Decreased Mir-150-5p and Mir-23A-3p Expressions in Patients with Active Pulmonary Tuberculosis (TB) Disease

Abstract:Background: The aim of this study is to explore the role of microRNAs (miR)-21/23a/146a/150/155 targeting the toll-like receptor (TLR) pathway in host immune response to active pulmonary tuberculosis (TB) disease. Material and Methods: Gene expression levels of the five miRs, TLR2/4/6, and their down-stream mediators were assessed by quantitative RT-PCR method in peripheral blood mononuclear cells from 38 patients with active pulmonary TB, 16 subjects with latent TB infection (LTBI), and 16 non-infected healthy subjects (NIHS). Results: Gene expression levels of miR-150-5p were decreased in patients with active TB group as compared with either LTBI subjects or HS. Subgroup analysis showed that gene expression levels of both miR-23a-3p and miR-146a-5p were decreased in active TB patients with high bacterial burden (n=12) versus those with low bacterial burden (n=26) or NIHS. TLR 4 expressions were increased in active TB patients as compared with NIHS, and negatively correlated with miR-150-5p, miR-146a-5p, and miR-155-5p gene expressions. Both miR-150-5p and miR-23a-3p were negatively correlated with IFN-γ, Notch3, and SOCS1 gene expressions. In vitro ESAT-6 and CFP-10 stimuli in THP-1 cells resulted in down-regulation of miR-23a-3p and miR-155-5p, and up-regulations of several predicted target genes, including TNF-α, IFN-γ, TLR2, TLR4, Notch3, SOCS1, and FOXO1. Conclusions: MiR-150-5p and miR-23a-3p were down-regulated in active TB disease. These findings provide biological insight into mechanisms by which Mycobacterium TB-suppressed miRs modulate human immunity, and indicate that over-expression of the miR-150 and miR-23a may be a new host-directed immunotherapy for active TB.
