Abstract:Biocompatible single‐component theranostic nanoagents instinctly affording multiple imaging modalities with satisfying therapeutic functions are highly desirable for anticancer treatments. Although cobalt‐based phosphides are well‐recognized as competent electrocatalysts, their potentials for biomedical applications remain unexplored. In this work, cobalt phosphide nanoparticles (CoP NPs) are developed to be a powerful theranostic agent for multimodal imaging and anticancer photothermal therapy. The uniform CoP NPs in a size of ≈21 nm are synthesized via a facile thermal decomposition method, followed by surface modification. The resultant CoP NPs exhibit excellent compatibility and stability in water as well as various physiological solutions. Supported by the good biocompatibility, strong near‐infrared absorption, and high photothermal conversion property, significant photothermal effect of the NPs is demonstrated, realizing efficient hyperthermia ablation on cancer cells. Importantly, the CoP NPs have shown considerable capabilities on high‐contrast in vitro and in vivo triple‐modal imaging, including infrared thermal (IRT), photoacoustic (PA), and T2‐weighted magnetic resonance (MR) imaging. This work has unraveled the promising potentials of CoP‐based nanoagent for precise diagnosis and efficient therapy.

Abstract:Multifunctional theranostic platforms, especially single component-based platforms, enable both cancer treatment and real-time imaging as well as enhance the efficiency of treatment. In this study, 50 nm Mo2C nanospheres were explored as a "one-for-all" theranostic agent. The light-harvesting of Mo2C covered the entire near infrared region, and NIR irradiation concurrently triggered hyperthermia and reactive oxygen species (ROS) production; thus, synergistic outcomes of photothermal and photodynamic therapy could be realized. Both in vitro and in vivo experiments have confirmed the superiority of the synergistic phototherapy in killing cancer cells and removing solid tumors; moreover, Mo2C proposed herein has been proven to be applicable as a photoacoustic imaging and CT imaging contrast agent for in vivo tumor depiction; furthermore, Mo2C demonstrates excellent biocompatibility, showing minimal hematotoxicity and tissue toxicity. A theoretical simulation performed by density functional theory revealed that the metallic character and the interband/intraband transition of Mo2C accounted for its broad photoabsorption. The antitumor mechanism of Mo2C was investigated on a solid tumor by B-mode ultrasonography (US) and magnetic resonance imaging (MRI), revealing a typical liquefactive necrosis process; hence, herein, the dual-imaging guided phototherapy was efficiently mediated by Mo2C.
In vivo near-infrared photothermal therapy and computed tomography imaging of cancer cells using novel tungsten-based theranostic probe.

Abstract:Photothermal therapy, as a physical therapeutic technique to kill cancer, has generated a great deal of interest. Photothermal agents hence play a critical role in this modern therapy. We report the use of transition metal oxides as photothermal agents based on PEGylated WO3-x nanoparticles. The well-prepared nanoparticles presented effective results during photothermal therapy both in vitro and in vivo by using near-IR laser irradiation (980 nm, 0.5 W cm(-2)). The tumor cells were effectively damaged using low power density during a short irradiation time without destroying healthy tissues. In vitro results of photothermal therapy with PEGylated WO3-x nanoparticles proved to be effective on 4T1 murine breast cancer cells via a confocal microscopy method and MTT assay. In vivo results were further confirmed by hematoxylin and eosin (H & E) histological staining. Additionally, PEGylated WO3-x nanoparticles were shown to be effective as a CT imaging contrast agent on a tumor-bearing mouse model. Our results suggest that this generation of PEGylated WO3-x nanoparticles can potentially be used in oncological CT imaging and photothermal therapy.
Porous Cobalt Sulfide Hollow Nanospheres with Tunable Optical Property for Magnetic Resonance Imaging-Guided Photothermal Therapy.

Abstract:Transition-metal chalcogenides with an imaging element and tunable optical property are strongly desired as ideal high-efficiency photothermal theranostic agents to diagnose and eliminate tumors. Herein, we report on a one-pot solvothermal strategy to synthesize various porous cobalt sulfide hollow nanospheres (PCSH NSs) and elucidate the relation between PCSH NSs and their optical absorption as a guide to obtain optimal photothermal therapy (PTT) agents. After PEG modification, PEG-PCSH NSs show superexcellent photothermal conversion efficiency (∼70.1%) which is higher than that of binary transition-metal chalcogenides materials reported to date. A low dose (100 μL, 25 ppm) could completely ablate tumors under an 808 nm laser power of 0.7 W cm-2, reducing in vivo long-term residual agent content and thus lowering the possibility of side effects. Additionally, they also exhibit excellent biocompatibility, good photostability and utility for magnetic resonance imaging. Our results indicate that PCSH NSs can be considered as an outstanding PTT agent and give guidance towards the design of other photothermal theranostic agents.
Polyethylene Glycol-Modified Cobalt Sulfide Nanosheets for High-Performance Photothermal Conversion and Photoacoustic/magnetic Resonance Imaging

Abstract:Theranostic nanoagents that integrate the diagnoses and therapies within a single nanomaterial are compelling in their use for highly precise and efficient antitumor treatments. Herein, polyethylene glycol (PEG)-modified cobalt sulfide nanosheets (CoS-PEG NSs) are synthesized and unitized as a powerful theranostic nanoagent for efficient photothermal conversion and multimodal imaging for the first time. We demonstrate that the obtained CoS-PEG NSs show excellent compatibility and stability in water and various physiological solutions, and can be effectively internalized by cells, but exhibit a low cytotoxicity. The CoS-PEG NSs exhibit an efficient photothermal conversion capacity, benefited from the strong near-infrared (NIR) absorption, high photothermal conversion efficiency (∼33.0%), and excellent photothermal stability. Importantly, the highly effective photothermal killing effect on cancer cells after exposure to CoS-PEG NSs plus laser irradiation has been confirmed by both the standard Cell Counting Kit-8 and live-dead cell staining assays, revealing a concentration-dependent photothermal therapeutic effect. Moreover, utilizing the strong NIR absorbance together with the T2-MR contrast ability of the CoS-PEG NSs, a high-contrast triple-modal imaging, i.e., photoacoustic (PA), infrared thermal (IRT), and magnetic resonance (MR) imaging, can be achieved, suggesting a great potential for multimodal imaging to provide comprehensive cancer diagnosis. Our work introduces the first bioapplication of the CoS-PEG nanomaterial as a potential theranostic nanoplatform and may promote further rational design of CoS-based nanostructures for precise/efficient cancer diagnosis and therapy.
A Semimetal-Like Molybdenum Carbide Quantum Dots Photoacoustic Imaging and Photothermal Agent with High Photothermal Conversion Efficiency

Abstract:Theranostic platforms integrating imaging diagnostic and therapeutic interventions into a single nanoplatform have attracted considerable attention for cancer-individualized therapies. However, their uncertain stability, complex pharmacokinetics, and intrinsic toxicology of multiple components hinder their practical application in clinical research. In this paper, stable and high-concentration molybdenum carbide quantum dots (Mo2C QDs) with a diameter of approximately 6 nm and a topographic height of about 1.5 nm were synthesized using a facile sonication-assisted liquid-phase exfoliation approach. The prepared Mo2C QDs exhibited a strong near-infrared (NIR) absorbance with a high molar extinction coefficient of 4.424 Lg−1cm−1 at 808 nm, a high photothermal conversion efficiency of 42.9%, and showed excellent performance on photoacoustic imaging. The Mo2C QDs had high stability and highly biocompatibility, with low cytotoxicity. Under NIR irradiation, a remarkable in vitro and in vivo therapeutic effect was obtained. Such a stable and biocompatible all-in-one theranostic nanoagent generated by facile synthesis that combines promising imaging guidance and effective tumor ablation properties may hold great potential for theranostic nanomedicine.

Abstract:A new generation of photothermal theranostic agents is developed based on PEGylated WS2 nanosheets. Bimodal in vivo CT/photoacoustic imaging reveals strong tumor contrast after either intratumoral or intravenous injection of WS2 -PEG. In vivo photothermal treatment is then conducted in a mouse tumor model, achieving excellent therapeutic efficacy with complete ablation of tumors. This work promises further exploration of transition-metal dichalcogenides for biomedical applications, such as cancer imaging and therapy.

Abstract:A novel multifunctional theranostic agent has been successfully fabricated by loading iron oxide nanoparticles into poly(lactic acid) (PLA) microcapsules followed by surface functionalization with graphene oxide. Both in vitro and in vivo experiments proved that the resulting microcapsules could serve as contrast agents to simultaneously enhance ultrasound, magnetic resonance and photoacoustic imaging. The composite microcapsules show good biocompatibility and rapid response to magnetic fields. Due to the strong absorption of the near-infrared light, the composite microcapsules could efficiently kill cancer cells upon NIR laser irradiation. In addition, it was found that such a photothermal effect could be obviously enhanced by applying an external magnetic field. In a nutshell, this multifunctional microcapsule can be developed as a promising platform that integrates multimodality imaging and therapy capabilities for effective cancer theranostics.

Abstract:The promising potential of photodynamic therapy (PDT) has fueled the development of minimally invasive therapeutic approaches for cancer therapy. However, overcoming limitations in PDT efficacy in the hypoxic tumor environment and light penetration depth remains a challenge. We report the engineering of tungsten carbide nanoparticles (W2C NPs) for 1,064 nm laser-activated dual-type PDT and combined theranostics. The synthesized W2C NPs allow the robust generation of dual-type reactive oxygen species, including hydroxyl radicals (type I) and singlet oxygen (type II), using only single 1,064 nm laser activation, enabling effective PDT even in the hypoxic tumor environment. The W2C NPs also possess high photothermal performance under 1,064 nm laser irradiation, thus enabling synergistically enhanced cancer therapeutic efficacy of PDT and photothermal therapy. Additionally, the photoacoustic and X-ray computed tomography bioimaging properties of W2C NPs facilitate the integration of tumor diagnosis and therapy. The developed W2C based theranostic nanoagents increase the generation of reactive oxygen species in hypoxic tumors, improve the light penetration depth, and facilitate combined photothermal therapy and photoacoustic/computed tomography dual-mode bioimaging. These attributes could spur the exploration of transition metal carbides for advanced biomedical applications.

Abstract:Multifunctional cancer treatments based on gas therapy combined with other cancer treatments have gained tremendous attention and hold great promise in biomedical applications. In this study, a carbon monoxide-releasing nanoplatform combined with near-infrared (NIR) laser-triggered photothermal therapy (PTT) was constructed. The nanoplatform was composed of manganese pentacarbonyl bromide (MnCO)-loaded g-carbon nitride/polypyrrole (CNPpy) nanomaterials (MnCO@CNPpy). MnCO can be triggered to produce CO under H2O2 conditions. Upon exogenous NIR light stimulation and tumor microenvironment-overexpressed H2O2, MnCO@CNPpy exhibited excellent CO generation performance and photothermal effect. The generation of CO induced intracellular oxidative stress and caused cell apoptosis. Additionally, photoacoustic (PA) imaging was performed to track the delivery and accumulation of the nanomaterial in tumor sites because of the great photothermal conversion of CNPpy. The presented MnCO@CNPpy nanoplatform displayed desirable PTT and CO therapy in the inhibition of tumor growth and may provide a promising strategy for multifunctional antitumor synergistic treatments.

Abstract:The booming development of nanomedicine offers great opportunities for cancer diagnostics and therapeutics. Herein, a magnetic targeting-enhanced cancer theranostic strategy using a multifunctional magnetic-plasmonic nano-agent is developed, and a highly effective in vivo tumor photothermal therapy, which is carefully planed based on magnetic resonance (MR)/photoacoustic (PA) multimodal imaging, is realized. By applying an external magnetic field (MF) focused on the targeted tumor, a magnetic targeting mediated enhanced permeability and retention (MT-EPR) effect is observed. While MR scanning provides tumor localization and reveals time-dependent tumor homing of nanoparticles for therapeutic planning, photoacoustic imaging with higher spatial resolution allows noninvasive fine tumor margin delineation and vivid visualization of three dimensional distributions of theranostic nanoparticles inside the tumor. Utilizing the near-infrared (NIR) plasmonic absorbance of those nanoparticles, selective photothermal tumor ablation, whose efficacy is predicted by real-time infrared thermal imaging intra-therapeutically, is carried out and then monitored by MR imaging for post-treatment prognosis. Overall, this study illustrates the concept of imaging-guided MF-targeted photothermal therapy based on a multifunctional nano-agent, aiming at optimizing therapeutic planning to achieve the most efficient cancer therapy.

Abstract:Cancer is a severe disease, which have troubled human being for a long time. The development of nanotechnology has provided a new way for cancer treatment. It is a promising strategy to integrate imaging and therapeutic functions into one single nanoplatform to achieve efficient combination of diagnosis and treatment. Herein, we exploited novel CuMo2S3-PEG-Gd nanocomposites (NCs) for magnetic resonance imaging (MRI), guiding the photothermal therapy (PTT)/photodynamic therapy (PDT)/chemodynamic therapy (CDT). The experimental results showed that CuMo2S3-PEG-Gd NCs have a high photothermal conversion efficiency (40.6%), excellent biocompatibility and good biosecurity. The CuMo2S3-PEG-Gd NCs exhibited a clear MRI performance for tumor due to connecting Gd, which can guide in vivo therapy to improve the therapeutic effect. Moreover, both in vitro and in vivo therapeutic results of CuMo2S3-PEG-Gd NCs exhibited that the PTT/PDT/CDT achieved a remarkably synergistic effect, which could efficiently inhibit the tumor growth. Thus, CuMo2S3-PEG-Gd NCs, which integrated imaging with multiple therapies, have a good potential as theranostic agent for tumor.Graphical abstract

Abstract:Fabrication of nanotheranostic agents attracted a growing interest in accurate diagnosis and therapy of cancer by the sustainable biomineralization strategy. Herein, it is reported that biomineralized cobalt sulfide nanodots (Co9S8 NDs) with intrinsic peroxidase-like activity are potentially useful for synergetic phototherapy with tumor multimodal imaging navigation. The multifunctional Co9S8 NDs are first synthesized in aqueous phase via a simple biomineralization route. Detailed characterization reveals that the constructed peroxidase-like Co9S8 NDs not only possess strong paramagnetism with near-infrared absorption but also generate reactive oxygen species via photocatalytic reaction. As a result, the Co9S8 NDs reveal excellent performance for magnetic resonance and photoacoustic imaging of tumors, and outstanding tumor ablation efficiency owing to the near-infrared responsive photodynamic and photothermal effect. To our knowledge, ultrasmall Co9S8 NDs via aqueous phase synthesis have not been reported for multimodal theranostic applications so far.

Abstract:Abstract: Carbon-based nanomaterials (CBNM)have been widely used in various fields due to their excellent physicochemical properties. In particular, in the area of tumor diagnosis and treatment, researchers have frequently reported them for their potential fluorescence, photoacoustic (PA), and ultrasound imaging performance, as well as their photothermal, photodynamic, sonodynamic, and other therapeutic properties. As the functions of CBNM are increasingly developed, their excellent imaging properties and superior tumor treatment effects make them extremely promising theranostic agents. This review aims to integrate the considered and researched information in a specific field of this research topic and systematically present, summarize, and comment on the efforts made by authoritative scholars. In this review, we summarized the work exploring carbon-based materials in the field of tumor imaging and therapy, focusing on PA imaging-guided photothermal therapy (PTT) and discussing their imaging and therapeutic mechanisms and developments. Finally, the current challenges and potential opportunities of carbon-based materials for PA imaging-guided PTT are presented, and issues that researchers should be aware of when studying CBNM are provided.

Snxwo3 As A Theranostic Platform For Realizing Multi-Imaging-Guided Photothermal/Photodynamic Combination Therapy

Abstract:Precise oncotherapy requires effective cancer treatments that are guided by clinical imaging techniques. One of the most representative cases is multi-imaging-guided phototherapy. This study presents a novel multifunctional theranostic agent of SnxWO3 tungsten bronze, which is an excellent light absorber in the near infrared (NIR) range. Theoretical calculations based on density functional theory confirm that the insertion of donor Sn atoms into orthorhombic WO3 gives rise to the broadband visible-NIR absorption. Accordingly, both the photothermal effect and reactive oxygen species (ROS) production could be realized under NIR light irradiation by SnxWO3 tungsten bronze nanocrystals, thereby triggering the potent in vivo photothermal and photodynamic synergistic therapy. Meanwhile, modified SnxWO3 tungsten bronze has the functions of photoacoustic imaging (PAI), X-ray computed tomography (CT) imaging and near-infrared fluorescence (NIRF) imaging for tumor detection as well. Finally, for investigating the antitumor mechanism of in vivo solid tumors, clinical imaging modalities of B-mode ultrasonography (US) and magnetic resonance imaging (MRI) are employed to monitor the tumor evolution process after the photo-treatment, verifying a typically liquefactive necrosis process. These results indicate that the SnxWO3 tungsten bronze nanostructure is a promising theranostic agent for imaging-guided cancer therapy.

Abstract:Fe5 C2 NPs exhibit a high contrast in magnetic resonance imaging (MRI), superior photoacoustic tomography improvements, and efficient photothermal therapy (PTT) due to their unique core/shell structure, with a magnetic core and carbon shell. By conjugating a new class of affinity proteins (ZHER2:342), they can target to tumor cells with low cytotoxicity, and kill them through laser irritation. It is also possible to ablate tumors under guidance by MRI and PTT without noticeable side effects.
