Abstract:Cadmium (Cd)-contamination impairs biological nitrogen fixation in legumes (BNF), threatening global food security. Innovative strategies to enhance BNF and improve plant resistance to Cd are therefore crucial. This study investigates the effects of graphitic carbon nitride nanosheets (g-C3N4 NSs) on soybean (Glycine max L.) in Cd contaminated soil, focusing on Cd distribution, chemical forms and nitrogen (N) fixation. Soybean plants were treated with 100 mg kg-1 g-C3N4 NSs, with or without 10 mg kg-1 Cd for 4 weeks. Soil addition of g-C3N4 NSs alleviated Cd toxicity and promote soybean growth via scavenging Cd-mediated oxidative stress and improving photosynthesis. Compared to Cd treatment, g-C3N4 NSs increased shoot and root dry weights under Cd toxicity by 49.5% and 63.4%, respectively. g-C3N4 NSs lowered Cd content by 35.7%-54.1%, redistributed Cd subcellularly by increasing its proportion in the cell wall and decreasing it in soluble fractions and organelles, and converted Cd from high-toxicity to low-toxicity forms. Additionally, g-C3N4 NSs improved the soil N cycle, stimulated nodulation, and increased the N-fixing capacity of nodules, thus increasing N content in shoots and roots by 12.4% and 43.2%, respectively. Mechanistic analysis revealed that g-C3N4 NSs mitigated Cd-induced loss of endogenous nitric oxide in nodules, restoring nodule development. This study highlights the potential of g-C3N4 NSs for remediating Cd-contaminated soil, reducing Cd accumulation, and enhancing plant growth and N fixation, offering new insights into the use of carbon nanomaterials for soil improvement and legume productivity under metal(loid)s stress.

Abstract:Cadmium (Cd) pollution poses a serious threat to plant growth and yield. Nanomaterials have shown great application potential for alleviation of Cd toxicity to plants. In this study, we applied graphitic carbon nitride nanosheets (g-C3N4 NSs) for alleviation of Cd-toxicity to soybean (Glycine max L.). The g-C3N4 NSs supplementation significantly improved plant growth and reduced oxidative damage in the Cd-toxicated soybean seedlings through hydroponic culture. Particularly, the g-C3N4 NSs dynamically regulated the root cell wall (RCW) components by increasing pectin content and modifying its demethylation via enhancing pectin methylesterase (PME) activity, therefore greatly enhanced stronger RCW-Cd retention (up to 82.8%) and reduced Cd migration to the shoot. Additionally, the g-C3N4 NSs reversed the Cd-induced chlorosis, increased photosynthetic efficiency because of enhancement in Fv/Fm ration, Y(II) and sugars content. These results provide new insights into the alleviation of Cd toxicity to plants by g-C3N4 NSs, and shed light on the application of low-cost and environmental-friendly carbon-based NMs for alleviating heavy metal toxicity to plants.

Interactive Dynamics Between Rhizosphere Bacterial and Viral Communities Facilitate Soybean Fitness to Cadmium Stress Revealed by Time-Series Metagenomics

Abstract:The distinctive symbiotic relationship between rhizobacteria and soybean (Glycine max) can enhance soil fertility and assist soybean defense heavy metal stress, such as cadmium (Cd), in soils. Soil viruses are critical for modulating microbial activities and enhancing host adaptability to soil contamination by encoding auxiliary metabolic genes (AMGs). However, the community structure, functional diversity and ecological roles of soil viruses and their dynamic interactions with soil-dwelling microbes under Cd stress remain unclear. Here, we characterized the bulk and rhizosphere bacterial community in a Cd-contaminated soybean field at five growth stages and investigated the rhizosphere microbial functional traits and the viral community after flowering using metagenomics. We showed that the rhizosphere bacterial community exhibited a stronger soybean development-dependent diversity pattern relative to bulk soil and had a more critical role in predicting Cd accumulation. The relative abundance of Actinobacteria in the rhizosphere increased with soybean development, particularly in the families Nocardioidaceae and Intrasporangiaceae, which were further identified as the main contributors of amino acid transport E and secondary metabolites Q genes. Viral analysis suggested an interactive and complementary assembly between viral and bacterial communities. Importantly, viruses exhibited a broader host range during the flowering period, and infected specific functional taxa such as Bradyrhizobium and Nocardioides, highlighting their potential impact on the rhizobia-soybean symbiosis and soybean growth and fitness. Furthermore, AMGs related to element cycling and microbial metabolism were identified and functionally and phylogenetically characterized, suggesting the contribution of these virus-encoded genes to biogeochemical cycles and soybean fitness under Cd stress. Together, our results expand our understanding of the ecological functions of rhizosphere viruses in Cd pollution-stressed soils, and provide insights in developing new strategies for bioremediation of contaminated soils.
Biogenic silicon nanoparticles mitigate cadmium (Cd) toxicity in rapeseed (Brassica napus L.) by modulating the cellular oxidative stress metabolism and reducing Cd translocation

Abstract:Nano-enabled strategies have emerged as promising alternatives to resolve heavy metals (HMs) related harms in an eco-friendly manner. Here, we explored the potential of biogenic silicon nanoparticles (SiNPs) in alleviating cadmium (Cd) stress in rapeseed (Brassica napus L.) plants by modulating cellular oxidative repair mechanisms. Biogenic SiNPs of spherical shapes with size ranging between 14 nm and 35 nm were synthesized using rice straw extract and characterized through advanced characterization techniques. A greenhouse experiment results showed that SiNPs treatment at 250 mg kg−1 significantly improved growth parameters, including fresh weight (33.3 %) and dry weight (32.6 %) of rapeseed plants than Cd-treated control group. Photosynthesis and leaf gas exchange parameters were also positively influenced by SiNPs treatment, indicating enhanced photosynthetic efficiency. Additionally, SiNPs treatment at 250 mg kg−1 increased the activities of antioxidant enzymes such as superoxide dismutase (19.1 %), peroxidase (33.4 %), catalase (14.4 %), and ascorbate peroxidase (33.8 %), which may play a crucial role in ROS scavenging and reduction in Cd-induced oxidative stress. TEM analysis revealed that SiNPs treatment effectively mitigated Cd-induced damage to leaf ultrastructure, while qPCR analysis showed that SiNPs treatment changed the expressions of the antioxidant defense and stress related genes. Moreover, SiNPs treatment significantly influenced the Cd accumulation and Si contents in plants. Overall, our findings revealed that biogenic SiNPs have great potential to serve as a sustainable, eco-friendly, and non-toxic alternative for the remediation of Cd toxicity in rapeseed plants.

Abstract:Cadmium (Cd) contamination has led to various harmful impacts on soil microbial ecosystem, agricultural crops, and thus human health. Nanomaterials are promising candidates for reducing the accumulation of heavy metals in plants. In this study, graphitic carbon nitride (g-C 3 N 4 ), a two-dimensional polymeric nanomaterial, was applied for ameliorating Cd phytotoxicity to soybean ( Glycine max (L.) Merr.). Its impacts on rhizosphere variables, microorganisms, and metabolism were examined. It was found that g-C 3 N 4 increased carbon/nitrogen/phosphorus (C/N/P) content, especially when N contents were averagely 4.2 times higher in the g-C 3 N 4 -treated groups. g-C 3 N 4 significantly induced alterations in microbial community structures ( P < 0.05). The abundance of the probiotics class Nitrososphaeria was enriched (on average 70% higher in the g-C 3 N 4 -treated groups) as was Actinobacteria (226% higher in the g-C 3 N 4 group than in the CK group). At the genus level, g-C 3 N 4 recruited more Bradyrhizobium (122% higher) in the Cd + g-C 3 N 4 group than in the Cd group and more Sphingomonas (on average 24% higher) in the g-C 3 N 4 -treated groups. The changes of microbial clusters demonstrated the potential of g-C 3 N 4 to shape microbial functions, promote plant growth, and enhance Cd resistance, despite observing less pronounced modifications in microbial communities in Cd-contaminated soil compared to Cd-free soil. Moreover, abundance of functional genes related to C/N/P transformation was more significantly promoted by g-C 3 N 4 in Cd-contaminated soil (increased by 146%) than in Cd-free one (increased by 32.8%). Therefore, g-C 3 N 4 facilitated enhanced microbial survival and adaptation through the amplification of functional genes. These results validated the alleviation of g-C 3 N 4 on the microbial communities in the soybean rhizosphere and shed a new light on the application of environmental-friendly nanomaterials for secure production of the crop under soil Cd exposure.

Abstract:Graphitic carbon nitride (g-C3N4) is a promising candidate for heavy metal remediation, primarily composed of carbon (C) and nitrogen (N). It has been demonstrated that g-C3N4 adjusts rhizosphere physicochemical conditions, especially N conditions, alleviating the absorption and accumulation of Cadmium (Cd) by soybeans. However, the mechanisms by which g-C3N4 induces N alterations to mitigates plant uptake of Cd remain unclear. This study investigated the impact of g-C3N4-mediated changes in N conditions on the accumulation of Cd by soybeans using pot experiments. It also explored the microbiological mechanisms underlying alterations in soybean rhizospheric N cycling induced by g-C3N4. It was found that g-C3N4 significantly increased N content in the soybean rhizosphere (p < 0.05), particularly in terms of available nitrogen (AN) of nitrate and ammonium. Plants absorbed more ammonium nitrogen (NH₄⁺-N), the content of which in the roots showed a significant negative correlation with Cd concentration in plant (p < 0.05). Additionally, g-C3N4 significantly affected rhizospheric functional genes associated with N cycling (p < 0.05) by increasing the ratio of the N-fixation functional gene nifH and decreasing the ratios of functional genes amoA and nxrA involved in nitrification. This enhances soybean's N-fixing potential and suppresses denitrification potential in the rhizosphere, preserving NH₄⁺-N. Niastella, Flavisolibacter, Opitutus and Pirellula may play a crucial role in the N fixation and preservation process. In summary, the utilization of g-C3N4 offers a novel approach to ensure safe crop production in Cd-contaminated soils. The results of this study provide valuable data and a theoretical foundation for the remediation of Cd polluted soils.
Cadmium effect on oxidative metabolism of pea (Pisum sativum L.) roots. Imaging of reactive oxygen species and nitric oxide accumulation in vivo

Abstract:Growth of pea (Pisum sativum L.) plants with 50 microM CdCl2 for 15 d produced a reduction in the number and length of lateral roots, and changes in structure of the principal roots affecting the xylem vessels. Cadmium induced a reduction in glutathione (GSH) and ascorbate (ASC) contents, and catalase (CAT), GSH reductase (GR) and guaiacol peroxidase (GPX) activities. CuZn-superoxide dismutase (SOD) activity was also diminished by the Cd treatment, although Mn-SOD was slightly increased. CAT and CuZn-SOD were down-regulated at transcriptional level, while Mn-SOD, Fe-SOD and GR were up-regulated. Analysis of reactive oxygen species (ROS) and nitric oxide (NO) levels by fluorescence and confocal laser microscopy (CLM) showed an over-accumulation of O2*- and H2O2, and a reduction in the NO content in lateral and principal roots. ROS overproduction was dependent on changes in intracellular Ca+2 content, and peroxidases and NADPH oxidases were involved. Cadmium also produced an increase in salicylic acid (SA), jasmonic acid (JA) and ethylene (ET) contents. The rise of ET and ROS, and the NO decrease are in accordance with senescence processes induced by Cd, and the increase of JA and SA could regulate the cellular response to cope with damages imposed by cadmium.

Abstract:Cadmium (Cd) is an extremely toxic heavy metal that can originate from industrial activities and accumulate in agricultural soils. This study investigates the potential of biologically synthesized silicon oxide nanoparticles (Bio-SiNPs) in alleviating Cd toxicity in bayberry plants. Bio-SiNPs were synthesized using the bacterial strain Chryseobacterium sp. RTN3 and thoroughly characterized using advanced techniques. A pot experiment results demonstrated that Cd stress substantially reduced leaves biomass, photosynthesis efficiency, antioxidant enzyme activity, and induced oxidative damage in bayberry ( Myrica rubra ) plants. However, Bio-SiNPs application at 200 mg kg −1 significantly enhanced plant biomass, chlorophyll content (26.4 %), net photosynthetic rate (8.6 %), antioxidant enzyme levels, and mitigated reactive oxygen species production under Cd stress. Bio-SiNPs modulated key stress-related phytohormones by increasing salicylic acid (13.2 %) and abscisic acid (13.7 %) contents in plants. Bio-SiNPs augmented Si deposition on root surfaces, preserving normal ultrastructure in leaf cells. Additionally, 16S rRNA gene sequencing demonstrated that Bio-SiNPs treatment favorably reshaped structure and abundance of specific bacterial groups (Proteobacteria, Actinobacteriota, and Acidobacteriota) in the rhizosphere. Notably, Bio-SiNPs application significantly modulated the key metabolites (phenylacetaldehyde, glycitein, maslinic acid and methylmalonic acid) under both normal and Cd stress conditions. Overall, this study highlights that bio-nanoremediation using Bio-SiNPs enhances tolerance to Cd stress in bayberry plants by beneficially modulating biochemical, microbial, and metabolic attributes.

Abstract:Most studies have focused on regulation in a metabolic pathway in response to exogenous selenium under cadmium stress, rather than the change of key factors in soil and pepper plants. In this study, the correlations in environmental variables, microorganisms, metabolic pathways, Se and Cd morphology under nano-Se intervention were examined using metabolomics and microbial diversity in rhizosphere soil and pepper plants. The principal forms of Se in the soils were Se (VI) and SeCys, while SeMet and MeSeCys were the main components in the root, stem, leaves, and fruits in the treatment of nano-Se (5 and 20 mg/L) relative to the control. Soil enzymes,metabolites (fluorescein diacetate, urease, brassinolide, and p-hydroxybenzonic acid), and plant metabolites (rutin, luteolin, brassinolide, and abscisic acid) were remarkably enhanced by nano-Se fortification. The bio-enhancement of nano-Se can boost the beneficial microorganisms of Gammaproteobacteria, Alphaproteobacteria, Bacteroidia, Gemmatimonadetes, Deltaproteobacteria, and Anaerolineae in rhizosphere soil. Changes in microbial community were found to be strongly linked to the environment index, enzymes, soil metabolites, Se forms, which reduced Cd bioavailability and Cd accumulation in pepper plants. In conclusion, the nano-Se application integrates soil-plant balance by improving soil qualities and assigning signaling molecule levels in rhizosphere soil and pepper plants.
Cadmium Exposure Alters Rhizospheric Microbial Community and Transcriptional Expression of Vetiver Grass

Abstract:Vetiver grass ( Chrysopogon zizanioides L.) has been used to remediate cadmium (Cd)-contaminated soil, while there have been few studies on the influence of Cd exposure on the rhizospheric microbial community and transcriptional expression of C. zizanioides . In this study, we investigated the response of the rhizospheric microbial community and transcriptional expression of C. zizanioides in 20 mg/kg Cd-contaminated soil. The results showed that Cd levels in the roots and shoots of C. zizanioides reached 250.80 and 73.40 mg/kg, respectively. The Cd exposure changed the rhizospheric bacterial community, resulting in the significant enrichment of Sphingomonas , Lysobacter , and Gemmatimonadetes in Cd-contaminated soil. In addition, 880 and 3,419 differentially expressed genes were identified in the plant roots and shoots, respectively, in response to Cd stress. Among these, the overexpressed genes associated with redox homeostasis, glutathione (GSH) metabolism, cell wall biosynthesis, and transmembrane transport pathways were found to participate in Cd detoxification in C. zizanioides. These findings could be useful for understanding the selective variation of the rhizospheric microbial community and the detoxification mechanisms of C. zizanioides in Cd phytoremediation.

Abstract:The use of nitrogen (N) fertilizers increases crop yield, but the accumulation of residual N in agricultural soils poses significant environmental risks. Improving the N use efficiency (NUE) of crops can help reduce N pollution. While nanomaterials have been shown to enhance crop agronomic traits, more research is needed to clarify the regulatory mechanisms involved. In this study, foliar spraying of carbon dots (CDs, 1 mg mL −1 ) derived from Salvia miltiorrhiza increased the activity of plasma membrane H + -ATPase in Arabidopsis thaliana roots, promoting the uptake, transport, and assimilation of NO 3 − and NH 4 + . The upregulation of N metabolism-related genes, such as AtAMTs and AtNRTs , was also observed in A. thaliana roots. Transcriptome analysis suggested that this regulatory effect is mediated by the shoot-to-root mobile polypeptide CEPD1 (C-terminally encoded peptide DOWNSTREAM 1) signaling pathway. Additionally, foliar application of CDs increased the NUE of sweetpotato ( Ipomoea batatas (L.) Lam.) from 2.5% to 8.1%. The upregulation of genes such as CEPD1 in leaves was observed following CDs application under different N conditions. Finally, foliar spraying of CDs significantly increased field yield and enhanced tolerance to low N stress in sweetpotato. Overall, this study demonstrated that foliar application of CDs improved NUE in plants through CEPD1-dependent signaling.

Abstract:Trifolium repens (T. repens) can accumulate significant amounts of heavy metal ions, and has strong adaptability to wide environmental conditions, and relatively large biomass, which is considered a potential plant for phytoremediation. However, the molecular mechanisms of T. repens involved in Cd tolerance have not yet been studied in detail. This study was conducted to examine the integrative responses of T. repens exposed to a high-level CdCl2 by investigating the physiological and transcriptomic analyses. The results suggested that T. repens seedlings had a high degree of tolerance to Cd treatment. The roots accumulated higher Cd concentration than leaves and were mainly distributed in the cell wall. The content of MDA, soluble protein, the relative electrolyte leakage, and three antioxidant enzymes (POD, SOD, and APX) was increased with the Cd treatment time increasing, but the CAT enzymes contents were decreased in roots. Furthermore, the transcriptome analysis demonstrated that the differentially expressed genes (DEGs) mainly enriched in the glutathione (GSH) metabolism pathway and the phenylpropanoid biosynthesis in the roots. Overexpressed genes in the lignin biosynthesis in the roots might improve Cd accumulation in cell walls. Moreover, the DEGs were also enriched in photosynthesis in the leaves, transferase activity, oxidoreductase activity, and ABA signal transduction, which might also play roles in reducing Cd toxicity in the plants. All the above, clearly suggest that T. repens employ several different mechanisms to protect itself against Cd stress, while the cell wall biosynthesis and GSH metabolism could be considered the most important specific mechanisms for Cd retention in the roots of T. repens.

However, the synergistic potential of combining 24-epibrassinosteroid (24-epiBRs) and titanium oxide nanoparticles (TiO 2 -NPs) to alleviate Cd toxicity and restore soybean ( Glycine max L.) production remains unexplored. Thus, a pot-based experimental trial was conducted to assess the effects of applying TiO 2 -NPs (15 mg L −1 ) and 24-epiBRs (10 −7 M), individually and in combination, on soybean growth in soil cultivated with 30 ppm of Cd. The study revealed that Cd toxicity significantly inhibited soybean root length (11.0 %), root dry biomass (63.5 %), root fresh biomass (84.9 %), shoot length (11.7 %), shoot dry biomass (49.0 %), and shoot fresh biomass (27.3 %), compared to the control. Additionally, the toxicity of Cd enhanced the oxidative stress and lowered the photosynthetic efficiency, gas exchange characteristics, and antioxidant defense system of soybeans. Interestingly, the combined application of TiO 2 -NPs and 24-epiBRs ameliorated the Cd toxic effects and improved the agronomic traits, photosynthesis efficiency, and antioxidant activity in soybeans by lowering oxidative stress. Specifically, the dual application of 24-epiBRs and TiO 2 -NPs effectively lowered the Cd levels in roots, shoots, and leaves of soybean plants by 62.5, 162.7, and 87.1 %, respectively, relative to the control soybean plants grown under Cd stress. Overall, the combined treatment of TiO 2 -NPs and 24-epiBRs synergistically reduced Cd uptake and restored soybean physiology in Cd-contaminated soils. Moving forward, further research should include field trials to assess the effectiveness and economic viability of this novel method.

Abstract:The application of nanoparticles (NPs) has been proved as an efficient and promising technique for mitigating a wide range of stressors in plants. The present study elucidates the synergistic effect of iron oxide nanoparticles (IONPs) and silicon nanoparticles (SiNPs) in the attenuation of Cd toxicity in Phaseolus vulgaris . Seeds of P. vulgaris were treated with IONPs (10 mg/L) and SiNPs (20 mg/L). Seedlings of uniform size were transplanted to pots for 40 days. The results demonstrated that nanoparticles (NPs) enhanced growth, net photosynthetic rate, and gas exchange attributes in P. vulgaris plants grown in Cd-contaminated soil. Synergistic application of IONPs and SiNPs raised not only K + content, but also biosynthesis of polyamines (PAs), which alleviated Cd stress in P. vulgaris seedlings. Additionally, NPs decreased malondialdehyde (MDA) content and electrolyte leakage (EL) in P. vulgaris plants exposed to Cd stress. These findings suggest that stress alleviation was mainly attributed to the enhanced accumulation of K + content, improved antioxidant defense system, and higher spermidine (Spd) and putrescine (Put) levels. It is suggested that various forms of NPs can be applied synergistically to minimize heavy metal stress, thus increasing crop production under stressed conditions.

Role of Soil and Foliar‐Applied Carbon Dots in Plant Iron Biofortification and Cadmium Mitigation by Triggering Opposite Iron Signaling in Roots

Abstract:The Fe3+‐mobilizing and light‐emitting properties of CDs are determinants for triggering opposite Fe signaling in roots, which results in a specific modulatory pattern on the uptake, translocation, and accumulation of Fe, Mn, Zn, and Cd in plant response to soil and foliar‐applied CDs. In China, iron (Fe) availability is low in most soils but cadmium (Cd) generally exceeds regulatory soil pollution limits. Thus, biofortification of Fe along with mitigation of Cd in edible plant parts is important for human nutrition and health. Carbon dots (CDs) are considered as potential nanomaterials for agricultural applications. Here, Salvia miltiorrhiza‐derived CDs are an efficient modulator of Fe, manganese (Mn), zinc (Zn), and Cd accumulation in plants. CDs irrigation (1 mg mL−1, performed every week starting at the jointing stage for 12 weeks) increased Fe content by 18% but mitigated Cd accumulation by 20% in wheat grains. This finding was associated with the Fe3+‐mobilizing properties of CDs from the soil and root cell wall, as well as endocytosis‐dependent internalization in roots. The resulting excess Fe signaling mitigated Cd uptake via inhibiting TaNRAMP5 expression. Foliar spraying of CDs enhanced Fe (44%), Mn (30%), and Zn (19%) content with an unchanged Cd accumulation in wheat grains. This result is attributed to CDs‐enhanced light signaling, which triggered shoot‐to‐root Fe deficiency response. This study not only reveals the molecular mechanism underlying CDs modulation of Fe signaling in plants but also provides useful strategies for concurrent Fe biofortification and Cd mitigation in plant‐based foods.

Abstract:Cadmium (Cd) is a nonessential element in plants and has adverse effects on the growth and development of plants. However, the molecular mechanisms of Cd phytotoxicity, tolerance and accumulation in hyperaccumulators Solanum nigrum L. has not been well understood. Here, physiology, transcriptome, and metabolome analyses were conducted to investigate the influence on the S. nigrum under 0, 25, 50, 75 and 100 μM Cd concentrations for 7 days. Pot experiments demonstrated that compared with the control, Cd treatment significantly inhibited the biomass, promoted the Cd accumulation and translocation, and disturbed the balance of mineral nutrient metabolism in S. nigrum , particularly at 100 μM Cd level. Moreover, the photosynthetic pigments contents were severely decreased, while the content of total protein, proline, malondialdehyde (MDA), H 2 O 2 , and antioxidant enzyme activities generally increased first and then slightly declined with increasing Cd concentrations, in both leaves and roots. Furthermore, combined with the previous transcriptomic data, numerous crucial coding-genes related to mineral nutrients and Cd ion transport, and the antioxidant enzymes biosynthesis were identified, and their expression pattern was regulated under different Cd stress. Simultaneously, metabolomic analyses revealed that Cd treatment significantly changed the expression level of many metabolites related to amino acid, lipid, carbohydrate, and nucleotide metabolism. Metabolic pathway analysis also showed that S. nigrum roots activated some differentially expressed metabolites (DEMs) involved in energy metabolism, which may enhance the energy supply for detoxification. Importantly, central common metabolism pathways of DEGs and DEMs, including the "TCA cycle", "glutathione metabolic pathway" and "glyoxylate and dicarboxylate metabolism" were screened using conjoint transcriptomics and metabolomics analysis. Our results provide some novel evidences on the physiological and molecular mechanisms of Cd tolerance in hyperaccumulator S. nigrum plants.

Abstract:The effects of cadmium stress on nodulation, N2-fixation capabilities of the root nodule, the change in ultrastructure of the root nodule, soybean growth, and the distribution of cadmium in plants were studied. The results obtained show that the nodulation of soybean roots was greatly inhibited by the addition of Cd, especially at the addition level of 10 and 20 mgkg−1 soil. The inhibition of plant growth, especially the root growth, increased as the cadmium concentration increased, with deleterious effects observed for the roots. The weight ratio of soybean root/leaf decreased as the Cd concentration increased, which might explain the reason for nodulation decreases. The results also indicate that N2-fixation of root nodule was stimulated to some extent at the low levels of Cd addition, but decreased sharply with further increase of the Cd concentration. High Cd levels were also associated with changes in the ultrastructure of root nodule, in which the effective N2-fixing area was reduced and the N2-fixing cells in the area also reduced. In addition, the results also reveal that the content of Cd in different parts of the plants was as follows: roots≫stems>seeds, indicating that the accumulation of Cd by roots is much larger than that by any other part of the soybean plant, and might cause deleterious effects to root systems.
Nanoselenium transformation and inhibition of cadmium accumulation by regulating the lignin biosynthetic pathway and plant hormone signal transduction in pepper plants

Abstract:Abstract Selenium (Se) can promote the growth and resistance of agricultural crops as fertilizers, while the role of nano-selenium (nano-Se) against Cd remains unclear in pepper plants ( Capsicum annuum L.). Biofortification with nano-Se observably restored Cd stress by decreasing the level of Cd in plant tissues and boosting the accumulation in biomass. The Se compounds transformed by nano-Se were primarily in the form of SeMet and MeSeCys in pepper tissues. Differential metabolites and the genes of plant signal transduction and lignin biosynthesis were measured by employing transcriptomics and determining target metabolites. The number of lignin-related genes ( PAL , CAD , 4CL , and COMT ) and contents of metabolites (sinapyl alcohol, phenylalanine, p -coumaryl alcohol, caffeyl alcohol, and coniferaldehyde) were remarkably enhanced by treatment with Cd1Se0.2, thus, maintaining the integrity of cell walls in the roots. It also enhanced signal transduction by plant hormones and responsive resistance by inducing the biosynthesis of genes ( BZR1 , LOX3 , and NCDE1 ) and metabolites (brassinolide, abscisic acid, and jasmonic acid) in the roots and leaves. In general, this study can enable a better understanding of the protective mechanism of nano-Se in improving the capacity of plants to resist environmental stress.

Transcriptional and Metabolic Mechanism of Carbon Dots Enhancing Rice Growth and Resistance by Promoting Root

Abstract:Increasing climate change and pollutant discharge induce constant challenges to crops, while crops are vulnerable to environmental and pollutant stresses. In this study, a carbon dots (CDs) was developed that significantly increased rice seedling growth, and successfully reduced the inhibition of heavy metal cadmium (Cd), salt (NaCl), and herbicide 2,4-D stresses on rice seed-ling growth by pre-spraying. The root of rice seedlings responded specifically to CDs exposure, with significant improvements in root biomass, architecture, cell wall thickness, mechanical strength, and metabolic vitality. Metabolomics and transcriptomics were combined to reveal the regulatory mechanism of CDs in rice seedlings. Transcriptome analysis indicated that CDs up-regulated genes related to cytokinin, jasmonic acid, salicylic acid, MAPK signaling pathway, calcium homeostasis, and peroxidase, and downregulated those related to auxin, abscisic acid, and ethylene. Metabolomic analysis suggested CDs improved the metabolites related to antioxidant (betalain, ascorbate, aldarate, and glutathione), formation of cell wall, plasma membrane, xylem, and root cortex (phenylpropanoids biosynthesis, stilbenoid, diarylheptanoid and gingerol biosynthesis, and sphingolipid), and energy metabolism (nicotinate, nicotinamide, glyoxylate, dicarboxylate, and nitrate cycle) in rice seedlings. Therefore, pre-spraying CDs reprogrammed stress signaling pathways and enhanced adaptive responses in rice seedlings, ultimately increasing growth potential and stress resistance. This study presents a promising nano-bio-stimulant of CDs for crop resilience in the context of increasing climate change and contributes to sustainable agriculture.

Abstract:The potential influences of cadmium (Cd) on the biochemical processes of the soil nitrogen (N) cycle, along with the dynamics of ammonification, nitrification, and denitrification processes in the rhizosphere and non-rhizosphere (bulk soil), respectively, were investigated in a Cd-stressed system during an entire soybean growing season. In terms of Cd pollution at the seedling stage, the ammonifying bacteria proved to be the most sensitive microorganisms, whereas the effects of Cd on denitrification were not obvious. Following the growth of soybeans, the influences of Cd on ammonification in the bulk soil were: toxic impacts at the seedling stage, stimulatory effects during the early flowering stage, and adaptation to the pollutant during the podding and ripening stages. Although nitrification and denitrification in the bulk soil decreased throughout the entire growth cycle, positive adaptation to Cd stress was observed during the ripening stage. Moreover, during the ripening stage, denitrification in the bulk soil under high Cd treatment (20 mg kg−1) was even higher than that in the control, indicating a probable change in the ecology of the denitrifying microbes in the Cd-stressed system. Changes in the activity of microbes in the rhizosphere following plant growth were similar to those in the non-rhizosphere in Cd treatments; however, the tendency of change in the rhizosphere seemed to be more moderate. This suggested that there was some mitigation of Cd stress in the rhizosphere.
