Isolation and Identification of the Antarctic Soil Bacillus and Its Biocontrol Effect Against Brown Stalk Rot on Maize

Abstract:[Objectives]This study was conducted to select the Bacillus strains from the Antarctic soil,which had good biocontrol effect against brown stalk rot on maize.[Methods]The Bacillus strains were isolated from penguin rookery in the vicinity of the Great Wall Station of the Antarctic soil.Combining with 16S rDNA and gyrB gene sequence,the strains were identified.We selected the Bacillus strains,which had antagonism effect significantly against the pathogenic bacteria of the brown stalk rot on maize,by the plate antagonistic test.It was confirmed by the field trials that the Bacillus strains which were selected from the plate antagonistic test had good potential in application.The identification of antibacterial lipopeptide compounds were conducted with MALDI-TOF-MS.[Results]Combining with 16S rDNA and gyrB gene sequence analysis,the 23 strains of Bacillus which were isolated from the Antarctic soil were identified as Bacillus amyloliquefaciens (9 isolates) and Bacillus licheniformis (14 isolates).Through the plate antagonistic test,we got 4 strains which inhibited the pathogen of the brown stalk rot on maize significantly.They were B.amyloliquefaciens EZ15-07,EZ15-09,and B.licheniformis EZ01-05,EZ15-08.The results of the field trial showed that the biocontrol effect of EZ15-07 and EZ01-05 reached to 59.3% and 55.3%,respectively.The lipopeptide compounds analysis showed that strain EZ15-07 produced Surfactin and Bacillomycin D,whereas strain EZ01-05 produced Bacillomycin D and Fengycin.[Conclusions]The Bacillus strains EZ15-07 and EZ01-05 obtained from the Antarctic soil had significant biocontrol effects against brown stalk rot on maize.Both of them had good potential in application.
