Abstract:A HFIP-promoted highly selective hydroxyalkylation of aniline derivatives with arylglyoxal hydrates has been realized. The reaction produces various N,N-dialkylanilines and their derivatives with α-hydroxy carbonyl units in good to excellent yields under mild conditions. Furthermore, the synthetic potential of this method has been demonstrated by the facile synthesis of several structurally interesting molecules such as benzil, 1,2,4-triazine, quinoxaline, hydantoin, and 2-thiohydantoin with aromatic amine units.

Abstract:An efficient, green and atom-economical iron-catalyzed hydroamination of vinylpyridines with azoles has been developed. This reaction works smoothly for a diverse range of vinylpyridines and azoles, including diazoles and triazoles. The reaction was catalyzed by 5 mol% FeCl3 in toluene at 110 degrees C for 3 h to afford the anti-Markovnikov hydroamination products in moderate to excellent yields and excellent selectivities.

Abstract:Asymmetric hydroxycarbonylation is one of the most fundamental yet challenging methods for the synthesis of carboxylic acids. Herein, we reported the development of a palladium-catalyzed highly enantioselective Markovnikov hydroxycarbonylation of vinyl arenes with CO and water. A monodentate phosphoramidite ligand L6 plays vital role in the reaction. The reaction tolerates a range of functional groups, and provides a facile and atom-economical approach to an array of 2-arylpropanoic acids including several commonly used non-steroidal anti-inflammatory drugs. The catalytic system has also enabled an asymmetric Markovnikov hydroalkoxycarbonylation of vinyl arenes with alcohols to afford 2-arylpropanates. Mechanistic investigations suggested that the hydropalladation is irreversible and is the regio- and enantiodetermining step, while hydrolysis/alcoholysis is probably the rate-limiting step.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:A simple and direct α-allylic alkylation of unmodified aldehydes with allylic alcohols catalyzed by primary amine/Br⊘nsted acid has been developed. The N,N-dimethylethylenediamine/TfOH was identified as the optimal catalyst to promote this transformation via an enamine process in high yields (up to 88%) and with good diastereoselectivities (up to 9:1).

Abstract:A combined theoretical and experimental approach was used to systematically study the Brønsted acid-promoted aziridination of electron-deficient olefins. It was found that Brønsted acid-promoted aziridination of electron-deficient olefins proceeded through the attack of the internal nitrogen of the azide to the terminal carbon of protonated olefin, which afforded an acyclic adduct that subsequently discharged N2 to produce the aziridine ring. The basicity of the electron-deficient olefins is an important parameter to determine the efficiency of Brønsted acid-promoted aziridination. More basic carbonyl compounds including vinyl ketones and acrylamides were predicted to be readily activated by Brønsted acid such as TfOH, whereas less basic carbonyl compounds were predicted to be poor substrates. Significantly, all these theoretical predictions were demonstrated to be consistent with the experimental data. Furthermore, a systematic evaluation of TfOH-promoted aziridination of acrylamides was performed, which established a new, single-step method for the preparation of a number of aziridine-2-carboxamides.

Abstract:Conventional hydrofunctionalization of α-olefins with mineral acids proceeds with Markovnikov selectivity to afford branched isomers. The direct formation of linear constitutional isomers is challenging, yet anti-Markovnikov addition would be valuable for the synthesis of commodity chemicals, such as primary alcohols, which are currently only accessible via stoichiometric redox reactions, with a full equivalent of waste of both oxidant and reductant. Strategies that utilize radical intermediates have been demonstrated, but only for activated alkenes, and the direct use of aqueous mineral acids remains elusive. Here we present anti-Markovnikov addition reactions of aqueous hydrochloric and nitric acid to unactivated alkenes. The transformation is enabled by the in situ generation of photoredox-active ion pairs, derived from acridine and the mineral acid, as a combined charge- and phase-transfer catalyst. The introduction of a hydrogen atom transfer catalyst enabled us to bypass the challenging chain propagation by hydrochloric and nitric acids that originates from the high bond dissociation energy.

Abstract:Reported herein is a chiral Bronsted acid-catalyzed asymmetric intramolecular allylic amination reaction, allowing facile access to a range of biologically interesting chiral 2-substituted hydroquinolines in up to 90% yield and with up to 93% ee. Furthermore, a significant effect of an N-protecting group was observed in this asymmetric process.

Abstract:A visible-light-driven sulfamate esters guided alkylation of unactivated C(sp3)-H bonds enabled by a 1,6-HAT/radical addition cascade is described. Not only structurally diverse Michael acceptors but also styrenes are amenable to this alkylation reaction. Notably, the N-H bonds activation radical relay refrained from prefunctionalization and using excess external oxidants.

Abstract:Utilization of formic acid directly as a carboxylic reagent with pre‐decomposition into CO and H2O was an attractive and promising protocol to scaffold carboxylic acids. Herein, with BINAP‐modified Pd(OAc)2 catalytic system free of Ac2O or DCC, the hydrocarboxylation of styrene (and its derivatives) with FA predominantly following anti‐Markovnikov's rule was fulfilled successfully to afford the linear carboxylic acids in the isolated yields of 60~88%. The external CO in catalytic amount was introduced as a co‐ligand to work together with BINAP to adjust the behavior of Pd‐catalyst. The in situ FT‐IR and 1H NMR analyses convince that this one‐step hydrocarboxylation goes by palladium‐hydride (PdII‐H) mechanism.

Abstract:C(sp3) centers adjacent to (hetero)aryl groups are widely present in physiologically active molecules. Metal‐hydride‐catalyzed hydroalkylation of alkenes represents an efficient means of forging C(sp3)‐C(sp3) bonds, boasting advantages as a wide source of substrates, mild reaction conditions, and facile selectivity manipulation. Nevertheless, the hydroalkylation of vinylarenes encounters constraints in terms of substrate scope, necessitating the employment of activated alkyl halides or alkenes containing chelating groups, remains a challenge. In this context, we report a general nickel‐hydride‐catalyzed hydroalkylation protocol for vinylarenes. Remarkably, this system enables α‐selective hydroalkylation of both aryl and heteroaryl alkenes under an extra ligand‐free condition, demonstrating excellent coupling efficiency and selectivity. Furthermore, through the incorporation of chiral bisoxazoline ligands, we have achieved regio‐ and enantioselective hydroalkylation of vinylpyrroles, thereby facilitating the synthesis of α‐branched alkylated pyrrole derivatives.

Abstract:Visible-light-induced three-component 1,2-alkylarylation of alkenes and alkyl radical addition/cyclization of acrylamides have been realized via a photocatalytic halogen-atom transfer (XAT) process. This metal-free protocol utilizes readily available tertiary alkylamine as both an electron donor and XAT reagent for the activation of alkyl halides using naphthalimide (NI)-based organic photocatalysts. Abstract:Direct α-C(sp3)-H alkylation of hydroaminoalkylation (HAA) catalyzed by tBuOK/DMSO/O2 from α-aryl benzylamines with styrenes is reported. The reaction has delivered diverse selectively substituted amines and N-heterocycles under mild conditions, showing high functional group tolerance. Control experiments have shown that α-amino radicals are proposed to be generated by hydrogen atom transfer (HAT) to dimsyl radical derived from SET oxidation in tBuOK/DMSO/O2 system.

Abstract:A visible light accelerated C-H functionalization reaction in palladium-catalyzed arylation of vinyl arenes with diaryliodonium salts is reported in the absence of additional photosensitizer. The kinetic isotope effect (k(H)/k(D)) was changed from 3.6 (under darkness) to 1.1 when irradiated by visible light, which indicated that the C-H functionalization step was the rate determining step under darkness and significantly accelerated by the irradiation of visible light. Finally the synthesis of ortho tetra-substituted vinylarene atropisomers with high enantiospecificity was realized via this protocol.
