Metabolomic Profiling Differences among Asthma, COPD and Healthy Controls: a LC-MS-based Metabolomics Analysis

Abstract:Backgrounds: Early diagnosis of asthma and distinguishing asthma from other airflow limitation disease, such as chronic obstructive pulmonary disease (COPD), is sometimes very difficult, although the inflammatory and cellular profile in the airways are much different between them. Metabolomics is the comprehensive assessment of metabolites generated by cellular metabolic pathways. We hypothesized that the metabolic condition of asthma would differ from COPD and healthy controls. Objectives: To compare the serum metabolomics profiles among mild persistent asthma, stable COPD and healthy controls and to explore potential metabolic biomarkers and pathways. Methods: The serum metabolomic profiles of 17 subjects with mild persistent asthma, 17 subjects with stable COPD and 15 healthy controls were determined by an untargeted metabolomic analysis, utilizing LC-MS coupled with a series of multivariate statistical analyses. Results: Orthogonal projections to latent structures-discriminate analysis (OPLS-DA) showed a distinct separation between asthma and healthy controls in electrospray positive and negative ions modes, respectively. A total of 19 differential metabolites were identified. Similarly, OPLS-DA showed a distinct separation between asthma and COPD in electrospray positive and negative ions modes, respectively. A total of 17 differential metabolites were identified. Of all the metabolites identified, serum level of hypoxanthine was markedly greater in asthmatic subjects, compared with COPD or healthy controls. Conclusion: Subjects with asthma have a unique serum metabolome which can distinguish them from those with COPD and healthy controls.
