Abstract:The reaction mechanism of palladium(0)-catalyzed reaction of aryl iodides, norbrnene, and di-tert-butyldiaziridinone has been studied theoretically. Two reaction mechanisms were calculated. The reaction mechanism involving Cs2CO3 combination first followed by alkene insertion is calculated to be preferred to the one in which alkene insertion occurs first followed by Cs2CO3 combination. The calculations suggest that the reaction proceeds via C-I bond oxidative addition, CsI center dot CsCO3 cluster anion formation, alkene insertion, C-H bond activation, N-N bond oxidative addition, and two successive C(sp(2))-N bond-forming, and C(sp(3))-N bond-forming reductive elimination steps. The N-N bond oxidative addition involves the rate-determining transition state with a free energy barrier of 29.4 kcal/mol. The role of base Cs2CO3 played during CsI center dot CsCO3 cluster anion formation and N-N bond oxidative addition process has been discussed according to the formation of relatively strong Pd-O bond. The role of base K2CO3 has been calculated and compared with Cs2CO3.

The calculation result shows that the reaction mechanism involves sequential steps of substrate coordination, ligand-to-ligand hydrogen transfer (LLHT), and C-C reductive elimination to produce the final product tricyclic imidazole. The LLHT step is calculated to be the rate-determining step. The oxidative addition of the benzimidazole C-H bond to the Ni center and the insertion of the alkene into the Ni-H bond occur concertedly in the LLHT step. The effects of N-heterocyclic carbene (NHC) ligands and AlMe3 on the reactivity and regioselectivity were also analyzed. These calculation results shed light on some ambiguous suggestions from experiments.

Abstract:Density functional theory (DFT) calculations with B3LYP and M06 functionals elucidated the reactivities of alkynes and Z/E selectivity of cyclodecatriene products in the Ni-catalyzed [4 + 4 + 2] cycloadditions of dienes and alkynes. The Ni-mediated oxidative cyclization of butadienes determines the Z/E selectivity. Only the oxidative cyclization of one s-cis to one s-trans butadiene is facile and exergonic, leading to the observed 1Z,4Z,8E-cyclodecatriene product. The same step with two s-cis or s-trans butadienes is either kinetically or thermodynamically unfavorable, and the 1Z,4E,8E- and 1Z,4Z,8Z-cyclodecatriene isomers are not observed in experiments. In addition, the competition between the desired cooligomerization and [2 + 2 + 2] cycloadditions of alkynes depends on the coordination of alkynes. With either electron-deficient alkynes or alkynes with free hydroxyl groups, the coordination of alkynes is stronger than that of dienes, and alkyne trimerization prevails. With alkyl-substituted alkynes, the generation of alkyne-coordinated nickel complex is much less favorable, and the [4 + 4 + 2] cycloaddition occurs.

Abstract:Detailed mechanisms of the intramolecular C(sp(3))-C(sp(2)) bond formation via C(sp(3))-H bond activation of a trimethylsilyl (TMS) substituent catalyzed by palladium(0) complexes have been investigated with the aid of density functional theory (DFT) calculations. The results reveal that the favorable catalytic cycle includes oxidative addition, ligand substitution, concerted metalation deprotonation (CMD) and reductive elimination steps. The CMD was found to be the rate-determining step with an overall free energy barrier of 26.4 kcal mol(-1). For the analogous CMe3-substituted substrate, the C(sp(3))-H bond activation of the CMe3 substituent was calculated to have a high free energy barrier of 29.9 kcal mol(-1). Our calculation results show that during the deprotonation process of the TMS C(sp(3))-H bond, the adjacent Si atom stabilizes the charge accumulated on the C(sp(3))-H carbon and facilitates the C(sp(3))-H bond activation due to the ability of Si to engage in Si-C hypervalent bonding.

Abstract:The reaction mechanism of the Pd(0)-catalyzed alkyne cyanothiolation reaction is investigated by MP2, CCSD(T) and the density functional method B3LYP. The overall reaction mechanism is examined. The B3LYP results are consistent with the results of CCSD(T) and MP2 methods for the isomerization, acetylene insertion and reductive elimination steps, but not for the oxidative addition step. For the oxidative addition, the bisphosphine and monophosphine pathways are competitive in B3LYP, while the bisphosphine one is preferred for CCSD(T) and MP2 methods. The electronic mechanisms for the oxidative addition of thiocyanate HS-CN to Pd(PH3)2 and Pd(PH3) and for the acetylene insertion into Pd-S and Pd-CN are discussed in terms of the electron-donation and back-donation. The chemo-selectivity that acetylene inserts into the Pd-S bond rather than into the Pd-CN bond is due to the involvement of the S p orbital. It is the doubly occupied S p unhybridized orbital that donates an electron to the alkylene pi* anti-bonding orbital, which makes insertion into Pd-S bond more favorable than into the Pd-CN bond. During the insertion into the Pd-S bond, the S sp(2) hybrid orbital and unhybridized p orbital transform into each other, while the C sp hybrid orbital shifts its direction for insertion into Pd-CN bond. By using the monosubstituted acetylenes (CN, Me and NH2), the influence of substituents at acetylene on the chemo- and regio-selectivities is analyzed.

Abstract:With the aid of D FT calculations, we have examined the mechanism of the Ni-catalyzed cycloaddition of 3-azetidinone with alkynes. Our DFT results did not support the originally proposed mechanism, which involves a ring expansion through beta-carbon elimination. Instead, our calculations supported a mechanism which involves oxidative addition of 1-Boc-3-azetidinone to a Ni(0) center to form an intermediate having both Ni-C(O) and Ni-C(sp(3)) bonds, then alkyne insertion into either the Ni-C(O) or Ni-C(sp(3)) bond depending on the alkyne substrates being studied, and finally reductive elimination to give the c-ycloaddition products. The nature of insertion of alkynes into a Ni-C bond was also discussed. When an alkyne substrate inserts into the Ni-C(O) bond, we found that the alkyne acts as a nucleophile to attack the electron-deficient, metal-bonded C=O carbon center. When an alkyne substrate inserts into the Ni-C(sp(3)) bond, the alkyne acts as an electrophile to interact with the Ni-C(sp(3)) bond.
Mechanistic Investigation into the Regio-Controllable Hydroallylations of Alkynes with Allylborons under Pd-Based Synergetic Catalyses

Abstract:Density functional theory calculations were employed to investigate the Pd-catalyzed regio-selective hydroallylations of alkynes with allylborons: cooperation of Cu(OAc)2 and dppe resulting in 1,4-dienes while combination of AdCO2H and PCy3 leading to 1,5-dienes. A unified rationalization mechanism called "Lewis-acid-base-interaction promoted deprotonation/3,3-rearrangement" was proposed. Compared with the commonly reported metathesis pathway to only afford the metal-allyl intermediate, in the newly established mechanism, an additional Brønsted acid (as an initiator of the Pd0 oxidative addition) is generated by the interaction of the allylboron (Lewis acid) B atom with the nBuOH (Lewis base) O atom, and subsequent 3,3-rearrangement ensures the thermodynamic feasibility of the reaction. In addition, it was found that excess Cu(OAc)2 plays two potential roles in the oxidative addition/alkyne insertion: (i) the participation of one AcO- of Cu(OAc)2 ensures a large orbital overlap between the migrating H and Pd atoms, facilitating the formal AcO-H cleavage and (ii) the extra (OAc)2Cu···O(carboxyl) σ-coordination indirectly contributes to the (Me)C≡C(Ph) insertion into the Pd-H bond. Further analysis showed that the origin of the regioselectivity is closely related to the employed phosphorus ligand. These revealed results, which have been overlooked in the previous documents, would aid the development of new related catalytic reactions.

Abstract:Density functional theory (DFT) method B3LYP with a dispersion term (B3LYP-D3BJ) has been used to clarify the regioselectivity of zinc mediated 1,3-dipolar cycloaddition of azides and alkynes. Computational results indicate that the dipolar cycloaddition takes place via a concerted five-membered-ring transition state, leading to a 1,5-disubstituted 1,2,3-triazole product, which is consistent with the experiment reported by Greaney's group. The coordination of imidazole ligand to zinc is reversible, and the regioselectivity is irrelevant to the coordination of imidazole ligand. Moreover, substituent effect of alkynes has also been studied. Finally, distortion–interaction analysis along the reaction pathways and frontier molecular orbital theory are used to explain the reactivity and 1,5-regioselectivity.

Abstract:Density functional theory (DFT) calculations were performed to investigate the detailed mechanisms of metal-catalyzed reactions of alkynyl thioethers with isoxazoles/anthranils. The origins of the product selectivities were also explored. The results show that when [IPrAu(CH3CN)]+ was used as the catalyst, β-selective addition gave a sulfenylated pyrrole/indole as the primary product. However, the major product was a sulfur-substituted β-keto enamide, which was formed via α-addition, when [Zn(OTf)]+ was used. The product selectivity was determined by the first nucleophilic attack step. We found that the catalyst and substrate both played crucial roles in guiding the product selectivity. The electronic effects of ligands on the catalyst and substituents on the substrate were the dominant factor. For the anthranil substrate, the preference for C1–C6 bond coupling rather than C1–C4 bond coupling can be attributed to effective conjugation between the carbonyl group and the phenyl moiety during C1–C6 bond coupling. These insights into the mechanisms of metal-catalyzed reactions of alkynyl thioethers with isoxazoles/anthranils will facilitate the design of new catalytic reactions of isoxazoles with electron-deficient alkynes.

Mechanistic Insights into Solvent and Ligand Dependency in Cu(I)-Catalyzed Allylic Alkylation-with Gem-Diborylalkanes

Abstract:The recent Cu-catalyzed allylic substitution reaction between gem-diboryalkane and allyl electrophiles shows intriguing solvent and ligand-controlled regioselectivity. The α-alkylation product was obtained in DMF solvent, while γ-alkylation product was obtained in dioxane solvent and the dioxane and NHC ligand situation. In the present study, density functional theory calculations have been used to investigate the reaction mechanism and origin of the regioselectivity. For both dioxane and DMF, γ-alkylation undergoes successive oxidative addition (CH2Bpin trans to leaving group) and direct Cγ–C reductive elimination. The α-alkylation is found to undergo oxidative addition (CH2Bpin trans to leaving group), isomerization, and Cα–C reductive elimination rather than the previously proposed oxidative addition (−CH2Bpin cis to the leaving group) and Cα–C reductive elimination. The γ-alkylation and α-alkylation is, respectively, favorable for dioxane and DMF solvent, which is consistent with the γ- and α-selectivity in experiment. The solvent interferes the isomerization step, thereby affects the relative facility of the α- and γ-alkylation. Further investigation shows that η1-intermediate formation promoted by solvent is the rate-determining step of the isomerization. The stronger electron-donating ability of DMF than dioxane facilitates the η1-intermediate formation and finally results in the easier isomerization in DMF. For dioxane and NHC situation, in the presence of neutral NHC ligand, the −PO4Et2 group tightly coordinates with the Cu center after the oxidative addition, preventing the isomerization process. The regioselectivity is determined by the relative facility of the oxidative addition step. Therefore, the favorable oxidative addition (in which −CH2Bpin trans to the leaving group) results in the facility of γ-alkylation.

Abstract:The density functional theory (DFT) has been employed to elucidate the mechanisms of ruthenium-catalyzed cycloaddition of enynes with alkynes. The calculated results suggest that there are similar reaction pathways for symmetrical and nonsymmetrical alkynes, including oxidative cyclization, insertion of the alkynes, and reductive elimination, respectively. The oxidative cyclization is calculated to be the rate-determining step. The regioselectivity-determining step for this reaction is the alkyne insertion and the observed regioselectivity could be interpreted through the HOMO coefficient analysis.

Abstract:Employing density functional theory (DFT) calculations, the study meticulously analyzes the mechanistic details of the Pd-catalyzed dual-γ-1,1-C(sp3)–H activation of aliphatic carboxylic acids. To explore the intrinsic nature of double C(sp3)–H in aliphatic carboxylic acids, we compared the reaction mechanisms of two coupling agents: allyl alcohol and methyl acrylate. The calculations indicated that allyl alcohol favors the second C(sp3)–H activation due to the absence of conjugation effects, which typically hinder C(sp3)–H activation. In contrast, methyl acrylate favors the Michael addition mechanism due to reduced steric hindrance and the formation of a stabilizing Na−O coordination interaction. We also computed the reaction mechanism of the coupling agent but-3-en-2-yl acetate and compared it with that of allyl alcohol to determine the reasons for the occurrence of β-H elimination. For allyl alcohol, β-H elimination is favored due to the lack of steric hindrance in the transition state, whereas for but-3-en-2-yl acetate, β-OAc elimination is preferred because of a stabilizing N-H⋯O hydrogen bond in the transition state. The study also highlights the significant role of Na2HPO4 base in enhancing the reaction's efficiency. Specifically, we found that the presence of Na2HPO4 base significantly facilitates the hydroxyl deprotonation process. To further explore the reaction's diastereoselectivity, we calculated the diastereoselectivity-determining steps for two substrates with significantly different d.r. ratios and analyzed the driving forces behind the high diastereoselectivity. Our analysis revealed that the diastereoselectivity of the reaction improves as the disparity in steric hindrance between the two substituents at the β-position of the carboxylic acid increases. Overall, the mechanistic insights gained in this study may prove valuable in the ongoing advancement and application of metal-catalyzed dual-1,1-C(sp3)−H activation.
