Bronchial Lavage Exosome miRNA Correlates to Metabolic Disturbance in Patients with Pulmonary Acinetobacter Baumannii Infection

Abstract:Aim: Critical pulmonary infection lead to metabolic disturbances. Acinetobacter Baumannii(Ab) Infection, due to the persistent course of the disease, obtains its distinct metabolic features. Yet few studies have discussed the global metabolic profile and the cause for its dysregulation in patients with pulmonary Ab Infection. In this study, we aim to characterize the metabolic profile and identifies its potential association with bronchial exosome miRNA release in patients with pulmonary Ab Infection. Methods: Patients admitted to hospital for pulmonary infection were enrolled. Demographic and biochemical measurements were collected and serum samples were obtained after overnight fasting. Nontargeted metabolomics was applied for serum samples. Bronchoscopy was performed and bronchial lavage was collected meanwhile. Exosome was extracted and miRNA was sequenced afterwards. Results: We have found significant alteration of serum metabolites in pulmonary infection patients as compared to control. Notably, patients with Ab Infection exhibited different metabolic characteristics. Moreover, exosome miRNAs from bronchial lavage of patients with Ab Infection were most significantly enriched in cellular metabolic pathways. At last, the altered miRNA is correlated with dysregulated metabolites in amino acids and glucose metabolism. Conclusions: Exosome miRNA alteration is correlated to dysregulated metabolism in patients with Ab Infection. The correlation between exosome miRNA and dysregulated metabolites highlight potential biomarkers and therapeutics for diagnosing and treating Baumannii Infection.
