Abstract:Clonal propagation of plants by induction of adventitious roots (ARs) from stem cuttings is a requisite step in breeding programs. A major barrier exists for propagating valuable plants that naturally have low capacity to form ARs. Due to the central role of auxin in organogenesis, indole-3-butyric acid is often used as part of commercial rooting mixtures, yet many recalcitrant plants do not form ARs in response to this treatment. Here we describe the synthesis and screening of a focused library of synthetic auxin conjugates in Eucalyptus grandis cuttings and identify 4-chlorophenoxyacetic acid– l -tryptophan-OMe as a competent enhancer of adventitious rooting in a number of recalcitrant woody plants, including apple and argan. Comprehensive metabolic and functional analyses reveal that this activity is engendered by prolonged auxin signaling due to initial fast uptake and slow release and clearance of the free auxin 4-chlorophenoxyacetic acid. This work highlights the utility of a slow-release strategy for bioactive compounds for more effective plant growth regulation.

Root Development Enhanced by Using Indole-3-butyric Acid and Naphthalene Acetic Acid and Associated Biochemical Changes of in Vitro Azalea Microshoots

Abstract:Based on the importance of producing in vitro adventitious roots, this study was carried out to investigate the effects of indole-3-butyric acid (IBA) and naphthalene acetic acid (NAA) at a concentration of 2 mg L−1 on the formation of adventitious roots of azalea and their impact on biochemical changes and endogenous hormones. The rooting percentage, root number, and root length were increased in the microshoots of both studied cultivars (‘Mingchao’ and ‘Zihudie’) when the growth medium was supplemented with IBA. Additionally, peroxidase, indole acetic acid oxidase, hydrogen peroxide, and soluble protein contents were improved in both cultivars by auxin treatments especially during the first 7 days of the rooting period. However, application of IBA and NAA increased catalase and polyphenol oxidase in both cultivars during the first 14 and 28 days of culture. The increase in endogenous indole acetic acid (IAA) levels was accompanied by low activity of IAAO during most periods of root induction of microshoots in all treatments. Endogenous gibberellic acid levels were increased after 7 days of culture and then increased again after 28 days of culture. In contrast, the levels of endogenous zeatin riboside and isopentenyl adenosine were decreased with auxin treatments in the first period of the rooting process and then increased after 21 and 28 days of culture. The present study demonstrated that IBA at a concentration of 2 mg L−1 has a strong effect on azalea rooting. Moreover, the efficiency of IBA and NAA effects on biochemical changes during adventitious root induction was investigated, which may provide new horizons of in vitro rooting production and provide valuable information for the micropropagation of Rhododendron plants.
Root-Derived Auxin Contributes To The Phosphorus-Deficiency-Induced Cluster-Root Formation In White Lupin (Lupinus Albus)

Abstract:Formation of cluster roots is a typical morphological response to phosphorus (P) deficiency in white lupin (Lupinus albus), but its physiological and molecular mechanisms are still unclear. We investigated the role of auxin in the initiation of cluster roots by distinguishing the sources of auxin, measuring the longitudinal distribution patterns of free indole-3-acetic acid (IAA) along the root and the related gene expressions responsible for polar auxin transport (PAT) in different developmental stages of cluster roots. We found that removal of shoot apex or primary root apex and application of auxin-influx or -efflux transport inhibitors, 3-chloro-4-hydroxyphenylacetic acid, N-1-naphthylphthalamic acid and 2,3,5-triiodobenzoic acid, to the stem did not affect the number of cluster roots and the free-IAA concentration in the roots of P-deficient plants, but when these inhibitors were applied directly to the growth media, the cluster-root formation was greatly suppressed, suggesting the fundamental role of root-derived IAA in cluster-root formation. The concentration of free IAA in the roots was higher in P-deficient plants than in P-adequate ones, and the highest in the lateral-root apex and the lowest in the mature cluster roots. Meanwhile the expression patterns of LaAUX1, LaPIN1 and LaPIN3 transcripts related to PAT was consistent with concentrations of free IAA along the lateral root, indicating the contribution of IAA redistribution in the cluster-root development. We proposed that root-derived IAA plays a direct and important role in the P-deficiency-induced formation of cluster roots.
Synthetically derived BiAux modulates auxin co-receptor activity to stimulate lateral root formation

Abstract:Abstract The root system plays an essential role in plant growth and adaptation to the surrounding environment. The root clock periodically specifies lateral root prebranch sites (PBS), where a group of pericycle founder cells (FC) is primed to become lateral root founder cells and eventually give rise to lateral root primordia or lateral roots (LRs). This clock-driven organ formation process is tightly controlled by modulation of auxin content and signaling. Auxin perception entails the physical interaction of TRANSPORT INHIBITOR RESPONSE 1 (TIR1) or AUXIN SIGNALING F-BOX (AFBs) proteins with AUXIN/INDOLE-3-ACETIC ACID (Aux/IAA) repressors to form a co-receptor system. Despite the apparent simplicity, the understanding of how specific auxin co-receptors are assembled remains unclear. We identified the compound bis-methyl auxin conjugated with N-glucoside, or BiAux, in Arabidopsis (Arabidopsis thaliana) that specifically induces the formation of PBS and the emergence of LR, with a slight effect on root elongation. Docking analyses indicated that BiAux binds to F-box proteins, and we showed that BiAux function depends on TIR1 and AFB2 F-box proteins and AUXIN RESPONSE FACTOR 7 activity, which is involved in FC specification and LR formation. Finally, using a yeast (Saccharomyces cerevisiae) heterologous expression system, we showed that BiAux favors the assemblage of specific co-receptors subunits involved in LR formation and enhances AUXIN/INDOLE-3-ACETIC ACID 28 protein degradation. These results indicate that BiAux acts as an allosteric modulator of specific auxin co-receptors. Therefore, BiAux exerts a fine-tune regulation of auxin signaling aimed to the specific formation of LR among the many development processes regulated by auxin.

Abstract:In the present study, the effects of indole-3-butyric acid (IBA) alone and in combination with myo-inositol under in vitro conditions on morphogenic and biochemical responses in the MxM 14 (Prunus avium × Prunus mahaleb) sweet cherry rootstock were investigated using shoot tips as explants. The best treatment that induced the better response for rooting in terms of root number per rooted explant (16.46), root length (23.12 mm), root fresh weight (0.372 g), root dry weight (0.037 g) and rooting percentage (100 %) was 9.84 μΜ IBA in the absence of myo-inositol. Thus, myo-inositol suppressed the positive effect of IBA on rooting. The simultaneous application of IBA and myo-inositol achieved the better response for shoots development regarding their length (22.5–25.6 mm) in relation to the control (15 mm). However, the optimum shoot biomass was obtained with 9.84 μΜ IBA resulting in a 4-fold increase in shoot fresh (0.455 g) and dry weight (0.045 g) compared to the control. Regarding biochemical parameters, it was shown that there is no evident relation between chlorophyll levels, endogenous leaf proline or carbohydrate content in both leaves and roots, and improvement of rooting and shoot biomass in the respective observed treatments. On the contrary, the enhancement of rooting and shoot biomass could be related to the elevated endogenous proline levels in roots. According to the results obtained, there may be a correlation between the augmented proline levels in both leaves and roots in the IBA + myo-inositol combination treatments and shoot growth of MxM 14 explants (i.e. microcuttings height). The survival percentage of the ex vitro MxM 14 plantlets was 93 %. An efficient protocol of in vitro rooting and vegetative growth for the cherry rootstock MxM 14 was established using IBA and myo-inositol which constitutes a very useful tool for future genetic transformation.

Abstract:Deeper and longer roots allow crops to survive and flourish, but our understanding of the plant growth regulators promoting root system establishment is limited. Here, we report that, a novel auxin receptor agonist, named K-10, had a remarkable promotive effect on root growth in both Arabidopsis thaliana and Oryza sativa through the enhancement of root-related signaling responses. Using computer-aided drug discovery approaches, we developed potent lead compound by screening artificial chemicals on the basis of the auxin receptor TIR1 (Transport Inhibitor Response 1), and a series of N-(benzo[d] [1,3] dioxol-5-yl)-2-(one-benzylthio) acetamides, K-1 to K-22, were designed and synthesized. The results of bioassay showed that K-10 exhibited an excellent root growth-promoting activity far exceeding that of NAA (1-naphthylacetic acid). A further morphological investigation of the auxin related mutants ( yucQ , tir1 ) revealed that K-10 had auxin-like physiological functions and was recognized by TIR1, and K-10 significantly enhanced auxin response reporter's ( DR5:GUS ) transcriptional activity. Consistently, transcriptome analysis showed that K-10 induced a common transcriptional response with auxin and down-regulated the expression of root growth-inhibiting genes. Further molecular docking analysis revealed that K-10 had a stronger binding ability with TIR1 than NAA. These results indicated that this class of derivatives could be a promising scaffold for the discovery and development of novel auxin receptor agonists, and the employment of K-10 may be effective for enhancing root growth and crop production.

Abstract:Abscisic acid (ABA) is a key signaling molecule promoting ripening of non-climacteric fruits such as sweet cherry (Prunus avium L.). To shed light on the role of other hormones on fruit development, ripening and anthocyanin production, the synthetic auxin 1-naphthaleneacetic acid (NAA) was applied to sweet cherry trees during the straw-color stage of fruit development. NAA-treated fruits exhibited higher concentrations of 1-aminocyclopropane-1-carboxylic acid (ACC) and ABA-glucose ester (ABA-GE), which are a precursor of ethylene and a primary storage form of ABA, respectively. Consistent with these observations, transcript levels of genes encoding ACC synthase and ACC oxidase, both involved in ethylene biosynthesis, were increased after 6 days of NAA treatment, and both ABA concentration and expression of the regulator gene of ABA biosynthesis (NCED1 encoding 9-cis-epoxycarotenoid dioxygenase) were highest during early fruit ripening. In addition, transcript levels of key anthocyanin regulatory, biosynthetic and transport genes were significantly upregulated upon fruit exposure to NAA. This was accompanied by an increased anthocyanin concentration and fruit weight whilst fruit firmness and cracking index decreased. Altogether our data suggest that NAA treatment alters ethylene production, which in turn induces ripening in sweet cherry and enhanced anthocyanin production, possibly through ABA metabolism. The results from our study highlight the potential to use a single NAA treatment for manipulation of cherry ripening.

Abstract:Growth regulation tailors development in plants to their environment. A prominent example of this is the response to gravity, in which shoots bend up and roots bend down1. This paradox is based on opposite effects of the phytohormone auxin, which promotes cell expansion in shoots while inhibiting it in roots via a yet unknown cellular mechanism2. Here, by combining microfluidics, live imaging, genetic engineering and phosphoproteomics in Arabidopsis thaliana, we advance understanding of how auxin inhibits root growth. We show that auxin activates two distinct, antagonistically acting signalling pathways that converge on rapid regulation of apoplastic pH, a causative determinant of growth. Cell surface-based TRANSMEMBRANE KINASE1 (TMK1) interacts with and mediates phosphorylation and activation of plasma membrane H+-ATPases for apoplast acidification, while intracellular canonical auxin signalling promotes net cellular H+ influx, causing apoplast alkalinization. Simultaneous activation of these two counteracting mechanisms poises roots for rapid, fine-tuned growth modulation in navigating complex soil environments.

Abstract:Acer rubrum is difficult to root during cutting propagation. Auxin/indole-acetic acids (Aux/IAA) proteins, which are encoded by the early response genes of auxin, are transcriptional repressors that play important roles in auxin-mediated root growth and development. In this study, ArAux/IAA13 and ArAux/IAA16, which were significantly differentially expressed after 300 mg/L indole butyric acid treatment, were cloned. Heatmap analysis revealed that they might be associated with the process of adventitious root (AR) growth and development mediated by auxin. Subcellular localization analysis showed that they performed their function in the nucleus. Bimolecular fluorescence complementation assays revealed the interactions between them and two auxin response factor (ARF) proteins, ArARF10 and ArARF18, confirming their relevance to AR growth and development. Overexpression of transgenic plants confirmed that the overexpression of ArAux/IAA13 and ArAux/IAA16 inhibited AR development. These results help elucidate the mechanisms of auxin-mediated AR growth and development during the propagation of A. rubrum and provide a molecular basis for the rooting of cuttings.

Abstract:Auxin, cytokinin, and abscisic acid (ABA) are important plant hormones that regulate many growth and developmental processes. In recent years, a number of genes involved in the metabolic and signaling pathways for auxin, cytokinin and ABA have been cloned and characterized. With organ- and tissue-specific or conditionally active gene promoters, it has become possible to manipulate concentrations of plant hormones in planta to create commercially desirable traits. Seedless fruit production and the extension of shelf life of green produce and ornamental plants are two successful examples of manipulating concentrations of these hormones in planta. In this review, we will focus our discussion on the effects of the over- or under-expression of genes involved in the biosynthetic and catabolic pathways of these hormones with an emphasis on their potential applications in ornamental crops. Role of auxin on adventitious root formation and subsequent growth of cutting raised plantlets of Ginkgo biloba L.

Abstract:Role of some auxins, indol-3-butyric acid (IBA) and α-naphthalene acetic acid (NAA) has been examined for their stimulatory effects on adventitious root formation in stem cuttings of Ginkgo biloba as well as on subsequent growth and survival of these cutting raised plantlets. Lower concentration of IBA (10.0 µM) was found to be the most effective treatment as it not only induced maximum rooting (88.89%) but also enhanced number of roots, length of roots and length of longest root to the maximum. Further, the growth performance of these cutting raised plantlets, via control and IBA treatment (best treatment), was compared. It was found that the IBA treated plantlets were morphologically healthy in terms of their shoot height, diameter of shoot, number of nodes per cutting, number of leaves/node and number of branches per cutting than the control plants. Besides this, the survival rate of IBA treated plantlets was 100% in comparison to control where it was 87.5%. Therefore it was concluded that IBA treatment not only improve the percent rooting but also improve the subsequent growth and survival rate of the plantlets of G. biloba. Abstract:Auxin elicits diverse cell behaviors through a simple nuclear signaling pathway initiated by degradation of Aux/IAA co-repressors. Our previous work revealed that members of the large Arabidopsis Aux/IAA family exhibit a range of degradation rates in synthetic contexts. However, it remained an unresolved issue whether differences in Aux/IAA turnover rates played a significant role in plant responses to auxin. Here, we use the well-established model of lateral root development to directly test the hypothesis that the rate of auxin-induced Aux/IAA turnover sets the pace for auxin-regulated developmental events. We did this by generating transgenic plants expressing degradation rate variants of IAA14, a crucial determinant of lateral root initiation. Progression through the well-established stages of lateral root development was strongly correlated with the engineered rates of IAA14 turnover, leading to the conclusion that Aux/IAAs are auxin-initiated timers that synchronize developmental transitions.

Ethylene Action Inhibition Improves Adventitious Root Induction in Adult Chestnut Tissues

Abstract:Phase change refers to the process of maturation and transition from the juvenile to the adult stage. In response to this shift, certain species like chestnut lose the ability to form adventitious roots, thereby hindering the successful micropropagation of adult plants. While auxin is the main hormone involved in adventitious root formation, other hormones, such as ethylene, are also thought to play a role in its induction and development. In this study, experiments were carried out to determine the effects of ethylene on the induction and growth of adventitious roots. The analysis was performed in two types of chestnut microshoots derived from the same tree, a juvenile-like line with a high rooting ability derived from basal shoots (P2BS) and a line derived from crown branches (P2CR) with low rooting responses. By means of the application of compounds to modify ethylene content or inhibit its signalling, the potential involvement of this hormone in the induction of adventitious roots was analysed. Our results show that ethylene can modify the rooting competence of mature shoots, while the response in juvenile material was barely affected. To further characterise the molecular reasons underlying this maturation-derived shift in behaviour, specific gene expression analyses were developed. The findings suggest that several mechanisms, including ethylene signalling, auxin transport and epigenetic modifications, relate to the modulation of the rooting ability of mature chestnut microshoots and their recalcitrant behaviour.

Abstract:Auxin is a key hormone that regulates plant growth and development, including plant shape and sensitivity to environmental changes. Auxin is biosynthesized and metabolized via many parallel pathways, and it is sensed and transduced by both normal and atypical pathways. The production, catabolism, and signal transduction pathways of auxin primarily govern its role in plant growth and development, and in the response to stress. Recent research has discovered that auxin not only responds to intrinsic developmental signals, but also mediates various environmental signals (e.g., drought, heavy metals, and temperature stresses) and interacts with hormones such as cytokinin, abscisic acid, gibberellin, and ethylene, all of which are involved in the regulation of plant growth and development, as well as the maintenance of homeostatic equilibrium in plant cells. In this review, we discuss the latest research on auxin types, biosynthesis and metabolism, polar transport, signaling pathways, and interactions with other hormones. We also summarize the important role of auxin in plants under abiotic stresses. These discussions provide new perspectives to understand the molecular mechanisms of auxin's functions in plant development.
Abscisic acid biosynthesis is necessary for full auxin effects on hypocotyl elongation

Abstract:In concert with other phytohormones, auxin regulates plant growth and development. However, how auxin and other phytohormones coordinately regulate distinct processes is not fully understood. In this work, we uncover an auxin-abscisic acid (ABA) interaction module in Arabidopsis that is specific to coordinating activities of these hormones in the hypocotyl. From our forward genetics screen, we determine that ABA biosynthesis is required for the full effects of auxin on hypocotyl elongation. Our data also suggest that ABA biosynthesis is not required for the inhibitory effects of auxin treatment on root elongation. Our transcriptome analysis identified distinct auxin-responsive genes in root and shoot tissues, which is consistent with differential regulation of growth in these tissues. Further, our data suggest that many gene targets repressed upon auxin treatment require an intact ABA pathway for full repression. Our results support a model in which auxin stimulates ABA biosynthesis to fully regulate hypocotyl elongation.

Abstract:Root parasitic plants in the Orobancheceae, such as and , cause significant damage to crop production. The germination step of these root parasitic plants is induced by host-root-derived strigolactones (SLs). After germination, the radicles elongate toward the host and invade the host root. We have previously discovered that a simple amino acid, tryptophan (Trp), as well as its metabolite, the plant hormone indole-3-acetic acid (IAA), can inhibit radicle elongation of . These results suggest that auxin plays a crucial role in the radicle elongation step in root parasitic plants. In this report, we used various auxin chemical probes to dissect the auxin function in the radicle growth of and . We found that synthetic auxins inhibited radicle elongation. In addition, auxin receptor antagonist, auxinole, rescued the inhibition of radicle growth by exogenous IAA. Moreover, a polar transport inhibitor of auxin, -1-naphthylphthalamic acid (NPA), affected radicle tropism. We also proved that exogenously applied Trp is converted into IAA in seeds, and auxinole partly rescued this radicle elongation. Our data demonstrate a pivotal role of auxin in radicle growth. Thus, manipulation of auxin function in root parasitic plants should offer a useful approach to combat these parasites.

Phloroglucinol Improves Direct Rooting of In Vitro Cultured Apple Rootstocks M9 and M26

Abstract:Advances in micropropagation techniques have helped produce true-to-type clones of many horticulturally important plants. However, several cultivars of apple are difficult to root in vitro. In these cases, adventitious roots are induced together with undesirable formation of callus, which decrease the acclimatization rate of in vitro produced plantlets. In this study, two apple rootstocks, M9 and M26, were subjected to different concentrations of indole-3-butyric acid (IBA) to induce root formation. Although addition of IBA to the medium induced root formation, rhizogenesis was accompanied by the undesirable formation of callus in both cultivars. On the other hand, in gene expression analysis, the indole-3-acetic acid (IAA) synthase genes AAO1 and YUC1 were expressed more highly in M9 than in M26. This suggests that endogenous auxin levels may be higher in M9, which may explain why M9 plantlets are difficult to root and experience high levels of callus formation during propagation. In addition, rooting medium containing 0.1 mg·L−1 IBA was supplemented with different concentrations of phloroglucinol (0, 0.5, 1.0, and 2.0 mM) to examine whether direct rooting efficiency in the M9 could be improved. Addition of 1.0 mM phloroglucinol increased rooting percentage and decreased callus formation in the M9 rootstock. The rootstock M9 is a desirable cultivar but presents a problem with true-to-type direct rooting. Addition of phloroglucinol may improve direct rooting and eliminate callus formation during propagation.

Abstract:To evaluate the effect of auxin and auxin analogues on in vitro rooting, and provide a useful suggestion for the establishment of efficient rooting protocol in plants, the roles of IAA, Indole-3-butyric acid (IBA), and alpha-naphthaleneacetic acid (NAA) in regulating root system development during the in vitro successive subculturing of mature trees in Pinus massoniana were studied. Firstly, results for variations in endogenous plant growth regulators (PGRs) levels and rooting capacity of shoots in vitro cultured on half-strength Murashige and Skoog medium (MMS) containing 1.2 mu M l(-1) NAA showed that the endogenous IAA level in shoots was significantly enhanced with the increase of number of subcultures, and there was a remarkably linear relationship between the IAA level and rooting (%) or root number. The culture of shoots on half-strength MMS medium enriched with 4 mu M l(-1) IAA resulted in increases in both rooting (%) and root number without any negative effect on root development, while 4 mu M l(-1) IBA caused the decrease in root number. IBA of lower concentration (2 mu M l(-1) enhanced rooting (%) and root number, but its promotive effects on root formation were delayed compared with effects of 4 mu M l(-1)IAA. These results suggest that the use of IAA for the in vitro rooting of mature P. massoniana trees is efficient.

Abstract:Plant cells possess the unique ability to revert differentiated somatic cells back to a pluripotent state, a trait utilized in in vitro tissue culture. This plant propagation strategy involves the exogenous supplementation of auxin and cytokinin, two phytohormones that promote cellular reprogramming and de novo organogenesis. When exposed to an auxin-rich medium, somatic cells proliferate and form the callus, a pluripotent cell mass. Following the attainment of pluripotency, the callus can be transferred to a cytokinin-rich medium to induce shoot regeneration.
