Abstract:Protein adsorption onto implant surfaces is of great importance for the regulation of implant bioactivity. Surface modification of implants is a promising way in the molecular design of biocompatible materials against nonspecific adsorption of proteins. On the basis of these fundamental facts, we focus in this work on the different behavior of protein adsorption on hydroxylated and nonhydroxylated rutile TiO2 (110) surfaces through molecular dynamics simulations. Our investigation indicates that the distribution of the water molecules at the interface induced by the surface modification plays an important role in the protein adsorption. The surface with modified hydroxyl groups was observed to have much greater affinity to the protein, as reflected by the larger protein-surface electrostatic interaction and by the larger amount of adsorbed residues. The highly ordered structure of the modified hydroxyl groups on the hydroxylated surface diminishes the possibility of hydrogen bond formation between the surface and the water molecules above it, which in turn makes it easier for the protein to move closer to the surface with hydroxyl modification.
Effects Due to Interadsorbate Interactions on the Dipeptide/TiO2 Surface Binding Mechanism Investigated by Molecular Dynamics Simulations

Abstract:Classical molecular dynamics simulations, supported by ab initio periodic calculations, were carried out to investigate peptide adsorption mechanisms onto a rutile (110) TiO2 layer in the presence of water molecules. Different binding modes, comprising multiple coordination to the titanium atoms, of several conformers, simultaneously adsorbed upon the surface, were analyzed in detail. In agreement with experimental and theoretical findings, peptide carbonyl oxygens and nitrogens were found to be possible coordination atoms. Local effects were responsible of adsorption and desorption events and intermolecular interactions induced conformational changes and reorientations of the molecules with respect to the surface that produced both strongly and weakly adsorbed species.

Abstract:Surface modification of titanium (Ti) using biomolecules has attracted much attention recently. In this study, a new strategy has been employed to construct a stable and bioactive coating on Ti. To this end, a derivative of hyaluronic acid (HA), i.e. HA-GRGDSPC-(SH), was synthesized. The disulfide-crosslinked Arg–Gly–Asp (RGD)-containing collagen/hyaluronic acid polyelectrolyte membrane (PEM) coating was then fabricated on Ti through the alternate deposition of collagen and HA-GRGDSPC-(SH) with five assembly cycles and subsequent crosslinking via converting free sulphydryl groups into disulfide linkages (RGD–CHC-Ti group). The assembly processes for PEM coating and the physicochemical properties of the coating were carefully characterized. The stability of PEM coating in phosphate-buffered saline solution could be adjusted by the crosslinking degree, while its degradation behaviors in the presence of glutathione were glutathione concentration dependent. The adhesion and proliferation of MC3T3-E1 cells were significantly enhanced in the RGD–CHC-Ti group. Up-regulated bone specific genes, enhanced alkaline phosphatase activity and osteocalcin production, the increased areas of mineralization were also observed in the RGD–CHC-Ti group. These results indicate that the strategy employed herein may function as an effective way to construct stable, RGD-containing bioactive coatings on Ti.
Preparation and biological evaluation of Nano-HA coating on tatinium surfaces

Abstract:Objective This study applied an electrochemical technology to prepare a nano-hydroxyapatite(nano-HA) coating and evaluated the effects of the nano-HA coating on osteoblast biological ability.Methods Titanium plates were roughened.Nano-HA coatings were deposited by the electrochemical technology.FSEM,FTIR and XRD analysis were used to observe topography and chemical composition of coatings.The biological activity was evaluated by the BMSCs cell culture.Results FSEM observation showed the rod-like crystals uniformity covered the porous surfaces,the diameter of which was 70～80nm.XRD and FTIR analysis showed this coating consisted of HA.Nano-HA coatings improved expression of ALP and OC.Conclusion Nano-HA coatings were deposited on rough titanium surfaces by the electrochemical technology and improved BMSCs into osteoblast.
Conformational Changes of Fibrinogen Adsorption onto Hydroxyapatite and Titanium Oxide Nanoparticles

Abstract:The effect of surface adsorption on protein structure is of increasing interest in the biological, medical, and industrial fields. The structure of fibrinogen, a major plasma protein, adsorbed onto TiO2 and HA was examined by employing differential scanning calorimetry (DSC), circular dichroism (CD), and fluorescence spectroscopy. It was found that the TiO2 surface slightly decreased the ordered secondary structure of the protein; the fibrinogen conformation further changed upo washing and desorption. The alpha-helix content decreased gradually during the adsorption, washing, and desorption processes. The results were also used to estimate the overall structure of the protein in the adsorbed and desorbed states. It is significant that the fibrinogen transition enthalpy increased in each case upon adsorption onto HA. The transition enthalpy increased upon adsorption onto TiO2 in two out of three cases tested. These results, combined with the DSC thermograms of fibrinogen at different ionic strengths, suggest that electrostatic interactions are the main mechanism controlling the adsorption of fibrinogen to TiO2 and HA. Abstract:Titanium dioxide (TiO2) nanorods and calcium phosphate (CaP) composite coatings were prepared and characterized. TiO2 nanorod films were grown on titanium substrates via a hydrothermal method. Then, calcium phosphate was deposited into TiO2 nanorod films electrochemically in an aqueous electrolyte containing calcium and phosphorus species. As a result, CaP/TiO2 nanorod coatings were obtained. The amount of calcium phosphate deposited could be controlled through varying temperature, voltage and duration. It was found that both the presence of calcium phosphate and UV treatment could affect the surface wettability of the composite coatings. A bovine serum albumin adsorption test showed that the protein adsorption ability of the composite coatings was also improved with the presence of calcium phosphate and UV treatment. Moreover, the presence of calcium phosphate could promote the deposition of bone-like minerals. It is suggested that such coatings may exhibit promising initial cellular responses and thus be a good alternate for bioactive coatings.
Behavior regulation of adsorbed proteins via hydroxyapatite surface texture control.

Abstract:The influence of nanometer-scale interfaces on proteins has received much attention in recent years. The dynamic behaviors of bone morphogenetic protein-7 (BMP-7) on a series of hydroxyapatite (HAP) surface textures were investigated to explore the influence of different surface textures using molecular dynamics (MD), steered molecular dynamics simulations (SMD), and quantum mechanics calculations. It is observed that the interaction energy curve from SMD simulations can exhibit the dynamic behavior of BMP-7 in detail. Both the type and the number difference of the adsorptive residues and the intensity discrepancy of interaction, which is induced by the specific texture of the HAP surface, could be uncovered from the energy curve qualitatively and semiquantitatively in this study. The largest conformational change occurs in the system 010+a. The quantum mechanics calculations suggest that there is a phenomenon of electron transfer from HAP to the groups of BMP-7 during the adsorption process. These findings suggest that surface-engineering techniques could be employed to directly control the texture of HAP surfaces in order to regulate the behavior of a protein adsorbed onto the nanometer-scale interface.
Effects of collagen modification on the osteogenic performance of different surface-modified titanium samples in vitro

Abstract:Objectives: The aim of this study was to evaluate the effects of collagen modification on the osteogenic performance of different surface-modified titanium, including alkaline etching, alkaline etching followed by silanization, and alkaline etching followed by dopamine modification. The proliferation, adhesion, and osteogenic differentiation abilities of MC3T3-E1 cells on the surfaces with collagen modification were analyzed and compared. Methods: Collagen was immobilized on the surfaces of pure titanium (Ti-C), alkaline-etched titanium (Ti-Na-C), alkaline-etched and silanized titanium (Ti-A-C), and alkaline-etched and dopamine-modified titanium (Ti-D-C), with pure titanium (Ti) as the control group. The surface morphology was observed by scanning electron microscopy (SEM), and the surface elemental composition was analyzed by X-ray photoelectron spectroscopy (XPS). Contact angle measurements were conducted to evaluate the hydrophilicity of the surfaces. MC3T3-E1 cells were cultured on the surfaces, and their proliferation, adhesion, and osteogenic differentiation abilities were assessed using CCK-8 assay, laser scanning confocal microscope, alkaline phosphatase (ALP) staining, Alizarin red staining and quantitative analysis, as well as real-time quantitative polymerase chain reaction (RT-qPCR) to evaluate the mRNA expression levels of osteogenic-related genes, including ALP, typeⅠcollagen (COL-1), osteocalcin (OCN), osteopontin (OPN). Results: SEM and XPS results confirmed the successful immobilization of collagen on the titanium surfaces, with the Ti-Na-C group exhibiting a higher amount of collagen modification. Contact angle measurements showed improved hydrophilicity of the surfaces after collagen modification. CCK-8 results indicated good compatibility of the materials with MC3T3-E1, with enhanced cell proliferation on the collagen-modified surfaces. Cell fluorescence staining revealed better cell spreading on the collagen-modified surfaces, and ALP and Alizarin red staining results suggested that the Ti-Na-C group exhibited the best osteogenic performance, with significantly higher absorbance values in the Alizarin red quantification analysis. RT-qPCR analysis showed that the Ti-Na-C group had the highest expression of the osteogenic-related gene OPN. Conclusions: Among the different collagen modification approaches employed in this study, collagen modification on alkaline-etched titanium surfaces showed the most conducive effects on MC3T3-E1 cell adhesion, spreading, proliferation, and osteogenic differentiation. This approach can be considered as the optimal collagen modification strategy for enhancing osteogenesis on titanium surfaces.
Preparation and Characterization of TiO2/silicate Hierarchical Coating on Titanium Surface for Biomedical Applications.

Abstract:In the current work, TiO2/silicate hierarchical coatings with various nanostructure morphologies were successfully prepared on titanium substrates through micro-arc oxidation (MAO) and subsequent hydrothermal treatment (FIT). Moreover, the nucleation mechanism and growth behavior of the nanostructures, hydrophilicity, protein adsorption and apatite-inducing ability of various coatings were also explored. The novel TiO2/silicate hierarchical coatings comprised calcium silicate hydrate (CSH) as an outer-layer and TiO2 matrix as an inner-layer.According to the morphological features, the nanostructures were classified as nanorod, nanoplate and nanoleaf. The morphology, degree of crystallinity and crystalline phases of CSH nanostructures could be controlled by optimizing the HT conditions. The nucleation of CSH nanostructures is caused by release and re-precipitation mechanism. The TiO2/CSH hierarchical coatings exhibited some enhanced physical and biological performances compared to MAO-fabricated coating. The improvement of the hydrophilicity, fibronectin adsorption and apatite-inducing ability was found to be morphological dependent according to the following trend: nanoleaf coating > nanoplate coating > nanorod coating > MAO coating. The results indicate that the tuning of physical and morphological properties of nanostructures coated on biomaterial surface could significantly influence the hydrophilicity, protein adsorption and in vitro bioactivity of biomaterial. Surface modification by pre-adsorption of proteins and polypeptides on Ti substrate with controlled hydrophilicity to improve biocompatibility

Abstract:The water contact angle (WCA) value of the Ti substrate surfaces was successfully controlled to high hydrophilicity (≤ 20 deg.) by the treatment of UV light irradiation, atmospheric plasma or hydrothermal treatment, and stored in different environments to control the variation in WCA over time. Proteins with various molecular mass were used to investigate the protein adsorption on the Ti substrate controlled WCA. ATR-FTIR was employed to quantify the protein adsorption amount. It was found that all proteins had relatively higher adsorption amount on superhydrophilic surface. The adsorption amount of proteins was similar under the same WCA value. PEGs , as organic molecules without peptide bonds, was unable to be adsorbed on Ti with WCA 90 deg. In particular, we have carried out in vivo tests upon the protein pre-adsorbed Ti specimens. The in vivo tests indicated that the Ti substrate which pre-adsorbed proteins with high molecular mass had much higher osteoconductivity than that without protein adsorption. On the other hand, the PEGs adsorbed substrates showed no effect on osteoconductivity enhancement. This was due to its lack of peptide bonds. It was considered that implants adsorbed polypeptides can greatly enhance the osseointegration, regardless the mass and type of molecules. In general, the adsorption of protein and polypeptide containing amino acid is an effective way to improve osseointegration, before implantation into body environment. It provides crucial guidance to the biocompatibility improvement of the metallic bone implant materials.

Abstract:The osteo-bioactive potential of biomaterials can be modulated by altering material properties such as chemical composition, surface topography, and geometry. The correlation between the physicochemical properties of biomaterials and their osteo-bioactivity is highly complex. As a material widely used in bone repair, the structure-activity/dose-effect relationship between titanium (Ti) surface features and osteo-bioactivity remains unclear. To quantitatively enhance the osteogenic activity of Ti, we employed femtosecond laser (FSL) technology to create Ti surfaces with gradient changes in bioactive (nucleation) sites. Based on the apatite deposition ability on the etched surfaces, the quantitative relationship between the osteo-bioactivity of Ti surfaces and their characteristic parameters was systematically explored. Concurrently, classical molecular dynamics (MD) simulations were utilized to investigate the aggregation behavior of calcium and phosphate ions on the Ti surfaces with different sites. The findings from mineralization experiments revealed that the type and density of bioactive site could influence the deposition of apatite. It further identifies TiO2 and Ti-OH as the crucial bioactive sites, with basic Ti-OH exhibiting superior efficacy as a bioactive site compared to its acidic counterpart. Moreover, the bioactive (nucleation) site densities should maintain at a minimum of 2.33 ± 0.55/nm2 for TiO2 and 2.50 ± 0.59/nm2 for -OH, ensuring satisfactory osteo-bioactivity on the Ti surface. The results provide the atomic/molecular features of Ti surfaces that effectively foster apatite deposition and bioactivity, promoting the rapid progression of titanium-based bone repair materials.

Abstract:The reference peptide's side groups (polylysine, PL(–NH 2 )) were changed to create the carboxy-grafted polylysine (PL-COOH), and Methyl-grafted polylysine (PL-CH 3 ) peptides. The influence of polypeptides with different side groups on the surface mineralization behavior of hydroxyapatite/tricalcium phosphate (HA/TCP) biphasic ceramics was investigated using the static immersion mineralization method of simulated bodily fluid and the dynamic mineralization method based on quartz crystal microbalance (QCM-D) technology. Experiments revealed that changing the side groups of the peptides altered their secondary structure as well as the adsorption behavior on the substrate surface. Furthermore, the hydrophilicity and hydrophobicity of the substrate surface were altered by peptide adsorption. These structural and property changes impacted Ca 2+ and PO 4 3- concentrations, and thus governed biomineralization. This study will offer fresh perspectives on bone implant material surface design.

Abstract:Surface structure and composition play essential roles in the osseointegration of titanium implants. In the present study, a nanoscale surface structure incorporated with calcium ions was fabricated on a titanium surface by hydrothermal treatment. The characteristics of the surfaces were analysed, and the bioactivity of the samples was evaluated in vitro and in vivo . nm-Ti and nm/Ca-Ti surfaces were significantly more hydrophilic than control-Ti surfaces. nm/Ca-Ti samples showed much faster bone-like apatite precipitation in simulated body fluid than the other samples. The results of MC3T3-E1 cell tests demonstrated that both nm-Ti and nm/Ca-Ti surfaces accelerated cell adhesion and proliferation. The highest level of osteogenesis-related genes (Runx2, bone morphogenetic protein-2, osteopontin and osteocalcin) were observed in nm/Ca-Ti samples, followed by nm-Ti samples. Alizarin red staining experiment showed that the amount of extracellular matrix mineralized nodules in nm/Ca-Ti group was significantly higher than others. In animal experiments using SD rats, nm/Ca-Ti showed the highest value of new bone formation at two and four weeks. The present study suggested that the nanostructure and calcium ions showed synergetic effects on accelerating bone-like apatite precipitation and osteoblast cell growth and differentiation. Animal experiment further indicated that such surface could promote early osteogenesis.

Abstract:The understanding of interfaces and interaction of organic molecules and inorganic materials are the important issues in biomineralization. Experimentally, it has been found that amino acids (AA) can regulate the morphology of hydroxyapatite (HAP) crystals significantly. In this study, molecular dynamics simulation is employed to investigate the detailed adsorption behavior of polar, ionic, and hydrophobic AA on the {100} face of HAP at the atomic level. The results indicate that various AA are adsorbed on the HAP crystal surface mainly by amino and carboxylate groups at the specific sites. Multiple interaction points are found for polar and ionic AA. The adsorbed AA molecules occupy the Ca and P sites of the HAP surfaces which may inhibit and regulate the HAP growth. The adsorbed amino acid layer can also change the interfacial hydration layer and influence the transportation of ions in and out of HAP, which may be another strategy of biological control in biomineralization.
Mesoscopic Model for Reversible Adsorption Stage of Albumin and Fibrinogen on TiO2 Surface.

Abstract:The competitive behavior of proteins in the reversible adsorption stage plays a crucial role in determining the composition of the protein layer and the subsequent biological responses to the biomaterial. However, such competitive adsorption is a mesoscopic process at physiological protein concentration, and neither a macroscopic experiment nor microscopic MD (molecular dynamics) simulation is suitable to clarify it. Here, we proposed a mesoscopic DPD (dissipative particle dynamics) model to illustrate the competitive process of albumin and fibrinogen on TiO2 surface with its parameters deduced from our previous MD simulation, and proved the model well retained the diffusion and adsorption properties of proteins in the competitive adsorption on the plane surface. We then applied the model to the competitive adsorption on the surfaces with different nanostructures and observed that when the nanostructure size is much larger than that of protein, the increase in surface area is the main influencing factor; when the nanostructure size is close to that of protein, the coordination between the nanostructure and the size and shape of protein significantly affects the competitive adsorption process. The model has revealed many mechanical phenomena observed in previous experimental studies and has the potential to contribute to the development of high-performance biomaterials.

Abstract:Functionalization of Titanium implants using adequate organic molecules is a proposed method to accelerate the osteointegration process, which relates to topographical, chemical, mechanical, and physical features. This study aimed to assess the potential of a peptide derived from cementum attachment protein (CAP‐p15) adsorbed onto aTiO2 surfaces to promote the deposition of calcium phosphate (CaP) minerals and its impact on the adhesion and viability of human periodontal ligament cells (hPDLCs). aTiO2 surfaces were synthesized by magnetron sputtering technique. The CAP‐p15 peptide was physically attached to aTiO2 surfaces and characterized by atomic force microscopy, fluorescence microscopy, and water contact angle measurement. We performed in vitro calcium phosphate nucleation assays using an artificial saliva solution (pH 7.4) to simulate the oral environment. morphological and chemical characterization of the deposits were evaluated by scanning electronic microscopy (SEM) and spectroscopy molecular techniques (Raman Spectroscopy, ATR‐FTIR). The aTiO2 surfaces biofunctionalized with CAP‐p15 were also analyzed for hPDLCs attachment, proliferation, and in vitro scratch‐healing assay. The results let us see that the homogeneous amorphous titanium oxide coating was 70 nanometers thick. The CAP‐p15 (1 μg/mL) displayed the ability to adsorb onto the aTiO2 surface, increasing the roughness and maintaining the hydrophilicity of the aTiO2 surfaces. The physical adsorption of CAP‐p15 onto the aTiO2 surfaces promoted the precipitation of a uniform layer of crystals with a flake‐like morphology and a Ca/P ratio of 1.79. According to spectroscopy molecular analysis, these crystalline deposits correspond to carbonated hydroxyapatite. Regarding cell behavior, the biofunctionalized aTiO2 surfaces improved the adhesion of hPDLCs after 24 h of cell culture, achieving 3.4‐fold when compared to pristine surfaces. Moreover, there was an increase in cell proliferation and cell migration processes. Physical adsorption of CAP‐p15 onto aTiO2 surfaces enhanced the formation of carbonate hydroxyapatite crystals and promoted the proliferation and migration of human periodontal ligament‐derived cells in in vitro studies. This experimental model using the novel bioactive peptide CAP‐p15 could be used as an alternative to increasing the osseointegration process of implants.

Abstract:The nanoscale surface topography of biomaterials can have strong effects on protein adsorption. While there are numerous surface statistical parameters for the characterization of nanorough surfaces, none of them alone provides a complete description of surface morphology. Herein, a selection of nanorough titanium oxide surfaces has been fabricated with root-mean-square roughness (Sq) values below 2.7 nm but very different surface morphologies. The adsorption of the proteins myoglobin (MGB), bovine serum albumin (BSA), and thyroglobulin (TGL) at these surfaces was investigated in situ by ellipsometry to assess the importance of six of the most common surface statistical parameters. For BSA adsorption, both protein film thickness and time constant of adsorption were found to scale linearly with Sq s. For TGL, however, the same adsorption characteristics depend linearly on the surface skewness (Ssk), which we attribute to the rather extreme size of this protein. Finally, a mixed behavior is observed for MGB adsorption, showing different linear correlations with Sq and Ssk. These results demonstrate the importance of a thorough morphological characterization of the surfaces employed in protein adsorption and possibly also cell adhesion studies.

Abstract:In serval experimental researches, UV-induced hydrophilicity enabled better hemocompatibility in the TiO2 surface, which was considered to be caused by the removal of the carboxylic acid contamination from the surface. In this paper, we altered the surface wetting property by applying the formate contamination on the rutile (110) surface, and systematically investigated the adsorption properties of albumin and fibrinogen on hydrophilic/hydrophobic TiO2 surface. Unique contacts were found between the charged residues and the hydrophilic surface, anchoring the protein on the surface. The small size and the heart shape of albumin make it easy to cross the stable water layers near the surface. Besides, albumin has a higher proportion of charged residues, so it can form more unique contacts on the hydrophilic surface. Therefore, the albumin tends to adsorb on the hydrophilic surface. For the hydrophobic surface, the water layers near the surface are weakened, which helps the fibrinogen diffusing to the surface and adjusting its orientation. Although the hydrophobic surface cannot form the unique contacts, the larger size of fibrinogen can provide more residues to form enough ordinary contacts after adjusting, and then achieves stable adsorption. Therefore, fibrinogen tends to adsorb on the hydrophobic surface.

Abstract:Surface modification of medical implants is considered as an effective method to improve cellular behaviors and the integration of tissues with materials. Titanium (Ti)-based materials with four different micro/nano-structures and compositions were prepared by acid etching, electrochemical anodization and alkali-heat treatment. The surface morphologies and compositions of the different surface-modified Ti materials were characterized by field-emission scanning electron microscopy (FE-SEM), atomic force microscopy (AFM) and X-ray diffraction (XRD). The effects of the micro/nano structured and compositions of the surfaces on cellular responses were investigated in vitro by observing the morphology, adhesion, proliferation and osteogenic differentiation of osteoblasts. To further investigate the underlying mechanisms, an RT-PCR assay was performed to analyze the expression levels of cell adhesion-related genes. Our results indicated that the nanosized structure and anatase composition could promote the adhesion and proliferation of MC3T3-E1 pre-osteoblast, as well as alkaline phosphatase activity and extracellular matrix mineralization via the integrin-FAK signaling pathway. Taken together, our innovation presented in this work demonstrated that the surface nano-structure design and composition of biomedical implants can be modified of for future orthopaedic applications.
