Abstract:A new synthesis of L-norvaline is described. Using valeric acid as the raw material, alpha-brominevaleryl chloride was achieved by acyl chlorination and alpha-position bromination in one-pot with the yield of 84.7%. The yields of the following ammoniation of alpha-brominevaleryl chloride and resolution of DL-norvaline were 88.7% and 26.7%, respectively. D-Norvaline was also obtained in a similar method from the waste resolution solution. Other optical active amino acids, valine, alpha-aminobutyric acid and alanine were also synthesized in similar ways.

Abstract:The syntheses and full characterization of chiral amine alcohol complexes of aluminum are reported. These complexes are of two general formulae, those resulting from the reaction of 1 equiv of an aluminum reagent with an amine alcohol, [AA-AlR(2)](2) (where AA = phenylglycinol (PGly), R = Me (1), Et (2), SiMe(3) (3); phenylalaninol (PAla), R = Me (4), Et (5), SiMe(3) (6); and diphenylalaninol (DPAla), R = Me (7), Et (8), SiMe(3) (9), and the series resulting from the reaction of 2 equiv of AlMe(3) with an amine alcohol, AA(AlMe(2))-AlMe(3) (where AA = PGly (10), PAla (11), and DPAla (12)). These compounds significantly increase the number of chiral group 13 complexes that have been reported in the literature. Additionally, the crystal structures of 5, 6, and 11 have been obtained.

Abstract:A green and fully enzymatic procedure for the resolution of chiral alcohols through lipase/esterase-catalyzed acylation and subsequent lipase-catalyzed aminolysis using anhydrous ammonia was demonstrated. Both enantiomers can be obtained in high ee values (up to >99%) under ambient reaction conditions. The solvent and acyl donors can be recycled, and the enzyme can be reused for up to five times.

Abstract:Two novel chrial bisaminoalcohols, 1, 3-bis-[(2' S)-(diphenylhydroxymethyl)pyrrolidin-1-yl]-2-propanone (3) and 1,3-bis-[(2'S)-(diphenylhydroxymethyl)pyrrolidin-1-yl]-2-propanol (4), were prepared using the 1, 3-dibromo-2-propanone and ( 21 S) - ( diphenylhydroxymethyl) pyrrolidine as the starting materials. The yields were 39% and 35%, respectively. The asymmetric direct aldol reactions of unmodified ketones and aldehydes were achieved with the ee up to 9% by using the bisaminoalcohol-diethylzinc as the catalyst.

Abstract:The sodium salts of(S)-2-(1-imidazolyl)alkanoic acids(2) were prepared by the cyclization of natural amino acids with formaldehyde aqueous solution and glyoxal aqueous solution under basic condition.The sodium salts(2) were esterified to form(S)-2-(1-imidazolyl)alkanoates(3a~3g).The(S)-2-(1-imidazolyl)alkanols(4a~4e) were obtaind by reduction of 3a~3e via NaBH_(4) with satisfactory yields.The structures of the compounds(3a~3g,4a~4e) were confirmed by IR,~(1)H NMR,MS,and elemental analysis.

Abstract:A family of ferrocenyl amino alcohols (14 examples) has been prepared from the corresponding planar chiral ketones and evaluated as a new class of chiral ligands for asymmetric ethyl‐ and phenylzinc additions to aldehydes. The highly enantioselective synthesis of these ligands is facile, scalable, robust and relies on the use of cheaply available starting materials. Asymmetric ethyl additions proceeded smoothly at ambient temperature, with reaction times as short as 1 hour furnishing the desired products in up to 90% yield and 99% ee. The corresponding phenyl additions afforded products in up to 81% yield and 88% ee. The observed stereochemical outcome for the addition of diethylzinc was rationalized and the key proposed transition state was supported by an analysis of the X‐ray structures of a series of amino alcohol ligands.

Abstract:Experimental studies on the modification of (1R,2S)-1,2-diphenyl-2-aminoethanol, which is used to promote the alkylation of N-diphenylphosphinoyl benzalimine with diethylzinc, revealed that N-monosubstituted amino alcohols exhibited higher enantioselectivities than their N,N-disubstituted counterparts and imino alcohols. Application of the optimal chiral ligand 3c to activate the reaction of N-diphenylphosphinoyl arylimines with diethylzinc and dibutylzinc resulted in excellent enantiomeric selectivities of up to 98% ee. The origin of the experimentally observed enantioselectivities was revealed by density functional calculations (B3LYP/6-31G*) on the transition structures of several model reactions.

Abstract:Under the catalysis of a Cu(I) or Ag(I) salt with a chiral diphosphine ligand, the enantioselective nucleophilic ring opening of racemic 2‐styrenylaziridines via kinetic resolution or dynamic kinetic asymmetric transformation (DyKAT) and of meso aziridines via desymmetrization with alcohols was realized. The reaction provided a range of chiral vicinal amino ether derivatives in 47%‐98% yields and with 50%‐98% ee. The reaction could be extended to aliphatic carboxylic acids as nucleophiles. The synthetic utility was demonstrated in a four‐step formal synthesis of a diastereomer of the antirythmetic agent vernakalant.

Abstract:Three novel sulfur-containing chiral β-amino alcohols 4a-4c have been prepared from S-alkylated L-cysteine 2a-2c. These compounds could be used as catalysts in the enantioselective reduction of prochiral ketones with the NaBH 4/I 2 system. The optically active secondary alcohols were obtained in moderate to high enantiomeric excesses and high chemical yields.

Abstract:Give Me an Ar, give Me an N! Arylation of the methyl group in a simple derivative of readily available alanine under palladium catalysis was followed by intramolecular amidation at the same position to give chiral α-amino-β-lactams with a wide range of aryl substituents (see scheme; Phth=phthaloyl). The α-amino-β-lactams were obtained in moderate to high yields with good functional-group tolerance and high diastereoselectivity.

Abstract:A novel chiral diallylic diamine has been synthesized through double addition of allyl zinc bromide to chiral imines in this contribution. Firstly, (S)-N-Boc-2-amino-3-phenylpropanaI was prepared through N-Boc protection and Dess-Martin oxidation. Then, it was condensed with (S)-valine methyl ester to form a chiral imine, which was reacted with allyl zinc bromide in the presence of CeC1(3)center dot 7H(2)O to afford the corresponding chiral homoallylic amine with high optical activity. After removing Boc group, the newly formed chiral homoallylic amine was condensed with 2-thiophenecarboxaldehyde to provide a novel chiral imine with high optical purity. The following asymmetric addition of allyl zinc bromide to the novel chiral imine provided the target compound. The structure and the purity of desired compound and the key intermediates were demonstrated by H-1 NMR, C-13 NMR and MS spectra.

Abstract:A highly stereoselective iodolactonization of 4-allenoic acids with a new sterically demanding electrophilic iodination reagent to afford optically active γ-butyrolactones has been developed. The reaction shows high efficiency of axial chirality transfer and excellent Z/E selectivity and has been applied to the synthesis of chiral cis-β,γ-disubstituted γ-butyrolactones to give very high diastereomeric and enantiomeric excess values. The reaction has been successfully utilized in the synthesis of naturally occurring compounds (+)-cis-whisky lactone and (+)-cis-3-methyl-4-decanolide.

Abstract:Chiral amino alcohols are prevalent synthons in pharmaceuticals and synthetic bioactive compounds. The efficient synthesis of chiral amino alcohols using ammonia as the sole amino donor under mild conditions is highly desired and challenging in organic chemistry and biotechnology. Our previous work explored a panel of engineered amine dehydrogenases (AmDHs) derived from amino acid dehydrogenase (AADH), enabling the one-step synthesis of chiral amino alcohols via the asymmetric reductive amination of α-hydroxy ketones. Although the AmDH-directed asymmetric reduction is in a high stereoselective manner, the activity is yet fully excavated. Herein, an engineered AmDH derived from a leucine dehydrogenase from Sporosarcina psychrophila (SpAmDH) was recruited as the starting enzyme, and the combinatorial active-site saturation test/iterative saturation mutagenesis (CAST/ISM) strategy was applied to improve the activity. After three rounds of mutagenesis in an iterative fashion, the best variant wh84 was obtained and proved to be effective in the asymmetric reductive amination of 1-hydroxy-2-butanone with 4-fold improvements in k cat /K m and total turnover number (TTN) values compared to those of the starting enzyme, while maintaining high enantioselectivity (ee >99%) and thermostability (T 50 15 >53°C). In preparative-scale reaction, the conversion of 100 and 200 mM 1-hydroxy-2-butanone catalyzed by wh84 was up to 91-99%. Insights into the source of an enhanced activity were gained by the computational analysis. Our work expands the catalytic repertoire and toolbox of AmDHs.

Asymmetric Deprotonation Using Polymer-Supported Chiral Lithium Amides

Abstract:Polymer-supported chiral lithium amides were prepared and used as chiral bases in the asymmetric deprotonation of prochiral cyclic ketones. The reaction of Merrifield resin 1 with chiral amines 2 in the presence of NaHCO3 gave the polymer-supported amines 3a and 3b (eq. 1). The polymer-supported chiral lithium amide 3a-Li was generated in situ from amine 3a with BuLi. The deprotonation of the prochiral cyclic ketones 4 with the chiral lithium amide 3a-Li and the reaction of the resulting lithium enolates with trimethylsilyl chloride gave the trimethylsilyl enol ethers 5 in 86-94% yield with moderate enantioselectivity (31-41% ee) (eq. 2). The reaction of 6 with 3-Li gave (S)-7 with high enantioselectivity (3a: 71% ee, 3b: 81% ee) (eq. 3). The polymer-supported amine 3b was recovered and reused five times.
