Abstract:Dissolved organic matter (DOM) affects arsenite [As(III)] toxicity by altering its sorption equilibrium at the cell wall interface. A better understanding of such mechanism is of great importance to assess As(III) ecotoxicity in aquatic systems. Batch experiments were conducted to study the effects of DOM on the regulation of As(III) sorption and toxicity in the diatom Navicula sp. The influence of humic acid (HA) on As(III) toxicity was assessed by measuring algal growth, chlorophyll a , and reactive oxygen species (ROS), whereas As(III) mobility across the cell wall was estimated by determining the concentration of intracellular, cell-wall-bound, and free As(III) ions in cell media. Results showed that the effects of HA on arsenite toxicity varied depending on various combinations of As(III)-HA concentrations. EC 50 had an approximate threefold increase from 8.32 (HA-free control) to 22.39 μM (at 20 mg L −1 HA) when Navicula sp. was exposed to 1.0–100.0 μM of As(III), compared to an overall low complexation ratio of HA-As(III) in a range of 0.91–6.00 %. The cell wall-bound and intracellular arsenic content decreased by 19.8 and 20.3 %, respectively, despite the lower arsenite complexation (2.10 ± 0.16 % of the total As). Meanwhile, intracellular ROS was decreased by 12.6 % in response to 10.0 μM As(III) and 10 mg L −1 HA vs. the HA-free control. The significant contrast indicated that complexation alone could not explain the HA-induced reduction in arsenite toxicity and other factors including HA–cell surface interactions may come into play. Isotherms describing adsorption of HA to the Navicula sp. cells combined with morphological data by scanning electron microscopy revealed a protective HA floccule coating on the cell walls. Additional Fourier transform infrared spectroscopic data suggested the involvement of carboxylic groups during the adsorption of both HA and As(III) on the Navicula sp. cell surface. Collective data from this study suggest that cell wall-bound HA can moderate As(III) toxicity through the formation of a protective floccule coating occupying As(III) sorption sites and decreased effective functional groups capable of binding As(III). Our findings imply that As(III) toxicity can be alleviated due to the increased hindrance to cellular internalization of As(III) in the presence of naturally abundant DOM in water.
Regulation mechanisms of humic acid on Pb stress in tea plant (<i>Camellia sinensis</i> L.)

Abstract:Though the interaction between humic acid (HA) and heavy metals has been widely reported, the effects of HA on the toxicity of heavy metals to plants are still in debate. In this study, the regulation mechanisms of HA on Pb stress in tea plant (Camellia sinensis L.) was investigated through hydroponic experiments, and the experimental results were explained by using transmission electron microscope (TEM), scanning transmission X-ray microscopes (STXM) and isobaric tags for relative and absolute quantitation (iTRAQ) differential proteomics. Significant alleviation of Pb stress was found with HA coexistence. TEM results showed that HA greatly mitigated the damage of cells caused by Pb stress. Compared with sole Pb treatment, the addition of HA increased the contents of pectin and pectic acid in the cell wall by 10.5% and 30.5%, while arabinose (Ara) and galactose (Gal) decreased by 20.5% and 15.9%, respectively, which were beneficial for increasing Pb adsorption capacity of the cell wall and promoting cell elongation. Moreover, iTRAQ differential proteomics analysis proved that HA strengthened the antioxidant system, promoted the synthesis of cell wall, and stabilized protein and sulfur-containing substance metabolism in molecular level. Notably, the concentration of calcium (Ca) in the cell wall of HA coexistence treatment was 47.4% higher than Pb treatment. STXM results also indicated that the distribution of Ca in the cell wall was restored with the presence of HA. This might promote the formation of the egg-box model, thus alleviating Pb stress in cells. Our results reveal the regulation mechanisms of HA on Pb detoxification in plants and provide useful information for improving the safety of agricultural products. (c) 2020 Elsevier Ltd. Abstract:In this study, influences of cations (Na+, K+, Ca2+, and Mg2+), anions (NO3-, Cl-, and SO42-), and humic acid (HA) on the antimicrobial efficacy of silver nanoparticles (AgNPs)/Ag+ against Phanerochaete chrysosporium were investigated by observing cell viability and total Ag uptake. K+ enhanced the antimicrobial toxicity of AgNPs on P. chrysosporium, while divalent cations decreased the toxicity considerably, with preference of Ca2+ over Mg2+. Impact caused by a combination of monovalent and divalent electrolytes was mainly controlled by divalent cations. Compared to AgNPs, however, Ag+ with the same total Ag content exhibited stronger antimicrobial efficacy towards P. chrysosporium, regardless of the type of electrolytes. Furthermore, HA addition induced greater microbial activity under AgNP stress, possibly originating from stronger affinity of AgNPs over Ag+ to organic matters. The obtained results suggested that antimicrobial efficacy of AgNPs was closely related to water chemistry: addition of divalent electrolytes and HA reduced the opportunities directly for AgNP contact and interaction with cells through formation of aggregates, complexes, and surface coatings, leading to significant toxicity reduction; however, in monovalent electrolytes, the dominating mode of action of AgNPs could be toxic effects of the released Ag+ on microorganisms due to nanoparticle dissolution.

Abstract:This work investigated the role of natural organic matter (NOM) in the environmental processes of silver nanoparticles (AgNP) and the uptake and accumulation of AgNP in wheat. Different NOMs (Suwannee River humic acids [SRHA], fulvic acid [FA]) and Ag elements (Ag-(0) and Ag+) were incubated in a hydroponic media for 15 days. The results showed that the NOM (10 mg C L-1) altered the dissolution, stabilization, uptake and accumulation of AgNP. The dissolution of AgNP declined in the presence of NOM. Compared with FA, the dissolved Ag+ decreased much more from 0.30 mg L-1 to 0.10 mg L-1 in the presence of SRHA. The fluorescence quenching results indicated that SRHA exhibited stronger binding to Ag+ than that of FA, and the quenching constants Ksv were 0.1309 (SRHA) and 0.0074 (FA), respectively. C-O, C-H, C-O-C, and Me-OH were involved in the interaction between NOM and AgNP. The NOM decreased the accumulated content of Ag in wheat. Hence, NOM alleviated the inhibition of AgNP to wheat growth. SRHA reduced the Ag content of wheat roots approximately 3-fold. These results clearly indicated the importance of NOM on altering the behavior, fate and toxicity of AgNP in an environment. (C) 2018 Elsevier Ltd. All rights reserved.
The influence of humic acid on the toxicity of nano-ZnO and Zn2+ to the Anabaena sp

Abstract:This study explored the effects of humic acid (HA) on the toxicity of ZnO nanoparticles (nano-ZnO) and Zn(2+) to Anabaena sp. Typical chlorophyll fluorescence parameters, including effective quantum yield, photosynthetic efficiency and maximal electron transport rate, were measured by a pulse-amplitude modulated fluorometer. Results showed that nano-ZnO and Zn(2+) could inhibit Anabaena sp. growth with the EC50 (concentration for 50% of maximal effect) of 0.74 ± 0.01 and 0.3 ± 0.01 mg/L, respectively. In the presence of 3.0 mg/L of HA, EC50 of nano-ZnO increased to 1.15 ± 0.04 mg/L and EC50 of Zn(2+) was still 0.3 ± 0.01 mg/L. Scanning electron microscopy observation revealed that HA prevented the adhesion of nano-ZnO on the algae cells due to the increased electrostatic repulsion. The generation of intracellular reactive oxygen species and cellular lipid peroxidation were significantly limited by HA. Nano-ZnO had more damage to the cell membrane than Zn(2+) did, which could be proven by the malondialdehyde content in Anabaena sp. cells. © 2014 Wiley Periodicals, Inc. Abstract:In the process of activated sludge against silver nanoparticles (AgNPs) present in wastewater, soluble microbial products (SMP) produced by microbial secretion or macromolecular cell lysis formed a reactive barrier against AgNPs. Proteins (PN) and humic acids (HA), the primary constituents of SMP, were extracted from activated sludge in this study. The adsorption mechanism of HA, which had excellent adsorption properties, on AgNPs was first analyzed. Then, Fourier transform infrared spectroscopy (FTIR) and X-ray photoelectron spectroscopy (XPS) were further used to explore the bonding mechanisms of AgNPs by HA and PN, respectively. The results showed that the surface structure of HA was cracked after resisting AgNPs, and the adsorption process on AgNPs was monomolecular layer adsorption, with the maximum adsorption capacity of 12.9 mg/g at 35 degrees C. Thus, HA was an adsorbent with medium adsorption capacity. The FTIR and XPS analyses demonstrated that HA chelated and chemisorbed with AgNPs through chemical bonds such as -OH, C-H, C--C, C-N, C--O, and C-O-C, thereby reducing the microbial cytotoxicity of AgNPs. The chemical reaction between PN and AgNPs occurred via -OH and C-H dependent hydrogen bonding, and N-H was oxidized to -NO3, which ultimately resulted in the destruction of the PN peptide chain structure and the change of PN conformation. The present study investigated the resistance mechanism of HA and PN, the main components of SMP in activated sludge system, under the stress of AgNPs, which can provide theoretical support for the in-situ bioremediation of AgNPs in wastewater and the potential application of SMP.

Abstract:The fate and toxicity of silver nanoparticles (AgNPs) and ions in water bodies is largely determined by the natural organic matter (NOM)-mediated redox cycling. However, the process of NOM-mediated redox cycling in the day/night cycles remains elusive. In this study, the inter-transformation between AgNPs and Ag+ ion caused by humic acid (HA) was investigated under controlled light and dark conditions. It was shown that HA induced the reduction of Ag+ into AgNPs in simulated sunlight, and also oxidize AgNPs to release Ag+ in darkness. Kinetics data demonstrated that the rates of AgNPs formation and dissolution increased along with the increment of HA concentrations. Along with the pH increase, the reduction of Ag+ accelerated, but the oxidative dissolution of AgNPs was inhibited. In day-night cycles, the AgNPs and Ag+ concentrations exhibited similar wave-shaped change curves. The peaks of AgNPs and Ag+ ion appeared at 7 p.m. and 7 a.m., respectively. The toxicity of AgNPs/Ag+ to Escherichia coli was determined primarily by the concentration of dissolved Ag+, but also affected by the particle-specific toxicity. The dual role of HA implied that previous reports about the photo-reduction of Ag+ to AgNPs by NOM should be reconsidered, and the oxidizability of HA in darkness strongly affect the transformation and toxicity of AgNPs in water.

Abstract:A hydroponic experiment combined with synchronous radiation X-ray fluorescence (SRXRF) analysis was designed to understand the influence of humic acid (HA) in tea plants under lead stress. The results showed that the quantitative relationship (QR) between humic acid and Pb is an important factor affecting the regulation of humic acid with respect to the accumulation of Pb in tea plants. Besides, excess humic acid might stimulate the accumulation of Pb in the root cell wall and transfer to the shoot organs through undifferentiated casparian band structure. This study could provide a theoretical basis for the scientific evaluation of the effect of humic acid on tea uptake and the accumulation of Pb and the practical application of humic acid in reducing Pb pollution in the field.

Abstract:Adsorption of dissolved organic matter (DOM) can alter the environmental fate, bioavailability and toxicity of silver nanoparticles (Ag NPs). However, a number of questions remain about DOM's ability to modify nanotoxicity. Here, we examine the impact of humic acid (HA, as a model DOM) on the toxicity of Ag NPs (10 μg L-1) in the marine clam Ruditapes philippinarum. Results showed that DOM additions to Ag NP treatments reduce clam silver tissue burdens and the oxidative stress response. However, HA does not significantly affect the impact of Ag NPs on clam acetylcholinesterase activity and feeding behavior (measured as filtration rate). Overall, the integrated biological response index supports the conclusion that humic acid reduces the toxicity of Ag NPs, clearly indicating the importance of considering environmental factors when assessing potential risks posed by nanomaterials in natural settings.

Abstract:In an aquatic environment, nanoparticles (NPs) acquire coatings of natural organic matter (NOM) that alter the fate and toxicity of NPs. However, the essential relationship between engineered NP properties and NOM assemblage that determines how NPs are modified and behave at biological interfaces remains unclear. Herein, we use a complementary experimental and molecular dynamics approach to elucidate the mechanisms underlying adsorption of humic acid (HA) on NPs of different surface properties and effects on NP interactions with bacteria. Our results demonstrate that hydrophobic and positively charged NPs are more capable of adsorbing HAs in unique standing or lying conformations that expose particular groups to modify NP properties. In particular, HA adsorption increases effective NP size, reduces NP's hydrophobicity, and reverses NP's surface charge. As a consequence of HA adsorption, the binding of NPs to bacteria is sterically hindered and suppressed by electrostatic repulsion between negatively charged groups in adsorbed HAs and lipopolysaccharide on the bacterial membrane. The toxicity of NPs is mitigated as seen by reductions of NP association with bacteria and NP-induced membrane damage. These findings provide compelling explanations on the cascading events from dictation of HA adsorption by engineered NP properties to how the modified NP surfaces change their toxicity.
The competition of humic acid aggregation and adsorption on clay particles and its role in retarding heavy metal ions

Abstract:Humic acid (HA) is of great importance in controlling the fate of heavy metals (HMs), however, the pivotal influence of HA aggregation within the HA-clay-HM ternary system on retarding HM mobility remains obscure. This study performed molecular dynamics simulations to delve into the consequences of HA aggregation on the environmental behavior of Cd2+ and Pb2+ (0.1-0.6 M) in the co-existence of illite particles. HA can readily aggregate into clusters, adhering to the illite surface or freely dispersing in the solution. These HA clusters significantly modulate HM mobility, contingent upon their location, arrangement, and interaction with illite. Consequently, HA exhibited a pronounced retardation effect on HM migration, stemming from the competition between HA aggregation and its adsorption on illite. Additionally, the retardation effect of HA aggregation was more obvious for Cd2+ (as compared to Pb2+), owing to its stronger interaction with the functional groups of HA. These findings contribute to the development of potential HA-based strategies for remediation of heavy metal-contaminated sites.

Abstract:Studies regarding the effect of environmental factors on the environmental behaviour and potential toxicity of nanoplastics (NPs) are limited but important. In this study, four polystyrene NPs with different functional groups and charges (PS, PS-COOH, n-PSNH2, p-PSNH2) were selected to investigate the effect of humic acid (HA) and salinity on their aggregation behaviour and toxicity. The results showed that salinity significantly accelerated the aggregation of the four NPs, while HA mainly exerted a stabilizing effect on the three negatively charged NPs. In contrast, the positively charged p-PSNH2 aggregated significantly at first but remained stable as HA concentration further increased. The joint effect mainly depended on their concentration ratio. The aggregation phenomena can be explained by the Derjaguin - Landau - Verwey - Overbeek (DLVO) theory. Also, the acute toxicity of NPs on Daphnia magna was affected by the surface charge of NPs, and the positively charged p-PSNH2 showed the lowest toxicity among the selected NPs. Furthermore, the presence of HA effectively alleviated the toxicity of PS and p-PSNH2, as the survival rates increased from 15% to 45%-95% and 100% respectively. Our results demonstrate that the surface properties of NPs significantly influence their aggregation and toxicity.

Abstract:In this research, several biochemical variations in plant of Lemna minor L. were investigated to reflect Ag+ toxicity. Lemna minor L. changed colorless AgNO3 to colloidal brown at doses equal to and greater than 1 mg L−1. Optical and fluorescence microscopy revealed the presence of bright spots in roots of tested plant related to Ag/Ag2O-NPs. Photosynthetic pigment contents of Lemna minor L. declined upon exposure to Ag+ with an evidently higher decrease in chlorophyll a than in chlorophyll b. Similarly, Ag+ treatment caused an evident reduction in the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). The reduction in antioxidase activity was significantly higher in POD than in SOD and CAT. Ag+ treatment resulted in a significant increment in the level of malondialdehyde (MDA) content as the judging criteria of cellular injury which showed sign of dose-related. The alterations occurred in RAPD profiles of treated samples following Ag+ toxicity containing loss of normal bands, appearance of new bands, and variation in band intensities compared with the normal plants. In addition, morphological character and biomass of Lemna minor L. subjected to increasing Ag+ concentrations were evaluated to reveal Ag+ toxicity. Our study demonstrated that Lemna minor L. have a high sensitivity to indicate fluctuation of water quality. It would be beneficial that modulating the genotype of Lemna minor L. to bear high proportion of contaminates.

Abstract:Nowadays nanomaterials have been widely used to remove heavy metals from water/wastewater due to their large surface area and high reactivity. Humic acid (HA) and fulvic acid (FA) exist ubiquitously in aquatic environments and have a variety of functional groups which allow them to complex with metal ions and interact with nanomaterials. These interactions can not only alter the environmental behavior of nanomaterials, but also influence the removal and transportation of heavy metals by nanomaterials. Thus, the interactions and the underlying mechanisms involved warrant specific investigations. This review outlined the effects of HA/FA on the removal of heavy metals from aqueous solutions by various nanomaterials, mainly including carbon-based nanomaterials, iron-based nanomaterials and photocatalytic nanomaterials. Moreover, mechanisms involved in the interactions were discussed and potential environmental implications of HA/FA to nanomaterials and heavy metals were evaluated.
Effects of Humic Acid on the Interactions Between Zinc Oxide Nanoparticles and Bacterial Biofilms

Abstract:The effects of humic acid (HA) on interactions between ZnO nanoparticles (ZnO NPs) and Pseudomonas putida KT2440 biofilms at different maturity stages were investigated. Three stages of biofilm development were identified according to bacterial adenosine triphosphate (ATP) activity associated with biofilm development process. In the initial biofilm stage 1, the ATP content of bacteria was reduced by more than 90% when biofilms were exposed to ZnO NPs. However, in the mature biofilm stages 2 and 3, the ATP content was only slightly decreased. Biofilms at stage 3 exhibited less susceptibility to ZnO NPs than biofilms at stage 2. These results suggest that more mature biofilms have a significantly higher tolerance to ZnO NPs compared to young biofilms. In addition, biofilms with intact extracellular polymeric substances (EPS) showed higher tolerance to ZnO NPs than those without EPS, indicating that EPS play a key role in alleviating the toxic effects of ZnO NPs. In both pure ZnO NPs and ZnO-HA mixtures, dissolved Zn2+ originating from the NPs significantly contributed to the overall toxicity. The presence of HA dramatically decreased the toxicity of ZnO NPs due to the binding of Zn2+ on HA. The combined results from this work suggest that the biofilm maturity stages and environmental constituents (such as humic acid) are important factors to consider when evaluating potential risks of NPs to ecological systems.
Influence of humic acid on algal uptake and toxicity of ionic silver.

Abstract:The biogeochemical cycle of silver has been profoundly disturbed by various anthropogenic activities. To better understand the relationship among silver speciation, bioavailability, and toxicity in freshwaters, we have studied the short-term uptake of silver by two species of green algae, Chlamydomonas reinhardtii and Pseudokirchneriella subcapitata, in the presence or absence of a well-characterized humic acid (Suwannee River Humic Acid, SRHA). The free Ag+ concentrations in the exposure solutions were determined using an equilibrium ion-exchange technique. According to the biotic ligand model, for a given free metal ion concentration, metal uptake should remain the same in the presence or absence of humic acid. However, short-term silver uptake in the presence of SRHA was greater than anticipated on the basis of free Ag+ concentration. Subsequent determination of silver subcellular distribution revealed that significantly more silver was present in the "cell debris" fraction (known to contain the cell wall and fragmented membranes) in the presence of SRHA than in its absence. Finally, this increase in silver uptake in the presence of humic acid did not result in decreased algal growth. These results suggest that the increase in silver uptake observed in the presence of SRHA is surface-bound, not truly internalized.
Role of Secondary Particle Formation in the Persistence of Silver Nanoparticles in Humic Acid Containing Water under Light Irradiation

Abstract:The wide use of silver nanoparticles (AgNPs) leads to the increasing release of AgNPs into the environment. Dissolved organic matter (DOM) is a key factor affecting the behaviors and fate of AgNPs in the aquatic environment. However, the mechanisms for the DOM-mediated transformations of AgNPs are still not fully understood. In this study, we investigated the persistence of AgNPs in the aquatic environment in the presence of different concentrations of humic acid (HA) over periods of time up to 14 days. The Ag species were monitored and characterized by absorption spectrometry, transmission electron microscopy (TEM), inductively coupled plasma mass spectrometry (ICP-MS), and multicollector ICP-MS (MC-ICP-MS). Results showed that the long-term persistence of AgNPs in HA-containing water was determined by two critical concentrations of HA. When the HA concentration exceeded a lower critical value, AgNPs could be persistent in the solution, and a large number of AgNPs were formed secondarily from the HA-induced reduction of the Ag+ ions released from the primary AgNPs, causing, a redistribution of the particle size. With the HA concentration above a higher critical value, AgNPs could persist in the solution without a significant change in particle size. Notably, we used Ag isotope fractionation to investigate the transformation mechanism of AgNPs. The natural isotopic analysis by MC-ICP-MS revealed that the size redistribution of AgNPs caused significant Ag isotope fractionation, which gave additional evidence for the proposed mechanisms. This study provides new insights into the environmental fate of engineered AgNPs and highlights the usefulness of stable isotope fractionation in environmental nanotechnology.
Transport, Fate, and Stimulating Impact of Silver Nanoparticles on the Removal of Cd(II) by Phanerochaete Chrysosporium in Aqueous Solutions.

Abstract:Despite the knowledge about increasing discharge of silver nanoparticles (AgNPs) into wastewater and its potential toxicity to microorganisms, the interaction of AgNPs with heavy metals in the biological removal process remains poorly understood. This study focused on the effect of AgNPs (hydrodynamic diameter about 24.3 ± 0.37 nm) on the removal of cadmium (Cd(II)) by using a model white rot fungus species, Phanerochaete chrysosporium. Results showed that the biological removal capacity of Cd(II) increased with the concentration of AgNPs increasing from 0.1 mg/L to 1 mg/L. The maximum removal capacity (4.67 mg/g) was located at 1 mg/L AgNPs, and then decreased with further increasing AgNPs concentration, suggesting that an appropriate concentration of AgNPs has a stimulating effect on the removal of Cd(II) by P. chrysosporium instead of an inhibitory effect. Results of Ag+ and total Ag concentrations in the solutions together with those of SEM and XRD demonstrated that added AgNPs had undergone oxidative dissolution and transported from the solution to the surface of fungal mycelia (up to 94%). FTIR spectra confirmed that amino, carboxyl, hydroxyl, and other reducing functional groups were involved in Cd(II) removal, AgNPs transportation, and the reduction of Ag+ to AgNPs.

Abstract:Humic acid (HA) exerts a variety of significant environmental and geochemical influences on the soils, sediments and aqueous environments. The interaction with metal ions induces strong HA-metal complexation, thus effecting the transport of the toxic metals as well as the colloidal aggregation of HA. In the present work, we systematically report and analyze the aggregation mechanisms of HAs in solutions filled with different heavy cations (Ag+, Cd2+ and Cr3+) or common metal ions (Na+, Ca2+ and Al3+) under neutral and low pH conditions by using molecular dynamic simulations. We aim to explore the effects of pH, metal ions type and other possible weak interactions on the aggregation capabilities of HA. Scrutiny of the simulation results showed that the aggregation of HAs under neutral condition was driven by the HA-metal complexation which combined the effects of electrostatic attraction and inter-molecular bridging between cations and COO- groups. Larger extent of aggregation was found in heavy metal ions compared with the common ones. On the other hand, under low pH condition, due to the protonation states of carboxyl and phenolic group, the aggregation of HAs was stabilized mainly by weak forces, such as hydrogen bonds between different functional groups. In addition, other weak interactions such as the hydrophilic and hydrophobic effects, the cation-π interactions have also been proposed to be progressive effects on the coagulation behavior. Our computational studies give supplement to the experimental observation and provide insights into the intrinsic mechanisms of the aggregation behavior of HAs and their complexation with metal ions. Such computational modelling supplied a highly effective tool for qualitatively evaluating their roles in environmental remediation.
