Abstract:Two bafilomycin derivatives with an unprecedented 6/5/5 tricyclic ring system (1 and 2), along with five known analogues (3-7), were identified from a marine Streptomyces sp. The structures of 1 and 2 were elucidated by analysis of HRESIMS, extensive NMR spectroscopic methods, GLAD DET C-13 NMR calculations and comparison of the spectroscopic data with those of known related compounds. Abstract:On the basis of the one strain-many compounds strategy, five compounds including two new holomycin derivatives 2 - 3, two new cyclopropaneacetic acid derivatives 4 - 5, together with one known compound holomycin (1) were isolated from a marine-derived bacterium Streptomyces sp. DT-A37. Their structures were elucidated using NMR and HR-ESI-MS analyses. All these compounds were evaluated for their antimicrobial activity, cytotoxic activity, and inhibitory activity against BRD4 protein. Compound 1 exhibited potent cytotoxicity against H1975 cells with IC50 value of 1 μm, and its minimal inhibitory concentration values against Escherichia coli and Staphylococcus aureus were both 64 μm.

Abstract:Cyclopiumolides A (1) and B (2), first representatives of two novel biosynthetic related 13-membered macrolides featuring an uncommon verrucosidinol unit condensed with a spiculisporic acidic moiety, were identified from the fungus Penicillium cyclopium SD-413, which was obtained from the deep-sea sediments collected in the East China Sea. The structures of cyclopiumolides A (1) and B (2) were identified on the basis of extensive NMR spectroscopic and mass spectrometric data analysis. Their relative and absolute configurations were determined by quantum mechanical calculations of ECD spectra comparing with that of experimental curves and by DP4 + NMR data calculations. Compounds 1 and 2 exhibited significant cytotoxic potencies against the tumor cell lines SF126, FaDu, and TE-1 with IC50 values ranging from 5.86 to 17.05 μM. The inhibition modes and binding sites of 1 and 2 were inspected using molecular docking simulations.

Abstract:Strain HM190, a moderate halophile, was isolated from rhizosphere soil of the mangroveKandelia obovatain Fugong village, China. The 16S ribosomal RNA (rRNA) gene sequence and the results of phylogenetic analysis revealed that strain HM190 belonged to the genusStreptomycesand had the highest sequence similarity of 99.79% toStreptomyces heilongjiangensisNEAU-W2(T). The complete genome of strain HM190 comprised 7,762,826 bp in a linear chromosome with 71.97% G + C content. According to antiSMASH analysis, a total of 30 biosynthetic gene clusters (BGCs) were predicted to be involved in secondary metabolism, 12 of which were responsible for the production of polyketide- and non-ribosomal peptide-derived secondary metabolites. Gene cluster 5 was responsible for macrolide biosynthesis in a strain-specific 126,331-bp genomic island belonging to the left-arm region. Combined genomics-metabolomics analysis led to the discovery of three 22-membered macrolides (compounds1-3). Their structures were elucidated by using spectroscopic techniques including high-resolution electrospray ionization mass spectroscopy (HRESIMS) and nuclear magnetic resonance (NMR). The absolute configurations of compounds1-3were determined by the X-ray single crystal diffraction and NMR data analysis. All three compounds displayed moderate cytotoxic activities toward tumor cell lines HepG2, A549, and HCT116.

Abstract:Alokicenones A-H (1-8), eight new tetrahydroanthracenes and one known okicenone (9) were identified from the secondary metabolites of mangrove-derived Streptomyces sp. HN-A101. Their structures were elucidated by HRESIMS and NMR spectroscopic data. The absolute configurations of them were determined by the calculated and experimental ECD curves. Compounds 1-2 and 9 showed moderate cytotoxicity against HCT116 and SW620 cancer cell lines with IC50 values from 0.63 to 7.73 mu M. Abstract:A macrocyclic polyketide with novel carbon skeleton, namely hainanmycin A, was isolated from the mangrove-derived Streptomyces sp. 219807. Abstract:Eight new cyclizidine-type alkaloids (1-8) and one known alkaloid (9) were identified from the chemical investigations of a marine-derived actinomycete, Streptomyces sp. HNA39. Among these alkaloids, compounds 3, 7, and 8 contain a chlorine atom, and the known alkaloid, (+)-ent-cyclizidine (9), is now first reported as a natural product. Their structures were elucidated by extensive NMR-spectroscopic analysis and HRESIMS data. The absolute configurations of all of the compounds were established by ECD calculations. Cytotoxicity evaluations of all of the compounds showed that compound 2 exhibited significant activity against the PC3 and HCT116 human-cancer-cell lines with IC50 values of 0.52 ± 0.03 and 8.3 ± 0.1 μM, respectively. Interestingly, compounds 2, 5, 7, and 8 exhibited moderate inhibition against the ROCK2 protein kinase with IC50 values from 7.0 ± 0.8 to 42 ± 3 μM.

Abstract:A pair of enantiomeric 4-oxabicyclic[4.3.0]lactam derivatives, (+)- and (-)-penicilactam A (1), and one new polyketide derivative penicitrinone F (2) were isolated from the marine-derived fungus Penicillium griseofulvum GT-10. Their structures and absolute configurations were elucidated through extensive spectroscopic analyses combined with the calculated ECD spectra. Penicitrinone F (2) had moderate inhibitory activity towards Bacillus subtilis with a MIC value of 6.3 μM.

Abstract:Polyene macrolactams are a special group of natural products with great diversity, unique structural features, and a wide range of biological activities. Herein, a cryptic gene cluster for the biosynthesis of putative macrolactams was disclosed from a sponge-associated bacterium, Streptomyces sp. DSS69, by genome mining. Cloning and heterologous expression of the whole biosynthetic gene cluster led to the discovery of weddellamycin, a polyene macrolactam bearing a 23/5/6 ring skeleton. A negative regulator, WdlO, and two positive regulators, WdlA and WdlB, involved in the regulation of weddellamycin production were unraveled. The fermentation titer of weddellamycin was significantly improved by overexpression of wdlA and wdlB and deletion of wdlO. Notably, weddellamycin showed remarkable antibacterial activity against various Gram-positive bacteria including MRSA, with MIC values of 0.10-0.83 mu g/mL, and antifungal activity against Candida albicans, with an MIC value of 3.33 mu g/mL. Weddellamycin also displayed cytotoxicity against several cancer cell lines, with IC50 values ranging from 2.07 to 11.50 mu M.

Abstract:The fungus strain SCSIO 40433 was isolated from an Arctic-derived glacier sediment sample and characterized as Tolypocladium cylindrosporum. A new compound, cylindromicin (1), and seven known secondary metabolites (2–8) were isolated from this strain. The chemical structures of these compounds were elucidated by comprehensive spectroscopic analyses. Cylindromicin (1) featured a 3,4-dihydro-2H-pyran skeleton. The absolute configuration of compound 1 was assigned via interpretation of key Nuclear Overhauser Effect Spectroscopy (NOESY) correlations and Electronic Circular Dichroism (ECD) calculation. Cylindromicin (1) exhibited significant tyrosinase inhibition activity. This study highlights Polar fungi as a potential resource for new bioactive natural products.

Abstract:Chemical fractionation of the EtOH extract of a medicinal macro fungus, Inonotus obliquus, afforded an array of lanostane-type triterpenoids (1-11) including two new ones (1 and 8). The structures of these compounds were determined on the basis of spectroscopic analyses, single crystal X-ray crystallography of 3-6 and biosynthetic considerations. With the confirmatory structural information provided by X-ray diffraction analysis in hand, several previously reported 21,24-cyclolanostanes, such as inonotsutriols A-C and (20R,21S,24S)-21,24-cyclopenta-3β,21,25-trihydroxylanosta-8-ene, were structurally corrected. In addition, the NMR data of other types of 21,24-cyclo triterpenoids were also re-examined and structural revisions were thus suggested. Compounds 2, 6 and 8 showed significant cytostatic effects against a panel of tumor cell lines with IC50 values ranging from 7.80 to 18.5 μM. Further assays established that compound 2 exerted promising in vitro anti-breast cancer potential by inhibiting the proliferation and migration of 4T1 cells.

Abstract:Introduction: One new levantilide D (2) with a 20-membered macrolide, along with one known levantilide C (1), was isolated from a rare marine actinomycetes Micromonospora sp. (FIMYZ52). Method: The structural elucidation of the compounds was based on integrated analyses involving 1D and 2D NMR spectroscopy and HR-ESI-MS. Result: The relative and absolute configurations of compounds 1 and 2 were ascertained by integrating multispectral ROESY and NOESY information from analogs in conjunction with theoretical TDDFT-ECD calculations. Levantilide D (2) exhibited antibacterial activity against Staphylococcus aureus. Conclusion: A plausible biosynthetic pathway of the macrolide compounds (1-2) was proposed.

Abstract:Introduction: Three novel macrolactams, FW8-1 (1), FW8-4 (2), and FW60-20 (3), were isolated from a culture of Micromonospora sp. The structures of these compounds were elucidated using Mass Spectrometry (MS) and comprehensive Nuclear Magnetic Resonance (NMR) analyses. Methods: The relative configurations of compounds 1-3 were assigned through theoretical calculations of their NMR spectra. Results: The isolation and determination of the relative configurations of these macrolactams have provided fresh perspectives on the biosynthetic pathways, leading to the formation of polyene macrolactams. Conclusion: Further, virtual screening and bioactivity predictions have suggested compounds 1-3 to possess potential anti-tumor, anti-inflammatory, and neuroprotective properties.

Abstract:Four compounds ( 1 – 4 ) featuring with an L -rhodinose and spiroketal, possess uncommon continuous hydroxy groups in the macrolide skeleton, and a dichloro-diketopiperazine ( 5 ) were isolated from a marine derived Micromonospora sp. FIMYZ51. The determination of the relative and absolute configurations of all isolates was achieved by extensive spectroscopic analyses, single-crystal X-ray diffraction analysis, and ECD calculations. According to structural characteristic and genomic sequences, a plausible biosynthetic pathway for compound 1 - 4 was proposed and a spirocyclase was inferred to be responsible for the formation of the rare spirocyclic moiety. Compounds 1 - 4 exhibited potent antifungal activities which is equal to itraconazole against Aspergillus niger . Compounds 1 - 5 exhibited different degree of inhibitory activities against opportunistic pathogenic bacteria of endocarditis ( Micrococcus luteus ) with MIC values ranging from 0.0625 μg/mL to 32 μg/mL. Compounds 2 and 3 showed moderate cytotoxicity against drug-resistant tumor cell lines (Namalwa and U266). The result not only provides active lead-compounds, but also reveal the potential of the spirocyclase gene resources from Micromonospora sp., which highlights the promising potential of the strain for biomedical applications.

Abstract:A new polycyclic tetramate macrolactam designated allostreptamide ( 1 ), together with four known congeners, were isolated from the culture extract of Allostreptomyces RD068384. The planar structure of the new compound was elucidated through interpretation of NMR and MS data. The absolute configuration was determined through ROESY and ECD analyses. The isolated compounds revealed antifungal potential against fourteen Candida albicans isolates with minimum inhibitory concentrations (MICs) ranging from 64 to 2048 μg ml -1 . Compound 3 showed antibiofilm action and considerably reduced the viability of five isolates (36%) in the formed biofilm. The qRT-PCR revealed that 3 downregulated the BCR1 , PLB2 , ALS1 , and SAP5 biofilm related gene expression. Therefore, 3 could be a promising antifungal therapy for C. albicans infections.

Abstract:Six new halogenated butenolides, tongalides A-C (1-3) and their acetylated congeners (4-6), were isolated from an extract of the Antarctic rhodophyte Delisea sp. that displayed significant antibiotic activity. The structures of the compounds were determined by analysis of data acquired by spectroscopic and spectrometric techniques including NMR, HRESIMS, optical rotation, and X-ray diffraction studies. The newly isolated compounds were assayed for antibacterial activity, but exhibited no growth inhibition of ESKAPE pathogens. The extract bioactivity was attributed to the previously reported Z-acetoxyfimbrolide A also isolated from the extract, providing further evidence that the exocyclic double bond is essential to the antibacterial activity of the structurally related fimbrolide class of metabolite.

Abstract:Three new 22-membered polyol macrolides, dactylides A−C ( 1−3 ), were isolated from Dactylosporangium aurantiacum ATCC 23491 employing repeated chromatographic separations, and their structures were established based on detailed analysis of NMR and MS data. The relative configurations at the stereocenters were established via vicinal 1 H– 1 H coupling constants, NOE correlations, and by application of Kishi's universal NMR database. In order to get insights into the biosynthetic pathway of 1−3 , the genome sequence of the producer strain D. aurantiacum was obtained and the putative biosynthetic gene cluster encoding their biosynthesis was identified through bioinformatic analysis using antiSMASH. Compounds 1−3 showed significant in-vitro antimycobacterial and cytotoxic activity.

Abstract:Marine natural products, characterized by fascinating drug-like functionalities and promising biological activities, are important base materials for innovative drugs and agrochemicals. Chemical investigations of the marine-algal-derived endophytic fungus Aspergillus sp. 1022LEF residing in the inner tissue of marine red alga yielded a novel polyketide-terpene hybrid metabolite, namely tennessenoid A ( 1 ), as well as six known biosynthetic congeners including two steroids, ergosta-4,6,8(14),22-tetraen-3-one ( 2 ) and (22 E ,24 R )-3α-ureido-ergosta-4,6,8(14),22-tetraene ( 3 ), and four sorbicillinoid-based compounds, saturnispol G ( 4 ), trichodimerol ( 5 ), and dihydrotrichodimer ethers A and B ( 6 and 7 ). Their structures were unambiguously determined based on extensive 1D/2D NMR and HRESIMS spectroscopic analyses. Tennessenoid A ( 1 ) was characterized as an unprecedented steroid−sorbicillinoid adduct via a C−C bond, which was rarely-observed in natural products. All of the isolated compounds were evaluated for their antifungal activities against eight plant pathogenetic fungi. 1 , in particular, demonstrated broad-spectrum activities against Sclerotium rolfsii Sacc., Fusarium oxysporum (Schl.) F.sp cucumerinum Owen, Coniella diplodiella Petrak et Sydow, Physalospora piricola Nose., Fusarium graminearum schw., Alternaria mali rob., Colletotrichum orbiculare Arx., and Alternaria porri (E11iott) Cifed., with the inhibition zone diameters ranging from 2 to 7 mm.

Cytotoxic 14-Membered Macrolides from a Mangrove-Derived Endophytic Fungus, Pestalotiopsis Microspora

Abstract:Seven new 14-membered macrolides, pestalotioprolides C (2), D-H (4-8), and 7-O-methylnigrosporolide (3), together with four known analogues, pestalotioprolide B (1), seiricuprolide (9), nigrosporolide (10), and 4,7-dihydroxy-13-tetradeca-2,5,8-trienolide (11), were isolated from the mangrove-derived endophytic fungus Pestalotiopsis microspora. Their structures were elucidated by analysis of NMR and MS data and by comparison with literature data. Single-crystal X-ray diffraction analysis was used to confirm the absolute configurations of 1, 2, and 10, while Mosher's method and the TDDFT-ECD approach were applied to determine the absolute configurations of 5 and 6. Compounds 3-6 showed significant cytotoxicity against the murine lymphoma cell line L5178Y with IC50 values of 0.7, 5.6, 3.4, and 3.9 μM, respectively, while compound 5 showed potent activity against the human ovarian cancer cell line A2780 with an IC50 value of 1.2 μM. Structure-activity relationships are discussed. Coculture of P. microspora with Streptomyces lividans caused a roughly 10-fold enhanced accumulation of compounds 5 and 6 compared to axenic fungal control.
