Abstract:We report a novel turn-on two-photon fluorescent probe (TPNF) for detecting formaldehyde based on aza-Cope rearrangement and intramolecular charge transfer (ICT). The probe, containing a 1,8-naphthalimide dye with a homoallylamino group, has was shown with high selectivity, sensitivity and low cytotoxicity and it was capable of detecting formaldehyde in live cells. Moreover, a time-dependent density functional theory (TD-DFT) calculation was performed to demonstrate the turn on mechanism of the probe via ICT effect. Finally, a zebrafish larvae imaging experiment reveals that TPNF displayed a significant signal change when treated with FA in vivo. Abstract:Formaldehyde (FA) is endogenously produced in live systems and has been implicated in a diverse array of pathophysiological processes. To disentangle the detailed molecular mechanisms of FA biology, a reliable method for monitoring FA changes in live cells would be indispensable. Although there have been several fluorescent probes reported to detect FA, most are limited by the slow detection kinetics and the intrinsic disadvantage of detecting FA in an irreversible manner which may disturb endogenous FA homeostasis. Herein we developed a coumarin-hydrazonate based fluorogenic probe (PFM) based on a finely-tailored stereoelectronic effect. PFM could respond to FA swiftly and reversibly. This, together with its desirable specificity and sensitivity, endows us to track endogenous FA in live neurovascular cells with excellent temporal and spatial resolution. Further study in the brain tissue imaging showed the first direct observation of aberrant FA accumulation in cortex and hippocampus of Alzheimer's mouse model, indicating the potential of PFM as a diagnostic tool.

Abstract:Formaldehyde (FA), as the simplest endogenous carbonyl molecule, participates in many biosynthesis and metabolism in living organisms, such as nucleotides and adenosine triphosphate (ATP). FA concentrations are sub-millimolar in the normal healthy body, but can rise significantly in a number of disease pathologies. As a result, detecting endogenous FA is critical for illness diagnosis and rehabilitation therapy monitoring. Recent studies have focused on the FA-responsive turn-on fluorescence probe, which has huge promise in the detection and visualization of FA in living cells and organisms, as well as exceptional use in disease diagnosis and therapeutic monitoring. This review summarizes the fluorescence luminescence mechanism and design concepts of FA fluorescent probes, as well as their recent applications in bioimaging and illness diagnostics. Additionally, this article indicates the present dilemma of FA-responsive fluorescent probe, including selectivity, specificity, and detection mode, which may provide references for the development of FA-responsive fluorescent probes.

Abstract:Understanding the spatiotemporal dynamics of formaldehyde (FA) is crucial for elucidating its pathophysiology. In this study, we designed a series of organelle-resolved probes to investigate FA dynamics. By incorporating various organelle anchors into a coumarin hydrazonate, we developed a series of probes with excellent organelle selectivity and FA specificity, enabling rapid and precise sensing of FA in an organelle-resolved way. Leveraging these probes, we captured the spatiotemporal dynamics of native FA in response to exogenous FA or oxidative stress challenges. In particular, we unveiled the distinct responses of various organelles to identical cellular stressors. Moreover, we observed the dynamic response within individual organelles when cells were exposed to stressors for varying durations. We envision these probes will serve as versatile tools for probing FA pathophysiology.

Abstract:The linear photophysical, excited state absorption (ESA), superfluorescence, and two-photon absorption (2PA) properties of 4,4'-(1E,1'E)-2,2'-(7,7'(1E,1'E)2,2'(4,4'-sulfonylbis(4,1-phenylene))-bis(ethane-2,1-diyl)bis(9,9-didecy-9H-fluorene7,2-diyl))bis(ethane-2,1-diyl) bis(N,N-diphenylaniline) (1) were investigated in organic and aqueous media with respect to its potential application in biological imaging. The analysis of linear photophysical properties revealed a rather complex nature of the main one-photon absorption band, strong solvatochromic effects in the steady-state fluorescence spectra, single-exponential fluorescence decay, and high fluorescence quantum yields in organic solvents (approximate to 1.0). The ESA spectra of 1 suggested potential for light amplification in nonpolar media while efficient superfluorescence in cyclohexane was demonstrated. The degenerate 2PA spectra of 1 were obtained over a broad spectral range (640-900 nm), using a standard two-photon induced fluorescence method under 1 kHz femtosecond excitation. Two well defined 2PA bands with maximum 2PA cross sections up to 1700 GM in the higher energy, short wavelength band and approximate to 1200 GM in the lower energy, long wavelength band of 1 were shown. The potential use of 1 in bioimaging was demonstrated via one-and two-photon in vitro fluorescence imaging of HCT 116 cells.

Abstract:An important challenge for reaction-based fluorescent probes is that they generally require analyte consumption for fluorescence signal generation, thus creating potential perturbation of native analyte homeostasis or change of local concentrations. Herein, we reported two formaldehyde (FA) regeneration fluorescent probes, NAP-FAP-1 and NAP-FAP-2. An unprecedented regiospecific FA-induced intramolecularity strategy is implemented in the probe design, which adopts 3-(benzylamino)-succinimide as the FA-selective reaction group. The probes are able to capture the analyte molecule, induce regiospecific imide bond cleavage, and then release the captured FA molecule with simultaneous fluorescence turn-on response via a unique dual PeT/ICT quenching mechanism. The probes have shown potentials in detection, comparison, and imaging of FA levels intracellularly and inside lysosomes. These features make them useful for the study of FA homeostasis and functions in biological systems with minimal perturbation.

Abstract:Formaldehyde (FA) is one of reactive carbonyl species (RCS), which is involved in various physiological and pathological processes of organisms. Herein, a novel fluorescent probe ( FAP1 ) containing lysosomal localization group for the detection of FA was designed and synthesized. The probe exhibited good fluorescence recognition performances for FA, such as large Stokes shift (130 nm), specific selectivity, good anti-interference ability, and low minimum detection limit (16 nM). It is worth mentioning that the probe has good biocompatibility and could be applied to the imaging detection of FA in lysosome.

The probe has the advantages of simple synthesis, convenient use, strong atomic economy, good biocompatibility, and photobleaching resistance. Abstract:Formaldehyde (FA) is one of the most important reactive carbonyl species. It is not only exogenously generated from the environment but also continuously produced in most organisms. Low concentrations of FA take part in several important physiological processes, while elevated levels of FA are associated with different kinds of diseases. Fluorescent probes have emerged as promising chemical tools for FA detection and real-time visualization in food, air, and living organisms, etc., enabling the discovery of the diverse functions of FA in physiology and pathology processes. This review will summarize recent progress in the development of FA probes, including both small-molecular organic FA probes and functional material-based probes. In addition, prospects for future design and research will be also discussed.

Abstract:Increasing evidence has highlighted the endogenous production of formaldehyde (FA) in a variety of fundamental biological processes and its involvement in many disease conditions ranging from cancer to neurodegeneration. To examine the physiological and pathological relevance and functions of FA, fluorescent probes for FA imaging in live biological samples are of great significance. Herein we report a systematic investigation of 2-aza-Cope reactions between homoallylamines and FA for identification of a highly efficient 2-aza-Cope reaction moiety and development of fluorescent probes for imaging FA in living systems. By screening a set of N-substituted homoallylamines and comparing them to previously reported homoallylamine structures for reaction with FA, we found that N-p-methoxybenzyl homoallylamine exhibited an optimal 2-aza-Cope reactivity to FA. Theoretical calculations were then performed to demonstrate that the N-substituent on homoallylamine greatly affects the condensation with FA, which is more likely the rate-determining step. Moreover, the newly identified optimal N-p-methoxybenzyl homoallylamine moiety with a self-immolative β-elimination linker was generally utilized to construct a series of fluorescent probes with varying excitation/emission wavelengths for sensitive and selective detection of FA in aqueous solutions and live cells. Among these probes, the near-infrared probe FFP706 has been well demonstrated to enable direct fluorescence visualization of steady-state endogenous FA in live mouse brain tissues and elevated FA levels in a mouse model of breast cancer. This study provides the optimal aza-Cope reaction moiety for FA probe development and new chemical tools for fluorescence imaging and biological investigation of FA in living systems.

Abstract:Formaldehyde (FA) is endogenously produced in living systems through a variety of biological processes and has been implicated in many pathological conditions. Detection tools for biological FA are therefore of great interest. Reported here are novel activity-based genetically encoded fluorescent and luminescent probes for detecting FA in aqueous solutions and living mammalian cells. A FA-reactive lysine analogue, PrAK, was site-specifically incorporated into the essential lysine sites of enhanced green fluorescent protein (EGFP) and firefly luciferase (fLuc) to afford fluorescent and luminescent FA probes, respectively. FA selectively reacts with PrAK residues on EGFP and fLuc through a 2-aza-Cope rearrangement, resulting in fluorescence and luminescence turn-on responses, respectively, to FA selectively over potentially interfering reactive species in aqueous buffer. Moreover, the genetically encoded probes are capable of visualizing FA at physiologically relevant levels in living mammalian cells by fluorescence and luminescence imaging, demonstrating their potential as new tools to explore FA biology.

Abstract:As a reactive carbonyl molecule, formaldehyde (FA) plays an important part in plant physiological and pathological activities. It is necessary to probe FA in plant for the study of its functions. In this paper, we report DISE , a dicyanoisophorone-based turn-on near-infrared fluorescent probe, which exhibits a highly sensitive response to FA. In addition, confocal imaging experiments showed that DISE could image endogenous and exogenous FA in Arabidopsis root tissue. It demonstrates that the probe has the potential to be an imaging tool to research the FA's biological roles in plant.

Abstract:Epilepsy is a chronic neurodegenerative disease, and accumulating evidence suggests its pathological progression is closely associated with peroxynitrite (ONOO-). However, understanding the function remains challenging due to a lack of in vivo imaging probes for ONOO- determination in epileptic brains. Here, the first near-infrared imaging probe (named ONP) is presented for tracking endogenous ONOO- in brains of kainate-induced epileptic seizures with high sensitivity and selectivity. Using this probe, the dynamic changes of endogenous ONOO- fluxes in epileptic brains are effectively monitored with excellent temporal and spatial resolution. In vivo visualization and in situ imaging of hippocampal regions clearly reveal that a higher concentration of ONOO- in the epileptic brains associates with severe neuronal damage and epileptogenesis; curcumin administration can eliminate excessively increased ONOO-, further effectively protecting neuronal cells. Moreover, by combining high-content analysis and ONP, a high-throughput screening method for antiepileptic inhibitors is constructed, which provides a rapid imaging/screening approach for understanding epilepsy pathology and accelerating antiseizure therapeutic discovery.

Abstract:A two-photon absorbing (2PA) and aggregation-enhanced near-infrared (MR) emitting pyran derivative, encapsulated in and stabilized by silica nanopartides (SiNPs), is reported as a nanoprobe for two-photon fluorescence microscopy (2PFM) bioimaging that overcomes the fluorescence quenching associated with high chromophore loading. The new SiNP probe exhibited aggregate-enhanced emission producing nearly twice as strong a signal as the unaggregated dye, a 3-fold increase in two-photon absorption relative to the DFP in solution, and approximately 4-fold increase in photostability. The surface of the nanoparticles was functionalized with a folic acid (FA) derivative for folate-mediated delivery of the nanoprobe for 2PFM bioimaging. Surface modification of SiNPs with the FA derivative was supported by zeta potential variation and H-1 NMR spectral characterization of the SiNPs as a function of surface modification. In vitro studies using HeLa cells expressing a folate receptor (FR) indicated specific cellular uptake of the functionalized nanopartides. The nanoprobe was demonstrated for FR-targeted one-photon in vivo imaging of HeLa tumor xenograft in mice upon intravenous injection of the probe. The FR-targeting nanoprobe not only exhibited highly selective tumor targeting but also readily extravasated from tumor vessels, penetrated into the tumor parenchyma, and was internalized by the tumor cells. Two-photon fluorescence microscopy bioimaging provided three-dimensional (3D) cellular-level resolution imaging up to 350 mu m deep in the HeLa tumor.

Abstract:Formaldehyde (FA) is endogenously generated via fundamental biological processes in living systems. Aberrant FA homeostasis in subcellular microenvironments is implicated in numerous pathological conditions. Fluorescent probes for detecting FA in specific organelles are thus of great research interest. Herein, we present a modular strategy to construct diverse organelle-targeting FA probes by incorporating selective organelle-targeting moieties into the scaffold of a 1,8-naphthalimide-derived FA fluorescent probe. These probes react with FA through the 2-aza-Cope arrangement and exhibit highly selective fluorescence increases for detecting FA in aqueous solutions. Moreover, these organelle-targeting probes, i.e., FFP551-Nuc, FFP551-ER, FFP551-Mito, and FFP551-Lyso, allow selective localization and imaging of FA in the nucleus, endoplasmic reticulum, mitochondria, and lysosomes of live mammalian cells, respectively. Furthermore, FFP551-Nuc has been successfully employed to monitor changes of endogenous FA levels in the nucleus of live mammalian cells. Overall, these probes should represent new imaging tools for studying the biology and pathology associated with FA in different intracellular compartments.

Abstract:Formaldehyde (FA) is an endogenous one-carbon metabolite and an environmental pollutant and carcinogen. Elevated FA levels are associated with many diseases. Methods for the convenient and in situ detection of FA levels are of great significance for understanding FA's biofunctions and signalling pathways. Herein, the NAP-FAP2 series of fluorescent probes for FA detection were developed based on FA-promoted C-N cleavage of 3-nitrophenylazanyl N-arylcarbamate via FA-induced intramolecularity, where the aryl group is the fluorophore 1,8-naphthalimide-4-yl. The 3-nitrophenylazanyl containing reactive group also functions as a fluorescence quenching group via a photo-induced electron transfer mechanism to generate turn-on fluorescence response upon reaction with FA. The probes were applied to explore FA level changes in erastin-induced ferroptosis, and it was found that the FA level increases intracellularly, but not in the endoplasmic reticulum, suggesting that the FA level increases in ferroptosis are not derived from lipid peroxidation.

Abstract:In neuroscience, fluorescence labeled two-photon microscopy is a promising tool to visualize ex vivo and in vivo tissue morphology, and track dynamic neural activities. Specific and highly photostable fluorescent probes are required in this technology. However, most fluorescent proteins and organic fluorophores suffer from photobleaching, so they are not suitable for long-term imaging and observation. To overcome this problem, we utilize tetraphenylethene-triphenylphosphonium (TPE-TPP), which possesses aggregation-induced emission (AIE) and two-photon fluorescence characteristics, for neuroimaging. The unique AIE feature of TPE-TPP makes its nanoaggregates resistant to photobleaching, which is useful to track neural cells and brain-microglia for a long period of time. Two-photon fluorescence of TPE-TPP facilitates its application in deep in vivo neuroimaging, as demonstrated in the present paper.

Herein, we have designed a robust fluorescent probe ( Nu-DH ) targeted the nucleus to recognize FA. Nu-DH has high selectivity, large turn-on fluorescence (up to 120-fold) signal and sensitive detection for recognizing FA. Furthermore, in molecular docking calculation and titration experiments, Nu-DH has good affinity with DNA and RNA. Most importantly, the unique probe Nu-DH targets nucleus well and has been effectively implemented in zebrafish or in decoration materials for FA imagery. We found that many decoration materials will release a certain amount of FA, among which the FA release amount of white latex is the largest. We expect Nu-DH to become an effective tool for the diagnosis and monitoring of related diseases due to the influence of FA in vivo and in environment.
