SNPS ASSOCIATED WITH VITAMIN D INSUFFICIENCY AND RISK OF ALZHEIMER’S DISEASE IN CHINESE POPULATION

Abstract:Vitamin D insufficiency has been implicated as a risk factor for Alzheimer disease (AD) in several studies. Recently, 4 single nucleotide polymorphisms (SNPs) to be genome-wide significant for 25(OH)D were identified to have association with risk of AD. These include rs2282679 in GC, rs10741657 near CYP2R1, rs12785878 near DHCR7 and rs6013897 in CYP24A1. But whether the 4 SNPs account for risks in Chinese AD remains unknown. A case–control cohort study was conducted in 533 AD patients and 551 normal controls. Genotyping was determined by PCR and SNaPshot sequencing. Statistical significance was analyzed by the Chi-squared test. We found the frequency of rs2282679 was significantly different between the AD cases and healthy controls (p=0.028 for genotype frequency; p=0.010 for allele frequency, OR=0.845) while there was no statistical significance of other 3 SNPs in genotype or allele frequencies between the two groups. After being stratified by gender and age at onset, we found rs2282679 was still related to male AD patients (p=0.025 for genotype frequency; p=0.007 for allele frequency, OR=0.750). In addition, we found rs12785878 to be significantly associated with increased risk of Early onset of AD(EOAD) in both genotype and allele frequencies (p=0.024 for genotype frequency; p=0.009 for allele frequency, OR=1.183). Our results suggest that rs12785878 might be associated with risk of EOAD while other SNPs related to Vitamin D insufficiency might not be associated with increased risk of AD in Chinese population.
