Abstract:Stimulated emission depletion microscopy (STED) holds great potential in biological science applications, especially in studying nanoscale subcellular structures. However, multi-color STED imaging in live-cell remains challenging due to the limited excitation wavelengths and large amount of laser radiation. Here, we develop a multiplexed live-cell STED method to observe more structures simultaneously with limited photo-bleaching and photo-cytotoxicity. By separating live-cell fluorescent probes with similar spectral properties using phasor analysis, our method enables five-color live-cell STED imaging and reveals long-term interactions between different subcellular structures. The results here provide an avenue for understanding the complex and delicate interactome of subcellular structures in live-cell.

Abstract:For improving the imaging velocity and simplifying the system of chemical imaging sensors, a novel design of multi-channel light addressable potentiometric sensor based on microlens array was proposed. With microlens array, the single laser was separated into a line-array focused laser. Every focused laser was modulated separately to a different settled frequency by an optical chopper. The surface potential at all illuminated regions could be measured simultaneously by analyzing the resulting photocurrent. The results indicate that this system has excellent frequency resolution, and the IV characteristics can be easily obtained by digital compensation. This system can be used to light addressable potentiometric sensor and scanning photo-induced impedance microscopy, and allows for fast imaging by line-scanning. And with different sensing materials, this device can be used to detect several components of sample in parallel.

Abstract:Conventional fluorescence scanners utilize multiple filters to distinguish different fluorescent labels, and problems arise because of this filter-based mechanism. In this work we propose a line-monitoring, hyperspectral fluorescence technique which is designed and optimized for applications in multi-channel microfluidic systems. In contrast to the filter-based mechanism, which only records fluorescent intensities, the hyperspectral technique records the full spectrum for every point on the sample plane. Multivariate data exploitation is then applied to spectra analysis to determine ratios of different fluorescent labels and eliminate unwanted artifacts. This sensor is designed to monitor multiple fluidic channels simultaneously, providing the potential for multi-analyte biosensing. The detection sensitivity is approximately 0.81 fluors/μm2, and this sensor is proved to act with a good homogeneity. Finally, a model experiment of detecting short oligonucleotides has demonstrated the biomedical application of this hyperspectral fluorescence biosensor.

Abstract:Real-time fluorescence sensing can provide insight into biodynamics. However, few fluorescent tools are available to overcome the tissue scattering and autofluorescence interference for high-contrast in vivo sensing with high spatiotemporal resolution. Here, we develop a molecular-based FRET nanosensor (MFN) capable of producing a dynamic ratiometric NIR-IIb (1500-1700 nm) fluorescence signal under a frequency-modulated dual-wavelength excitation bioimaging system. The MFN provides reliable signals in highly scattering tissues and enables in vivo real-time imaging at micrometer-scale spatial resolution and millisecond-scale temporal resolution. As a proof of concept, a physiological pH-responsive nanosensor (MFNpH) was designed as a nanoreporter for intravital real-time monitoring of the endocytosis dynamics of nanoparticles in the tumor microenvironment. We also show that MFNpH allows the accurate quantification of pH changes in a solid tumor through video-rate ratiometric imaging. Our study offers a powerful approach for noninvasive imaging and sensing of biodynamics with micrometer-scale spatial resolution and millisecond-scale temporal resolution.

Abstract:Herein, we develop a novel method for designing electrochemical biosensors with both current and potential signal outputs for the simultaneous determination of two species in a living system. Oxygen (O 2 ) and pH, simple and very important species, are employed as model molecules. By designing and synthesizing a new molecule, Hemin‐aminoferrocene (Hemin‐Fc), we create a single electrochemical biosensor for simultaneous detection and ratiometric quantification of O 2 and pH in the brain. The reduction peak current of the hemin group increases with the concentration of O 2 from 1.3 to 200.6 μ m . Meanwhile, the peak potential positively shifts with decreasing pH from 8.0 to 5.5, resulting in the simultaneous determination of O 2 and pH. The Fc group can serve as an internal reference for ratiometric biosensing because its current and potential signals remain almost constant with variations of O 2 and pH. The developed biosensor has high temporal and spatial resolutions, as well as remarkable selectivity and accuracy, and is successfully applied in the real‐time quantification of O 2 and pH in the brain upon ischemia, as well as in tumor during cancer therapy.
Luminescent Silica Nanosensors for Lifetime Based Imaging of Intracellular Oxygen with Millisecond Time Resolution.

Abstract:Intracellular oxygen concentration was quantitatively imaged and rapidly traced with millisecond time resolution. We have demonstrated a new kind of oxygen nanosensors based on a ruthenium complex doped solid silica nanoparticles, which showed high oxygen sensing performance (I0/I100 = 3.29, t95 < 3 s) and ease of surface functionalization. Their sensing performance can be tuned by changing types of oxygen-sensitive probes and particle morphology. The nanosensors showed excellent control in both sensor size (from 30 to 200 nm), monodispersity, morphology, surface chemistry, and batch to batch consistency. Their uniform size distribution and good biocompatibility made them suitable for intracellular studies. Because the sensor surface can be easily functionalized with arbitrary units (such as transmembrane motifs, drugs, organelle-targeting groups, imaging reagent, and multiple sensor probes), these nanosensors provide a general platform to build easy-to-use tools for intracellular applications. The ease of surface functionalization was demonstrated by modifying the sensors outer surface with morpholinopropylamine and (3-carboxypropyl) triphenyl phosphonium, to actively target intracellular lysosomes and mitochondria of the tested cell lines (HeLa, MCF-7, and MCF-10A). Applying the mitochondria-targeting oxygen nanosensor together with our custom-built rapid phosphorescent lifetime imaging system, variations of intracellular oxygen have been quantitatively imaged and traced (in millisecond intervals) in real time and in situ.

Abstract:We report on the first dual nanosensors for imaging of pH values and oxygen partial pressure in cells. The sensors have a unique nanostructure in that a soft core structure is rigidized with a silane reagent, while poly(ethylene glycol) chains form an outer shell. Lipophilic oxygen-sensitive probes and reference dyes are encapsulated inside the hydrophobic core, while a pH-sensitive probe is covalently attached to the poly(ethylene glycol) end-group on the shell. The core/shell structure renders the nanosensors well dispersed and highly stable in various kinds of aqueous media. Their average size is 12 nm, and they respond to both pH and oxygen in the physiological range. They do not pass cell membranes, but can be internalized into the cellular cytosol by electroporation, upon which they enable sensing and imaging of pH values and oxygen with high spatial resolution. The nanosensor strategy shown here is expected to be applicable to the development of various other kinds of multiple nanosensors for in vivo studies.

Abstract:Living cells can rapidly adjust their metabolic activities in response to external stimuli, leading to fluctuations in intracellular temperature and reactive oxygen species (ROS) levels. Monitoring these parameters is essential for understanding cellular metabolism, particularly during dynamic biological processes. In this study, we present a bifunctional nanoprobe capable of simultaneous measurement of ROS levels and temperature within single cells. The nanoprobe features two individually addressable nanoelectrodes, with platinum (Pt) and nickel (Ni) coatings on both sides. At the tip, these two metal layers form a nano-thermocouple, enabling precise intracellular temperature measurements, while the Pt layer facilitates selective ROS detection. This dual functionality allows for real-time monitoring of cellular responses during synergistic chemo-photothermal therapy of cancer cells and zebrafish embryos subjected to mitochondrial toxic stress. Our results demonstrate that the nanoprobe effectively measures increases in temperature and ROS levels in HeLa cells undergoing chemo-photothermal therapy, as well as in chemically stimulated zebrafish embryos. By providing detailed analysis of submicrometer-scale temperature and ROS variations within living cells, this nanoprobe offers valuable insights into cellular processes and holds promise for early disease detection and drug development.

Abstract:We report the design of a phosphorescence/fluorescence dual-emissive nanoscale metal-organic framework (NMOF), R-UiO, as an intracellular oxygen (O2) sensor. R-UiO contains a Pt(II)-porphyrin ligand as an O2-sensitive probe and a Rhodamine-B isothiocyanate ligand as an O2-insensitive reference probe. It exhibits good crystallinity, high stability, and excellent ratiometric luminescence response to O2 partial pressure. In vitro experiments confirmed the applicability of R-UiO as an intracellular O2 biosensor. This work is the first report of a NMOF-based intracellular oxygen sensor and should inspire the design of ratiometric NMOF sensors for other important analytes in biological systems.

Abstract:An effective strategy is developed to realize a ratiometric oxygen nanoprobe with two-channel phosphorescence signals based on a core–shell silica nanoparticle. This nanoprobe exhibits good water dispersibility and biocompatibility. Bioimaging experiments demonstrate that the nanoprobe can be used for monitoring the change in intracellular O2 concentration and performing quantitative O2 measurement through ratiometric and time-resolved luminescence bioimaging. Abstract:Oxygen plays a crucial role in many biological processes. Accurate monitoring of oxygen level is important for diagnosis and treatment of diseases. Autofluorescence is an unavoidable interference in luminescent bioimaging, so that an amount of research work has been devoted to reducing background autofluorescence. Herein, a phosphorescent iridium(III) complex-modified nanoprobe is developed, which can monitor oxygen concentration and also reduce autofluorescence under both downconversion and upconversion channels. The nanoprobe is designed based on the mesoporous silica coated lanthanide-doped upconversion nanoparticles, which contains oxygen-sensitive iridium(III) complex in the outer silica shell. To image intracellular hypoxia without the interferences of autofluorescence, time-resolved luminescent imaging technology and near-infrared light excitation, both of which can reduce autofluorescence effectively, are adopted in this work. Moreover, gradient O2 concentration can be detected clearly through confocal microscopy luminescence intensity imaging, phosphorescence lifetime imaging microscopy, and time-gated imaging, which is meaningful to oxygen sensing in tissues with nonuniform oxygen distribution.

Abstract:Mitochondrial reactive oxygen species (ROS) and pH fluctuations are closely correlated with mitochondrial dysfunctions, which are implicated in various human diseases including neurodegenerative disorders and cancers. Simultaneously monitoring the changes of ROS and pH of mitochondria remains a major challenge in the mitochondrial biology. In this study, we develop a novel mitochondria-targeted fluorescent nanosensor for real-time imaging of the fluctuations of hydrogen peroxide (H2O2) and pH in living cells. The fluorescence probes for detecting pH and H2O2 were loaded in the small-sized mesoporous silica nanoparticles (MSN). Then the polyethylenimine was attached to cap the pores of MSN, the triphenylphosphonium was further modified to target mitochondria in living cells. Confocal fluorescence imaging indicated that the nanosensor could effectively target mitochondria and successfully achieved real-time imaging of mitochondrial H2O2 and pH fluctuations in living cells. Notably, this is a single nanosensing system that is capable of visualizing multiple subcellular analytes at the same time and position by multicolor fluorescence imaging. The current approach can provide a promising tool to investigate the interplaying roles of various subcellular analytes in living cells.

Abstract:A mitochondria-targeted fluorescent sensor (Mito-ST), constructed by integrating a sulfamoylbenzoxadiazole fluorophore with a phosphonium group, displays the specific Zn2+ -induced hypsochromic shifts of both excitation (69 nm) and emission (35 nm) maxima. Its ratiometric Zn2+ imaging ability via dual excitation mode has been applied in MCF-7 cells to clarify the different behaviours of mitochondrial Zn2+ release stimulated by H2O2 and SNOC.

Abstract:A novel and compact ion sensor was developed by combining 2D plasmonic K x MoO 3 and Na x WO 3 with hexagonal channel and photonic crystal nanobeam cavity (PCNC) with Fano resonance. Fano resonance PCNC with a deterministic air mode (mirror period > gradient periodic region) and dielectric mode (mirror period < gradient periodic region) with a period of 180 nm were designed, which can be fabricated on multi project wafer (MPW) processing. The sensing system exhibits an refractive index (RI) and temperature sensitivity of 234.48 nm / RIU and 96 nm / K, respectively. Three PCNCs adopted wavelength division multiplexing over a wide range of wavelengths. Utilizing the near-infrared absorption of degenerated plasma K x MoO 3 and Na x WO 3 and its hexagonal ring-like ion sieve, a temperature decoupled Sodium (Na + ) and potassium (K + ) was achieved. Here, Fano resonance originates from interference between a bus waveguide with continue mode, inserted with two air holes, and the discrete state photonic crystal nanobeam cavity. This work paves the way to achieve an ultra-compact lab-on-chip resonance-based photonic building block.

Abstract:A fluorescent capillary sensor for detecting the bioanalytically and biologically relevant analytes pH, O-2, and CO2 has been designed. The sensor is based on single capillary tube with 2 mm inner diameter, which is simultaneous doped with emissive O-2-sensitive indicator Ru(dpp)(3)(PF6)(2) (RuDP), pH-sensitive and CO2-sensitive indicator 8-hydroxypyrene-1, 3,6-trisulfonic acid trisodium salt (HPTS). The multifunctional sensor can be excited at the same wavelength and realize multi-parameter optical monitoring at the different sensing position. Studies in phosphate-buffered solutions display the excellent feasibility of the capillary sensor for fluorescence detection of pH, O-2, and CO2. Overall, the multifunctional capillary sensor exhibits great potential in real-time blood gas analysis, and clinical application of multi-parameter biological detection.

Abstract:Nanopore measurement has advanced in single-molecule analysis by providing a transient time and confined space window that only allows one interested molecule to exist. By optimization and integration of the electrical and optical analysis strategies in this transient window, the acquisition of comprehensive information could be achieved to resolve the intrinsic properties and heterogeneity of a single molecule. In this work, we present a roadmap to build a unified optical and electrochemical synchronous measurement platform for the research of a single molecule. We design a low-cost ultralow-current amplifier with low noise and high-bandwidth to measure the ionic current events as a single molecule translocates through a nanopore and combine a multi-functional optical system to implement the acquisition of the fluorescence, scattering spectrum, and photocurrent intensity of single molecule events in a nanopore confined space. Our system is a unified and unique platform for the protein nanopore, the solid-state nanopore, and the glass capillary nanopore, which has advantages in the comprehensive research of nanopore single-molecule techniques.

Abstract:In this paper, we propose a multi-channel photonic crystal fiber sensor, which adopts dual-polarization and multiple materials to effectively reduce the mutual interference between channels and enhance the surface plasmon resonance, thus achieving simultaneous detection of a multi-channel with low interference. Four channels are polished around the cylindrical fiber, and then different metal films (gold or silver) and plasmonic materials (titanium dioxide, thallium pentoxide, or graphene) are added to the sensing area of each channel. All channels detect refractive indices in the range of 1.34 to 1.42. The sensing performance of the fiber optic sensor was numerically investigated using the full vector finite element method. After the optimization of structural parameters, the maximum wavelength sensitivity of channel-1, channel-2, channel-3, and channel-4 are 49,800 nm/RIU, 49,000 nm/RIU, 35,900 nm/RIU, and 36,800 nm/RIU, respectively. We have theoretically analyzed the sensor’s capabilities for partial bio-detection and simulated its detection capability with a wavelength sensitivity of 11,500 nm/RIU for normal red blood cells and 12,200 nm/RIU for MCF-7 cancerous cells. Our proposed sensor has a novel design, can detect multiple channels simultaneously, has strong anti-interference capability and high sensitivity, and has good sensing characteristics.
