Characterization of Fe Deficiency Induced RING Finger Family Members in Apple Species

Abstract:Iron (Fe) is a critical element for plants and Fe deficiency can cause adverse effects on their growth, development, yield and quality. RING finger family is a large group known for their role under abiotic stresses in plants. The main aim of this research work was to identify the RING finger family members that responded to Fe deficiency in apple species. Their genetic structure, molecular evolution, and expression levels were analyzed to study the underlying mechanism of RING finger genes under Fe deficiency stress. A total of 76 RING finger genes that responded to Fe deficiency were identified. Based on their structural characteristics and phylogenetic relationships, the identified genes were divided into four subfamilies: Group I, Group II, Group III and Group IV. Chromosomal localization demonstrated that the 76 RING finger genes were randomly distributed on 17 chromosomes. The genetic structure and amino-acid-motif encoded varied among different gene members. Fifteen pairs of RING finger genes were identified as segmental duplicated. Further, synteny analysis with Circos was performed to examine the evolutionary relationship between Malus domestica and other Rosaceae genomes. The results of qRT-PCR in Malus xiaojinensis showed expression levels of these genes under Fe deficiency, indicating that Fe deficiency could induce the expression of these genes, thus verifying the accuracy of transcriptome data. These results provided useful information about the evolutionary relationship of RING finger gene family, potentially important under Fe deficiency. Moreover, it will help to identify the function of these RING finger genes in future research.
