Abstract:An investigation of Trichoderma biodiversity involving a large-scale environmental gradient was conducted to understand the Trichoderma distribution in China. A total of 3,999 isolates were isolated from forestry, grassland, wetland and agriculture ecosystems, and 50 species were identified based on morphological characteristics and sequence analysis of genetic markers. Trichoderma harzianum showed the largest proportion of isolates and the most extensive distribution. Hypocrea semiorbis, T. epimyces, T. konilangbra, T. piluliferum, T. pleurotum, T. pubescens, T. strictipilis, T. hunua, T. oblongisporum and an unidentified species, Trichoderma sp. MA 3642, were first reported in China. Most Trichoderma species were distributed in Jilin and Heilongjiang Provinces in northeast China and the fewest were distributed in Qinghai Province. Based on the division of ecological and geographic factors, forestry ecosystems and low-altitude regions have the greatest species biodiversity of Trichoderma.

Abstract:Trichoderma spp. are diverse fungi with wide distribution. In this study, we report on three new species of Trichoderma, namely T. nigricans, T. densissimum and T. paradensissimum, collected from soils in China. Their phylogenetic position of these novel species was determined by analyzing the concatenated sequenc-es of the second largest nuclear RNA polymerase subunit encoding gene (rpb2) and the translation elonga-tion factor 1- alpha encoding gene (tef1). The results of the phylogenetic analysis showed that each new species formed a distinct clade: T. nigricans is a new member of the Atroviride Clade, and T. densissimum and T. paradensissimum belong to the Harzianum Clade. A detailed description of the morphology and cultural characteristics of the newly discovered Trichoderma species is provided, and these characteristics were compared with those of closely related species to better understand the taxonomic relationships within the Trichoderma.

Abstract:Trichoderma is widely used as a biocontrol microbe against plant diseases in the world,which is mainly applied for controlling a variety of soil borne plant diseases and some plant aboveground diseases.With the advent of MAMPs(microbe-associated molecular patterns) theory,the study on mechanism of plant disease control and improved plant resistance to stress factors with Trichoderma as a biocontrol agent has come into a new stage,especially as the development of technologies based on systems biology,it has become possible to understand the nature of interaction of plant-Trichoderma-pathogen from point of omics view,which therefore would intensively enrich the theory basis of Trichoderma as biocontrol agent.Meanwhile,a diversified technical system for applying Trichoderma products developed in national agricultural industry would provide great promotion to sustainable development of agriculture in the future.This review will focus on new research progress on biocontrol mechanism with Trichoderma against plant diseases at molecular level.

Abstract:Trichoderma is a dominant component of the soil mycoflora. During the field investigations of northern, central, and southwestern China, three new species in the Stromaticum clade were encountered from soil, and named as T. hebeiense, T. sichuanense, and T. verticillatum. Their phylogenetic positions were determined by analyses of the combined two genes: partial sequences of translation elongation factor 1-alpha and the second largest RNA polymerase subunit-encoding genes. Distinctions between the new species and their close relatives were discussed. Trichoderma hebeiense appeared as a separate terminal branch. The species is distinctive by its oblong conidia and aggregated pustules in culture. Trichoderma sichuanense features in concentric colony and produces numerous clean exudates on aerial mycelium in culture. Trichoderma verticillatum is characterized by its verticillium-like synanamorph and production of abundant chlamydospores. In vitro antagonism towards the new species was tested by dual culture technique.

Abstract:Trichoderma is mainly used to control soil-borne diseases as well as some leaf and panicle diseases of various plants. Trichoderma can not only prevent diseases but also promotes plant growth, improves nutrient utilization efficiency, enhances plant resistance, and improves agrochemical pollution environment. Trichoderma spp. also behaves as a safe, low-cost, effective, eco-friendly biocontrol agent for different crop species. In this study, we introduced the biological control mechanism of Trichoderma in plant fungal and nematode disease, including competition, antibiosis, antagonism, and mycoparasitism, as well as the mechanism of promoting plant growth and inducing plant systemic resistance between Trichoderma and plants, and expounded on the application and control effects of Trichoderma in the control of various plant fungal and nematode diseases. From an applicative point of view, establishing a diversified application technology for Trichoderma is an important development direction for its role in the sustainable development of agriculture.

Abstract:Saprotrophy on plant biomass is a recently developed nutrition strategy for Trichoderma. However, the physiology and evolution of this new nutrition strategy is still elusive. We report the deep sequencing and analysis of the genome of Trichoderma longibrachiatum, an efficient cellulase producer. The 31.7-Mb genome, smallest among the sequenced Trichoderma species, encodes fewer nutrition-related genes than saprotrophic T. reesei (Tr), including glycoside hydrolases and nonribosomal peptide synthetase-polyketide synthase. Homology and phylogenetic analyses suggest that a large number of nutrition-related genes, including GH18 chitinases, beta-1,3/1,6-glucanases, cellulolytic enzymes, and hemicellulolytic enzymes, were lost in the common ancestor of T. longibrachiatum (Tl) and Tr. dN/dS (omega) calculation indicates that all the nutrition-related genes analyzed are under purifying selection. Cellulolytic enzymes, the key enzymes for saprotrophy on plant biomass, are under stronger purifying selection pressure in Tl and Tr than in mycoparasitic species, suggesting that development of the nutrition strategy of saprotrophy on plant biomass has increased the selection pressure. In addition, aspartic proteases, serine proteases, and metalloproteases are subject to stronger purifying selection pressure in Tl and Tr, suggesting that these enzymes may also play important roles in the nutrition. This study provides insights into the physiology and evolution of the nutrition strategy of Trichoderma.
Identification and classification of Trichoderma spp. by morphology and soluble protein gel electrophoresis analysis

Abstract:Sixty one isolates of Trichoderma spp. isolated from different plant rhizosphere were identified morphologically as 47 Trichoderma harzianum, 10 Trichoderma viride, 2 Trichoderma koningii and 2 Trichoderma virens respectively. UPGMA dendrogram of all the isolates was produced by soluble protein gel electrophoresis(SPGE) data and showed 3 groups (A, B, and C) at the taxonomic distance index 13.8. Group A was divided into subgroups A1, A2, A3, A4, A5 at the distance 9.1, and included 47 Trichoderma harzianum which was the main members, 8 Trichoderma viride, 1 Trichoderma koningii and 2 Trichoderma virens. Group B included only 1 Trichoderma viride. Group C included 1 Trichoderma viride and 1 Trichoderma koningii. These results showed that Trichoderma spp. diversed morphologically and sub-taxonomic groups might exist in Trichoderma harzianum. The grouping obtained with SPEG method was found to be in accordance to geographic variation of the isolates.
The Study on Identification and Biological Characteristics of Trichoderma longibrachiatum TlCC

Abstract:A Trichoderma (TlCC) isolated from soil was identified as Trichoderma longibrachiatum on the basis of their morphology and DNA sequence analysis of ITS rDNA. The biological characteristics of TlCC were screened and investigated. Though it could grow on PSA, PMA, NA, Czapek and basic cultrure medium, PDA was the most favorable medium for the hypha growth. It could grow and produce spore from 15℃ to 40℃, but the optimal temperature for the growth was 35℃, and for the sporulation was from 25℃ to 35℃. For the basic culture medium, fructose was optimal carbon source for hypha growth, fructose and tragantine were propitious to spore production. The best nitrogen source for hypha growth and spore production was glutamate. The pH for hyphae growth and spore production ranged between 2 and 10, the optimal pH for hyphae growth was 5, from 3 to 7 were propitious to spore production.

Abstract:As widely development and utilization of cellulose in the field of energy,materials and chemistry industry,Trichoderma reesei has been caught more and more attention for its being a kind of important cellulase strain for industry.For enhancing its cellulase product,people have done a lot of work on it,and obtained several considerably good mutant strains.To learn the genome of Trichoderma reesei and its mutant strains is helpful to us understand its system of cellulase hyperproduction,also helpful to people construct its genetic engineering strain in the future.This article introduced the background of Trichoderma reesei and part of its hypercellulase-product strains,also elaborated the research development of its mutant strains genome in the recent years.

Abstract:Clubroot disease is caused by obligate biotrophic protist Plasmodiophora brassicae and is one of the most damaging for crucifer plants.Recently it has become a severe problem globally,especially in Europe,North America,East Asia and other regions.It is also threatening the normal vegetable supply in China in the near future.This review summarizes recent advances in the field of the physiological,biochemical and molecular basis of root gall formation,inheritance of clubroot resistance,molecular markers and its genetic maps in crucifer crops.Finally,future research direction of clubroot resistance are proposed.
Trichoderma Biodiversity in China: Evidence for a North to South Distribution of Species in East Asia.

Abstract:Towards assessing the biodiversity and biogeography of Trichoderma, we have analyzed the occurrence of Trichoderma species in soil and litter from four areas in China: North (Hebei province), South-East (Zhejiang province), West (Himalayan, Tibet) and South-West (Yunnan province). One hundred and thirty five isolates were grouped according to tentative morphological identification. A representative 64 isolates were verified at the species level by the oligonucleotide barcode program TrichO Key v.1.0 and the custom BLAST server TrichoBLAST, using sequences of the ITS1 and 2 region of the rRNA cluster and from the longest intron of the tef1 (translation elongation factor 1-α) gene. Eleven known species (Trichoderma asperellum, T. koningii, T. atroviride, T. viride, T. velutinum, T. cerinum, T. virens, T. harzianum, T. sinensis, T. citrinoviride, T. longibrachiatum) and two putative new species (T. sp. C1, and T. sp. C2), distinguished from known species both by morphological characters and phylogenetic analysis, were identified. A significant difference in the occurrence of these species was found between the North (Hebei) and South-West (Yunnan) areas, which correlates with previously reported species distributions in Siberia and South-East Asia, respectively. As in previous studies, T. harzianum accounted for almost half of the biodiversity; although, in this study, it was exclusively found in the North, and was predominantly represented by an ITS1 and 2 haplotype, which has so far been rarely found elsewhere. This study therefore reveals a North–South gradient in species distribution in East Asia, and identifies Northern China as a potential center of origin of a unique haplotype of T. harzianum.

Abstract:Trichogramma species make up one of the most commonly used groups of natural enemies for biological control programs worldwide. Given the major successes in using Trichogramma to control economically important lepidopterous pests on agricultural crops in China, the biology and ecology of these wasps have been intensively studied to identify traits that contribute to successful biological control. Since the 1960s, improved mass production of Trichogramma and better augmentative release methods to suppress agricultural pests have been achieved. We review the history of research and development; current knowledge on biodiversity and bio-ecology of the species used; and achievements in mass-rearing methods, release strategies, and current large-scale applications in China. In addition, we discuss potential issues and challenges for Trichogramma research and applications in the future.

Abstract:The genus Trichoderma is a diverse group of free-living fungi in the family Hypocreaceae (class Ascomycetes), that are living at different ecosystems in a wide range of climatic zones and that can be found all over the globe. The common ecological nich of the genus Trichoderma includes Natural soils, Decaying wood, Agricultural Habitats, Endophytes and mushroom related substrates. Trichoderma classified base on their morphological and cultural characteristics, conidiophore structure as well as the size of conidia. Now adays, molecularmethods including DNA fingerprinting and sequence analysis of multiple genes (ITS1- 5.8 rDNA-ITS2 and genes encoding translation-elongation factor 1-alpha (tef1) as well as some specific molecular databases have been used for taxonomic classification of this fungi. Trichodermahave been used agriculture biological control of plant diseases and as plant growth promoter, in industry as sources of cellulolytic and hemicellulolytic enzymes used in different industries, in human health producer of secondary metabolites that have clinical importance and in enviromt they are as a potential biodegrader of toxic compounds and soil bioremediation.Therefore, this review article presented the ecological diversiy, taxonomic classification and application of genus Trichoderma.

Abstract:The trichome is one of the core factors to protect plant from the environmental stress conditions and acts as buffer zone between plant surface and environment.Furthermore,the specificity of developmental and differentiation of the trichome cell make it one of the optimal system to study the regulation of cell development and cell fate determination.Recently,the development of the molecular regulation mechanism of trichome developed quickly with the improvement of the techniques in life science,specially in Molecular Biology.Some genes related to trichome have been cloned from modern plant Arabiopsis and their functions are also known well now.Those results provide the theoretical guidance for complex trichomes in plants and agricultural breeding.In this review,we want to show the molecular regulation of trichome development by its developing time,the factors that effecting trichome development,and the development of the trichome in other plants.Thus we hope this review would offer the recent developmental information of the plant trichome without secretary gland to people and researchers.

Abstract:A new strain was isolated from 21 samples that collected from the subtropical broad-leaved forest in Guangzhou,Guangdong Province.It was found that the strain can produce laccase and decolorise methylene blue.The morphology of colony,physiological and biochemical properties were studied.The PDA medium plates were fleetly covered by Xylh colonies on the third day with the bearing of green spores.The ultra-structure of Xylh mycelium and spores was characterized by Atomic Force Microscopy.The surface of mycelium was smooth and presenting some rills,while the dormant spores was rough and covered with many small protuberances like drusens.Contrast to the germinating spore(diameter of 5.0 ~ 6.0 μ m),the dormant spore was much smaller(diameter of 3.0 ~ 3.5 μ m).The interesting point is that Xylh can grow on a medium containing kraft lignin or bagasse without other carbon resources.The peak laccase activity of 50IU/L appeared on the 32h during the liquid fermentation.With the identification of 18S rDNA,the strain of Xylh was classified as a Trichoderma atroviride.

Abstract:In order to identify the bio-control related genes from Trichoderma harzianum, Chaetomium cuperium and C. globosum, the expressed sequence tags (ESTs) of these plant disease bio-control fungi were determined. The cDNA libraries of those fungi were constructed from their mycelia and induced cultures. ESTs of the fungi were sequenced. Analysis of those sequences showed that more than 100 genes from the fungi are plant disease bio-control related genes although more possible bio-control related genes are still to be investigated. The biocontrol related genes are very important for new bio-fungicide development in the future.

Abstract:Tr-92 that induces systemic resistance in cucumber to Botrytis cinerea was isolated from the soils surrounding the plant roots.The strain was identified by morphology and analysis of the 18S rDNA sequence.The ability of induced systemic resistance in cucumber was preliminarily studied.The results showed that the colony morphology and microscopic morphology of Tr-92 strain was consistent with those of Trichoderma.The similarity in 18S sequences was 99%.Thus,Tr-92 was identified as Trichoderma harzianum.By inoculating the strain onto cucumber seedlings at 5-leaf stage,and then challenged with B.cinerea after 24 h,the control effect index to B.cinerea was up to 56%.The activity of resistance-related enzymes,including peroxidase,polyphenol oxidase,beta-1,3-glucanase,phenylalanine aminotransferase and chitinase,in cucumber leaves increased significantly.The study provides appropriate strain resources for the biological control of plant diseases.
