An Osmosensitive Cation Channel Required for Hearing

Abstract:Life has evolved appropriate adaptive responses to maintain cell shape and volume when facing osmotic stress. Ca2+-dependent K+ or Cl- efflux plays a critical role in the regulatory volume decrease (RVD) under hypotonic stress; however, the molecular identities of the osmosensitive ion channels mediating initial Ca2+ influx remain unclear. Here, we identified an osmosensitive nonselective cation channel in the inner ear sensory hair cells. Genetic deletion results in necroptosis of outer hair cells (OHCs) and progressive hearing loss. We further analyzed its subcellular localization and functions in inner ear hair cells.
