?knockout mice were unable to increase urine-concentrating ability. Compared with all-UT?knockout mice, the UT-A1?knockout mice exhibited similarly elevated daily urine output and decreased urine osmolality, indicating impaired urea-selective urine concentration. Our experimental findings reveal that UT-A1 has a predominant role in urea-dependent urine-concentrating mechanisms, suggesting that UT-A1 represents a promising diuretic target.
Deubiquitinase USP47 ameliorates cardiac hypertrophy through reducing protein O-GlcNAcylation

Abstract:Cardiac hypertrophy is a crucial risk factor for heart failure when the heart is confronted with physiological or pathological stimuli. The ubiquitin-proteasome system (UPS) plays a critical role in the pathogenesis of cardiac hypertrophy. However, as a key component of the UPS, the role of deubiquitinating enzymes (DUBs) in cardiac hypertrophy is not well understood. Here, we observed that the expression level of deubiquitinase USP47 was increased in hypertrophic hearts and angiotensin II (Ang II)-stimulated neonatal rat cardiomyocytes (NRCMs). Adenovirus-mediated gain- and loss-of-function approaches indicated that USP47 overexpression significantly attenuated Ang II-induced cardiac hypertrophy in vitro and in vivo, whereas endogenous USP47 deficiency promoted the pro-hypertrophic effect of Ang II. Further investigation demonstrated that USP47 inhibited O-GlcNAcylation in cardiomyocytes by controlling the expression of O-GlcNAcase (OGA). Mechanistically, USP47 bound, deubiquitinated, and stabilized protein arginine methyltransferase 5 (PRMT5), thus upregulating OGA expression. We found that the restoration of PRMT5 abolished the pro-hypertrophic effects of USP47 silence in vitro. Therefore, our results provide the first evidence of the involvement of USP47 in cardiac hypertrophy and identify USP47 as a potential target for hypertrophic therapy.
