GENE VARIANTS INVOLVING THE METABOLISM PATHWAYS OF AMYLOID BETA (aβ) AND TAU PROTEIN IN ALZHEIMER’S DISEASE IN HAN CHINESE

Abstract:The typical pathological features of Alzheimer's disease (AD) are amyloid beta (Aβ) plaques and phosphorylated tau protein. The genes involving the metabolism pathways of Aβ and tau protein were reported to play a crucial role in AD. Five different pathways of genes related to Aβ and tau protein metabolism were screened in 372 sporadic LOAD patients and 345 normal controls, including Aβ-degrading (ECE1, MMEL1, NEP, IDE, MMP9, ACE), Aβ-generating (BACE1, BACE2), tau-kinases (GSK3B, CDK5, MARK1), tau-pathology (FERMT2, BIN1), and tau -phosphatases (PPP2CA, PPP2R2A, ANP32A, LCMT1, PPME1, PIN1). A total of 15 common SNPs in NEP, MARK1, BIN1, PPP2R2A and PPP2CA had significantly different frequencies between cases and controls, mainly involving Aβ-degrading, tau-kinases, tau-pathology and tau-phosphatases pathway. Further analysis using burden test found the rare variants in ECE1 and MMP9 were associated with sporadic LOAD risk. We found 15 common SNPs in four different pathways of genes related to Aβ and tau protein metabolism as well as rare variants in ECE1 and MMP9 were associated with sporadic LOAD risk. These results suggested the roles of genes related to Aβ and tau protein metabolism in sporadic LOAD etiology.
