Abstract:Chitosan, the cationic polysaccharide derived from the natural polysaccharide chitin, has been studied as a biomaterial for more than two decades. As a polycationic polymer with favorable properties, it has been widely used to form polyelectrolyte complexes with polyanions for various applications in drug delivery fields. In recent years, a growing number of studies have been focused on the preparation of polyelectrolyte complexes based on chitosan and its water soluble derivatives. They have been considered well-suited as biomaterials for a number of vital drug carriers with targeted/controlled release profiles, e.g., films, capsules, microcapsules. In this work, an overview highlights not only the favorable properties of chitosan and its water soluble derivatives but also the good performance of the polyelectrolyte complexes produced based on chitosan. Their various types of applications as drug carriers are reviewed in detail.

Abstract:Objective: To prepare porous medical chitosan membranes and do their performance characterization.Method: Membranes were prepared by solvent evaporation gel method,degradation of chitosan enzyme to prepare porous chitosan membranes,then their morphology was observed by scanning electron microscope,weighing method was adopted to detect relationship between moisture absorption effect and hydrolysis time-remaining weight,and test adsorption of porous chitosan membranes to bovine serum albumin and controlled release properties to silver sulfadiazine.Result: Ratio of chitosan and endonuclease was 0.1 g·U-1,enzymolysis time was 3 h,both surface and internal of membranes generated porous structure which was suitable for pharmaceutical carriers.Conclusion: In situ endonuclease digestion of porous chitosan membranes had advantages of mild reaction conditions,pore rate controlled and non-toxic substance residues etc,which had a good application prospect.

Abstract:A new type of hydrophilic chitosan nanoparticles encapsulated with BSA(BSA-CS NPs) was prepared through ionic cross-linking technique based on CS and sodium tripolyphosphate(TPP) at room temperature.The influence of formulation conditions on the size of the BSA-CS NPs was investigated.Results showed that BSA-CS NPs with diameter of below 100 nm were prepared when the pH value was 5.0,the mass ratio of CS to TPP was 4,the molecular weights of CS was 40 kDa,and the BSA encapsulation efficiency was more than 50%.This system was also applied in the preparation of chitosan nanoparticles encapsulated with Gamma Seroglobulin(IG-CS NPs).

Abstract:Chitosan nanoparticles fabricated via different preparation protocols have been in recent years widely studied as carriers for therapeutic proteins and genes with varying degree of effectiveness and drawbacks. This work seeks to further explore the polyionic coacervation fabrication process, and associated processing conditions under which protein encapsulation and subsequent release can be systematically and predictably manipulated so as to obtain desired effectiveness. BSA was used as a model protein which was encapsulated by either incorporation or incubation method, using the polyanion tripolyphosphate (TPP) as the coacervation crosslink agent to form chitosan-BSA-TPP nanoparticles. The BSA-loaded chitosan-TPP nanoparticles were characterized for particle size, morphology, zeta potential, BSA encapsulation efficiency, and subsequent release kinetics, which were found predominantly dependent on the factors of chitosan molecular weight, chitosan concentration, BSA loading concentration, and chitosan/TPP mass ratio. The BSA loaded nanoparticles prepared under varying conditions were in the size range of 200-580nm, and exhibit a high positive zeta potential. Detailed sequential time frame TEM imaging of morphological change of the BSA loaded particles showed a swelling and particle degradation process. Initial burst released due to surface protein desorption and diffusion from sublayers did not relate directly to change of particle size and shape, which was eminently apparent only after 6h. It is also notable that later stage particle degradation and disintegration did not yield a substantial follow-on release, as the remaining protein molecules, with adaptable 3-D conformation, could be tightly bound and entangled with the cationic chitosan chains. In general, this study demonstrated that the polyionic coacervation process for fabricating protein loaded chitosan nanoparticles offers simple preparation conditions and a clear processing window for manipulation of physiochemical properties of the nanoparticles (e.g., size and surface charge), which can be conditioned to exert control over protein encapsulation efficiency and subsequent release profile. The weakness of the chitosan nanoparticle system lies typically with difficulties in controlling initial burst effect in releasing large quantities of protein molecules.

Abstract:The objective of this work was to characterize a novel quaternary chitosan derivative [O-(2-hydroxyl) propyl-3-trimethyl ammonium chitosan chloride (O-HTCC)] nanoparticle system. O-HTCC nanoparticles were prepared with a simple and mild ionic gelation method upon the addition of a sodium tripolyphosphate solution to a low-molecular-weight O-HTCC solution. Highly cationic chitosan nanoparticles were prepared. Bovine serum albumin (BSA), a model protein drug, was incorporated into the nanoparticles. The physicochemical properties of the nanoparticles were determined with transmission electron microscopy (TEM), scanning electron microscopy (SEM), Fourier transform infrared analysis, differential scanning calorimetry, and X-ray diffraction (XRD) patterns. The results showed that increasing the BSA concentration from 1.5 to 2.5 mg/mL promoted the BSA encapsulation efficiency from 57.3% to 87.5% and the loading capacity from 70.2%, to 99.5%. Compared with the chitosan nanoparticles, the O-HTCC nanoparticles had lower burst release. TEM revealed that the BSA-loaded O-HTCC nanoparticles were smaller than the O-HTCC nanoparticles when the BSA concentration was 1.5 mg/mL; SEM showed that the size of the BSA-loaded O-HTCC nanoparticles was mostly affected by the BSA concentration, and the increase in size occurred with the concentration increasing. Thermograms and XRD of the BSA-loaded nanoparticles suggested that polyelectrolyte-protein interactions increased with the BSA concentration increasing and greater chain realignment in the BSA-loaded nanoparticles. Abstract:Several novel functionalized graft copolymer nanoparticles consisting of chitosan (CS) and the monomer methyl methacrylate (MMA), N-dimethylaminoethyl methacrylate hydrochloride (DMAEMC), and N-trimethylaminoethyl methacrylate chloride (TMAEMC), which show a higher solubility than chitosan in a broader pH range, have been prepared by free radical polymerization. The nanoparticles were characterized in terms of particle size, zeta potential, TEM, and FT-IR. These nanoparticles were 150-280 nm in size and carried obvious positive surface charges. Protein-loaded nanoparticles were prepared, and their maximal encapsulation efficiency was up to 100%. In vitro release showed that these nanoparticles provided an initial burst release followed by a slowly sustained release for more than 24 h. These graft copolymer nanoparticles enhanced the absorption and improved the bioavailability of insulin via the gastrointestinal (GI) tract of normal male Sprague-Dawley (SD) strain rats to a greater extent than that of the phosphate buffer solution (PBS) of insulin.

Abstract:OBJECTIVETo prepare insulin-loaded chitosan nan op articles.METHODSChitosan with different molecular weights w as prepared by enzymatic degradation and ultrafiltration separation.Their molecu lar weights were determined by gel permeation chromatography (GPC).Chitosan nano particles were prepared by solvent diffusion method.Several characteristics of c hitosan nanoparticles such as morphology,particle size,zeta potential,drug entra pment efficiency and drug release in vitro were evaluated.RESULTS Chitosan with three different molecular weights were obtained by contr olling the conditions of enzymatic degradation and ultrafiltration separation.Th e molecular weights of chitosan determined by gel permeation chromatography (GPC ) were 10 289,41 500, 101 870 ,respectively.The chitosan nanoparticles prepare d by solvent diffusion technique showed a spheric shape and exhibited a narrow p article size distribution,and became smaller after increasing the concentration of alcohol in the system or increasing the sonication treatment times.The distri bution of particle size also turned to narrower when using lower molecular weigh t chitosan.Due to the high zeta potentials of chitosan nanoparticle systems (ove r +30 mV),these systems were relatively stable.The average entrapment efficiency of insulin was 72.66% determined by radioimmunoassay.Insulin release from nanop articles in the delivery vehicle containing chitosanase showed a rapid delivery in the initial hour and then became slow in the next five hours.CONCL USIONIt was easy to prepare the insulin-loaded chitosan nanoparticles with chitosan with optional molecular weights.A high envelopment and certain su stained release effect could be obtained.

Abstract:Chitosan (CS) nanoparticles (NPs) prepared by ionotropic gelation technique are one of the forcefully studied nanosystems used for drug delivery. Though, a deficiency of inter-laboratory reproducibility and a poor physicochemical understanding of particle formation process have been decelerating their potential market applications. To report these shortcomings, the current work offerings the effect of primary formulation parameters on CSNPs formation by a single factor test. The stability of CSNPs at various pH and ionic strength was also analyzed. The mean particle size, zeta potential (ZP), and polydispersity index (PDI) of optimum CSNPs were 67.60±0.11 nm, +33.23±1.20 mV and 0.26±0.00, respectively. The SEM and TEM studies proved that the CSNPs were well dispersed spherical shape with good particle size distributions. The FTIR spectrum indicated that cross-linking was successfully carried out. To sum up, we found that by controlling the formulations parameters, it is possible to control in a reproducible manner the production and characteristics of CSNPs in the range of nanomaterial size.

Abstract:We developed a water-soluble Gliadin /sodium caseinate nanoparticle( Gliadin /SC nanoparticle) from gliadin,an abundant vegetal protein via an antisolvent precipitation method. Sodium caseinate( SC),a food grade natural protein acted as electrostatic and sterical stabilizer. The formation of the Gliadin /SC nanoparticle and microstructure were explored. The role of proportion of SC-to-gliadin and pH,ionic strength effect on particle size distribution,zeta potential and stablization were investigated. The Gliadin /SC nanoparticles are spherical. The average size of the Gliadin /SC nano-particle was 190 to 310 nm. Zeta potential of the PG-SC was around- 28. 6 to- 32. 8mV at pH7,and has storage stability over 2 weeks. The proposed hypothesis in this work could be applied to carry hydrophobic bioactive molecules in food industry.
Preparation and Characterization of N-succinyl-chitosan

Abstract:Poisonous solvents were widely used in the preparation of N-succinyl-chitosan,which were easy to cause environmental pollution and difficult to remove completely from the products.These disadvantages limited their uses in biomedical materials.In the present work,a novel method was proposed to successfully synthesize N-succinyl-chitosan,which was safe and environmentally friendly.A series of N-succinyl-chitosan with different degrees of substitution were prepared in absolute ethanol by controlling the reaction time.The different degrees of substitution,molecular weight and polydispersity index of the products were measured.The products were characterized by Fourier transform infrared(FTIR) spectroscopy,X-ray diffractometry(XRD) and thermogravimetry(TG).Furthermore,the water-solubility and biocompatibility were studied.When the reaction time is 15 h,the degree of substitution and molecular weight of the product are highest.Moreover,polydispersity index of the product is smallest among all the products(10 h,12 h,15 h,18 h,20 h).In addition,the results indicate N-succinyl-chitosan has been synthesized with decreased degree of crystallization and thermostability.Water-solubility of modified chitosan is improved due to the decrease of crystallization.N-succinyl-chitosan prepared in absolute ethanol has good biocompatibility,which has great potential in biomedical fields.

Abstract:Abstract In this paper, the β-cyclodextrin (β-CD) and biotin (Bi) were successfully grafted onto carboxymethyl chitosan (CMCS). And then the β-CD-Bi-CMCS nanoparticles (NPs) were prepared as oral nano-delivery carrier of protein drugs by ionic gelation method. The morphological feature of fabricated drug carrier was determined by dynamic light scattering and transmission electron microscopy. The result showed that the prepared NPs presented spherical structure with an average diameter of 138 nm. Bovine serum albumin (BSA) was selected as model protein drug that was entrapped in prepared drug carrier with satisfactory entrapment efficiency (79.18%) and loading content (3.96%). The drug release profiles of BSA/β-CD-Bi-CMCS NPs were studied at different pH environment for simulated gastric fluid (SGF), simulated intestinal fluid (SIF) and simulated colonic fluid (SCF). It was found that the BSA/β-CD-Bi-CMCS NPs displayed a pH dependent drug release profiles. After 72 h, the cumulative release amount of BSA in SGF, SIF, and SCF was about 20.57, 74.46, and 91%, respectively. Furthermore, the enzymatic degradation and cytotoxicity studies showed the synthesized β-CD-Bi-CMCS NPs had high chemical stability and biocompatibility. This work indicated that the β-CD-Bi-CMCS NPs had the potentiality as promising nanocarriers for oral delivery of protein drugs.

Abstract:Natural lutein is liposoluble and has powerful antioxidant activity, which can be self-aggregated to form H-type aggregates in organic-water systems. However,its application is limited by poor solubility and high instability. Here, whey proteins/chitosan-coated H-type aggregates lutein nanoparticles (NPs) were fabricated via a bottom-up layer-by-layer self-assembly technique. The optimal conditions for the preparation of the NPs (173.3 nm) were determined by the Dynamic Light Scattering and UV–vis spectroscopy. Briefly, three proteins (WPI and WPC and BSA) were used to fabricate polysaccharide-protein nanocarriers to encapsulate the lutein. These spectroscopy studies indicated that chitosan and BSA formed hydrophobic microdomain by the intermolecular electrostatic attraction force with remarkable changes of secondary structure in protein. The morphology revealed that the NPs were nearly spherical. The EE of the NPs was ≥90.4 %, with a LC of up to 28.3 %. Lutein in NPs can be stabilized as H-type aggregates at different temperatures and pH-values. Additionally, the cytotoxicity test of the NPs on L929 and Caco-2 cells showed that NPs had low cytotoxicity in a limited concentration range. Results indicated that whey protein/chitosan-coated lutein nanoparticles significantly improved water dispersion and stability of lutein and its aggregates, thus broadening their application in nutrient delivery system.

Abstract:Abstract Background The purpose of this study was to develop protein-/peptide-loaded nanoparticle-based delivery system, which can efficiently deliver therapeutic molecules to the lung via pulmonary delivery. The chitosan nanoparticles were prepared by the ionic gelation method, and bovine serum albumin was used as a model protein. These nanoparticles were characterized for size, zeta potential, encapsulation efficiency, cell cytotoxicity, uptake study, release profile and size distribution and uniformity. The chemical interaction of chitosan and protein was studied by XRD and FTIR. The integrity assessment of encapsulated protein into nanoparticle was studied by native and SDS-PAGE gel electrophoresis. Results The size and zeta potential of BSA nanoparticles were 193.53 ± 44.97 to 336.36 ± 94.63 and 12.73 ± 0.41 to 18.33 ± 0.96, respectively, with PDI values of 0.35–0.45. The encapsulation efficiency was in the range of 80.73 ± 6.37% to 92.34 ± 1.72%. The cumulative release of the BSA from the nanoparticles was 72.56 ± 6.67% in 2 weeks. The BSA-loaded nanoparticles showed good uptake and no significant cytotoxicity observed into the A549 cell line. In this study, it was also observed that during nanoparticles’ synthesis protein structure and integrity is not compromised. The nanoparticles showed controlled and sustained release with initial burst release. In TEM images, it was shown that nanoparticles’ distribution is uniform within nanometre range. Conclusion From this study, it was concluded that nanoparticles prepared by this method are suitable to deliver protein/peptide into the cells without any degradation of protein during process of nanoparticle fabrication.

Abstract:Liposomes are an important colloidal carrier system for controlled drug delivery. However some highly hydrophilic small molecules are difficult to entrap into liposomes and store stably, resulting in poor encapsulation efficiency and fast leakage. In the present work, fluorescein sodium (FS) was used as a model drug that was loaded into chitosan nanoparticles and then encapsulated into liposomes by reverse-phase evaporation (RPV). The encapsulation efficiency, particle size, zeta potential, release in vitro and pharmacokinetics in rats were determined in order to characterize the novel drug delivery system. The entrapment efficiency was above 80% in nanoparticles (Np) and 95% in liposomes encapsulating the nanoparticles (Lip-Np). The Lip-Np was composed of soybean phospholipids, cholesterol and chitosan, which the average diameter was 202.6 nm and zeta potential was -34.8 mV The release rate of fluorescein sodium from Lip-Np was slower than from Np and liposomes. FS in Lip-Np administered to rats exhibited prolonged circulation and higher bioavailability than FS in Np. The results indicated that liposomal release kinetics can be controlled by encapsulating nanoparticles and thus solid-cored liposomes can be used as a potential drug delivery system.

Abstract:Chitosan,in particular with a format of microspheres,has been extensively employed in many applications,that is,treatment of wastewater,chromatographic support,enzyme immobilization,drug delivery,dental application,and hemostasis.In this paper,porous chitosan microspheres were prepared by oil-in-water-in-oil(O/W/O) multiple emulsion crosslinking method.The low toxic and nontoxic solvents,ethyl acetate and castor oil,were applied as a cavity generating agent and a water-insoluble dispersion medium,respectively,to replace the commonly used toxic oil phase such as petroleum ether,toluene and paraffin.The low molecular weight chitosan was obtained by the depolymerization of raw chitosan with hydroperoxide.The viscosity molecular weight(Mw) of the chitosan was investigated by using an Ubbelohde viscosimeter and calculated based on Mark Houwink equation.For preparation of the porous chitosan microspheres,ethyl acetate was first emulsified in chitosan acetic acid solution,and subsequently the emulsion was inversely dispersed in castor oil.The chitosan micro-droplets were solidified and hardened by glutaraldehyde as crosslinking agent.All the preparation process was carried out at room temperature.The stirring rate of the emulsion,chitosan concentration,ethyl acetate/chitosan solution ratio,chitosan solution/castor oil ratio,and glutaraldehyde concentration were studied as effective preparation parameters on the size,size distribution and porosity of the chitosan microspheres.The chitosan microspheres were filtrated and rinsed with ethanol and water to remove the organic solvent and the surfactants.Finally,the porous chitosan microspheres were obtained by freeze-drying.The number average diameter of the optimized example of chitosan microspheres was 51.2 μm.The porous structures distributed in the chitosan microspheres were clearly observed by the SEM.

Abstract:The control of size and size distribution of microspheres is necessary for obtaining repeatable controlled release behavior. The chitosan microspheres were prepared by a membrane emulsification technique in this study. Chitosan was dissolved in 1 wt.% aqueous acetic acid containing 0.9 wt.% sodium chloride, which was used as a water phase. A mixture of liquid paraffin and petroleum ether 7:5 (v/v) containing PO-500 emulsifier was used as an oil phase. The water phase was permeated through the uniform pores of a porous glass membrane into the oil phase by the pressure of nitrogen gas to form W/O emulsion. Then GST (Glutaraldehyde Saturated Toluene) as crosslinking agent was slowly dropped into the W/O emulsion to solidify the chitosan droplets. The preparation condition for obtaining uniform-sized microspheres was optimized. The microspheres with different size were prepared by using the membranes with different pore size, and there was a linear relationship between the diameter of microspheres and pore size of the membranes when the microspheres were in the range of micron size. The smallest chitosan microspheres obtained was 0.4 mum in diameter. This is the first report for preparing the uniform-sized chitosan microspheres by membrane emulsification technique. Uniform chitosan microspheres were further used as a carrier of protein drug. Bovine serum albumin (BSA) as a model drug was loaded in the microspheres and released in vitro. The effects of pH value, diameter and crosslinking degree of microspheres, and BSA concentration on loading efficiency and release behavior were discussed.

Fabrication and characterization of novel water-soluble chitosan hydrochloride microspheres

Abstract:Microspheres of water-soluble chitosan hydrochloride were fabricated by emulsion cross-linking method.The properties including surface morphology,interaction,crystallization,size distribution,thermal stability and cytocompatibility were analyzed by different analysis methods and cell culture with MG63 cells.The results show that schiff's base reaction takes place between chitosan hydrochloride and glutaraldehyde.The chitosan hydrochloride microspheres have a spherical shape with smooth surface,poor crystallinity and small size(about 5-10μm by SEM).Compared with the chitosan microspheres,the chitosan hydrochloride microspheres had better balling property and thermal stability.Cell culture indicates that the chitosan hydrochloride microspheres have good cytocompatibility and are beneficial for adhesion,migration and growth of MG63 cells.Because of its water soluble property,there is a broad foreground for its application in drug loading and releasing.

Abstract:Chitosan is a rather abundant material, which has been widely used in food industrial and bioengieering aspects, including in encapsulating active food ingredients, in enzyme immobilization, and as a carrier for drug delivery, due to its significant biological and chemical properties such as biodegradable, biocompatible, bioactive, and polycationic. This review discussed preparation and applications of chitosan nanoparticles in the biomedical engineering field, namely as a drug delivery carrier for biopharmaceuticals.
Formulation and Characterization of Alginate Coated Chitosan Nanoparticles as Therapeutic Protein for Oral Delivery System

Abstract:Nanoparticles have been promptly studied and developed for oral protein delivery. Selection of excipients and formulation method depends on the physicochemical characteristics of the protein carried. Therefore, this study aims to design a formulation and characterize the alginate coated chitosan nanoparticles which carried different protein. Alginate coated chitosan nanoparticles were formed using an ionic gelation method. Optimization was done by varying the parameter such as crosslinker concentration, agitation method, rate, and time. The results show that chitosan nanoparticles formed by sonication using sodium tripolyphosphate (STPP): chitosan (1:0.8) was the best method to form nanoparticles. The particle size and polydispersity index (PDI) of bovine serum albumin (BSA) and lysozyme nanoparticles were 812.2 nm and 0.412 for BSA while 793.3 nm and 0.438 for lysozyme. Encapsulation efficiency (EE) of BSA is 52 % and lysozyme is 68 %. In vitro tests on acidic/gastric conditions showed that lysozyme (±32 %) was released faster than BSA (±10 %) during the 24 h incubation. Under neutral/terminal intestine conditions, percentage of BSA (±17 %) release is slightly higher than lysozyme (±13 %) for 8 h incubation period. It was concluded that the formulation of alginate coated chitosan nanoparticles in this study appears to be more effective for BSA delivery than lysozyme.
